| 4 | Effect of Marsdenia tenacissima extract on G2/M cell cycle arrest by upregulating 14-3-3σ and downregulating c-myc in vitro and in vivo显示文摘Objective: Marsdenia tenacissima extract(MTE) is a traditional Chinese herbal medicine with anti-cancer activity. In some previous studies, different mechanism actions of the anti-cancer effect of MTE have been revealed. In this study, we first observed that MTE exhibited G2/M cell cycle arrest on two different human breast cancer cell lines, MDA-MB-231 and MCF-7 by mediating 14-3-3σ and c-myc.Methods: The effect of MTE on G2/M cell cycle arrest was evaluated in MDA-MB-231 and MCF-7 cell lines. MTT assay was done for evaluation of cell viability. Flow cytometry was employed for cell cycle analysis. Western blotting analysis and immunohistochemistry were performed to analyze the expression of G2/M cell cycle-related key protein in cells and tissue samples. Animal studies have been conducted to elucidate the anti-tumor effect of MTE.Results: Cell cycle is the backbone for developing cancer. Cell cycle proteins play a major role in the progression of cell cycle and cell proliferation. However, some key protein directly or indirectly modulate the action of cell cycle protein that highly affect cell cycle regulation. In order to investigate cellular proliferation of cancer, we observed that MTE induced the upregulation of 14-3-3σ and downregulation of c-myc,and then reduced the expression of G2/M cell cycle associated key protein, leading to the inhibition of cellular entry into mitosis phase. | 5 | A systematic survey of LU domain-containing proteins reveals a novel human gene,LY6A,which encodes the candidate ortholog of mouse Ly-6A/Sca-1 and is aberrantly expressed in pituitary tumors显示文摘The Ly-6 and uPAR(LU)domain-containing proteins represent a large family of cell-surface markers.In particular,mouse Ly-6A/Sca-1 is a widely used marker for various stem cells;however,its human ortholog is missing.In this study,based on a systematic survey and comparative genomic study of mouse and human LU domain-containing proteins,we identified a previously unannotated human gene encoding the candidate ortholog of mouse Ly-6A/Sca-1.This gene,hereby named LY6A,reversely overlaps with a lncRNA gene in the majority of exonic sequences.We found that LY6A is aberrantly expressed in pituitary tumors,but not in normal pituitary tissues,and may contribute to tumorigenesis.Similar to mouse Ly-6A/Sca-1,human LY6A is also upregulated by interferon,suggesting a conserved transcriptional regulatory mechanism between humans and mice.We cloned the full-length LY6A cDNA,whose encoded protein sequence,domain architecture,and exon‒intron structures are all well conserved with mouse Ly-6A/Sca-1.Ectopic expression of the LY6A protein in cells demonstrates that it acts the same as mouse Ly-6A/Sca-1 in their processing and glycosylphosphatidylinositol anchoring to the cell membrane.Collectively,these studies unveil a novel human gene encoding a candidate biomarker and provide an interesting model gene for studying gene regulatory and evolutionary mechanisms. | Dan Liu Chunhui Xu Yanting Liu Wen Ouyang Shaojian Lin Aining Xu Yuanliang Zhang Yinyin Xie Qiuhua Huang Weili Zhao Zhu Chen Lan Wang Saijuan Chen Jinyan Huang Zhe Bao Wu Xiaojian Sun | 2023 | Frontiers of Medicine2023,17,3: | 0 |
| 6 | 硼替佐米治疗多发性骨髓瘤的临床初步研究(英文)显示文摘Objective:To explore the outcome and side-effects in patients with multiple myeloma treated with velcade and combined chemotherapy. Methods: 6 patients with multiple myeloma were treated with the combination of velcade and dexamethasone or DVD (V-DVD regimen) chemotherapy. Results: 6 patients responded to treatment. 4 patients achieved complete remission (CR), 1 achieved partial remission (PR) and 1 achieved micro responded (MR). Major toxicities consisted of peripheral neuropathy, thrombocytopenia and hepatic impairment. | 7 | Clinical research of biphenotypic acute leukemia witht(8;21)(q22;q22)显示文摘Objective:To report 4 cases of biphenotypic acute leukemia(BAL)with t(8;21)(q22;q22),and analyze the characteristics of morphology,immune phenotype,chromosome karyotype(MIC)and clinical manifestations.Methods:The BAL patients with t(8;21)(q22;q22)(group A)were compared with the randomly selected BAL patients with other clonical chromo- somal changes(group B)and acute myeloid leukemia M2 cases with t(8;21)(q22;q22)(group C)in MIC and clinical features.
