| 1 | BYSL contributes to tumor growth by cooperating with the mTORC2 complex in gliomas显示文摘Objective:BYSL,which encodes the Bystin protein in humans,is upregulated in reactive astrocytes following brain damage and/or inflammation.We aimed to determine the role and mechanism of BYSL in glioma cell growth and survival.Methods:BYSL expression in glioma tissues was measured by quantitative real-time PCR,Western blot,and immunohistochemistry.In vitro assays were performed to assess the role of BYSL in cell proliferation and apoptosis.Protein interactions and co-localization were determined by co-immunoprecipitation and double immunofluorescence.The expression and activity of the AKT/m TOR signaling molecules were determined by Western blot analysis,and the role of BYSL in glioma growth was confirmed in an orthotopic xenograft model.Results:The BYSL m RNA and protein levels were elevated in glioma tissues.Silencing BYSL inhibited glioma cell proliferation,impeded cell cycle progression,and induced apoptosis,whereas overexpressing BYSL protein led to the opposite effects.We identified a complex consisting of BYSL,RIOK2,and m TOR,and observed co-localization and positive correlations between BYSL and RIOK2 in glioma cells and tissues.Overexpressing BYSL or RIOK2 increased the expression and activity of AKT/m TOR signaling molecules,whereas downregulation of BYSL or RIOK2 decreased the activity of AKT/m TOR signaling molecules.Silencing BYSL or RIOK2 decreased the growth of the tumors and prolonged the lifespan of the animals in an orthotopic xenograft model.Conclusions:High expression of BYSL in gliomas promoted tumor cell growth and survival both in vitro and in vivo.These effects could be attributed to the association of BYSL with RIOK2 and m TOR,and the subsequent activation of AKT signaling. | Shangfeng Gao Zhuang Sha Junbo Zhou Yihao Wu Yunnong Song Cheng Li Xuejiao Liu Tong Zhang Rutong Yu | 2021 | Cancer Biology & Medicine2021,18,1: | 3 |
| 2 | Anisotropic Total Variation Regularization Based NAS-RIF Blind Restoration Method for OCT Image显示文摘Based on anisotropic total variation regularization(ATVR), a nonnegativity and support constraints recursive inverse filtering(NAS-RIF) blind restoration method is proposed to enhance the quality of optical coherence tomography(OCT) image. First, ATVR is introduced into the cost function of NAS-RIF to improve the noise robustness and retain the details in the image.Since the split Bregman iterative is used to optimize the ATVR based cost function, the ATVR based NAS-RIF blind restoration method is then constructed. Furthermore, combined with the geometric nonlinear diffusion filter and the Poisson-distribution-based minimum error thresholding, the ATVR based NAS-RIF blind restoration method is used to realize the blind OCT image restoration. The experimental results demonstrate that the ATVR based NAS-RIF blind restoration method can successfully retain the details in the OCT images. | 4 | Deciphering the pharmacological mechanism of Guan-Jie-Kang in treating rat adjuvant-induced arthritis using omics analysis显示文摘Traditional Chinese medicine (TCM) formulas have attracted increasing attention worldwide in the past few years for treating complex disease including rheumatoid arthritis. However, their mechanisms are complex and remain unclear. Guan-Jie-Kang (GJK), a prescription modified from “Wu Tou Decoction,” was found to significantly relieve arthritis symptoms in rats with adjuvant-induced arthritis after 30-day treatment, especially in the 24 g/kg/day group. By analyzing 1749 targets related to 358 compounds in the five herbs of GJK, we identified the possible anti-arthritis pathways of GJK, including the calcium signaling and metabolic pathways. Bone damage levels were assessed by micro-computed tomography, and greater bone protective effect was observed with GJK treatment than with methotrexate. Receptor activator of nuclear factor kB ligand (RANKL)-RANK signaling, which is related to calcium signaling, was significantly regulated by GJK. Moreover, a target metabolomics assay of serum was conducted;17 metabolic biomarkers showed significant correlations with treatment. An integrated pathway analysis revealed that pyruvate metabolism, purine metabolism, and glycolysis metabolism were significantly associated with the effects of GJK in arthritis treatment. | 5 | Isolation,cultivation and identification of brain glioma stem cells by magnetic bead sorting显示文摘This study describes a detailed process for obtaining brain glioma stem cells from freshly dissected human brain glioma samples using an immunomagnetic bead technique combined with serum-free media pressure screening.Furthermore,the proliferation,differentiation and self-renewal biological features of brain glioma stem cells were identified.Results showed that a small number of CD133 positive tumor cells isolated from brain glioma samples survived as a cell suspension in serum-free media and proliferated.Subcultured CD133 positive cells maintained a potent self-renewal and proliferative ability,and expressed the stem cell-specific markers CD133 and nestin.After incubation with fetal bovine serum,the number of glial fibrillary acidic protein and microtubule associated protein 2 positive cells increased significantly,indicating that the cultured brain glioma stem cells can differentiate into astrocytes and neurons.Western blot analysis showed that tumor suppressor phosphatase and tensin homolog was highly expressed in tumor spheres compared with the differentiated tumor cells.These experimental findings indicate that the immunomagnetic beads technique is a useful method to obtain brain glioma stem cells from human brain tumors. | Xiuping Zhou Chao Zheng Qiong Shi Xiang Li Zhigang Shen Rutong Yu | 2012 | Neural Regeneration Research2012,7,13: | 1 |
| 6 | Transcriptome analysis of Clematis lanuginosa: Novel features of the molecular events occurring under heat-shock stress显示文摘The weak heat tolerance of Clematis ornamental varieties negatively affects their ornamental qualities in the summer.To elucidate heat resistance mechanisms,Clematis lanuginosa,which is an important original parent of the Clematis large-flowered group of ornamental varieties,was selected for use in this study.Here,six libraries,including three biological replicates each of control and heat-shock stress samples,were determined using RNA-sequencing technology.In total,62,050 unigenes were obtained,and 6,439 unigenes exceeded 1 kb in length.A total of 42,377 unigenes were annotated using six databases.Between the two treatments,2,165 differentially expressed genes were identified,with 1,565 being up-regulated and 600 down-regulated.In addition,51 heat-shock protein-encoding genes were identified,among which the small heat-shock proteins accounted for 68.63%.In total,two heat-shock factors and 12 ribosomal proteins were significantly up-regulated under heatstress conditions.The differential expressions of ethylene-responsive transcription factor,chalcone synthase,cysteine-rich receptor-like kinase and aspartic protease unigenes in guard cells were induced by heat-shock.The data obtained will assist the elucidation of the molecular events underlying heat-stress responses in C.lanuginosa. | Linfang Li Shuan Wang Qiao Li Linghe Sun Xin Han Lulu Gao Peng Wang Fenni Lv Rutong Yang Ya Li | 2022 | Ornamental Plant Research2022,2,1: | 0 |

| 10 | Diversity Assessment of an Endemic Carpinus oblongifolia(Betulaceae)Using Specific-Locus Amplified Fragment Sequencing and Implications for Conservation显示文摘Carpinus oblongifolia is an endemic species and the extant wild populations show a fragmentation distribution in the Baohua Mountain of Jiangsu Province in eastern China.Understanding of genetic diversity plays an important role in C.oblongifolia survival and sustainable development.The wild C.oblongifolia population was artificially divided into four subpopulations according to the microhabitats,and another two subpopulations were constructed by progeny seedlings cultivated with the mature seeds.Then,the leaf buds of 80 individuals from six subpopulations were sampled to develop single nucleotide polymorphisms(SNPs)using specific-locus amplified fragment sequencing(SLAF-seq).Based on these SNPs,we aimed to characterize the genetic diversity and population structure of C.oblongifolia and provide an illumination and reference for effective management of such a small endemic population.The level of genetic diversity was low at the species level,and the progeny subpopulations had a relatively higher genetic diversity than the wild subpopulations.This may be attributed to a high gene flow and an excess heterozygosity to reduce the threat of genetic drift-based hazards.Moreover,the progeny subpopulations had the ability to form new clusters and a great contribution to the genetic structure variation of C.oblongifolia.These results will assist with the development of conservation and management strategies,such as properly evacuating competitive trees to provide more chance for pollen and seed flow in situ conservation,and establishing sufficient seedling plantlets under laboratory conditions for reintroduction to enlarge the effective population size. | Sumei Li Qing Wang Peng Wang Shuan Wang Rutong Yang Ya Li | 2022 | Phyton-International Journal of Experimental Botany2022,91,3: | 0 |
