1Yb-doped large-pitch fibres: effective single-mode operation based on higher-order mode delocalisation显示文摘Rare earth-doped fibres are a diode-pumped,solid-state laser architecture that is highly scalable in average power.The performance of pulsed fibre laser systems is restricted due to nonlinear effects.Hence,fibre designs that allow for very large mode areas at high average powers with diffraction-limited beam quality are of enormous interest.Ytterbium-doped,rod-type,large-pitch fibres(LPF)enable extreme fibre dimensions,i.e.,effective single-mode fibres with mode sizes exceeding 100 times the wavelength of the guided radiation,by exploiting the novel concept of delocalisation of higher-order transverse modes.The non-resonant nature of the operating principle makes LPF suitable for high power extraction.This design allows for an unparalleled level of performance in pulsed fibre lasers.Jens Limpert Fabian Stutzki Florian Jansen Hans-Jurgen Otto Tino Eidam Cesar Jauregui Andreas Tunnermann 2012Light(Science & Applications)2012,1,1:16

3Marking RNA:m^6A writers,readers,and functions in Arabidopsis显示文摘N^6-methyladenosine(m^6A)emerges as an important modification in eukaryotic mRNAs.m^6A has first been reported in 1974,and its functional significance in mammalian gene regulation and importance for proper development have been well established.An arsenal of writer,eraser,and reader proteins accomplish deposition,removal,and interpretation of the m^6A mark,resulting in dynamic function.This led to the concept of an epitranscriptome,the compendium of RNA species with chemical modification ofthe nucleobases in the cell,in analogy to the epigenome.While m^6A has long been known to also exist in plant mRNAs,proteins involved in m^6A metabolism have only recently been detected by mutant analysis,homology search,and mRNA interactome capture in the reference plant Arabidopsis thaliana.Dysregulation ofthe m^6A modification causes severe developmental abnormalities of leaves and roots and altered timing of reproductive development.Furthermore,m^6A modification affects viral infection.Here,we discuss recent progress in identifying m^6A sites transcriptome-wide,in identifying the molecular players involved in writing,removing,and reading the mark,and in assigning functions to this RNA modification in 4.thaliana.We highlight similarities and differences to m^6A modification in mammals and provide an outlook on important questions that remain to be addressed.Marlene Reichel Tino Koster Dorothee Staiger 2019Journal of Molecular Cell Biology2019,11,10:13
4A concept for multiterawatt fibre lasers based on coherent pulse stacking in passive cavities显示文摘Since the advent of femtosecond lasers,performance improvements have constantly impacted on existing applications and enabled novel applications.However,one performance feature bearing the potential of a quantum leap for high-field applications is still not available:the simultaneous emission of extremely high peak and average powers.Emerging applications such as laser particle acceleration require exactly this performance regime and,therefore,challenge laser technology at large.On the one hand,canonical bulk systems can provide pulse peak powers in the multi-terawatt to petawatt range,while on the other hand,advanced solid-state-laser concepts such as the thin disk,slab or fibre are well known for their high efficiency and their ability to emit high average powers in the kilowatt range with excellent beam quality.In this contribution,a compact laser system capable of simultaneously providing high peak and average powers with high wall-plug efficiency is proposed and analysed.The concept is based on the temporal coherent combination(pulse stacking)of a pulse train emitted from a high-repetition-rate femtosecond laser system in a passive enhancement cavity.Thus,the pulse energy is increased at the cost of the repetition rate while almost preserving the average power.The concept relies on a fast switching element for dumping the enhanced pulse out of the cavity.The switch constitutes the key challenge of our proposal.Addressing this challenge could,for the first time,allow the highly efficient dumping of joule-class pulses at megawatt average power levels and lead to unprecedented laser parameters.Sven Breitkopf Tino Eidam Arno Klenke Lorenz von Grafenstein Henning Carstens Simon Holzberger Ernst Fill Thomas Schreiber Ferenc Krausz Andreas Tunnermann Ioachim Pupeza Jens Limpert 2014Light(Science & Applications)2014,3,1:11
5全球婴幼儿辅助食品发展状况与趋势显示文摘在全球婴幼儿辅食产业发展历程中,欧美国家的婴幼儿辅食工业化生产和研究起步早,而我国婴幼儿配方谷粉的基础研究和发展的历史较短。亚洲地区是全球最大的婴幼儿辅食市场,区域销售金额占到总体销量金额的42.86%。目前我国婴幼儿辅食产品按市场销售的产品主要分为4类:(1)以米粉和面条为主的谷物类辅食(占55%);(2)以饼干、磨牙棒为主的健康零食(25%);(3)以果泥、菜泥为主的佐餐辅食(12%);(4)以营养包为主的营养补充品(8%)。其中谷物类辅食在婴幼儿辅食产品消费中占比最高。中国婴幼儿辅食的发展方向应充分重视食品安全、婴幼儿辅食品种细分化,加速婴幼儿有机辅食和高端化产品的发展,重视国有企业品牌的发展。王俏 王骁音 Tino Landl 郝斌 厉梁秋 杨燕涛 荫士安 2021中国妇幼健康研究2021,32,12:5
6Additive manufacturing in radiation oncology:a review of clinical practice,emerging trends and research opportunities显示文摘The additive manufacturing(AM)process plays an important role in enabling cross-disciplinary research in engineering and personalised medicine.Commercially available clinical tools currently utilised in radiotherapy are typically based on traditional manufacturing processes,often leading to non-conformal geometries,time-consuming manufacturing process and high costs.An emerging application explores the design and development of patient-specific clinical tools using AM to optimise treatment outcomes among cancer patients receiving radiation therapy.In this review,we:•highlight the key advantages of AM in radiotherapy where rapid prototyping allows for patient-specific manufacture•explore common clinical workflows involving radiotherapy tools such as bolus,compensators,anthropomorphic phantoms,immobilisers,and brachytherapy moulds;and•investigate how current AM processes are exploited by researchers to achieve patient tissuelike imaging and dose attenuations.Finally,significant AM research opportunities in this space are highlighted for their future advancements in radiotherapy for diagnostic and clinical research applications.Rance Tino Martin Leary Adam Yeo Elizabeth Kyriakou Tomas Kron Milan Brandt 2020International Journal of Extreme Manufacturing2020,2,1:3

9Use of siRNA molecular beacons to detect and attenuate mycobacterial infection in macrophages显示文摘Tuberculosis is one of the leading infectious diseases plaguing mankind and is mediated by the facultative pathogen, Mycobacterium tuberculosis(MTB). Once the pathogen enters the body, it subverts the host immune defenses and thrives for extended periods of time within the host macrophages in the lung granulomas, a condition called latent tuberculosis(LTB). Persons with LTB are prone to reactivation of the disease when the body's immunity is compromised. Currently there are no reliable and effective diagnosis and treatment options for LTB, which necessitates new research in this area. The mycobacterial proteins and genes mediating the adaptive responses inside the macrophage is largely yet to be determined. Recently, it has been shown that the mce operon genes are critical for host cell invasion by the mycobacterium and for establishing a persistent infection in both in vitro and in mouse models of tuberculosis. The Yrb E and Mce proteins which are encoded by the MTB mce operons display high degrees of homology to the permeases and the surface binding protein of the ABC transports, respectively. Similarities in structure and cell surface location impute a role in cell invasion at cholesterol rich regions and immunomodulation. The mce4 operon is also thought to encode a cholesterol transport system that enables the mycobacterium to derive both energy and carbon from the host membrane lipids and possibly generating virulence mediating metabolites, thus enabling the bacteria in its long term survival within the granuloma. Various deletion mutation studies involving individual or whole mce operon genes have shown to be conferring varying degrees of attenuation of infectivity or at times hypervirulence to the host MTB, with the deletion of mce4 A operon gene conferring the greatest degree of attenuation of virulence. Antisense technology using synthetic si RNAs has been used in knocking down genes in bacteria and over the years this has evolvedinto a powerful tool for elucidating the roles of various genes mediating infectivity and survival in mycobacteria.
