| 1 | Efficacy and safety of a novel anti-HER2 therapeutic antibody RC48 in patients with HER2-overexpressing,locally advanced or metastatic gastric or gastroesophageal junction cancer:a single-arm phase II study显示文摘Background:Current treatment options for human epidermal growth factor receptor 2(HER2)-overexpressing gastric cancer at third-line have shown limited clinical benefit.Further,there is no specific treatment for HER2 immunohistochemistry(IHC)2+and fluorescence in-situ hybridization-negative patients.Here,we report the efficacy and safety of a novel anti-HER2 antibody RC48 for patients with HER2-overexpressing,advanced gastric or gastroesophageal junction cancer.Methods:Patients with HER2-overexpressing(IHC 2+or 3+),locally advanced or metastatic gastric or gastroesophageal junction cancer who were under at least second-line therapy were eligible and received RC482.5 mg/kg alone every 2 weeks.The primary endpoint was the objective response rate(ORR)assessed by an independent review committee.Secondary endpoints included progressionfree survival(PFS),overall survival(OS),duration of response,time to progression,disease control rate,and safety.Results:Of 179 patients screened,125 were eligible and received RC48 treatment.The ORR was 24.8%(95%confidence interval[CI]:17.5%-33.3%).The median PFS and OS were 4.1 months(95%CI:3.7-4.9 months)and 7.9 months(95%CI:6.7-9.9 months),respectively.The most frequently reported adverse events were decreased white blood cell count(53.6%),asthenia(53.6%),hair loss(53.6%),decreased neutrophil count(52.0%),anemia(49.6%),and increased aspartate aminotransferase level(43.2%).Serious adverse events(SAEs)occurred in 45(36.0%)patients,and RC48-related SAEs were mainly decreased neutrophil count(3.2%).Seven patients had adverse events that led to death were not RC48-related.Conclusions:RC48 showed promising activity with manageable safety,suggesting potential application in patients with HER2-overexpressing,advanced gastric or gastroesophageal junction cancer who have previously received at least two lines of chemotherapy. | Zhi Peng Tianshu Liu Jia Wei Airong Wang Yifu He Liuzhong Yang Xizhi Zhang Nanfeng Fan Suxia Luo Zhen Li Kangsheng Gu Jianwei Lu Jianming Xu Qingxia Fan Ruihua Xu Liangming Zhang Enxiao Li Yuping Sun Guohua Yu Chunmei Bai Yong Liu Jiangzheng Zeng Jieer Ying Xinjun Liang Nong Xu Chao Gao Yongqian Shu Dong Ma Guanghai Dai Shengmian Li Ting Deng Yuehong Cui Jianmin Fang Yi Ba Lin Shen | 2021 | Cancer Communications2021,41,11: | 29 |
| 2 | Mining geographic episode association patterns of abnormal events in global earth science data显示文摘Abnormal events in earth science have great influence on both the natural envi-ronment and the human society. Finding association patterns among these events has great significance. Because data in earth science has characteristics of mass,high dimension,spatial autocorrelation and time delay,existing mining technolo-gies cannot be directly used on it. We propose a RSNN (range-based searching nearest neighbors) spatial clustering algorithm to reduce the data size and auto-correlation. Based on the clustered data,we propose a GEAM (geographic episode association pattern mining) algorithm which can deal with events time lags and find interesting patterns with specific constraints,to mine the association patterns. We carried out experiments on global climate datasets and found many interesting association patterns. | 3 | Observation and optimization of 2 μm mode‐locked pulses in all‐fiber net anomalous dispersion laser cavity显示文摘We integrally demonstrate 2μm mode-locked pulses performances in all-fiber net anomalous dispersion cavity.Stable mode-locking operations with the center wavelength around 1950–1980 nm can be achieved by using the nonlinear polarization rotation structure and properly designing the dispersion management component.Conventional soliton is firstly obtained with a total anomalous dispersion cavity.Due to the contribution of commercial ultra-high numerical aperture fibers,net dispersion is reduced to-0.077 ps2.So that stretched pulse with 19.4 nm optical bandwidth is obtained and the de-chirped pulse-width can reach 312 fs using extra-cavity compression.Under pump power greater than 890 mW,stretched pulse can evolve into noise-like pulse with 41.3 nm bandwidth.The envelope and peak of such broadband pulse can be compressed with up to 2.2 ps and 145 fs,respectively.The single pulse energy of largely chirped stretched and noise-like pulse can reach 1.785 nJ and 1.53 nJ,respectively.Furthermore,extra-cavity compression can also contribute to a significant increase of peak power. | Wanzhuo Ma Desheng Zhao Runmin Liu Tianshu Wang Quan Yuan Hao Xiong Haiying Ji Huilin Jiang | 2020 | Opto-Electronic Advances2020,3,11: | 2 |
| 4 | Ginger-indirect moxibustion plus acupuncture versus acupuncture alone for chronic fatigue syndrome:a randomized controlled trial显示文摘OBJECTIVE:To assess the efficacy and safety of gingerindirect moxibustion for chronic fatigue syndrome(CFS).METHODS:In this central randomized,controlled trial,290 CFS participants were recruited and randomly allocated to group A(ginger-indirect moxibustion plus acupuncture)or group B(acupuncture alone).The study consisted of a treatment period of 8 weeks with a total of 24 treatments(3 sessions per week,every other day),and a follow-up period of 12 weeks.The outcome was measured by Fatigue Severity Scale(FSS),Psychological Health Report(SPHERE),the Self-rating depression scale(SDS)and the Hamilton anxiety scale(HAMA)at baseline,2,4,6,8,12 and 20 weeks.RESULTS:With the treatment undergoing,the changes of FSS,SPHERE,SDS and HAMA scores in both groups increased gradually,and the effect maintained at the 12th week.Between groups,significantly higher score changes were seen in group A in FSS after 4 weeks treatment(11.94 vs 9.12,95%CI:0.94,4.7)and in SPHERE after 2 weeks treatment(3.7 vs 2.27,95%CI:0.56,2.31).But for SDS and HAMA,the improvement did not differ significantly between groups.No severe adverse events were reported.CONCLUSION:Ginger-indirect moxibustion is a safe and effective intervention to relieve fatigue and accompanying physical symptoms of CFS. | MA Tingting WU Jie YANG Lijie FENG Fen YANG Huilin ZHANG Jinhua ZHONG Yanjin NING Qing HUANG Lirong LIN Youbing YAN Jue CHEN Guiquan HOU Tianshu WANG Li REN Yuanfang TAN Jing | 2022 | Journal of Traditional Chinese Medicine2022,42,2: | 2 |
| 5 | Visualized and cascade-enhanced gene silencing by smart DNAzyme-graphene nanocomplex显示文摘BCL-2 gene as well as its products is recognized as a promising target for the molecular targeted therapy of tumors.However,due to certain defense measures of tumor cells,the therapeutic effect based on the gene silencing of BCL-2 is greatly reduced.Here we fabricate a smart response nucleic acid therapeutic that could silence the gene effectively through a dual-targeted and cascade-enhanced strategy.In brief,nano-graphene oxide(GO),working as a nano-carrier,is loaded with a well-designed DNAzyme,which can target and silence the BCL-2 mRNA.Furthermore,upon binding with the BCL-2 mRNA,the enzymatic activity of the DNAzyme can be initiated,cutting a substrate oligonucleotide to produce an anti-nucleolin aptamer AS1411.Nucleolin,a nucleolar phosphoprotein,is known as a stabilizer of BCL-2 mRNA.Via binding and inactivating the nucleolin,AS1411 can destabilize BCL-2 mRNA.By this means of simultaneously targeting mRNA and its stabilizer in an integrated system,effective silencing of the BCL-2 gene of tumor cells is achieved at both the cellular and in vivo levels.After being dosed with this nucleic acid therapeutic and without any chemotherapeutics,apoptosis of tumor cells at the cellular level and apparent shrinkage of tumors in vivo are observed.By labeling a molecular beacon on the substrate of DNAzyme,visualization of the enzymatic activity as well as the tumor in vivo can be also achieved.Our work presents a pure bio-therapeutic strategy that has positive implications for enhancing tumor treatment and avoiding side effects of chemotherapeutics. | Lingjie Ren Xiaoxia Chen Chang Feng Lei Ding Xiaomin Liu Tianshu Chen Fan Zhang Yanli Li Zhongliang Ma Bo Tian Xiaoli Zhu | 2020 | Nano Research2020,13,8: | 1 |

| 7 | JUNB mediates oxaliplatin resistance via the MAPK signaling pathway in gastric cancer by chromatin accessibility and transcriptomic analysis显示文摘tients,and chemotherapy resistance is one of the main reasons for treatment failure.Thus,it is important to reveal the mechanism of oxaliplatin resistance and to seek effective intervention strategies to improve chemotherapy sensitivity,thereby improving the survival and prognosis of gastric cancer patients.To understand the molecular mechanisms of oxaliplatin resistance,we generate an oxaliplatin-resistant gastric cancer cell line and conduct assay for transposase-accessible chromatin sequencing(ATAC-seq)and RNA sequencing(RNA-seq)for both parental and oxaliplatin-resistant AGS cells.A total of 3232 genomic regions are identified to have higher accessibility in ox-aliplatin-resistant cells,and DNA-binding motif analysis identifies JUNB as the core transcription factor in the regulatory network.JUNB is overexpressed in oxaliplatin-resistant gastric cancer cells,and its upregulation is associated with poor prognosis in gastric cancer patients,which is validated by our tissue microarray data.Moreover,chromatin immunoprecipitation sequencing(ChIP-seq)analysis reveals that JUNB binds to the tran-scriptional start site of key genes involved in the MAPK signaling pathway.Knockdown of JUNB inhibits the MAPK signaling pathway and restores sensitivity to oxaliplatin.Combined treatment with the ERK inhibitor piperlongu-mine or MEK inhibitor trametinib effectively overcomes oxaliplatin resistance.This study provides evidence that JUNB mediates oxaliplatin resistance in gastric cancer by activating the MAPK pathway.The combination of MAPK inhibitors with oxaliplatin overcomes resistance to oxaliplatin,providing a promising treatment opportunity for oxaliplatin-resistant gastric cancer patients. | Suyao Li Yichou Wei Xun Sun Mengling Liu Mengxuan Zhu Yitao Yuan Jiayu Zhang Yu Dong Keshu Hu Sining Ma Xiuping Zhang Bei Xu Hesheng Jiang Lu Gan Tianshu Liu | 2023 | Acta Biochimica et Biophysica Sinica2023,55,11: | 0 |
| 8 | Control of the ultrafast photo-electronic dynamics of a chemical-vapor-deposited-grown graphene by ozone oxidation显示文摘Diverse ultrafast dynamics have been reported on different graphene prepared by different methods.Chemicalvapor-deposited(CVD)growth is regarded as a very promising method for highly efficient production of graphene.However,CVD-grown graphene usually presents only one of the diverse ultrafast dynamics.Thus,control of the ultrafast photo-electronic dynamics of CVD-grown graphene is vital to present diversity for different photodetection applications of CVD-grown graphene.In this paper,we report on the first realization to our knowledge of control of the ultrafast dynamics of CVD-grown graphene and the manifestation of diverse ultrafast dynamics on sole CVD-grown graphene.We study the ultrafast photoelectronic dynamics of CVD-grown graphene with different degrees of oxidation caused by ozone oxidation using femtosecond time-resolved transient differential transmission spectroscopy,and we find that the ultrafast dynamics can evolve obviously with the time of ozone oxidation.The diverse ultrafast dynamics reported previously on different monolayer graphenes prepared by different methods are achieved on the sole CVD-grown graphene by controlling oxidation time.The mechanism for manipulation of the ultrafast dynamics by ozone oxidation is revealed by Raman spectroscopy as the control of the Fermi level of CVD-grown graphene.Simulations of dynamics based on the optical conductivity model of graphene and Fermi level change well reproduce the observed diverse ultrafast dynamics.Our results are very important for the diverse applications of graphene and open a new path toward the diverse ultrafast dynamics on sole graphene prepared by any method. | MIN GUO LAI-PENG MA WENCAI REN TIANSHU LAI | 2020 | Photonics Research2020,8,1: | 0 |

| 11 | Negative correlation between acetyl-CoA acyltransferase 2 and cetuximab resistance in colorectal cancer显示文摘The emergence of anti-EGFR therapy has revolutionized the treatment of colorectal cancer(CRC).However,not all patients respond consistently well.Therefore,it is imperative to conduct further research to identify the molecular mechanisms underlying the development of cetuximab resistance in CRC.In this study,we find that the expressions of many metabolism-related genes are downregulated in cetuximab-resistant CRC cells compared to their sensitive counterparts.Specifically,acetyl-CoA acyltransferase 2(ACAA2),a key enzyme in fatty acid metabolism,is downregulated during the development of cetuximab resistance.Silencing of ACAA2 promotes proliferation and increases cetuximab tolerance in CRC cells,while overexpression of ACAA2 exerts the opposite effect.RTK-Kras signaling might contribute to the downregulation of ACAA2 expression in CRC,and ACAA2 predicts CRC prognosis in patients with Kras mutations.Collectively,our data suggest that modulating ACAA2 expression contributes to secondary cetuximab resistance in Kras wild-type CRC patients.ACAA2 expression is related to Kras mutation and demonstrates a prognostic role in CRC patients with Kras mutation.Thus,ACAA2 is a potential target in CRC with Kras mutation. | Yitao Yuan Xun Sun Mengling Liu Suyao Li Yu Dong Keshu Hu Jiayu Zhang Bei Xu Sining Ma Hesheng Jiang Pengcong Hou Yufu Lin Lu Gan Tianshu Liu | 2023 | Acta Biochimica et Biophysica Sinica2023,55,9: | 0 |
