| 1 | Antisense MMP-9 RNA inhibits malignant glioma cell growth in vitro and in vivo显示文摘The matrix-degrading metalloproteinases (MMPs), particularly MMP-9, play important roles in the pathogenesis and development of malignant gliomas. In the present study, the oncogenic role of MMP-9 in malignant glioma cells was investigated via antisense RNA blockade in vitro and in vivo. TJ905 malignant glioma cells were transfected with pcDNA3.0 vector expressing antisense MMP-9 RNA (pcDNA-AS-MMP9), which significantly decreased MMP-9 expression, and cell proliferation was assessed. For in vivo studies, U251 cells, a human malignant glioma cell line, were implanted subcutaneously into 4-to 6-week-old BALB/c nude mice. The mice bearing well-established U251 gliomas were treated with intratumoral pcDNA-AS-MMP9-Lipofectamine complex (AS-MMP-9-treated group), subcutaneous injection of endostatin (endostatin-treated group), or both (combined therapy group). Mice treated with pcDNA (empty vector)-Lipofectamine served as the control group. Four or eight weeks later, the volume and weight of tumor, MMP-9 expression, microvessel density and proliferative activity were assayed. We demonstrate that pcDNA-AS-MMP9 significantly decreased MMP-9 expression and inhibited glioma cell proliferation. Volume and weight of tumor, MMP-9 expression, microvessel density and proliferative activity in the antisense-MMP-9-treated and therapeutic alliance groups were significantly lower than those in the control group. The results suggest that MMP-9 not only promotes malignant glioma cell invasiveness, but also affects tumor cell proliferation. | 4 | Comparison of Viromes in Ticks from Different Domestic Animals in China显示文摘Ticks are involved in the transmission of various arboviruses and some tick-borne viruses pose significant threats to the health of humans or livestock.This study aimed to investigate the geographical distribution of tick species and tickassociated viruses in central and eastern China.Total 573 ticks from domestic animals including dogs,sheep and cattle were collected in 2017.Two genera of ticks were identified including Rhipicephalus and Haemaphysalis.Sequencing was performed on Miseq Illumina platform to characterize the tick viromes from the four different sampling locations.Following trimming,13,640 reads were obtained and annotated to 19 virus families.From these sequences,above 37.74%of the viral reads were related to the RNA viruses.Virome comparison study revealed that the tick viral diversity was considerably different in the two identified tick genera.The viral diversity of R.microplus was significantly different from that of other Rhipicephalus species.On the other hand,substantial overlap in viral species was observed between the same genera.In addition,we found no evidence that the natural host played a major role in shaping virus diversity based on the comparison of their viromes.Rather,the geographic location seems to significantly influence the viral families.Phylogenetic study indicated that the novel negative-sense RNA viruses identified in this study was closely related to Bole tick virus 1 and 3 viruses.In conclusion,the present study provides a baseline for comparing viruses detected in ticks,according to species,natural hosts,and geographic locations. | Tingting Zhao Haiyan Gong Xiaojuan Shen Wen Zhang Tongling Shan Xiangqian Yu Seong Jin Wang Li Cui | 2020 | Virologica Sinica2020,35,4: | 2 |

| 10 | Synthesis of metallic glasses and metallic glass based composites in the Cu-Mo-Hf system by ion beam mixing显示文摘Single-phase and dual-phase metallic glasses as well as metallic glass based composites were synthesized in the Cu-Mo-Hf ternary metal system by 200 keV xenon ion beam mixing of far-from-equilibrium. It was found that Mo-Hf-based and Cu-Mo-based single-phase metallic glasses could be obtained at compositions around Cu17Mo20Hf63 and Cu34Mo57Hf9, respectively. Interestingly, at the nearly equal-atomic stoichiometry of Cu38Mo31Hf31, a dual-phase Cu-Mo-Hf metallic glass, consisting both of the Mo-Hf-based and Cu-Mo-based phases, was first obtained at relatively low irradiation doses ranging from (1-5)×1015 Xe+/cm2, and a single-phase metallic glass was eventually obtained at a dose of 7×1015 Xe+/cm2. In addition, two glass-based composites were obtained at the compositions of Cu14Mo62Hf24 and Cu77Mo14Hf9, and they consisted of the Mo-Hf based and Cu-Mo based metallic glasses, dissolved with some uniformly distributed BCC Mo-based and FCC Cu-based crystalline solid solutions, respectively. | 15 | Ultra-sensitive Dirac-point-based biosensing on terahertz metasurfaces comprising patterned graphene and perovskites显示文摘Biosensors are a focus of research on terahertz metasurfaces. However, reports of ultra-sensitive biosensors based on Dirac points are rare. Here, a new terahertz metasurface is proposed that consists of patterned graphene and perovskites. This serves as an ultra-sensitive Dirac-point-based biosensor for qualitative detection of sericin.Theoretically, sericin may make graphene n-doped and drive the Fermi level to shift from the valence band to the Dirac point, causing a dramatic decrease in conductivity. Correspondingly, the dielectric environment on the metasurface undergoes significant change, which is suited for ultra-sensitive biosensing. In addition, metal halide perovskites, which are up-to-date optoelectronic materials, have a positive effect on the phase during terahertz wave transmission. Thus, this sensor was used to successfully detect sericin with a detection limit of 780 pg/m L, achieved by changing the amplitude and phase. The detection limit of this sensor is as much as one order of magnitude lower than that of sensors in published works.
