1Pricking and penetrating moxibustion therapy in patients with refractory insomnia: a randomized and controlled clinical trial显示文摘OBJECTIVE: To observe the clinical effect of pricking and penetrating moxibustion therapy on refractory insomnia.METHODS: Totally 60 subjects were randomly divided into treatment group and control group, 30 in each group. The treatment group was treated with pricking and penetrating moxibustion therapy, the control group with penetrating moxibustion therapy, the treatment was given once every day, two groups of patients were treated 20 times,compared the efficacy, pinsburgh sleep quality index(PSQI) and Traditional Chinese Medicine symptom, safety evaluation between the two groups.RESULTS: Compared with 10, 20 times after the treatment, the effective rate of the treatment group(93.3%) higher than that of the control group(80.0%)(P < 0.05); Compared with beforetreatment, 20 times after treatment, the PSQI and TCM symptom pattern scores of the two groups were significantly decreased(P < 0.01); Compared with 10, 20 times after the treatment, the treatment group are better than the control group in improving the total score and the integral of PSQI(P <0.05 or P < 0.01); The treatment group are better than the control group in improving the TCM symptom pattern score, difficulty in going to sleep, palpitation, amnesia, fidget, sweating, impaired concentration(P < 0.01 or P < 0.05).CONCLUSION: The pricking and penetrating moxibustion therapy is safe and effective in the treatment of refractory insomnia and the treatment can improve the curative effect of the therapy that uses penetrating moxibustion only.Chen Yan Gao Xiyan Sun Cuiying 2018Journal of Traditional Chinese Medicine2018,38,5:4
2Adiponectin attenuates lipopolysaccharide-induced cell injury of H9c2 cells by regulating AMPK pathway显示文摘Adiponectin,an adipokine synthesized and secreted majorly by adipose tissue,is reported to exert cardioprotective properties via anti-inflammation and antiapoptosis.Lipopolysaccharide(LPS)is a common inflammation and apoptosis inducer of cardiomyocytes.However,few studies have reported the roles of adiponectin on LPS-induced inflammation as well as apoptosis of H9c2 cells,and the possible mechanisms of these effects.In the present study,we found that adiponectin significantly relieved LPS-induced cytotoxicity including decreased viability and elevated LDH release,inhibited LPS-triggered inflammation,which is evidenced by increases in release of TNF-α,IL-1β as well as IL-6,and attenuated the enhanced rates of apoptotic cells as well as increased caspase-3 activity caused by LPS in H9c2 cells.In addition,our data demonstrated that adiponectin upregulated AMP-activated protein kinase(AMPK)activation of H9c2 cells with or without LPS administration.Moreover,we found that blocking AMPK pathway by compound c attenuated the protective effects of adiponectin against the cytotoxicity,inflammatory response,and apoptosis of H9c2 cells resulted from LPS.Our observations bring novel insights for understanding the mediatory role of AMPK pathway implicated in the protective effects of adiponectin against LPS-induced cardiotoxicity.Rui Hao Guoying Su Xiaolin Sun Xiangran Kong Cuiying Zhu Guohai Su 2019Acta Biochimica et Biophysica Sinica2019,51,2:4
3Adsorption desulfurization of model gasoline by metal–organic framework Ni3(BTC)2显示文摘This work presented the synthesis of Ni-based metal-organic framework material with a paddle-wheel structure Ni3(BTC)2 (Ni-BTC) and its application in thiophene (TP) adsorption from gasoline distillate by batch method. Adsorption isotherms of TP, cyclohexene, and toluene in cyclohexane onto Ni-BTC were conducted at 298-308 K to interpret the different effect of cyclohexene and toluene on TP adsorption. The results showed that, compared w让h cyclohexene, toluene addition in model gasoline led to a more evident decline in sulfur capacity of Ni-BTC, which is opposite to isostructural HKUST-1. The adsorption isotherms of TP, cyclohexene and toluene fit Langmuir model, S-type model and Temkin model well, respectively, indicating that the adsorption mechanisms of TP and the two competitors are different from one another The adsorption capacities on Ni-BTC followed the order of cyclohexene < toluene < TP at the same equilibrium concentrations, implying the order of the adsorption affinities, which is in good agreement with the different extent of influence by the two competitors. The enthalpy of TP adsorption on Ni-BTC was estimated to be -80.01 kj/mol, almost twice that on HKUST-1. The poor reusability of Ni-BTC in batch experiment, which is owing to its sensitivity to the air, can be prevented from regenerating used Ni-BTC in fixed-bed reactor by N2 flow. 4Effect of enzymatic extraction conditions on the yield and the polymerization degree of procyanidins显示文摘Grape seeds are rich sources of procyanidin(PCs)known for potential health benefi ts.In this study,PCs were extracted from defatted grape seeds by enzymatic method in which pectinase and cellulase were used.The enzyme extraction process was further optimized by single factor experiment and response surface methodology.The optimal conditions were as follows:ethanol concentration of 70%,extraction time of 70 min,extraction temperature of 35℃,liquid/solid ratio of 103:1(mL/g),pectinase/cellulase ratio of 1:1,enzyme/solid ratio of 1:314 w/w.Under the above conditions,the extraction yields and mean degree of polymerisation(mDP)of PCs reached 47.18 mg/g dry material weight and 11.2,respectively.Compared with other extraction methods,enzyme extraction can obtain PCs with higher yield and lower mDP.According to the antioxidant activity test,PCs extracts with lower mDP showed better ability to clear 1,1-Diphenyl-2-picrylhydrazyl radical(DPPH).Enzymatic extraction was an effi cient method to obtain oligomeric procyanidin which has stronger antioxidant activity.Shijie Sun Xiong Chen Zhaoyang Pei Lulu Xu Cuiying Qin Jing Han 2020Journal of Polyphenols2020,2,1:0
5Comprehensive bioinformatics analysis and experimental validation:An anoikis-related gene prognostic model for targeted drug development in head and neck squamous cell carcinoma显示文摘We analyzed RNA-sequencing(RNA-seq)and clinical data from head and neck squamous cell carcinoma(HNSCC)patients in The Cancer Genome Atlas(TCGA)Genomic Data Commons(GDC)portal to investigate the prognostic value of anoikis-related genes(ARGs)in HNSCC and develop new targeted drugs.Differentially expressed ARGs were screened using bioinformatics methods;subsequently,a prognostic model including three ARGs(CDKN2A,BIRC5,and PLAU)was constructed.Our results showed that the model-based risk score was a good prognostic indicator,and the potential of the three ARGs in HNSCC prognosis was validated by the TISCH database,the model’s accuracy was validated in two independent cohorts of the Gene Expression Omnibus database.Immune correlation analysis and half-maximal inhibitory concentration were also performed to reveal the different landscapes of TIME between risk groups and to predict immuno-and chemo-therapeutic responses.Potential small-molecule drugs for HNSCC were subsequently predicted using the L1000FWD database.Finally,in vitro experiments were used to verify the database findings.The relative ARG mRNA expression levels in HNSCC and surrounding normal tissues remained consistent with the model results.BIRC5 knockdown inhibited anoikis resistance in WSU-HN6 and CAL-27 cells.Molecular docking,real-time PCR,cell counting kit-8(CCK-8),plate clone,and flow cytometry analyses showed that small-molecule drugs predicted by the database may target the ARGs in the prognostic model,inhibit HNSCC cells survival rate,and promote anoikis in vitro.Therefore,we constructed a new ARG model for HNSCC patients that can predict prognosis and immune activity and identify a potential small-molecule drug for HNSCC,paving the way for clinically targeting anoikis in HNSCC.LIN QIU ANQI TAO XIAOQIAN SUN FEI LIU XIANPENG GE CUIYING LI 2023Oncology Research2023,31,5:0
6Structural insights into ligand recognition and selectivity of somatostatin receptors显示文摘Somatostatin receptors(SSTRs)play versatile roles in inhibiting the secretion of multiple hormones such as growth hormone and thyroid-stimulating hormone,and thus are considered as targets for treating multiple tumors.Despite great progress made in therapeutic development against this diverse receptor family,drugs that target SSTRs still show limited efficacy with preferential binding affinity and conspicuous side-effects.Here,we report five structures of SSTR2 and SSTR4 in different states,including two crystal structures of SSTR2 in complex with a selective peptide antagonist and a non-peptide agonist,respectively,a cryo-electron microscopy(cryo-EM)structure of Gi1-bound SSTR2 in the presence of the endogenous ligand SST-14,as well as two cryo-EM structures of Gi1-bound SSTR4 in complex with SST-14 and a small-molecule agonist J-2156,respectively.By comparison of the SSTR structures in different states,molecular mechanisms of agonism and antagonism were illustrated.Together with computational and functional analyses,the key determinants responsible for ligand recognition and selectivity of different SSTR subtypes and multiform binding modes of peptide and non-peptide ligands were identified.Insights gained in this study will help uncover ligand selectivity of various SSTRs and accelerate the development of new molecules with better efficacy by targeting SSTRs.Wenli Zhao Shuo Han Na Qiu Wenbo Feng Mengjie Lu Wenru Zhang Mu Wang Qingtong Zhou Shutian Chen Wei Xu Juan Du Xiaojing Chu Cuiying Yi Antao Dai Liaoyuan Hu Michelle YShen Yaping Sun Qing Zhang Yingli Ma Wenge Zhong Dehua Yang Ming-Wei Wang Beili Wu Qiang Zhao 2022Cell Research2022,32,8:0
