1Identification of a novel coronavirus causing severe pneumonia in human:a descriptive study显示文摘Background:Human infections with zoonotic coronaviruses(CoVs),including severe acute respiratory syndrome(SARS)-CoV and Middle East respiratory syndrome(MERS)-CoV,have raised great public health concern globally.Here,we report a novel batorigin CoV causing severe and fatal pneumonia in humans.Methods:We collected clinical data and bronchoalveolar lavage(BAL)specimens from five patients with severe pneumonia from Wuhan Jinyintan Hospital,Hubei province,China.Nucleic acids of the BAL were extracted and subjected to next-generation sequencing.Virus isolation was carried out,and maximum-likelihood phylogenetic trees were constructed.Results:Five patients hospitalized from December 18 to December 29,2019 presented with fever,cough,and dyspnea accompanied by complications of acute respiratory distress syndrome.Chest radiography revealed diffuse opacities and consolidation.One of these patients died.Sequence results revealed the presence of a previously unknownβ-CoV strain in all five patients,with 99.8%to 99.9%nucleotide identities among the isolates.These isolates showed 79.0%nucleotide identity with the sequence of SARS-CoV(GenBank NC_004718)and 51.8%identity with the sequence of MERS-CoV(GenBank NC_019843).The virus is phylogenetically closest to a bat SARS-like CoV(SL-ZC45,GenBank MG772933)with 87.6%to 87.7%nucleotide identity,but is in a separate clade.Moreover,these viruses have a single intact open reading frame gene 8,as a further indicator of bat-origin CoVs.However,the amino acid sequence of the tentative receptor-binding domain resembles that of SARS-CoV,indicating that these viruses might use the same receptor.Conclusion:A novel bat-borne CoV was identified that is associated with severe and fatal respiratory disease in humans.Li-Li Ren Ye-Ming Wang Zhi-Qiang Wu Zi-Chun Xiang Li Guo Teng Xu Yong-Zhong Jiang Yan Xiong Yong-Jun Li Xing-Wang Li Hui Li Guo-Hui Fan Xiao-Ying Gu Yan Xiao Hong Gao Jiu-Yang Xu Fan Yang Xin-Ming Wang Chao Wu Lan Chen Yi-Wei Liu Bo Liu Jian Yang Xiao-Rui Wang Jie Dong Li Li Chao-Lin Huang Jian-Ping Zhao Yi Hu Zhen-Shun Cheng Un-Lin Liu Zhao-Hui Qian Chuan Qin Qi Jin Bin Cao Jian-Wei Wang 2020Chinese Medical Journal2020,,9:99
2Clinical and experimental study on regional administration of phosphorus32 glassmicrospheres in treating hepatic carcinoma显示文摘AIM To study the therapeutical effectiveness, dosage range and toxic adverse effects of domestic phosphorus 32 glass microsphere and evaluate its clinical significance. METHODS Ⅰ. Fifty two BALB/*!c tumor bearing male nude mice were allocated into treatment group( n =38) and control group( n =14). In the former group different doses of 32 P GMS were injected into the tumor mass, while in the latter 31 P GMS or no treatment was given. The experimental animals were sacrificed in batches, and then the tumors and their nearby tissues were examined by light and electron microscopy. Ⅱ. Through selective catheterization of hepatic artery, 32 P GMS was infused to 5 healthy domestic pigs in a dosage equivalent to the therapeutic dose for human being, and 31 P GMS was infused to another 5 healthy domestic pigs. Two pigs infused with contrast medium served as whole course blank controls. One pig from each group was surrendered to euthanasia at week 1, 4, 8 and 16 respectively. The ultrastructural histopath ological changes in liver tissues taken from different sites were evaluated semiquan titatively. Ⅲ. One hundred and twenty seven times of 32 P GMS intrahepatic artery interventional therapies were performed on 93 patients with hepatic carcinoma, including 79 cases of primary hepatic carcinoma and 14 cases of secondary hepatic carcinoma. 32 P GMS ( n =30), and group B, 32 P GMS and half dose of trans hepatic artery embolization (TAE) ( n =49) , and 18 patients with HCC by TAE only as control group C. Fourteen patients with secondary hepatic carcinoma were treated in the same way as group B or C. RESULTS Ⅰ. Comparing with the control group, the treatment group of tumor bearing nude mice attained the tumor inhibition rates of 59 7%-93 7% ( F =579 62, P <0 01) at 14*!d . At an absorbed dose of 7320Gy, the tumor cells were completely destroyed. When the absorbed doses ranged from 1830Gy to 3660Gy, most of the tumor cells showed the evidences of injury or necrosis, but there appeared some well differentiated tumor cells and enhanced effect of the autoimmunocytes. At an absorbed dose of 366Gy or less, some tumor cells still remained active proliferative ability. The definite anticancer effect appeared as early as 3d after intratumoral injection of 32 P GMS. Ⅱ. The cumulative amount of 32 P GMS in the target tissue after trans hepatic artery instillation attained more than 90% of the total dose administrated. Semiquantitative analysis of ultrastructral morphology in the experimental group showed no statistical difference between the nuclear abnormality (N abn ) and mitochondrial variability (M var ) at week 1 or 2, but revealed prominent difference (χ 2=6 70-9 68, P <0 01 , χ 2=65 09-115 09, P <0 001 ) as compared with those in the other groups. In the experimental group the N abn in tissues showed no significant difference between week 8 and week 16. No apparent changes were found in the stomach, spleen, kidney and lung tissues of the experimental pigs. Ⅲ. The therapeutical results of HCC patients in group A were closely approximated to those of group C, no hematological toxic side effects were noted, and the systemic reaction was mild. In some patients 2*!mos - 3*!mos after treatment some secondary foci appeared around the periphery of the primary lesion. In general better effectiveness was obtained in patients with small lesion. After analyzing by RIDIT method, the therapeutic result in group B was significantly better than that in group C, and secondary foci around the original lesion were rarely seen at 3*!mos after treatment. In group C the collateral circulation was reestablished along the periphery of primary foci and the secondary foci appeared more frequently, and required to undergo several courses of treatment. 3The rice OsV4 encoding a novel pentatricopeptide repeat protein is required for chloroplast development during the early leaf stage under cold stress显示文摘Pentatricopeptide repeat(PPR)proteins,characterized by tandem arrays of a 35 amino acid motif,have been suggested to play central and broad roles in modulating the expression of organelle genes in plants.However,the molecular mechanisms of most rice PPR genes remains unclear.In this paper,we isolated and characterized a temperature-conditional virescent mutant,OsV4,in rice(Oryza sativa cultivar Jiahua1(WT,japonica rice variety)).The mutant displays albino phenotype and abnormal chloroplasts at the three leaf stage,which gradually turns green after the four leaf stage at a low temperature(20°C).But the mutant always develops green leaves and well-developed chloroplasts at a high temperature(32°C).Genetic and molecular analyses uncovered that OsV4 encodes a novel chloroplast-targeted PPR protein including four PPR motifs.Further investigations show that the mutant phenotype is associated with changes in chlorophyll content and chloroplast development.The OsV4transcripts only accumulate to high levels in young leaves,indicating that its expression is tissue-specific.In addition,transcript levels of some ribosomal components and plastid-encoded polymerase-dependent genes are dramatically reduced in the albino mutants grown at 20°C.These findings suggest that OsV4 plays an important role during early chloroplast development under cold stress in rice.Xiaodi Gong Qianqian Su Dongzhi Lin Quan Jiang Jianlong Xu Jianhui Zhang Sheng Teng Yanjun Dong 2014Journal of Integrative Plant Biology2014,56,4:19
4Predictive factors for the local recurrence and distant metastasis of phyllodes tumors of the breast: a retrospective analysis of 192 cases at a single center显示文摘The local recurrence rate of phyllodes tumors of the breast varies widely among different subtypes, and distant metastasis is associated with poor survival. This study aimed to identify factors that are predictive of local recurrence-free survival(LRFS), distant metastasis-free survival(DMFS), and overall survival(OS) in patients with phyllodes tumors of the breast. Clinical data of all patients with a phyllodes tumor of the breast(n = 192) treated at Sun Yat-sen University Cancer Center between March 1997 and December 2012 were reviewed. The Pearson χ2 test was used to investigate the relationship between clinical features of patients and histotypes of tumors. Univariate and multivariate Cox regression analyses were performed to identify factors that are predictive of LRFS, DMFS, and OS. In total, 31(16.1%) patients developed local recurrence, and 12(6.3%) developed distant metastasis. For the patients who developed local recurrence, the median age at the diagnosis of primary tumor was 33 years(range, 17-56 years), and the median size of primary tumor was 6.0 cm(range, 0.8-18 cm). For patients who developed distant metastasis, the median age at the diagnosis of primary tumor was 46 years(range, 24-68 years), and the median size of primary tumor was 5.0 cm(range, 0.8-18 cm). In univariate analysis, age, size, hemorrhage, and margin status were found to be predictive factors for LRFS(P = 0.009, 0.024, 0.004, and 0.001, respectively), whereas histotype, epithelial hyperplasia, margin status, and local recurrence were predictors of DMFS(P = 0.001, 0.007, 0.007, and < 0.001, respectively). In multivariate analysis, independent prognostic factors for LRFS included age [hazard ratio(HR) = 3.045, P = 0.005], tumor size(HR = 2.668, P = 0.013), histotype(HR = 1.715, P = 0.017), and margin status(HR = 4.530, P< 0.001). Histotype(DMFS: HR = 4.409, P = 0.002; OS: HR = 4.194, P = 0.003) and margin status(DMFS: HR = 2.581, P = 0.013; OS: HR = 2.507, P = 0.020) were independent predictors of both DMFS and OS. In this cohort, younger age, a larger tumor size, a higher tumor grade, and positive margins were associated with lower rates of LRFS. 5Numerical simulation of the influences of sinker weight on the deformation and load of net of gravity sea cage in uniform flow显示文摘A numerical model for determining fishing net configuration and load is developed, based on the lumped masses method. The model is used to analyze the influences on the net of gravity sea cage by different sinker weights in uniform flow. The net of gravity sea cage is simulated under different current velocities and sinker weights. In order to verify the validity of the numerical results, model test results made by Lader and Enerhaug are cited and compared with the numerical ones. The results of numerical simulation agree well with the experimental ones and the agreement is within an error range of 13% under different velocities and sinker weights. The numerical results indicate that increasing the sinker weight can effectively reduce the net deformation, but the total drag force on the net will increase accordingly. 6Metamodel-based Global Optimization Using Fuzzy Clustering for Design Space Reduction显示文摘High fidelity analysis are utilized in modern engineering design optimization problems which involve expensive black-box models.For computation-intensive engineering design problems,efficient global optimization methods must be developed to relieve the computational burden.A new metamodel-based global optimization method using fuzzy clustering for design space reduction(MGO-FCR) is presented.The uniformly distributed initial sample points are generated by Latin hypercube design to construct the radial basis function metamodel,whose accuracy is improved with increasing number of sample points gradually.Fuzzy c-mean method and Gath-Geva clustering method are applied to divide the design space into several small interesting cluster spaces for low and high dimensional problems respectively.Modeling efficiency and accuracy are directly related to the design space,so unconcerned spaces are eliminated by the proposed reduction principle and two pseudo reduction algorithms.The reduction principle is developed to determine whether the current design space should be reduced and which space is eliminated.The first pseudo reduction algorithm improves the speed of clustering,while the second pseudo reduction algorithm ensures the design space to be reduced.Through several numerical benchmark functions,comparative studies with adaptive response surface method,approximated unimodal region elimination method and mode-pursuing sampling are carried out.The optimization results reveal that this method captures the real global optimum for all the numerical benchmark functions.And the number of function evaluations show that the efficiency of this method is favorable especially for high dimensional problems.Based on this global design optimization method,a design optimization of a lifting surface in high speed flow is carried out and this method saves about 10 h compared with genetic algorithms.This method possesses favorable performance on efficiency,robustness and capability of global convergence and gives a new optimization strategy for engineering design optimization problems involving expensive black box models.LI Yulin LIU Li LONG Teng DONG Weili 2013Chinese Journal of Mechanical Engineering2013,26,5:13

8Acupuncture through Baihui (DU20) to Qubin (GB7) mitigates neurological impairment after intracerebral hemorrhage显示文摘Inflammation plays an important role in nerve defects caused by intracerebral hemorrhage. Repairing brain damage by inhibiting the macrophage-inducible C-type lectin/spleen tyrosine kinase(Mincle/Syk) signaling pathway is a potential new target for treating cerebral hemorrhage. In this study, we aimed to determine whether acupuncture through Baihui(DU20) to Qubin(GB7) is an effective treatment for intracerebral hemorrhage through the Mincle/Syk signaling pathway. An intracerebral hemorrhage rat model was established by autologous blood infusion into the caudate nucleus. Acupuncture through Baihui to Qubin was performed for 30 minutes, once every 12 hours, for a total of three times. Piceatannol(34.62 mg/kg), a Syk inhibitor, was intraperitoneally injected as a control. Modified neurological severity score was used to assess neurological function. Brain water content was measured. Immunohistochemistry and western blot assay were used to detect immunoreactivity and protein expression levels of Mincle, Syk, and CARD9. Real-time polymerase chain reaction was used to determine interleukin-1β m RNA levels. Hematoxylin-eosin staining was performed to observe histopathological changes. Our results showed that acupuncture through Baihui to Qubin remarkably improved neurological function and brain water content, and inhibited immunoreactivity and expression of Mincle, Syk, CARD9, and interkeukin-1β. Moreover, this effect was similar to piceatannol. 9Isolation and genetic charac-terization of a fragile plant mutant in rice (Oryza sativa L.)显示文摘The fragile rice mutant was isolated from an M2 population of indica variety Shuang Ke Zao (SKZ) treated with γ-rays, and designated as fp1 {fragile plant 1) because of its fragile leaves and culms. To map FP1 locus, an F2 mapping population was derived from a cross between the fp1 and C-bao, a polymorphic japonic variety. The primary mapping result places the FP1 locus in an interval between two molecular markers, microsatellite marker RM16 (3.1 cM proximal to FP1) and STS marker G144a (9.1 cM distal to FP1) in the centromere region of chromosome 3. A CAPS marker C524a was further developed between RM16 and G144a, with 0.4 cM genetic distances to the FP1 locus, providing a practical starting point for constructing a BAC con-tig spanning the FP1 locus and cloning the fp1 gene. 13The enhanced X-ray Timing and Polarimetry mission—eXTP显示文摘In this paper we present the enhanced X-ray Timing and Polarimetry mission—eXTP. eXTP is a space science mission designed to study fundamental physics under extreme conditions of density, gravity and magnetism. The mission aims at determining the equation of state of matter at supra-nuclear density, measuring effects of QED, and understanding the dynamics of matter in strong-field gravity. In addition to investigating fundamental physics, eXTP will be a very powerful observatory for astrophysics that will provide observations of unprecedented quality on a variety of galactic and extragalactic objects. 15Preparation of Tolterodine Metabolite Loaded Biodegradable PLGA Microspheres显示文摘Biodegradable polymer poly(lactic-co-glycolic acid)(PLGA) was used to encapsulate the pharmacological activity metabolite of tolterodine by means of O/W emulsion solvent evaporation method via homogenization in the emulsification process. The influences of preparation parameters were investigated. The results indicate that increasing PLGA concentration from 15% to 40% made the encapsulation efficiency of 5-hydroxymethyl derivative of tolterodine(5-HMT) increased from 55.39% to 76.32%, and the particle size increased from 34.33 μm to 70.78 μm. In addition, when homogenization speed increased from 850 r/min to 2300 r/min, both particle size and encapsulation efficiency of microspheres decreased. An increase in the volume of aqueous phase led to higher encapsulation efficiency and bigger particle size. Increasing temperature made encapsulation efficiency and particle size change significantly. While reaction temperature increased from 20 °C to 50 °C, the encapsulation efficiency decreased from 70.44% to 24.07%, and particle size increased from 38.66 μm to 69.38 μm. High reaction temperature(over 40 °C) may lead to porous surface of microspheres. 17Association of Overlapped and Un-overlapped Comorbidities with COVID-19 Severity and Treatment Outcomes: A Retrospective Cohort Study from Nine Provinces in China显示文摘Objective Several COVID-19 patients have overlapping comorbidities. The independent role of each component contributing to the risk of COVID-19 is unknown, and how some non-cardiometabolic comorbidities affect the risk of COVID-19 remains unclear.Methods A retrospective follow-up design was adopted. A total of 1,160 laboratory-confirmed patients were enrolled from nine provinces in China. Data on comorbidities were obtained from the patients’ medical records. Multivariable logistic regression models were used to estimate the odds ratio(OR) and 95% confidence interval(95% CI) of the associations between comorbidities(cardiometabolic or non-cardiometabolic diseases), clinical severity, and treatment outcomes of COVID-19.Results Overall, 158(13.6%) patients were diagnosed with severe illness and 32(2.7%) had unfavorable outcomes. Hypertension(2.87, 1.30–6.32), type 2 diabetes(T2 DM)(3.57, 2.32–5.49),cardiovascular disease(CVD)(3.78, 1.81–7.89), fatty liver disease(7.53, 1.96–28.96), hyperlipidemia(2.15, 1.26–3.67), other lung diseases(6.00, 3.01–11.96), and electrolyte imbalance(10.40, 3.00–26.10)were independently linked to increased odds of being severely ill. T2 DM(6.07, 2.89–12.75), CVD(8.47,6.03–11.89), and electrolyte imbalance(19.44, 11.47–32.96) were also strong predictors of unfavorable outcomes. Women with comorbidities were more likely to have severe disease on admission(5.46,3.25–9.19), while men with comorbidities were more likely to have unfavorable treatment outcomes(6.58, 1.46–29.64) within two weeks.Conclusion Besides hypertension, diabetes, and CVD, fatty liver disease, hyperlipidemia, other lung diseases, and electrolyte imbalance were independent risk factors for COVID-19 severity and poor treatment outcome. 18High Resolution Radar Real-Time Signal and Information Processing显示文摘Radar is an electronic device that uses radio waves to determine the range, angle, or velocity of objects. Real-time signal and information processor is an important module for real-time positioning, imaging, detection and recognition of targets. With the development of ultra-wideband technology, synthetic aperture technology, signal and information processing technology, the radar coverage, detection accuracy and resolution have been greatly improved, especially in terms of one-dimensional(1D) high-resolution radar detection, tracking, recognition, and two-dimensional(2D) synthetic aperture radar imaging technology. Meanwhile, for the application of radar detection and remote sensing with high resolution and wide swath, the amount of data has been greatly increased. Therefore, the radar is required to have low-latency and real-time processing capability under the constraints of size, weight and power consumption. This paper systematically introduces the new technology of high resolution radar and real-time signal and information processing. The key problems and solutions are discussed, including the detection and tracking of 1D high-resolution radar, the accurate signal modeling and wide-swath imaging for geosynchronous orbit synthetic aperture radar, and real-time signal and information processing architecture and efficient algorithms. 19Protective effect of Saccharomyces boulardii on intestinal mucosal barrier of dextran sodium sulfate-induced colitis in mice显示文摘Background: The effect and mechanism of Saccharomyces boulardii (Sb) in inflammatory bowel disease are unclear. The objective of the study was to evaluate the impact of Sb on intestinal mucosal barrier and intestinal flora in a colitis mouse model. Methods: Forty C57BL/6J male mice were randomly assigned to five groups: normal control group (A), pathologic control group (B), Sb treatment group (C), mesalazine treatment group (D), and Sb combined with mesalazine treatment group (E). Colitis was induced by the addition of 2.5%(wt/vol) dextran sodium sulfate (DSS) in the drinking water ad libitum for 7 days. The general condition, weight change, stool property, and bloody stool level of mice were observed to evaluate the disease activity index. The expression of zona occludens-1 (ZO-1) and occludin in intestinal tissue were measured by immunohistochemistry. The level of tumor necrosis factor-α(TNF-α) and interleukin (IL)-8 in plasma was measured by enzyme linked immunosorbent assay. Inter-cellular tight junctions were observed by transmission electron microscopy. The feces and intestinal contents were collected sterilely, and intestinal flora was analyzed by 16S rRNA sequencing. Results: Compared with group B, Sb reduced the disease activity index and histological score of group C (disease activity index: group B 2.708 ± 0.628, group C 1.542 ± 0.616, PBC = 0.005;histological score: group B 9.875 ± 3.271, group C 4.750 ± 1.832, PBC = 0.005) in DSS-induced colitis in mice. Sb exerted a protect effect on the expression of ZO-1 (group B 2.075 ± 1.176, group C 4.225 ± 1.316, PBC = 0.019) and occludin (group B 2.200 ± 0.968, group C 3.525 ± 1.047, PBC = 0.023). Compared with group B, Sb decreased the level of TNF-α and IL-8 of group C (TNF-α: group B 716.323 ± 44.691 ng/L, group C 521.740 ± 90.121 ng/L, PBC = 0.001;IL-8: group B 128.992 ± 11.475 pg/mL, group C 106.283 ± 15.906 pg/mL, PBC = 0.012). Treatment with Sb preserved the tight junctions and ameliorated microvilli and inter-cellular space. Treatment with Sb also showed its own characteristics: a higher percentage of Bacteroidetes and a lower percentage of Firmicutes, with significant differences or a significant trend. The proportion of the S24-7 family was increased significantly in the Sb treatment group. Conclusions: Sb shows an anti-inflammatory effect and has a protective effect on the intestinal mucosal mechanical barrier. 20Biofilm Microenvironment-Responsive Nanotheranostics for Dual-Mode Imaging and Hypoxia-Relief-Enhanced Photodynamic Therapy of Bacterial Infections显示文摘The formation of bacterial biofilms closely associates with infectious diseases.Until now,precise diagnosis and effective treatment of bacterial biofilm infections are still in great need.Herein,a novel multifunctional theranostic nanoplatform based on MnO_(2)nanosheets(MnO_(2)NSs)has been designed to achieve pH-responsive dual-mode imaging and hypoxia-relief-enhanced antimicrobial photodynamic therapy(aPDT)of bacterial biofilm infections.In this study,MnO_(2)NSs were modified with bovine serum albumin(BSA)and polyethylene glycol(PEG)and then loaded with chlorin e_(6)(Ce_(6))as photosensitizer to form MnO_(2)-BSA/PEG-Ce_(6)nanosheets(MBP-Ce_(6)NSs).After being delivered into the bacterial biofilm-infected tissues,the MBP-Ce_(6)NSs could be decomposed in acidic biofilm microenvironment and release Ce_(6)with Mn^(2+),which subsequently activate both fluorescence(FL)and magnetic resonance(MR)signals for effective dual-mode FL/MR imaging of bacterial biofilm infections.Meanwhile,MnO_(2)could catalyze the decomposing of H2O_(2)in biofilm-infected tissues into O_(2)and relieve the hypoxic condition of biofilm,which significantly enhances the efficacy of aPDT.An in vitro study showed that MBP-Ce_(6)NSs could significantly reduce the number of methicillin-resistant Staphylococcus aureus(MRSA)in biofilms after 635nm laser irradiation.Guided by FL/MR imaging,MRSA biofilm-infected mice can be efficiently treated by MBP-Ce_(6)NSs-based aPDT.Overall,MBP-Ce_(6)NSs not only possess biofilm microenvironment-responsive dual-mode FL/MR imaging ability but also have significantly enhanced aPDT efficacy by relieving the hypoxia habitat of biofilm,which provides a promising theranostic nanoplatform for bacterial biofilm infections.Weijun Xiu Siyu Gan Qirui Wen Qiu Qiu Sulai Dai Heng Dong Qiang Li Lihui Yuwen Lixing Weng Zhaogang Teng Yongbin Mou Lianhui Wang 2020Research2020,,1:7
