1Impairment of pre-mRNA splicing in liver disease: Mechanisms and consequences显示文摘Pre-mRNA splicing is an essential step in the process of gene expression in eukaryotes and consists of the removal ofintrons and the linking of exons to generate mature mRNAs. This is a highly regulated mechanism that allows the alternative usage of exons, the retention ofintronic sequences and the generation of exonic sequences of variable length. Most human genes undergo splicing events, and disruptions of this process have been associated with a variety of diseases, including cancer. Hepatocellular carcinoma (HCC) is a molecularly heterogeneous type of tumor that usually develops in a cirrhotic liver. Alterations in pre-mRNA splicing of some genes have been observed in liver cancer, and although still scarce, the available data suggest that splicing defects may have a role in hepatocarcinogenesis. Here we briefly review the general mechanisms that regulatepre-mRNA splicing, and discuss some examples that illustrate how this process is impaired in liver tumorigenesis, and may contribute to HCC development. We believe that a more thorough examination of pre-mRNA splicing is still needed to accurately draw the molecular portrait of liver cancer. 2Effect of methanolic extract of Tetrapleura tetraptera(Schum and Thonn)Taub leaves on hyperglycemia and indices of diabetic complications in alloxan-induced diabetic rats显示文摘Objective:To investigate the ameliorative role of Tetrapleura tetraplera(Schum and Thonn)Taub(T.tetraplera)leaf in hyperglycemia with associated conditions like oxidative stress,kidney damage and disorders in lipid metabolism.Methods:Five groups of five rats each intraperitoneally received the following treatment schedules for 7 d:untreated normal control,untreated alloxan-diabetic control,diabetic treated with glibenclamide,normal rats treated with extract(50 mg/kg)and diabetic rats treated with the extract.Evaluations were made for fasting blood sugar,body weight changes,malondialdehyde,aspartate aminotransferase,alanine aminotransferase,bilirubin,superoxide dismutase,catalase,lipid profile,packed cell volume,hemoglobin,urea and creatinine in all the rats.Results:Whereas the untreated diabetic rats showed a significant decrease(P<0.05)in packed cell volume,superoxide dismutase,catalase and high-density lipoprotein-cholesterol with a concomitant increase in the levels of malondialdehyde,fasting blood sugar,aspartate aminotransferase,alanine aminotransferase,bilirubin,urea and creatinine,administration of methanolic extract of T.tetraplera leaf or glibenclamide alleviated these altered parameters in the treated rats.Conclusions:Methanolic extract of T.tetraplera leaves possesses a potent capacity for treatment of diabetes and the accompanying complications,including oxidative stress and hyperlipidemia.Sunday Ene-Ojo Atawodi Ojochenemi Ejeh Yakubu Mubarak Labaran Liman Dorothy Uju iliemene 2014Asian Pacific Journal of Tropical Biomedicine2014,4,4:1

16Anticariogenic potentials of clove,tobacco and bitter kola显示文摘Objective:To investigate three tropical plant materials - clove seeds[Syzygium aromalicum(S. aromaticum)],bitter kola fruits[Garcinia kola(G.kola)]and tobacco leaves(Nicotiana species) as potential targeted killers of Streptococcus mutans(S.mutatis),a cavity-causing bacterium(gram-positive, facultative anaerobe) that resides in a multispecies microbial community(dental plaque) for the treatment of dental caries(tooth decay).Methods:Thirty one(31) teeth samples were collected from patients with obvious signs of tooth decay(swollen gum,weak or fallen tooth,etc.) using sterile swab sticks.These samples were collected from two major denial clinics in Nsukka, Enugu State,Nigeria and investigated by spread inoculation onto sterile blood agar and Mueller Hinton agar(MHA) respectively and incubated at 37℃for 24 h.The discrete colonies obtained were further re-inoculated onto sterile Mitis salivarius agar(MSA) plates and incubated as above. The isolates were characterized by gram staining and calalase lesl.Tobacco leaves,clove seeds and bitter kola fruits were ground into powder,extracted with three different solvents(n-hexane, hot water and ethanol),filtered,dried and stored in clean containers,corked and kept until used. The plant extracts were investigated for phytochemistry,minimum inhibitory concentration(MIC), minimum cidal concentration(MCC)and compared with some conventional antibiotics commonly used against tooth decay.Antibiotic sensitivity test was also carried out.The results were statistically analyzed.Results:The extracts showed varied phytochemical composition but most abundantly the flavonoids.Our result also shows that females(16) have more tooth decay than males(15) and that 16 samples were very bloody while 15 were slightly bloody.The microbial characterization showed that 18 samples were catalase-positive indicating the presence of S. mutans while 13 were catalase-negative suspected to be Staphylococcus spp.The Gram reaction confirmed 13 Gram-negative and 18 Gram-positive organisms.The n-hexane extract had the best antimicrobial activity followed by the ethanol and lastly hot water.MIC showed that n-hexane clove extract had the largest inhibition zone diameter,followed by bitter kola extract and lastly tobacco extract.The antibiotic sensitivity test credited ciprofloxacin the best because it exhibited broad spectrum of action.Conclusions:Since the n-hexane extract of clove seeds demonstrated preferential growth-inhibitory activity against the causal cariogenic pathogens(S.mutans) in dental caries,we therefore,report here that clove extract be henceforth considered as a potential ingredient in toothpaste preparation.Dibua Esther Uju Nnamani Petra Obioma 2011Asian Pacific Journal of Tropical Medicine2011,4,10:0

18A Systematic Review on Antituberculosis Drug Discovery and Antimycobacterial Potential of Biologically Synthesized Silver Nanoparticles:Overview and Future Perspectives显示文摘Rapid emergence and quick evolution of drug-resistant and aggressive mycobacterial strains have resulted in the present antimycobacterial drug crisis and the persistence of tuberculosis as a major public health problem.Green/biological nanotechnologies constitute an interesting area of research for discovering antimycobacterial agents.This review focused on the biological(green)synthesis of silver nanoparticles(AgNPs)as an alternative source of antimycobacterial agents.Data for this study were searched and screened from three electronic databases(Google Scholar,PubMed and ScienceDirect)following the Preferred Reporting Items for Systematic Reviews and Meta-analyses flowchart.Data from in total 17 eligible studieswere reported in this systematic review.Twelve of the 17 studies used plants to fabricate AgNPs,whereas the remaining five studies used microorganisms(bacteria and/or fungi).Silver as part of silver nitrate(AgNO3)was themetal precursor reported for the synthesis of AgNPs in these studies.Silver nanoparticles were mostly spherical,with sizes ranging from12 to140nm.Resultsbasedon minimum inhibitory concentrations varied between studies and were divided into three groups:(i)those more effective than the antibiotic(controls),(ii)those more effective than plant extracts,and(iii)those less effective than the antibiotic controls.In addition,little or no cytotoxicity effects were reported.Silver nanoparticles were also shown to be highly specific or selective toward mycobacterial strains.This systematic review highlights the antimycobacterial potential of biologically synthesized AgNPs,underscoring the possibility of discovering/developing new antimycobacterial agents using biological synthesis approaches with less toxicity and high selectivity.Christian K.Ezeh Chibuzor N.Eze Uju M.E.Dibua Stephen C.Emencheta 2022Infectious Microbes & Diseases2022,4,4:0
19Is pass‑through of the exchange rate to restaurant and hotel prices asymmetric in the US?Role of monetary policy uncertainty显示文摘This study examines the exchange rate pass-through to the United States(US)restaurant and hotel prices by incorporating the effect of monetary policy uncertainty over the period 2001:M12 to 2019:M01.Using the nonlinear autoregressive distributed lag(NARDL)model,empirical evidence indicates asymmetric pass-through of exchange rate and monetary policy uncertainty.Moreover,a stronger pass-through effect is observed during depreciation and a negative shock in monetary policy uncertainty,corroborating asymmetric pass-through predictions.Our results further show that a positive shock in energy prices leads to an increase in restaurant and hotel prices.Furthermore,asymmetric causality indicates that a positive shock in the exchange rate causes a positive shock to restaurant and hotel prices.We found feedback causal effects between positive and negative shocks in monetary policy uncertainty and positive and negative shocks in the exchange rate.Additionally,we detected a one-way asymmetric causality,flowing from a positive(negative)shock to a positive(negative)shock in energy prices.Therefore,these findings provide insights for policymakers to achieve low and stable prices in the US restaurant and hotel industry through sound monetary policy formulations.Highlights.The drivers of restaurant and hotel business in tourism destinations are examined.There is asymmetric pass-through of exchange rate and monetary policy uncertainty.A stronger pass-through is observed during appreciation and a negative shock to monetary policy uncertainty.There is asymmetric causality from positive shock in exchange rate to postive shock in restaurant and hotel prices.Uju Violet Alola Ojonugwa Usman Andrew Adewale Alola 2023Financial Innovation2023,9,1:0
20Intrahepatic cholestasis of pregnancy is associated with increased risk of hepatobiliary disease and adverse fetal outcomes: A systematic review and meta-analysis显示文摘Background and aims:The aim of this study was to review the literature and perform a meta-analysis to clarify the association between intrahepatic cholestasis of pregnancy and risks of long-term maternal hepatobiliary disease as well as adverse fetal outcomes including preterm birth,meconium-stained amniotic fluid,and stillbirth.Methods:This systematic review and meta-analysis was conducted in accordance with the Preferred Reporting Items for Systematic Reviews and Meta-Analyses guidelines.A comprehensive literature search was performed using Cochrane,Embase,and PubMed databases to identify observational or cohort studies comparing pregnant women with intrahepatic cholestasis of pregnancy(ICP)to pregnant women without ICP.Data from the included studies were analyzed using the Review Manager 5.4.1 software.Results:The meta-analysis showed a significant association between ICP and the risk of hepatobiliary diseases(pooled risk ratio[RR]:2.81,95%confidence interval[CI]:2.66–2.97,p<0.00001),hepatitis C(HC):a significant association between ICP and risk of HC(pooled RR:4.02,95%CI:3.12–5.19,p<0.00001),meconium-stained amniotic fluid(MSAF):ICP was significantly associated with an increased risk of MSAF(pooled RR:1.91,95%CI:1.65–2.21,p<0.00001),and preterm birth:the meta-analysis demonstrated a significant association between ICP and preterm birth(pooled RR:2.11,95%CI:2.01–2.21,p<0.00001).Conclusion:ICP demonstrated statistically significant associations with increased risks of hepatobiliary disease,HC,MSAF,and preterm birth.Peter Olujimi Odutola Peter Oluwatobi Olorunyomi Olanrewaju Oalmide Olatawura Ifeoluwapo Olorunyomi Olukayode Madojutimi Ayomide O.Fatunsin Uju Okeke 2023iLIVER2023,2,4:0
