| 1 | Host metabolism dysregulation and cell tropism identification in human airway and alveolar organoids upon SARS-CoV-2 infection显示文摘The coronavirus disease 2019(COVID-19)pandemic is caused by infection with the severe acute respiratory syndrome coronavirus 2(SARS-CoV-2),which is spread primary via respiratory droplets and infects the lungs.Currently widely used cell lines and animals are unable to accurately mimic human physiological conditions because of the abnormal status of cell lines(transformed or cancer cells)and species differences between animals and humans.Organoids are stem cell-derived selforganized three-dimensional culture in vitro and model the physiological conditions of natural organs.Here we showed that SARS-CoV-2 infected and extensively replicated in human embryonic stem cells(hESCs)-derived lung organoids,including airway and alveolar organoids which covered the complete infection and spread route for SARS-CoV-2 within lungs.The infected ceils were ciliated,club,and alveolar type 2(AT2)cells,which were sequentially located from the proximal to the distal airway and terminal alveoli,respectively.Additionally,RNA-seq revealed early cell response to virus infection including an unexpected downregulation of the metabolic processes,especially lipid metabolism,in addition to the well-known upregulation of immune response.Further,Remdesivir and a human neutralizing antibody potently inhibited SARS-CoV-2 replication in lung organoids.Therefore,human lung organoids can serve as a pathophysiological model to investigate the underlying mechanism of SARS-CoV-2 infection and to discover and test therapeutic drugs for COVID-19. | Rongjuan Pei Jianqi Feng Yecheng Zhang Hao Sun Lian Li Xuejie Yang Jiangping He Shuqi Xiao Jin Xiong Ying Lin Kun Wen Hongwei Zhou Jiekai Chen Zhili Rong Xinwen Chen | 2021 | Protein & Cell2021,12,9: | 7 |
| 2 | Differently sized cuckoos pose different threats to hosts显示文摘Hole-nesting tits Parus spp.have been classified as'unsuitable'hosts for cuckoo parasitism because cuckoos cannot enter a cavity if the entrance is too small.However,Chinese tits could re-ject alien eggs and egg ejection rate increased with the local diversity of parasitic cuckoo species.Antiparasitic behavior among Chinese tits may have evolved due to greater size variation among sympatric cuckoo species.This raises the question of whether differently sized parasitic cuckoos pose different threats to Chinese tits.A green-backed tit Parus monticolus population that is sym-patric with Asian emerald cuckoo Chrysococcyx maculatus(eme-cuckoo,small-sized parasite)and common cuckoo Cuculus canorus(com-cuckoo,large-sized parasite),and a cinereous tit P.cinereus population that is only sympatric with com-cuckoo were chosen as study organisms.We observed behavioral response and recorded alarm calls of the 2 tit species to eme-cuckoo,com-cuckoo,chipmunk Tamias sibiricus(a nest predator)and dove Streptopelia orientalis(a harm-less control),and subsequently played back alarm calls to conspecific incubating females.In dummy experiments,both tit species performed intense response behavior to chipmunk,but rarely responded strongly to the 3 avian species.In playback experiments,both tit species responded strongly to conspecific chipmunk alarm calls,but rarely responded to dove alarm calls.The inten-sity of response of incubating female green-backed tits to eme-cuckoo and com-cuckoo alarm calls were similar to that of chipmunk alarm calls,while the intensity to eme-cuckoo alarm calls was higher than the intensity to dove alarm calls which was similar to that of com-cuckoo alarm calls.In contrast,few female cinereous tits responded to eme-cuckoo and com-cuckoo alarm calls.These findings indicated that the threat level of eme-cuckoo was slightly greater than that of com-cuckoo for sympatric green-backed tits,but not for allopatric cinereous tits. | Jiangping YU Mingju E Wei SUN Wei LIANG Haitao WANG Anders Pape MΦLLER | 2020 | Current Zoology2020,66,3: | 4 |
| 3 | Pan-genome of Raphanus highlights genetic variation and introgression among domesticated,wild,and weedy radishes显示文摘Post-polyploid diploidization associated with descending dysploidy and interspecific introgression drives plant genome evolution by unclear mechanisms.Raphanus is an economically and ecologically important Brassiceae genus and model system for studying post-polyploidization genome evolution and introgres-sion.Here,we report the de novo sequence assemblies for 11 genomes covering most of the typical sub-species and varieties of domesticated,wild and weedy radishes from East Asia,South Asia,Europe,and America.Divergence among the species,sub-species,and South/East Asian types coincided with Quaternary glaciations.A genus-level pan-genome was constructed with family-based,locus-based,and graph-based methods,and whole-genome comparisons revealed genetic variations ranging from single-nucleotide polymorphisms(SNPs)to inversions and translocations of whole ancestral karyotype(AK)blocks.Extensive gene flow occurred between wild,weedy,and domesticated radishes.High fre-quencies of genome reshuffling,biased retention,and large-fragment translocation have shaped the genomic diversity.Most variety-specific gene-rich blocks showed large structural variations.Extensive translocation and tandem duplication of dispensable genes were revealed in two large rearrangement-rich islands.Disease resistance genes mostly resided on specific and dispensable loci.Variations causing the loss of function of enzymes modulating gibberellin deactivation were identified and could play an important role in phenotype divergence and adaptive evolution.This study provides new insights into the genomic evolution underlying post-polyploid diploidization and lays the foundation for genetic improve-ment of radish crops,biological control of weeds,and protection of wild species'germplasms. | Xiaohui Zhang Tongjin Liu Jinglei Wang Peng Wang Yang Qiu Wei Zhao Shuai Pang Xiaoman Li Haiping Wang Jiangping Song Wenlin Zhang Wenlong Yang Yuyan Sun Xixiang Li | 2021 | Molecular Plant2021,14,12: | 3 |
| 4 | Multi-proxy evidence of environmental change related to collapse of the Liangzhu Culture in the Yangtze Delta,China显示文摘The Liangzhu Culture(5300–4300 cal.a BP)has been regarded as the culmination of the Neolithic Age in the Yangtze Delta and as an archaic state.However,the reason behind the collapse of the Liangzhu Culture remains controversial.Some scholars attributed it to social conflicts or foreign invasions,while others suggested that environmental deterioration,such as cold episodes,inundation and palaeo-typhoon events,disrupted the civilization.Notably,the yellowish silty sediments widely overlying the Liangzhu group sites have been presumed to be one of the vital clues to resolving these issues.The process and cause of their formation are key to understanding the decline in the Liangzhu Culture.In this study,two typical profiles composed of the Liangzhu cultural layer and yellowish silty sediments were collected in Liangzhu Ancient City.Multiple proxies,including pollen,phytoliths and diatoms,along with radiocarbon and optically stimulated luminescence dating and synthesized geochemical evidence from different sediments,were applied to reconstruct ecological and hydrological changes and discuss their linkage to cultural collapse.The regional ecology was typical freshwater wetlands during 4800–4400 cal.a BP,which facilitated rice farming and large-scale architectural engineering in Liangzhu Ancient City.Evidence from microfossils and dating of the yellowish silty sediments indicated that Liangzhu Ancient City was significantly affected by seawater after abandonment during 4300–3400 cal.a BP.Furthermore,geochemical evidence,including trace element and Sr-Nd isotopic compositions,suggested that the primary provenance of the yellowish silt was a mixture of sediments from the Qiantang River,East China Sea,Yangtze River and Yellow River,which had been transported to Liangzhu Ancient City by the Yellow Sea Coastal Current and a marine transgression.Therefore,based on evidence from archaeological sites and numerical simulation in the Yangtze Delta,it was supposed that the short-term accelerated relative sea-level rise at approximately 4400 cal.a BP caused a marine transgression around Taihu Lake,formed a large range of yellowish silty sediments,destroyed rice farming production,and ultimately led to the collapse of the Liangzhu Culture. | Keyang HE Houyuan LU Guoping SUN Xiang JI Yonglei WANG Kaikai YAN Xinxin ZUO Jiangping ZHANG Bin LIU Ningyuan WANG | 2021 | Science China Earth Sciences2021,64,6: | 3 |

| 10 | Significantly enhanced photocatalytic hydrogen production performance of g-C_(3)N_(4)/CNTs/CdZnS with carbon nanotubes as the electron mediators显示文摘The electron mediator can effectively improve the performance of the direct Z-scheme heterojunction photocatalysts. However, it is still a great challenge to select cheap and efficient electron mediators and to design them into the Z-scheme photocatalytic system. In the present paper, the g-C_(3)N_(4)/CNTs/CdZnS Z-scheme photocatalyst was prepared using carbon nanotubes(CNTs) as the electron mediators, and its photocatalytic hydrogen production performance was studied. Compared with single-phase g-C_(3)N_(4),CdZnS and biphasic g-C_(3)N_(4)/CdZnS photocatalysts, the photocatalytic hydrogen production performance of the prepared g-C_(3)N_(4)/CNTs/CdZnS has been significantly enhanced. Meanwhile, g-C_(3)N_(4)/CNTs/CdZnS possesses very good photocatalytic hydrogen production stability. The enhanced photocatalytic hydrogen production performance of g-C_(3)N_(4)/CNTs/CdZnS is attributed to the fact that CNTs, as an electron mediator,can accelerate the recombination of the photogenerated holes in the valence band of g-C_(3)N_(4) and the photogenerated electrons in the conduction band of CdZnS, which makes the g-C_(3)N_(4)/CNTs/CdZnS Zscheme photocatalyst be easier to escape the photogenerated electrons, increases the concentration of the photogenerated carriers and prolongs the lifetime of the photogenerated carriers. | 11 | A Heterozygous Phospholamban Variant(p.R14del)Leads to Left Ventricular Involvement and Heart Failure Phenotypes in Arrhythmogenic Right Ventricular Cardiomyopathy显示文摘This study aimed to determine the prevalence and clinical features of Arrhythmogenic Right Ventricular Cardiomyopathy(ARVC)caused by pathogenic mutations in the Phospholamban(PLN)gene.The study included 170 patients who had a confrmed diagnosis of ARVC and underwent PLN genetic screening using next-generation sequencing.The fndings of this study provide valuable insights into the association between PLN mutations and ARVC,which can aid in the development of more efective diagnostic and treatment strategies for ARVC patients.Out of the patients evaluated,six had a rare pathogenic mutation in PLN with the same p.R14del variant.Family screening revealed that heterozygous carriers of p.R14del exhibited a defnite ARVC phenotype.In clinical studies,individuals with the p.R14del mutation experienced a similar rate of malignant arrhythmia events as those with classic desmosome mutations.After adjusting for covariates,individuals with PLN mutations had a two point one seven times greater likelihood of experiencing transplant-related risks compared to those who did not possess PLN mutations(95%CI 1.08–6.82,p=0.035).The accumulation of left ventricular fat and fbers is a pathological marker for ARVC patients with p.R14del mutations.In a cohort of 170 Chinese ARVC patients,three point fve percent of probands had the PLN pathogenic variant(p.R14del)and all were female.Our data shows that PLN-related ARVC patients are at high risk for ventricular arrhythmias and heart failure,which requires clinical diferentiation from classic ARVC.Furthermore,carrying the p.R14del mutation can be an independent prognostic risk factor in ARVC patients. | Han Mo Xiumeng Hua Mengni Bao Zhe Sun Xiao Chen Mengda Xu Jiangping Song | 2024 | Phenomics2024,4,1: | 0 |

| 13 | Chikungunya in Zhejiang Province,Southeast China显示文摘Background:Chikungunya is emerging and reemerging word-widely in the past decades.It is non-endemic in Zhejiang Province,Southeast China.Aedes albopictus,one of major vectors of chikungunya,is widely-distribution in Zhejiang,and autochthonous transmission is possible after introducing chikungunya virus.Methods:Retrospectively collected the epidemiological,clinical and genetic data of chikungunya and conducted the descriptive analysis and gene sequence analysis.Results:From 2008 to 2022,29 chikungunya cases,including 26 overseas imported and 3 local cases,were reported and no cases died of chikungunya.More than half of the imported cases(53.85%)were from Southeast Asia.Seasonal peak of the imported cases was noted between August and September,and 42.31%cases onset in those 2 months.Eight prefecture-level cities and 16 counties reported cases during the study period,with Jinghua(27.59%)and Hangzhou(24.14%)reporting the largest number of cases.The 3 local cases were all reported in Qujiang,Quzhou in 2017.For imported cases,the male-female gender ratio was 2.71:1,20-30 years old cases(46.15%)and commercial service(42.31%)accounted for the highest proportion.Clinically,fever(100%),fatigue(94.44%),arthralgia(79.17%),headache(71.43%)and erythra(65.22%)were the most common reported symptoms.Eight whole-genome sequences were obtained and belonged to East/Central/South African(ECSA)or Asian genotype.Conclusions:With the change of immigration policy,the surveillance of chikungunya should be strengthened and the ability of the case discovery and diagnosis should be improved in Zhejiang in the post-COVID-19 era. | Jiangping Ren Feng Ling Ying Liu Jimin Sun | 2023 | Infectious Medicine2023,2,4: | 0 |

| 15 | Numerical investigation of viscous fingering in Hele-Shaw cell with spatially periodic variation of depth显示文摘Viscous fingering in a modified Hele-Shaw cell is numerically investigated.The cell allows periodic variation of depth in the lateral direction. The wavenumber n of the depth perturbation has great influence on fingering patterns. For n = 1, the fingering pattern due to the interface instability remains the same as that in the conventional Hele-Shaw cell, while the depth variation causes the steady finger to be a little narrower. For n = 2, four different fingering patterns are captured, similar to the available experimental observations in a modified Hele-Shaw cell containing a centered step-like occlusion. It is found that new fingering patterns appear as n further increases, among which, two patterns with spatial oscillation along both edges of the finger are particularly interesting.One is a symmetric oscillatory finger for n = 3, and the other is an asymmetric one for n = 4. The influence of capillary number on fingering patterns is studied for n = 3 and 4.We find that spatial oscillation of the finger nearly ceases at moderate capillary numbers and occurs again as the capillary number increases further. Meanwhile, the wide finger shifts to the narrow one. It is accompanied by a sudden decrease in the finger width which otherwise decreases continuously as the capillary number increases.
