1Human endogenous retrovirus W env increases nitric oxide production and enhances the migration ability of microglia by regulating the expression of inducible nitric oxide synthase显示文摘Human endogenous retrovirus W env(HERV-W env) plays a critical role in many neuropsychological diseases such as schizophrenia and multiple sclerosis(MS). These diseases are accompanied by immunological reactions in the central nervous system(CNS). Microglia are important immunocytes in brain inflammation that can produce a gasotransmitter – nitric oxide(NO). NO not only plays a role in the function of neuronal cells but also participates in the pathogenesis of various neuropsychological diseases. In this study, we reported increased NO production in CHME-5 microglia cells after they were transfected with HERV-W env. Moreover, HERV-W env increased the expression and function of human inducible nitric oxide synthase(hi NOS) and enhanced the promoter activity of hi NOS. Microglial migration was also enhanced. These data revealed that HERV-W env might contribute to increase NO production and microglial migration ability in neuropsychological disorders by regulating the expression of inducible NOS. 2Human leukemia antigen-A0201-restricted epitopes of humanendogenous retrovirus W family envelope(HERV-W env)inducestrong cytotoxic T lymphocyte responses显示文摘Human endogenous retrovirus W family(HERV-W) envelope(env) has been reported to be related to several human diseases, including autoimmune disorders, and it could activate innate immunity.However, there are no reports investigating whether human leukemia antigen(HLA)-A~*0201^+restriction is involved in the immune response caused by HERV-W env in neuropsychiatric diseases. In the present study, HERV-W env-derived epitopes presented by HLA-A~*0201 are described with the potential for use in adoptive immunotherapy. Five peptides displaying HLAA~*0201-binding motifs were predicted using SYFEPITHI and BIMAS, and synthesized. A CCK-8 assay showed peptides W, Q and T promoted lymphocyte proliferation. Stimulation of peripheral blood mononuclear cells from HLA-A~*0201^+ donors with each of these peptides induced peptidespecific CD8^+ T cells. High numbers of IFN-γ-secreting T cells were also detectable after several weekly stimulations with W, Q and T. Besides lysis of HERV-W env-loaded target cells, specific apoptosis was also observed. These data demonstrate that human T cells can be sensitized toward HERV-W env peptides(W, Q and T) and, moreover, pose a high killing potential toward HERV-W env-expressing U251 cells. In conclusion, peptides W Q and T, which are HERV-W env antigenic epitopes, have both antigenicity and immunogenicity, and can cause strong T cell immune responses. Our data strengthen the view that HERV-W env should be considered as an autoantigen that can induce autoimmunity in neuropsychiatric diseases, such as multiple sclerosis and schizophrenia. 3Glucose metabolism controls humanγδ-cell-mediated tumor immunosurveillance in diabetes显示文摘Patients with type 2 diabetes mellitus(T2DM)have an increased risk of cancer.The effect of glucose metabolism onγδT cells and their impact on tumor surveillance remain unknown.Here,we showed that high glucose induced Warburg effect type of bioenergetic profle in Vy9vδ2 T cells,leading to excessive lactate accumulation,which further inhibited lytic granule secretion by impairing the traffcking of cytolytic machinery to the Vy9vδ2 T-cell-tumor synapse by suppressing AMPK activation and resulted in the loss of antitumor activity in vitro,in vivo and in patients.Strikingly,activating the AMPK pathway through glucose control or metformin treatment reversed the metabolic abnormalities and restored the antitumor activity of Vy9vδ2 T cells.These results suggest that the impaired antitumor activity of Vy9vδ2 T cells induced by dysregulated glucose metabolism may contribute to the increased cancer risk in T2DM patients and that metabolic reprogramming by targeting the AMPK pathway with metformin may improve tumor immunosurveillance.Xiaofeng Mu Zheng Xiang Yan Xu Jing He Jianwen Lu Yuyuan Chen Xiwei Wang Chloe Ran Tu Yanme Wenyue Zhang Zhinan Yin Wing-hang Leung Yu-Lung Lau Yinping Liu Wenwei Tu 2022Cellular & Molecular Immunology2022,19,8:2

9PAS domain of the deduced Org35 protein mediates the interaction with NifA显示文摘NifA in Azospirillum brasilense plays a key role in regulating the synthesis of nitrogenase in response to ammonia and oxygen available. Recently, our laboratory has identified four clones, whose gene prodcuts interact with NifA, from A. brasilense Sp7 genomic libraries by using the yeast two-hybrid sys- tem with NifA as bait. We are interested in clone S35, one of the four clones, because it contains a PAS-domain coding region. The entire open reading frame (ORF) for the PAS domain-containing protein was isolated and designated as org35 here. org35 gene is 2211-bp long and encodes a protein of 736 aa with a predicted molecular weight of about 78.4 kD. The predicted amino acid sequence of org35 has similarity to some two-component sensor kinase/response regulator hybrids of bacteria. Struc- tural analyses showed that Org35 comprises at least three discrete conserved domains: the N-terminal PAS, the central histidine protein kinase (HPK) and the C-terminal response regulator (RR). The PAS domain of the deduced Org35 protein was found to interact directly with NifA, but the central HPK and the C-terminal RR domains of Org35 were not. 10Succession of bacterial community during composting:dissimilarity between compost mixture and biochar additive显示文摘Previous research showed that biochar addition facilitated composting and elevated nutrient retention.However,few of these studies explored bacterial structure and abundance in the compost mixture and biochar additive.Thus,this study aims to distinguish bacterial communities in both compost and bamboo biochar(BB)additive.Results indicated that the dynamics of nutrient contents in compost and BB samples were in a similar pattern,although there were lower levels of nutrients and metals(i.e.,Cu and Zn)in BB additives.The total number of operational taxonomic units(OTUs)in both compost and BB additives peaked on day 7 and then gradually reduced during composting.There was more abundance of bacteria in compost,whereas the diversity of bacteria was more in BB additives.Furthermore,the dominant bacteria in compost and BB samples were distinct at the different stages of composting.The Firmicutes steadily decreased in compost samples(from 34.78%to 7.65%),while it was the dominant phylum in BB additives during the whole composting period.Furthermore,Ruminofilibacter,Pseudoxanthomonas,and Actinomadura were the most abundant genera in compost samples than Pseudoxanthomonas,Azoarcus,and Paenibacillus in BB additives at the final stage of composting.Results from this study could provide a theoretical reference for the sound performance of biochar-added composting.Muyuan Zhang Weng Liang Zhineng Tu Ronghua Li Zengqiang Zhang Amjad Ali Ran Xiao 2021Biochar2021,3,2:1

16Experimental demonstration of suppressing residual geometric dephasing显示文摘The geometric phase is regarded as a promising strategy in fault tolerance quantum information processing(QIP) domain due to its phase only depending on the geometry of the path executed. However, decoherence caused by environmental noise will destroy the geometric phase. Traditional dynamic decoupling sequences can eliminate dynamic dephasing but can not reduce residual geometric dephasing, which is still vital for high-precision quantum manipulation. In this work, we experimentally demonstrate effective suppression of residual geometric dephasing with modified dynamic decoupling schemes,using a single trapped171 Ybtion. The experimental results show that the modified schemes can reduce dephasing rate up to more than one order of magnitude compared with traditional dynamic decoupling schemes, where residual geometric dephasing dominates. Besides, we also investigate the impact of intensity and correlation time of the low-frequency noise on coherence of the quantum system. 18Designer cell-self-implemented labeling of microvesicles in situ with the intracellular-synthesized quantum dots显示文摘Cell-derived microvesicles(MVs) are secreted from almost all kinds of mammalian cells into the extracellular space, and play crucial roles in intercellular communication and transporting biomolecules between cells. However, there is a great challenge for visualizing and monitoring of MVs’ bio-behaviors due to the limitations of existing labeling methods. Herein, we report the first paradigm of designer cell-self-implemented labeling of MVs secreted from living mammalian MCF-7 cells in situ using the intracellular-synthesized fluorescent quantum dots(QDs) during the formation of MVs. By elaborately coupling intracellular biochemical reactions and metabolism pathways, the MCF-7 cells can be illuminated brightly by intracellular-biosynthesized fluorescent CdSe QDs. Simultaneously, intracellular-synthesized QDs can be in situ encapsulated by the secreted MVs budding from the plasma membrane of the fluorescing cells to label the MVs with an efficiency of up to 89.9%. 20Chloroquine enhances the efficacy of chemotherapy drugs against acute myeloid leukemia by inactivating the autophagy pathway显示文摘Current therapy for acute myeloid leukemia (AML) is largely hindered by the development of drug resistance of commonly used chemotherapy drugs, including cytarabine, daunorubicin, and idarubicin. In this study, we investigated the molecular mechanisms underlying the chemotherapy drug resistance and potential strategy to improve the efficacy of these drugs against AML. By analyzing data from ex vivo drug-response and multi-omics profiling public data for AML, we identified autophagy activation as a potential target in chemotherapy-resistant patients. In THP-1 and MV-4-11 cell lines, knockdown of autophagy-regulated genes ATG5 or MAP1LC3B significantly enhanced AML cell sensitivity to the chemotherapy drugs cytarabine, daunorubicin, and idarubicin. In silico screening, we found that chloroquine phosphate mimicked autophagy inactivation. We showed that chloroquine phosphate dose-dependently down-regulated the autophagy pathway in MV-4-11 cells. Furthermore, chloroquine phosphate exerted a synergistic antitumor effect with the chemotherapy drugs in vitro and in vivo.
