| 3 | Single-cell RNA-seq highlights intra-tumoral heterogeneity and malignant progression in pancreatic ductal adenocarcinoma显示文摘Pancreatic ductal adenocarcinoma (PDAC) is the most common type of pancreatic cancer featured with high intra-tumoral heterogeneity and poor prognosis. To comprehensively delineate the PDAC intra-tumoral heterogeneity and the underlying mechanism for PDAC progression, we employed single-cell RNA-seq (scRNA-seq) to acquire the transcriptomic atlas of 57,530 individual pancreatic cells from primary PDAC tumors and control pancreases, and identified diverse malignant and stromal cell types, including two ductal subtypes with abnormal and malignant gene expression profiles respectively, in PDAC. We found that the heterogenous malignant subtype was composed of several subpopulations with differential proliferative and migratory potentials. Cell trajectory analysis revealed that components of multiple tumor-related pathways and transcription factors (TFs) were differentially expressed along PDAC progression. Furthermore, we found a subset of ductal cells with unique proliferative features were associated with an inactivation state in tumor-infiltrating T cells, providing novel markers for the prediction of antitumor immune response. | 4 | Late Mesozoic magmatism and tectonic evolution in the Southern margin of the North China Craton显示文摘Late Mesozoic granitic magmatism(158–112 Ma) are widespread in the southern margin of the North China Craton(NCC), contemporary with many world-class Mo-Au-Ag-Pb-Zn polymetallic deposits. There are abrupt changes in the elements and isotopic compositions of these granites at about 127 Ma. The early stage(158–128 Ma) granites show slightly or no negative Eu anomalies, large ion lithophile elements enriched and heavy REE depleted(such as Y and Yb), belonging to typical I-type granite. The late stage(126–112 Ma) granites are characterized by A-type and/or highly fractionated I-type granite, with higher contents of SiO2, K2 O, Y, Yb and Rb/Sr ratio and lower contents of Sr, δEu value and Sr/Y ratio than that of the early-stage granites.Moreover, the whole rock Nd and Hf isotopic compositions of the granites younger than 127 Ma show more depleted than those of the older one. The two stages of Late Mesozoic granites were derived from a source region of the ancient basement of the southern margin of the NCC incorporated the mantle material. The late stage(126–112 Ma) granites contain more fractions of mantle material with depleted isotopic composition than the early ones. The granites record evidence for a strong crust-mantle interaction. | 6 | Metformin inhibits pancreatic cancer metastasis caused by SMAD4 deficiency and consequent HNF4G upregulation显示文摘Pancreatic ductal adenocarcinoma(PDAC)has poor prognosis due to limited therapeutic options.This study examines the roles of genome-wide association study identified PDAC-associated genes as therapeutic targets.We have identified HNF4G gene whose silencing most effectively repressed PDAC cell invasiveness.HNF4G overexpression is induced by the deficiency of transcriptional factor and tumor suppressor SMAD4.Increased HNF4G are correlated with SMAD4 deficiency in PDAC tumor samples and associated with metastasis and poor survival time in xenograft animal model and in patients with PDAC(log-rank P=0.036;HR=1.60,95%CI=1.03–2.47).We have found that Metformin suppresses HNF4G activity via AMPK-mediated phosphorylation-coupled ubiquitination degradation and inhibits in vitro invasion and in vivo metastasis of PDAC cells with SMAD4 deficiency.Furthermore,Metformin treatment significantly improve clinical outcomes and survival in patients with SMAD4-deficient PDAC(log-rank P=0.022;HR=0.31,95%CI=0.14–0.68)but not in patients with SMAD4-normal PDAC.Pathway analysis shows that HNF4G may act in PDAC through the cell-cell junction pathway.These results indicate that SMAD4 deficiency-induced overexpression of HNF4G plays a critical oncogenic role in PDAC progression and metastasis but may form a druggable target for Metformin treatment. | Chengcheng Wang Taiping Zhang Quan Liao Menghua Dai Junchao Guo Xinyu Yang Wen Tan Dongxin Lin Chen Wu Yupei Zhao | 2021 | Protein & Cell2021,12,2: | 5 |
| 7 | Provenance and tectonic setting transition as recorded in the Neoproterozoic strata, western Jiangnan Orogen:Implications for South China within Rodinia显示文摘The Neoproterozoic Tonian strata (ca.870-725 Ma) in the western Jiangnan Orogen archive the records of sedimentary provenance and tectonic setting which can be used to understand the geological evolution of the South China Continent.These strata are separated into the basement and cover sequences by a regional angular unconformity.The basement sequence can be subdivided into the lower and the upper parts by the widespread interbedded ca.840 Ma basalt with pillow structure.In the present work,234 concordant detrital zircon analyses are obtained from three Tonian sandstone samples in the Fanjingshan district,Guizhou Province.Combined with previous results,a total of 1736 analyses of detrital zircon U-Pb ages derived from 12 formations of Tonian strata in the western Jiangnan Orogen are used to decipher the integrated sedimentary and tectonic histories.The zircons from the lowermost part of the basement sequence (the Yujiagou Formation) show oval morphology and display two Paleoproterozoic age peaks at 2325 Ma and 1845 Ma which are similar with the detrital zircon age peaks from the Late Paleoproterozoic to Early Mesoproterozoic Dongchuan/Dahongshan/Hekou groups,suggesting a passive margin basin in which the sediments were mainly sourced from the southwestern Yangtze Block.However,the zircon age population of the lower part of the basement sequence (the Xiaojiahe,Huixiangping formations and their equivalents) indicates the sedimentary derivation from bidirectional sources (the ca.870 Ma arc materials in the south and the old detritus from the southwestern Yangtze Block) which is consistent with a back arc setting for the deposition of the sediments.Zircons from the upper part of the basement sequence (the Duyantang Formation and its equivalent) show euhedral and subangular morphology and display a unimodal age peak at ca.835 Ma.This sequence was possibly deposited in a convergent setting and the detritus were came from the locally distributed syn-collisional igneous rocks.The lower part of the cover sequence (the Xinzhai and Wuye formations and their equivalents) shows a distinct zircon age peak at 815-809 Ma and two subordinate peaks at 2485 Ma and 2018 Ma,suggesting that the basin had gradually transformed into a continental rift basin and received the detritus from the ca.815 Ma post-collisional magmatic rocks as well as from different Paleoproterozoic source rocks in the northern Yangtze Block.We propose a tectonic evolution model that envisages eruption of ca.840 Ma basalt in a back arc basin that existed during ca.870-835 Ma,an angular unconformity was formed during amalgamation of the Yangtze Block and the Cathaysia Block at ca.835-820 Ma and the rifting of the South China Continent was initiated at ca.800 Ma.Our study concludes that the South China Continent was formed on the periphery of the Rodinia supercontinent. | Jiawei Zhang Taiping Ye Yaran Dai Jianshu Chen Hui Zhang Chuangu Dai Guohua Yuan Kaiyuan Jiang | 2019 | Geoscience Frontiers2019,10,5: | 4 |

| 10 | Zircon U-Pb geochronology of tuffite beds in the Baishugou Formation:Constraints on the revision of Ectasian System at the southern margin of the North China Craton显示文摘The Xiamaling Formation of 1.4–1.35 Ga in Jixian Section and adjoining areas represents a unique Ectasian Period(Mesoproterozoic)sedimentary sequence in the North China Craton(NCC).Studies carried out during the last decade have contributed significantly in improving our understanding about the Ectasian sedimentation tectonics in the NCC during this transitional interval between the breakup of Supercontinent Columbia and the assembly of Rodinia.The present study reports,for the first time,tuffite beds interlayered with carbonaceous-siliceous slate-phyllite in the upper Baishugou Formation(western Henan Province),at the southern margin of NCC.The LA-MC-ICPMS U-Pb geochronology of zircons from two tuffite samples has yielded almost identical ages of 1330±10 Ma(MSWD=0.43,N=38)and 1332±10 Ma(MSWD=0.90,N=24),that constrain a 1.33 Ga depositional age(Ectasian Period)for the Baishugou Formation.A close similarity in lithological characteristics and their respective superimposed lithostratigraphic relationships of the Baishugou and Xiamaling formations,underline a comparable depositional environment for them.The high-precision geochronology data on the Baishugou Formation would stimulate a relook into the Meso-Neoproterozoic chronostratigraphic framework of the NCC,and further research would lead to a comprehensive understanding of the geological evolution of the NCC during Ectasian Period and its correlation with analogous global events. | Xiyan ZHU Shiyan WANG Wenbo SU Taiping ZHAO Lanyin PANG Mingguo ZHAI | 2020 | Science China Earth Sciences2020,63,11: | 4 |

| 12 | Protective effects of ginkgo biloba leaves extract on peroxide-induced oxidative stress damage in PC12 cells显示文摘BACKGROUND: Extracts of ginkgo biloba leaves (EGB) and its metabolites have been reported to enhance brain function and nerve behavior. It has also been hypothesized that they can protect neurons from oxidative stress. OBJECTIVE: To investigate protective effects of EGB on peroxide (H2O2)-induced oxidative stress damage in PC12 cells. DESIGN: Observational contrast study. SETTING: Department of Pathophysiology,Guangdong Pharmacological College. MATERIALS: EGB was provided by Xi'an Fujie Biotechnological Development Company; 1640 culture medium,methylthiazolyl tetrazolium (MTT),trypsin and dimathyl sulfoxide (DMSO) by Sigma Company; PC12 cell strain by Cell Center of Medical College of Zhongshan University; calf serum by Hangzhou Sijiqing Bioengineering Company; lactate dehydrogenase (LDH) kit by Nanjing Jiancheng Bioengineering Research Institute. METHODS: The experiment was carried out in Department of Cell Biology of Guangdong Pharmacological College from June to December 2005. ① Cell culture: PC12 cells were cultured in 1640 medium containing 200 g/L fetal calf serum. The cells were diluted to 1×107 L-1 and washed every two days. Those cells were used to experiment until they grew in logarithm on solid wall. ② Grouping and intervention: PC12 cells (1×108 L-1) were plated in 96-well plates with the density of 200 μL/hole and divided into three groups: normal control group (routinely adding media),H2O2 group (treating with media and H2O2 for 20 hours) and EGB group (adding media,100 μmol/L EGB and 100 μmol/L H2O2). ③ MTT assay: PC12 cells (1× 108 L-1) were plated in 96-well plates and divided into three groups with 8 holes for each group. Under sterile condition,cells were added with 5 g/L MTT (100 μL) and cultured for 4 hours. And then,200 μL DMSO fluid was added and shaken for 30 minutes until blue crystal products formed were dissolved soundly. ④ Experimental evaluation: Absorbance (A) at 630 nm was measured and LDH activity was measured at the same time. MAIN OUTCOME MEASURES: Results of MTT assay and LDH activity. RESULTS: ① Results of MTT assay: A value was lower in the H2O2 group than that in the normal control group (P < 0.01),while A value was higher in the EGB group than that in the H2O2 group (P < 0.01). ② LDH activity: LDH activity was higher in the H2O2 group than that in the normal control group (P < 0.01),while LDH activity was lower in the EGB group than that in the H2O2 group (P < 0.01). | 15 | PHYSICAL MODELING OF HYDRAULIC DESILTATION IN TAPU RESERVOIR显示文摘A movable bed physical model was constructed to investigate hydraulic desiltation by flushing and lateral erosion in the Tapu reservoir, Taiwan. The model scaling is based on the requirement for dynamic similarity of cohesive sediment deposit initiation in flushing processes. For model scaling, flume experiments investigating the initiation of cohesive sediment deposits were carried out to establish the relationship between critical shear stress of the flow and dry density of the deposit. Experiments in the physical model were then performed to measure the variations of the reservoir water level, the outlet discharge and the outflow sediment discharge. The processes of emptying and flushing were observed and analyzed in the main flushing channel. One of the experiments was conducted to simulate the on-site flushing operations on June 11, 1997. The results showed that the total 0cumulative flushed sediment volume by physical modeling was close to that by numerical simulation. | 16 | The Effect of CD3-Specific Monoclonal Antibody on Treating Experimental Autoimmune Myasthenia Gravis显示文摘CD3-specific monoclonal antibody was the first one used for clinical practice in field of transplantation. Recently,renewed interests have elicited in its capacity to prevent autoimmune diabetes by inducing immune tolerance. In this study, we tested whether this antibody can also be used to treat another kind of autoimmune disease myasthenia gravis (MG) and explored the possible mechanisms. MG is caused by an autoimmune damage mediated by antibody- and complement-mediated destruction of AChR at the neuromuscular junction. We found that administration of CD3-specific antibody (Fab)2 to an animal model with experimental autoimmune myasthenia gravis (EAMG) (B6 mice received 3 times of AChR/CFA immunization) could not significantly improve the clinical signs and clinical score. When the possible mechanisms were tested, we found that CD3 antibody treatment slightly down-regulated the T-cell response to AChR, modestly up-regulation the muscle strength. And no significant difference in the titers of IgG2b was found between CD3 antibody treated and control groups. These data indicated that CD3-specific antibody was not suitable for treating MG, an antibody- and complementmediated autoimmune disease, after this disease has been established. | 18 | Reconsideration of the sediment characteristics of the second member of the Triassic Xujiahe Formation in Sanhuiba,Huaying Mountain显示文摘The second member of the Triassic Xujiahe Formation in the Sanhuiba area in Huaying Mountain is an important section of sedimentary delta facies in the Chongqing Tianfu Geological Survey and Training Base. In spite of some initial efforts on its lithology and lithofacies, the existing research is yet to be completed and suffers from contradictory conclusions and inadequate evidence. Starting from the sedimentary microfacies types and cyclic features, we analyzed the vertical sequence of a sedimentary delta model and discovered that this section is a constructional delta deposit. Eight incomplete and asymmetric regressive cycles are marked off, given that frequent erosion surfaces appeared in the section. Based on the analysis of structural characteristics of every monocycle, we summarized the system of cycles, tracts and sequence characteristics of the overall section. In view of this systematic series of cycles, we discussed the features of the syndepositional tectonic movements in the area and conclude that, due to frequent erosion, migration and diversion of a sub-channel branch was brought about by oscillating movements of the crust. In a comparison with neighboring sections, we explored the cause of erosion and the relations between erosion and cycles. | 19 | Sr speciation in producing SrCO_3 with celestite显示文摘Strontium speciation transferring in strontium residue and mixing strontium ore are investigated with extracting experiment, X-ray powder diffraction(XRD), X-ray fluorescence spectrometer(XRF) and Atomic Absorption Spectroscopy(AAS). The results show that there are strontium carbonate, strontium sulfate, strontium sulfide, and a little amount of strontium silicate and strontium ferrite in strontium residue. Impurities in strontium residue mainly contain carbonate and oxide of calcium, iron, magnesium and aluminum etc. The amount of strontium sulfide in strontium residue is larger than that in strontium ore. And a little strontium silicate and strontium ferrite are found in strontium ore. It is concluded that acid soluble method is used firstly in the process of producing strontium carbonate from mixing strontium ore. In addition, mechanical smash method and surface active treatment to strontium samples are beneficial to the ratio-leaching increase of strontium.
