1Application of next-generation sequencing technology to precision medicine in cancer: joint consensus of the Tumor Biomarker Committee of the Chinese Society of Clinical Oncology显示文摘Next-generation sequencing(NGS) technology is capable of sequencing millions or billions of DNA molecules simultaneously.Therefore, it represents a promising tool for the analysis of molecular targets for the initial diagnosis of disease, monitoring of disease progression, and identifying the mechanism of drug resistance. On behalf of the Tumor Biomarker Committee of the Chinese Society of Clinical Oncology(CSCO) and the China Actionable Genome Consortium(CAGC), the present expert group hereby proposes advisory guidelines on clinical applications of NGS technology for the analysis of cancer driver genes for precision cancer therapy. This group comprises an assembly of laboratory cancer geneticists, clinical oncologists, bioinformaticians,pathologists, and other professionals. After multiple rounds of discussions and revisions, the expert group has reached a preliminary consensus on the need of NGS in clinical diagnosis, its regulation, and compliance standards in clinical sample collection. 2Modified FOLFIRINOX for unresectable locally advanced or metastatic gallbladder cancer,a comparison with GEMOX regimen显示文摘Background:The first-line chemotherapy regimen for advanced gallbladder cancer(GBC)is gemcitabine plus platinum(GP),despite its efficacy is limited.The current investigation is a retrospective study to compare the safety and efficacy between the modified FOLFIRINOX(mFOLFIRINOX)and gemcitabine plus oxaliplatin(GEMOX)as the first-line chemotherapy for unresectable locally advanced or metastatic GBC.Methods:The data of patients with unresectable locally advanced or metastatic GBC,who were treated with mFOLFIRINOX or GEMOX as the first-line therapy between April 2014 and April 2018 at Xinhua Hospital Affiliated to Shanghai Jiaotong University School of Medicine,were retrieved.This retrospective study evaluated the clinical characteristics,survival outcomes and adverse events.Results:A total of 44 patients(n=25 in mFOLFIRINOX,n=19 in GEMOX)were included.There were no significant differences between groups in baseline characteristics.The median progression free survival(mPFS)was 5.0 months in the mFOLFIRINOX group and 2.5 months in the GEMOX group[P=0.021;hazard ratio(HR),0.499;95%CI,0.266 to 0.937].The median overall survival(mOS)was 9.5 months in the mFOLFIRINOX group and 7.0 months in the GEMOX group(P=0.019;HR,0.471;95%CI,0.239 to 0.929).Disease control rate(DCR)was 76.0%in the mFOLFIRINOX group and 47.4%in the GEMOX group(P=0.051).The rate of grade 3-4 adverse events was 48%in the mFOLFIRINOX group and 36.8%in the GEMOX group(P=0.459).The incidence of grade 3-4 neutropenia and diarrhea were more common in the mFOLFIRINOX group,while the incidence of grade 3-4 thrombocytopenia and peripheral neuropathy were more common in the GEMOX group.Conclusions:mFOLFIRINOX might improve the poor prognosis of unresectable locally advanced or metastatic GBC,and the results need to be further verified by prospective clinical studies.Xu-Ya Cui Xue-Chuan Li Jiu-Jie Cui Xiang-Song Wu Lu Zou Xiao-Ling Song Tai Ren Yi-Di Zhu Huai-Feng Li Yang Yang Ke Liu Xu-Sheng Han Zi-Yao Jia Wen-Guang Wu Xu-An Wang Wei Gong Li-Wei Wang Mao-Lan Li Ying-Bin Liu 2021Hepatobiliary Surgery and Nutrition2021,10,4:10
3Correlation between BRAF^(V600E) mutation and clinicopathological features in pediatric papillary thyroid carcinoma显示文摘In adults, the presence of the BRAF^(V600E) mutation in papillary thyroid cancer(PTC) has been demonstrated to be strongly associated with aggressive cancer-cell characteristics and poor patient prognosis. In contrast, the frequency of this mutation in pediatric PTC has undergone limited study, and the few available estimates range from 0 to 63%. Furthermore, the role of the BRAF^(V600E) mutation in pediatric PTC is controversial; thus, the present study aimed to investigate the prevalence and role of the BRAF^(V600E) mutation in48 pediatric patients with PTC, aged 3–13 years. Of these patients, 41 were diagnosed with classic PTC, five were found to have a follicular variant of PTC, and two to exhibit a diffuse sclerosing PTC variant. The BRAF^(V600E) mutation was identified to be present in 35.4% of the 48 analyzed patients, and in 41.5% of the patients diagnosed with classical PTC. Furthermore, the presence of the BRAF^(V600E) mutation was found to be associated with a patient age at diagnosis of less than ten years(P=0.011), the performance of a thyroidectomy(P=0.03), exhibited tumor multifocality(P=0.02) and/or extra-thyroidal invasion(P=0.003), and both a low MACIS(Metastases, Age, Completeness of resection, Invasion, Size)(P=0.036) and AMES(Age, Metastasis, Extent of tumor,Size)(P=0.001)score. 4Lignin biosynthesis by suppression of two O-methyl-transferases显示文摘Caffeic acid O-methyltransferase (COMT) and caffeoyl-CoA-3-O-methyltransferase (CCoAOMT) genes encode two methyltransferases at different substrate levels in lignin biosynthesis. We constructed two single antisense expression vectors containing the cDNA of COMT or CCoAOMT genes and one dual antisense expression vector containing cDNAs of both OMTs genes. The antisense constructs were transferred into tobacco mediated by Agrobac-terium tumefacience. PCR-Southern analysis indicated that antisense cDNAs had been integrated into the genome of the transgenic tobacco. The antisense genes were also expressed at transcriptional level displayed by Northern dot analysis. Klason lignin assay of 3-month-old transgenic tobaccos showed that repression in COMT or CCoAOMT alone could result in reduction of lignin content, more reduction was caused by suppression in CCoAOMT than in COMT. 5Protective role of hydrogen-rich water on aspirin-induced gastric mucosal damage in rats显示文摘AIM: To investigate the role of the hydrogen-rich water(HRW) in the prevention of aspirin-induced gastric mucosal injury in rats. METHODS: Forty male rats were allocated into four groups: normal control group, HRW group, aspirin group, and HRW plus aspirin group. The protective efficacy was tested by determining the gastric mucosal damage score. Malondialdehyde(MDA), superoxide dismutase(SOD), myeloperoxidase(MPO), interleukin(IL)-06 and tumor necrosis factor(TNF)-α in gastric tissues were evaluated. The serum levels of IL-1β and TNF-α were also detected. Histopathology of gastric tissues and localization of Cyclooxygenase 2(COX-2) were detected using hematoxylin and eosin staining and immunohistochemistry, respectively. RESULTS: Pretreatment with HRW obviously reduced aspirin-induced gastric damage scores(4.04 ± 0.492 vs 2.10 ± 0.437, P < 0.05). The oxidative stress levels of MDA and MPO in the gastric tissues increased significantly in the aspirin-treated group compared with the HRW group(2.43 ± 0.145 vs 1.79 ± 0.116 nmol/mg prot, P < 0.05 and 2.53 ± 0.238 vs 1.40 ± 0.208 U/g tissue, P < 0.05, respectively). HRW could obviously elevated the SOD levels in the gastric tissues(37.94 ± 8.44 vs 59.55 ± 9.02 nmol/mg prot, P < 0.05). Pretreatment with HRW significantly reduced IL-06 and TNF-α in the gastric tissues(46.65 ± 5.50 vs 32.15 ± 4.83 pg/mg, P < 0.05 and 1305.08 ± 101.23 vs 855.96 ± 93.22 pg/mg, P < 0.05), and IL-1β and TNF-α in the serum(505.38 ± 32.97 vs 343.37 ± 25.09 pg/mL, P < 0.05 and 264.53 ± 28.63 vs 114.96 ± 21.79 pg/mL, P < 0.05) compared to treatment with aspirin alone. HRW could significantly decrease the COX-2 expression in the gastric tissues(staining score: 8.4 ± 2.1 vs 2.9 ± 1.5, P < 0.05). 6Cloud point extraction and flame atomic absorption spectrometry analysis of palladium, platinum, and gold ions from industrial polluted soil显示文摘Cloud point extraction (CPE) with Tergitol TMN-6 was applied for the extraction of trace amounts of palladium (Pd(Ⅱ)), platinum (Pt(Ⅳ)), and gold (Au(Ⅲ)) in the soil of industrial sewage. Ammonium pyrolysine dithiocarbamate (APDC) was adopted as the chelating agent prior to CPE and then was detected by atomic absorption spectrometry (AAS). Different parameters such as the concentration of surfactants, chelating agent and salt, sample pH, equilibration temperature and time, centrifugation time and rates, and the effect of foreign ions were studied. Under optimum conditions, the low limits of detections are 1.4, 2.8 and 1.2 ng·ml^-1 and the enrichment factors are 21, 12, and 24 for Pd(Ⅱ), Pt(Ⅳ), and Au(Ⅲ, respectively. The relative standard deviations vary from 0.6% to 1.0% (n=11). All correlation coefficients of the calibration curves are >0.9960. 7Curved copper nanowires-based robust flexible transparent electrodes via all-solution approach显示文摘Curved Cu nanowire (CCN)-based high-performance flexible transparentconductive electrodes (FTCEs) were fabricated via a fully solution-processedapproach, involving synthesis, coating, patterning, welding, and transfer. Eachstep involved an innovative technique for completing the all-solution processes.The high-quality and well-dispersed CCNs were synthesized using a multi-polyolmethod through the synergistic effect of specific polyol reduction. To preciselycontrol the optoelectrical properties of the FTCEs, the CCNs were uniformlycoated on a polyimide (PI) substrate via a simple meniscus-dragging depositionmethod by tuning several coating parameters. We also employed a polyurethane(PU)-stamped patterning method to effectively produce 20/Jm patterns on CCNthin films. The CCN thin films exhibited high electrical performance, which isattributed to the deeply percolated CCN network formed via a solvent-dippedwelding method. 8Effects of Different Covering Ways on Emergence Rate of Sophora japonica L.Afforested by Direct Seeding显示文摘In order to maintain higher emergence rate with direct seeding in Shandong rocky mountain, effects of different covering ways on emergence rate of Sophora japonica L. direct seeding were studied. The results showed that both mulching film and grass cover could effectively increase the moisture content of the soil at the depth of 0-30 cm, contributing to soil moisture conservation and the inhibition of soil evaporation, which created favorable moisture environment for the growth of S. japonica seeds. Both the emergence rate and the seedling height under mulching film had greater improvement and partly enhanced by 26.65% and 6.85 cm respectively, and mulching film also helped S. japonica seeds emerge and ensured the seedling growth. The emergence rate and the seedling height under grass cover had been also raised in some degree, but they were much worse than those under mulching film. The emergence rate and the seedling height under block cover were not significantly increased. 11Down-regulation of FoxM1 inhibits viability and invasion of gallbladder carcinoma cells, partially dependent on inducement of cellular senescence显示文摘AIM:To investigate the effect of knockdown of forkhead box M1(foxM1)on the proliferation and invasion capacities of human gallbladder carcinoma(Gbc)-sd cells.METHODS:four foxM1 shRNAs were transfected into Gbc-sd cells with lipofectamine 2000 to select the appropriate shRNA for down-regulation of foxM1.A recombinant lentivirus for shfoxM1(lv-shfoxM1),which expresses FoxM1-specific shRNA,and a negative control carrying green fluorescent protein,which expresses a scrambled RNA,were constructed.After transfection with the recombinant adenovirus and screened with puromycin,RT-PcR and Western blot were utilized to evaluate the inhibition efficiency.cell viability was evaluated by MTT assay,and cell migration and invasion were assessed using the Transwell system.cells were suspended in serum-free medium and seeded into Transwell inserts either uncoated(for migration assay)or coated(for invasion assay)with growth factor-reduced Matrigel.To verify the involvement of foxM1 in the senescence of tumor cells,staining of senescenceβ-galactosidase(sAβ-gal),the widely used biomarker of cellular senescence,was also performed.RESULTS:After successful transfection of four foxM1small interfering RNAs(shRNAs)with lipofectamine2000,the shf1822 was selected as the most appropriate shRNA according to its obvious inhibitory effect.The recombinant adenovirus was then constructed with the shf1822 and successfully transfected into the Gbcsd cells,resulting in the significant inhibition of foxM1expression at both the mRNA and protein levels,compared with the negative control(P<0.05).After transfection,down-regulation of foxM1 significantly inhibited cell viability according to the MTT assay(P<0.05).In addition,Transwell migration and invasion assays also suggested the suppression of invasion ability of the transfected cells.sAβ-gal staining showed that downregulation of foxM1 could induce more senescent Gbc cells(P<0.05),suggesting the possible involvement of the senescence process of the foxM1-deficient cells in Gbc.CONCLUSION:foxM1 is functionally involved in viability of Gbc cells,partially dependent on the inducement of cellular senescence,and is a potential target for Gbc therapy.Jie Tao Xin-sen Xu Yan-zhou song Kai Qu Qi-fei Wu Rui-Tao Wang fan-di Meng Ji-chao Wei shun-bin dong Yue-lang zhang Min-hui Tai Ya-feng dong Lin Wang Chang liu 2014World Journal of Gastroenterology2014,20,28:4

13Correlation between TERT C228T and clinic-pathological features in pediatric papillary thyroid carcinoma显示文摘The aims of the present study were to reveal the prevalence of the TERT C228 T mutation in pediatric papillary thyroid carcinoma(PPTC) and to further investigate the role of the TERT C228 T mutation in PPTC. We also tested another TERT mutation, TERT C250 T, although this was not detected in PPTC patients. In this study, 48 patients with PPTC(41 with classic PPTC) were enrolled. DNA was extracted from PPTC tissues and TERT C228 T mutation analysis was performed. Chi-squared analysis,Fisher’s exact test, and a t-test were applied to test the significance of differences. The TERT C228 T mutation presented in 13(27.1%) of the 48 PPTC patients and 10(24.4%) of the 41 classical PPTC patients. There were significant differences between PPTC patients with the TERT C228 T mutation and those without in terms of modified radical neck dissection, multifocality,capsular invasion, extrathyroidal invasion, and American Joint Committee on Cancer(AJCC) tumor stage(P<0.05). 14Design of wide-range energy material beamline at the Shanghai Synchrotron Radiation Facility显示文摘We report the design of a wide-range energy material beamline(E-line) with multiple experimental techniques at the Shanghai Synchrotron Radiation Facility.The undulators consisted of an elliptically polarizing undulator and in-vacuum undulator that generate the soft and hard X-rays, respectively. The beamline covered a wide energy range from 130 to 18 ke V with both a high photon flux([ 10^(12) phs/s with exit silt 30 lm in soft X-ray and [ 5 9 10^(12) phs/s in hard X-ray within 0.1%BW bandwidth) and promising resolving power(maximum E/DE [ 15,000 in soft X-ray with exit silt 30 lm and [6000 in hard X-ray). 15Thrombocytopenia for prediction of hepatocellular carcinoma recurrence: Systematic review and meta-analysis显示文摘AIM: To investigate the association between thrombocytopenia and relapse after treatment for hepatocellular carcinoma(HCC).METHODS: We searched the Pub Med, EMBASE, and Web of Science databases to obtain eligible studies. The hazard ratios(HRs) values and 95% confidence intervals(CIs) were pooled by random effects model. Subsequently, we estimated the heterogeneity, performed a sensitivity analysis, determined the publication bias, and performed subgroup and meta-regression analyses. Study quality was assessed by using the Oxford Center for Evidence Based Medicine tool.RESULTS: We identified 18 eligible studies by retrieval(published during 2000-2014). Out of the 4163 patients with HCC who were recruited, 2746(66.0%) experienced recurrence. In general, our meta-analysis suggested that low platelet count(PLT) before therapy significantly increased the probability of postoperative recurrence(HR = 1.53, 95%CI: 1.29-1.81). PLT was also valuable in the prediction of intrahepatic distant recurrence(HR = 1.49, 95%CI: 1.25-1.77). Subgroupand meta-regression analyses identified various therapeutic modalities as the source of a high degree of heterogeneity. The pooled HR values showed no obvious change when a single study was removed, but otherwise, an opposite-effects model was used.
