2Neutrophil-mimicking therapeutic nanoparticles for targeted chemotherapy of pancreatic carcinoma显示文摘Due to the critical correlation between inflammation and carcinogenesis, a therapeutic candidate with anti-inflammatory activity may find application in cancer therapy. Here, we report the therapeutic efficacy of celastrol as a promising candidate compound for treatment of pancreatic carcinoma via na?ve neutrophil membrane-coated poly(ethylene glycol) methyl ether-block-poly(lactic-co-glycolic acid)(PEG-PLGA) nanoparticles. Neutrophil membranecoated nanoparticles(NNPs) are well demonstrated to overcome the blood pancreas barrier to achieve pancreas-specific drug delivery in vivo. Using tumor-bearing mice xenograft model, NNPs showed selective accumulations at the tumor site following systemic administration as compared to nanoparticles without neutrophil membrane coating. In both orthotopic and ectopic tumor models,celastrol-loaded NNPs demonstrated greatly enhanced tumor inhibition which significantly prolonged the survival of tumor bearing mice and minimizing liver metastases. 5Cancer stem cells:therapeutic implications and perspectives in cancer therapy显示文摘The cancer stem cell(CSC)theory is gaining increasing attention from researchers and has become an important focus of cancer research.According to the theory,a minority population of cancer cells is capable of self-renewal and generation of differentiated progeny,termed cancer stem cells(CSCs).Understanding the properties and characteristics of CSCs is key to future study on cancer research,such as the isolation and identification of CSCs,the cancer diagnosis,and the cancer therapy.Standard oncology treatments,such as chemotherapy,radiotherapy and surgical resection,can only shrink the bulk tumor and the tumor tends to relapse.Thus,therapeutic strategies that focus on targeting CSCs and their microenvironmental niche address the ineffec-tiveness of traditional cancer therapies to eradicate the CSCs that otherwise result in therapy resistance.The combined use of traditional therapies with targeted CSC-specific agents may target the whole cancer and offer a promising strategy for lasting treatment and even cure.Lu Han Sanjun Shi Tao Gong Zhirong Zhang Xun Sun 2013Acta Pharmaceutica Sinica B2013,3,2:4

8Implantable sandwich PHBHHx film for burst-free controlled delivery of thymopentin peptide显示文摘Sustained release and non-parental formulations of peptides and protein drugs are highly desirable because of enhanced therapeutic effects as well as improved patient compliance. This is especially true for small peptides such as thymopentin(TP5). To this end, implantable sandwich poly(hydroxybutyrate-co-hydroxyhexanoate)(PHBHHx) films were designed to prolong release time and to inhibit burst release phenomenon of TP5 by a simple volatilization method. In vitro release studies revealed that sandwich films had nearly no burst release. In vivo release time of sandwich films was prolonged to 42 days. Pharmacodynamic evaluation demonstrated that TP5 sandwich films significantly increased survival rates in a rat immunosuppressive model and normalized CD4^+/CD8^+ values. These results suggest that TP5 released from sandwich films can attenuate cyclophosphamide's immunosuppressive activity, and possibly achieve results comparable to daily TP5 injection therapy. 9Glucose-responsive erythrocyte-bound nanoparticles for continuously modulated insulin release显示文摘Glucose-responsive closed-loop insulin delivery systems represent a promising treatment strategy for diabetes,but current systems generally cannot achieve long-term effects.In this study,we designed an erythrocyte-biomimetic glucose-responsive system(EGRS)by coupling glucose-responsive nanoparticles(GRNs)to red blood cells;these nanoparticles exhibited the dual functions of glucose-responsiveness and persistent presence in circulation.GRNs are generated by encapsulating with insulin through ion crosslinking,followed by coloading with glucose oxidase(GOx)and catalase(CAT),a process that endows the nanoparticles with glucose-responsiveness.Simultaneously,the GRNs are coupled with red blood cells to camouflage them from the immune system,therefore,these erythrocyte-coupled GRNs can circulate in the blood for a long time.Under conditions of hyperglycemia,GOx acts on blood glucose to produce gluconic acid,which causes the rupture of GRNs and efficient release of insulin.Conversely,insulin is only released at the basic rate during hypoglycemia.Thus,EGRS can efficiently and continuously respond to hyperglycemia to maintain blood glucose levels within the normal range.Xiaomin Xu Yani Xu Yuai Li Min Li Leilei Wang Qiang Zhang Bingjie Zhou Qing Lin Tao Gong Xun Sun Zhirong Zhang Ling Zhang 2022Nano Research2022,15,6:2

11Asymptomatic patients and asymptomatic phases of Coronavirus Disease 2019(COVID-19):a population-based surveillance study显示文摘In this population-based study,we identified 307 confirmed COVID-19 cases from massive surveillance,including 129551 individuals screened at fever clinics or returning from Hubei and 3710 close contacts of confirmed COVID-19 patients.Among them,17 patients were asymptomatic at initial clinical assessment.These asymptomatic patients on admission accounted for a small proportion of all patients(5.54%)with relatively weak transmissibility,and the detection rate was 0.35 per 100 close contacts.Moreover,the dynamics of symptoms of the 307 patients showed that the interval from symptom remission to the final negativity of viral nucleic acid was 5.0 days(interquartile range 2.0 to 11.0 days),with 14 patients(4.56%)having re-detectable viral RNA after discharge.Overall;our findings suggested asymptomatic carriers and presymptomatic patients only accounted for a small proportion of COVID-19 patients.Also,the asymptomatic phase during recovery from COVID-19 implied that negativity in viral RNA is necessary as a de-isolation criterion and follow-up is recommended.Xueying Zheng Sihui Luo Yong Sun Mingfeng Han Jian Liu Liangye Sun Liangming Zhang Ping Ling Yu Ding Tengchuan Jin Zhirong Liu Jianping Weng 2020National Science Review2020,7,10:2
12Biodegradation Kinetics for Pre-treatment of Klebsiella pneumoniae Waste with Autothermal Thermophilic Aerobic Digestion显示文摘Biodegradation parameters and kinetic characteristics for pre-treating waste strains of Klebsiella pneu-moniae were studied in laboratory scale with an insulated reactor by an innovative technique,autothermal thermo-philic aerobic digestion(ATAD) . Based on an Arrhenius-type equation,an empirical model was developed to corre-late the removal of total suspended solid(TSS) with the initial TSS concentration,influent reaction temperature,aeration rate and stirring rate. The reaction temperatures of the ATAD system could be raised from the ambient temperatures of 25 °C to a maximum temperature of 65 °C. The exponentials for the initial TSS concentration,aeration rate and stirring rate were 1.579,-0.8175 and-0.6549,respectively,and the apparent activation energy was 6.8774 kJ·mol-1. The correlation coefficient for the pre-exponential factor was 0.9223. The TSS removal effi-ciency predicted by the model was validated with an actual test,showing a maximum relative deviation of 10.79%. 13Bioinformatical study on the proteomics and evolution of SARS-CoV显示文摘A novel coronavirus has been identified as the causative agent of the severe acute respiratory syndrome(SARS). For all the SARS-CoV associated proteins derivatedfrom the SARS-CoV genome, the physiochemical propertiessuch as the molecular weight, isoelectric point and extinction coefficient of each protein were calculated. Thetransmembrane segments and subcellular localization(SubLocation) prediction and conserved protein motifssearch against database were employed to analyze thefunction of SARS-CoV proteins. Also, the homology protein sequence alignment and evolutionary distance matrixcalculation between SARS-CoV associated proteins and the corresponding proteins of other coronaviruses wereemployed to identify the classification and phylogeneticrelationship between SARS-CoV and other coronaviruses.The results showed that SARS-CoV is a novel coronavirus which is different from any of the three previously knowngroups of coronviruses, but it is closer to BoCoV and MHV than to other coronaviruses. 14Applying genetic algorithms to the study of protein mutation theory显示文摘Genetic algorithms were applied to the study of simulation of protein mutation carried out on two_dimensional lattice model; to the study of effects of single mutation and double mutation on protein folding and protein structure stability. It is found that in two_dimensional lattice models, replacement of inner core hydrophobic residue by hydrophilic residue will result in reduction of protein stability; at the same time the number of residues of protein surface relatively grows with increase of protein chain length; and most mutations occur in residues of protein surface, these mutations are all neutral and have no effects on protein natural structure. 19A new method of research on molecular evolution of proteinase superfamily显示文摘The molecular evolutionary tree, also known as a phylogenetic tree, of the serine proteinase superfamily was constructed by means of structural alignment. Three-dimensional structures of proteins were aligned by the SSAP program of Orengo and Taylor to obtain evolutionary distances. The resulting evolutionary tree provides a topology graph that can reflect the evolution of structure and function of homology proteinase. Moreover, study on evolution of the serine proteinase superfamily can lead to better understanding of the relationship and evolutionary difference among proteins of the superfamily, and is of significance to protein engineering, molecular design and protein structure prediction. 20Shape-selective alkylation of benzene with ethylene over a core–shell ZSM-5@MCM-41 composite material显示文摘A series of ZSM-5@MCM-41 core-shell composite materials prepared via a multi-cycle-sol-gel coating strategy is investigated as the catalyst for benzene alkylation with ethylene,in which both ethylbenzene and para-diethylbenzene(p-DEB)are aimed as the target products.With multi-cycle-sol-gel coating,the external acid sites on the samples are gradually passivated by the inert MCM-41 shell.As a result,the shape selectivity to p-DEB is greatly enhanced.Nevertheless,the coating of mesoporous MCM-41 shell on ZSM-5 accelerates deactivation of the catalyst only due to the dilution effect of ZSM-5 content in the catalyst at the same space velocity,which is a reason that core-shell ZSM-5@MCM-41 will potentially be a practical catalyst in shape selective alkylation of benzene.In order to enhance the yield of p-DEB on ZSM-5@MCM-41,the reaction conditions at the fixed bed reactor including temperature,the molar rate of benzene to ethylene and GHSV,are also optimized.Zhenhao Shen Chongwei Ma Darui Wang Junlin He Hongmin Sun Zhirong Zhu Weimin Yang 2021Chinese Journal of Chemical Engineering2021,34,9:1
