1Antigenic variation of the human influenza A(H3N2) virus during the 2014–2015 winter season显示文摘The human influenza A(H3N2) virus dominated the 2014–2015 winter season in many countries and caused massive morbidity and mortality because of its antigenic variation. So far, very little is known about the antigenic patterns of the recent H3N2 virus. By systematically mapping the antigenic relationships of H3N2 strains isolat ed since 2010, we discov ered that two groups with obvious antigenic divergence, named SW13(A/Switzerland/9715293/2013-like strains) and HK14(A/Hong Kong/5738/2014-like strains), co-circulated during the 2014–2015 winter season. HK14 group co-circulated with SW13 in Europe and the United States during this season, while there were few strains of HK14 in China's Mainland, where SW13 has dominated since 2012. Furthermore, we found that substitutions near the receptor-binding site on hemagglutinin played an important role in the antigenic variation of both the groups. 2Ubiquitination of Rheb governs growth factor-induced mTORC1 activation显示文摘Mechanistic target of rapamycin mTOR complex 1 (mTORC1)plays a key role in the integration of various environmental signals to regulate cell growth and metabolism,mTORC1 is recruited to the lysosome where it is activated by its interaction with GTP-bound Rheb GTPase.However,the regulatory mechanism of Rheb activity remains largely unknown.Here,we show that ubiquitination governs the nucleotide-bound status of Rheb.Lysosome-anchored E3 ligase RNF152 catalyzes Rheb ubiquitination and promotes its binding to the TSC complex.EGF enhances the deubiquitination of Rheb through AKT-dependent USP4 phosphorylation,leading to the release of Rheb from the TSC complex.Functionally,ubiquitination of Rheb is linked to mTORC1-mediated signaling and consequently regulates tumor growth.Thus,we propose a mechanistic model whereby Rheb-mediated mTORC1 activation is dictated by a dynamic opposing act between Rheb ubiquitination and deubiquitination that are catalyzed by RNF152 and USP4 respectively.Lu Deng Lei Chen Linlin Zhao Yan Xu Xiaoping Peng Xinbo Wang Lin Ding Jiali Jin Hongqi Teng Yanming Wang Weijuan Pan Fei Yu Lujian Liao Li Li Xin Ge Ping Wang 2019Cell Research2019,29,2:16

4Variants of the arachidonate 5-lipoxygenase-activating protein (ALOX5AP) gene and risk of ischemic stroke in Han Chinese of eastern China显示文摘Variants of the arachidonate 5-lipoxygenase-activating protein (ALOX5AP) gene have been suggested to play an important role in the pathogenesis of atherosclerosis and ischemic stroke.This study was aimed to explore the association of ALOX5AP variants with ischemic stroke risk in Han Chinese of eastern China.A total of 690 ischemic stroke cases and 767 controls were recruited.The subjects were further subtyped according to the Trial of Org 10172 in Acute Stroke Treatment (TOAST) criteria.On the basis of that,two polymorphisms of the ALOX5AP gene (rs10507391 and rs12429692) were determined by TaqMan genotyping assay.In addition,plasma leukotriene B4 (LTB4) levels were analyzed in these subjects.There was no evidence of association between the two variants of ALOX5AP and the risk of ischemic stroke or its TOAST-subtypes.Haplotype analysis and stratification analysis according to sex,age,body mass index,hypertension,and diabetes also showed negative association.Analysis of LTB4 levels in a subset of cases and controls revealed that LTB4 levels were significantly higher in ischemic stroke cases than in the controls (70.06±14.75 ng/L vs 57.34±10.93 ng/L;P=0.000) and carriers of the T allele of the rs10507391 variant were associated with higher plasma LTB4 levels (P=0.000).The present study suggests there is no association of the two polymorphisms in the ALOX5AP gene with ischemic stroke risk in Han Chinese of eastern China.Gannan Wang Yao Wang Hao Sun Weijuan Cao Jing Zhang Hang Xiao Jinsong Zhang 2011The Journal of Biomedical Research2011,25,5:12

6Comparison of breast-conserving surgery and mastectomy in early breast cancer using observational data revisited:a propensity score-matched analysis显示文摘Recent observational studies showed that breast-conserving surgery(BCS) resulted in superior survival compared to mastectomy in breast cancer patients. This study compared the clinical outcomes of BCS and mastectomy using propensity score(PS)matching analysis, which had advantages over conventional methods in reducing bias. Nonmetastatic breast cancer patients who underwent BCS and mastectomy were matched 1:1 based on their PS. We used the Kaplan-Meier method and Cox-regression model to estimate the treatment effects. A total of 2,866 patients with a median follow-up time of 67 months were included in the original study population. Although the mastectomy cohort(N=1,219) had more advanced disease compared to the BCS cohort(N=1,647), LRFS was similar between the two groups(93.8% vs. 92.4%, P>0.05). BCS(vs. mastectomy) was associated with improved DFS(73.8% vs. 58.7%, P<0.01) and CSS(91% vs. 78.2%, P<0.01) in the original population. In the PS-matched population(N=1,668), clinicopathological features were equally distributed between the two cohorts. BCS(vs. mastectomy) was not associated with improved DFS(70.7% vs. 66.9%, P>0.05) or CSS(87.5% vs. 84.9%, P>0.05). We found that PS methods reduce bias when estimating treatment effects using observational data. 7Microelement analysis of source of raw materials of terracotta warriors and horses of Qin's Mausoleum in pits No.1 and No.2显示文摘In order to search for sources of the raw material of the Qin’s terracotta warriors and horses, we selected 20 samples of pottery warriors and pottery horses of the Qin’s Mausoleum from pit No. 1,19 samples of pottery warriors and brick from pit No. 2, 20 samples of clays near the Qin’s Mausoleum and 2 samples of Yaozhou porcelain bodies. The 32 kinds of element content of each sample were measured by neutron activation analysis (NAA). These data were analyzed by fuzzy cluster analysis, the trend cluster analysis diagram was obtained. The results showed that samples of terracotta warriors and horses from pits No.1 and No. 2 were closely related to the loam layer near Qin’s Mausoleum, but their relations with the loess layers were not so close, and they were out of all relation to Yaozhou porcelain bodies. 8Astragalus injection inhibits c-Jun N terminal kinase mRNA expression following oxygen-glucose deprivation and reintroduction in rat hippocampal neurons显示文摘BACKGROUND:In studies concerning cell injury induced by cerebral ischemia-reperfusion,current experiments have primarily focused on altered protein levels.In addition,the apoptotic proteins Bax and Bcl-2 have been thoroughly studied with regard to initiating neuronal apoptosis. OBJECTIVE:To establish an in vitro model of oxygen-glucose deprivation and reintroduction in the rat hippocampus to simulate cerebral ischemia-reperfusion injury;to observe c-Jun N-terminal kinase 3(JNK3) mRNA expression in hippocampal neurons following Astragalus injection;and thus to determine changes in the signaling and downstream pathways of neuronal apoptosis at the cellular and molecular level. DESIGN,TIME AND SETTING:A randomized,controlled,cellular and molecular experiment was performed at the Department of Central Laboratory,Chengde Medical College from February to June 2008. MATERIALS:Astragalus injection,the main ingredient of astragaloside,was purchased from Chengdu Di'ao Jiuhong Pharmaceutical Manufactory,China.JNK3 mRNA probe and in situ hybridization kit were purchased from Tianjin Haoyang Biological Technology,China,and JNK3 RT-PCR primers were designed by Shanghai Bio-engineering,China. METHODS:Primary cultures of hippocampal neurons derived from Sprague Dawley rats,aged 1-2 days,were established.After 8 days,the hippocampal neurons were assigned to the following interventions:model group,Astragalus group,and vehicle control group,cells were subjected to oxygen-glucose reintroduction after oxygen-glucose deprivation for 30 minutes in sugar-free Earle's solution and a hypoxia device,which contained high-purity nitrogen.The normal control group was subjected to primary culture techniques and was not treated using above-mentioned interventions. In addition,the Astragalus and vehicle control groups were treated with Astragalus injection(0.5 g/L raw drug) or sterile,deionized water at 2 hours prior to oxygen-glucose deprivation,respectively. MAIN OUTCOME MEASURES:JNK3 mRNA expression was measured by in situ hybridization and RT-PCR at 0,0.5,2,6,24,72,and 120 hours after oxygen-glucose reintroduction. RESULTS:Hippocampal neuronal morphology was normal in the normal control group. Hippocampal neurons exhibited apparent apoptosis-like pathological changes in the model,as well as the vehicle control,groups.The apoptosis-like pathological changes in the hippocampal neurons were less in the Astragalus group.Results from in situ hybridization and RT-PCR showed that JNK3 mRNA expression significantly increased in hippocampal neurons from model group,as well as the vehicle control group,compared with the normal control group(P<0.05).In addition,JNK3 mRNA expression significantly decreased in hippocampal neurons of the Astragalus group,compared with the model group and vehicle control group(P<0.05). 9Nondestructive 3D Image Analysis Pipeline to Extract Rice Grain Traits Using X-Ray Computed Tomography显示文摘The traits of rice panicles play important roles in yield assessment,variety classification,rice breeding,and cultivation management.Most traditional grain phenotyping methods require threshing and thus are time-consuming and labor-intensive;moreover,these methods cannot obtain 3D grain traits.In this work,based on X-ray computed tomography,we proposed an image analysis method to extract twenty-two 3D grain traits.After 104 samples were tested,the R^(2) values between the extracted and manual measurements of the grain number and grain length were 0.980 and 0.960,respectively.We also found a high correlation between the total grain volume and weight.In addition,the extracted 3D grain traits were used to classify the rice varieties,and the support vector machine classifier had a higher recognition accuracy than the stepwise discriminant analysis and random forest classifiers.In conclusion,we developed a 3D image analysis pipeline to extract rice grain traits using X-ray computed tomography that can provide more 3D grain information and could benefit future research on rice functional genomics and rice breeding.Weijuan Hu Can Zhang Yuqiang Jiang Chenglong Huang Qian Liu Lizhong Xiong Wanneng Yang Fan Chen 2020Plant Phenomics2020,2,1:6

11Establishment and Characterization of a Cell Based Artificial Antigen-Presenting Cell for Expansion and Activation of CD8^+ T Cells Ex Vivo显示文摘Artificial antigen-presenting cells are expected to stimulate the expansion and acquisition of optimal therapeutic features of T cells before infusion. Here CD32 that binds to a crystallizable fragment of IgG monoclonal antibody was genetically expressed on human K562 leukemia cells to provide a ligand for T-cell receptor. CD86 and 4-1BBL,which are ligands of co-stimulating receptors of CD28 and 4-1BB,respectively,were also expressed on K562 cells. Then we accomplished the artificial antigen-presenting cells by coupling K32/CD86/4-1BBL cell with OKT3 monoclonal antibody against CD3,named K32/CD86/4-1BBL/OKT3 cells. These artificial modified cells had the abilities of inducing CD8+ T cell activation,promoting CD8+ T cell proliferation,division,and long-term growth,inhibiting CD8+ T cell apoptosis,and enhancing CD8+ T cell secretion of IFN-γ and perforin. Furthermore,antigen-specific cytotoxic T lymphocytes could be retained in the culture stimulated with K32/CD86/4-1BBL/OKT3 cells at least within 28 days. 12In vitro effects of sodium hyaluronate on the proliferation and the apoptosis in chondrocytes from patients with Kashin-Beck disease and osteoarthritis显示文摘Objective:To identify the in vitro effects of sodium hyaluronate(HA) on the proliferation and the apoptosis of chondrocytes from patients with Kashin-Beck disease(KBD) and osteoarthritis(OA). Methods:Samples of articular cartilages from KBD and OA patients, as well as healthy volunteers(6 subjects in each of the 3 groups) were dissected, digested with collagenase and the cells cultured in monolayers. Chondrocytes from each sample were assigned to an untreated group and two HA-treated groups: H0(no HA), H100(HA, 0.1 g/L) and H500(HA, 0.5 g/L). The first passage chondrocytes were used to observe proliferation using the MTT assay, and apoptosis by flow cytometry through Annexin V/PI staining. Results:HA promoted proliferation of chondrocytes in all the three groups, and in KBD and OA groups, for cells cultured for 4 and 6 days, H500 significantly promoted the cell proliferation. The apoptotic rates of both KBD and OA group chondrocytes were in the order H500 < HA100 < H0. 13Genome-wide identification of agronomically important genes in outcrossing crops using OutcrossSeq显示文摘Many important crops(e.g.,tuber,root,and tree crops)are cross-pollinating.For these crops,no inbred lines are available for genetic study and breeding because they are self-incompatible,clonally propagated,or have a long generation time,making the identification of agronomically important genes difficult,particularly in crops with a complex autopolyploid genome.In this study,we developed a method,OutcrossSeq,for mapping agronomically important loci in outcrossing crops based on whole-genome low-coverage resequencing of a large genetic population,and designed three computation algorithms in OutcrossSeq for different types of outcrossing populations.We applied OutcrossSeq to a tuberous root crop(sweet potato,autopolyploid),a tree crop(walnut tree,highly heterozygous diploid),and hybrid crops(double-cross populations)to generate high-density genotype maps for the outcrossing populations,which enable precise identification of genomic loci underlying important agronomic traits.Candidate causative genes at these loci were detected based on functional clues.Taken together,our results indicate that OutcrossSeq is a robust and powerful method for identifying agronomically important genes in heterozygous species,including polyploids,in a cost-efficient way.The OutcrossSeq software and its instruction manual are available for downloading at www.xhhuanglab.cn/tool/OutcrossSeq.html.Mengjiao Chen Weijuan Fan Feiyang Ji Hua Hua Jie Liu Mengxiao Yan Qingguo Ma Jiongjiong Fan Qin Wang Shufeng Zhang Guiling Liu Zhe Sun Changgeng Tian Fengling Zhao Jianli Zheng Qi Zhang Jiaxin Chen Jie Qiu Xin Wei Ziru Chen Peng Zhang Dong Pei Jun Yang Xuehui Huang 2021Molecular Plant2021,14,4:4
14High-throughput screening identifies established drugs as SARS-CoV-2 PLpro inhibitors显示文摘A new coronavirus(SARS-CoV-2)has been identified as the etiologic agent for the COVID-19 outbreak.Currently,effective treatment options remain very limited for this disease;therefore,there is an urgent need to identify new anti-COVID-19 agents.In this study,we screened over 6,000 compounds that included approved drugs,drug candidates in clinical trials,and pharmacologically active compounds to identify leads that target the SARS-CoV-2 papain-like protease(PLpro).Together with main protease(Mpro),PLpro is responsible for processing the viral replicase polyprotein into functional units.There-fore,it is an attractive target for antiviral drug develop-ment.Here we discovered four compounds,YM155,cryptotanshinone,tanshinone I and GRL0617 that inhibit SARS-CoV-2 PLpro with IC50 values ranging from 1.39 to 5.63 pmol/L.These compounds also exhibit strong antiviral activities in cell-based assays.YM155,an anti-cancer drug candidate in clinical trials,has the most potent antiviral activity with an EC50 value of 170 nmol/L.In addition,we have determined the crystal structures of this enzyme and its complex with YM155,revealing a unique binding mode.YM155 simultaneously targets three'hot'spots on PLpro,including the substrate-binding pocket,the interferon stimulating gene product 15(ISG15)binding site and zinc finger motif.Our results demonstrate the efficacy of this screening and repur-posing strategy,which has led to the discovery of new drug leads with clinical potential for COVID-19 treatments.Yao Zhao Xiaoyu Du Yinkai Duan Xiaoyan Pan Yifang Sun Tian You Lin Han Zhenming Jin Weijuan Shang Jing Yu Hangtian Guo Qianying Liu Yan Wu Chao Peng Jun Wang Chenghao Zhu Xiuna Yang Kailin Yang Ying Lei Luke W.Guddat Wenqing Xu Gengfu Xiao Lei Sun Leike Zhang Zihe Rao Haitao Yang 2021Protein & Cell2021,12,11:4
15Epidemiological and Virological Surveillance of Seasonal Influenza Viruses—China,2020–2021显示文摘Introduction:During the coronavirus disease 2019(COVID-19)pandemic,the circulation of seasonal influenza virus declined globally and remained below previous seasonal levels.We analyzed the results of the epidemiology,antigenic,and genetic characteristics,and antiviral susceptibilities of seasonal influenza viruses isolated from the mainland of China during October 5,2020 through September 5,2021,to better assess the risk of influenza during subsequent influenza season in 2021–2022.Methods:Positive rates of influenza virus detection during this period were based on real-time polymerase chain reaction(PCR)detection by the Chinese National Influenza Surveillance Network laboratories,and isolated viruses from influenza positive samples were submitted to the Chinese National Influenza Center.Antigenic analyses for influenza viruses were conducted using the hemagglutination inhibition assay.Next-generation sequencing was used for genetic analyses.Viruses were tested for resistance to antiviral medications using a phenotypic assay and next-generation sequencing.Results:In southern China,the influenza positivity rate was elevated especially after March 2021 and was higher than the same period the previous year with the COVID-19 pandemic.In northern China,influenza positive rate peaked at Week 18 in 2021 and has declined since then.Nearly all isolated viruses were B/Victoria lineage viruses during the study period,and 37.3%of these viruses are antigenically similar to the reference viruses representing the vaccine components for the 2020–2021 and 2021–2022 Northern Hemisphere influenza season.All seasonal influenza viruses were susceptible to neuraminidase inhibitors and endonuclease inhibitors.Conclusions:Influenza activity has gradually increased in the mainland of China in 2021,although the intensity of activity is still lower than before the COVID-19 pandemic.The diversity of circulating influenza types/subtypes decreased,with the vast majority being B/Victoria lineage viruses.The surveillance data from this study suggest that we should strengthen influenza surveillance during the upcoming traditional influenza season.It also provided evidence for vaccine recommendations and prevention and control of influenza and clinical use of antiviral drugs.Weijuan Huang Xiyan Li Minju Tan Yanhui Cheng Tao Chen Hejiang Wei Xiaoxu Zeng Yiran Xie Jia Liu Ning Xiao Lei Yang Dayan Wang 2021China CDC weekly2021,3,44:4

17Effect of particle size and oxygen content on ignition and combustion of aluminum particles显示文摘Particle size and oxygen content are two of the key factors that affect the ignition and combustion properties of aluminum particles. In this study, a laser ignition experimental system and flame test system were built to analyze the ignition and combustion characteristics and the flame morphology of aluminum particles. A thermobalance system was used to analyze the thermal oxidation characteristics. In addition, the microstructure of aluminum was analyzed by scanning electron microscopy. It was found that the oxidized products were some of the gas phase products agglomerated. Smaller particle size samples showed better combustion characteristics. The combustion intensity, self-sustaining combustion time and the burn-off rate showed a rising trend with the decrease in the particle size. Increasing the oxygen content in the atmosphere could improve the ignition and combustion characteristics of the samples. Four distinct stages were observed in the process of ignition and combustion. Small particle size samples had a larger flame height and luminance, and the self-sustaining combustion time was much longer.Three distinct stages were observed during the thermal oxidation process. 18Preparation of hapten-specific monoclonal antibody for cadmium and its ELISA application to aqueous samples显示文摘High-affinity and specific monoclonal antibodies against cadmium-ethylene diamine tetraacetic acid(EDTA)complex have been produced using the hybridoma technique.A hapten was synthesized and characterized by Fourier Transform Infrared Spectroscopy(FT-IR)and UVVis.Competitive enzyme-linked immunosorbent assay(ELISA)for quantitative detection of cadmium in aqueous sample was developed.The monoclonal antibody with high level of binding affinity for Cd-IEDTA-BSA and high specificity for soluble Cd-EDTA complex showed less than 0.99%cross-reactivity with other 11 metals.The limit of detection was 0.10μg·L^(-1),and the effective linear range was 10^(-1)-10^(3)μg·L^(-1).The intra-and inter-assay coefficient variations were 1.5%-6.3%and 3.2%-7.4%,respectively.The spike recovery in different water samples were between 98.5%and 110.3%.The detection limit of this assay was well below the allowable concentration of cadmium(3μg·L^(-1)),and the working range was wider than that of other methods which showed the range of 2.19-86.38 and 0-10^(3)μg·L^(-1).The competitive ELISA established in this paper was sensitive and accurate in the screening of cadmium in aqueous samples.The results will lay a solid foundation for construction of an immunoassay kit for cadmium.Huan HE Bo TANG Cheng SUN Shaogui YANG Weijuan ZHENG Zichun HUA 2011Frontiers of Environmental Science & Engineering2011,5,3:4
19Comprehensive interactome analysis of the spike protein of swine acute diarrhea syndrome coronavirus显示文摘Swine acute diarrhea syndrome coronavirus(SADS‐CoV)is a recently discovered coronavirus that causes severe and acute diarrhea and rapid weight loss in piglets.SADS‐CoV was reported to be capable of infecting cell lines derived from diverse species,including bats,mice,hamsters,rats,chickens,pigs,nonhuman primates,and humans,implying its high risk of cross‐species infection.However,its receptor is still unknown.In this study,the receptor‐binding domain of the SADS‐CoV spike(S)protein was purified and then subjected to affinity purification(AP)‐coupled mass spectrometry(MS)‐based proteomic analysis to identify the interactors of the SADS‐CoV S protein.Forty‐three host proteins were identified,and a Gene Ontology analysis indicated that these interactors can be grouped into categories such as“cell‐cell adhesion”,“translation”“viral transcription”,suggesting that these processes may participate in the SADS‐CoV life cycles.RNA interference‐based screening of these interactors indicated that PPIB and vimentin can affect SADS‐CoV replication.Our study provides an overarching view into the host interactome of the SADS‐CoV S protein and highlights potential targets for the development of therapeutics against SADS‐CoV.Qingxing Wang Yun Luo Weijuan Shang Zhengli Shi Gengfu Xiao Leike Zhang 2021Biosafety and Health2021,3,3:3
20Complex Precipitation Mechanism of Ti-Nb-V Microalloyed Bainitic Base High Strength Steel显示文摘The addition of high Ti(>0.1%) in microalloyed bainitic high strength steel was designed, and the precipitation morphology of steels with different Ti, Nb, and V contents was studied by utilizing transmission electron microscopy(TEM). Based on the classical nucleation-crystal growth theory and the Johnson-Mehl-Avrami equation, the precipitation thermodynamic and kinetic model of second phase particles in austenite was established in the form of(Nbx,Vy,Tiz)C, and the complex precipitation mechanism of second phase particles was emphatically studied. The experimental results show that the complex precipitation particles could be divided into two categories: the coarser particles with about 100 nm grain size and the independent complex precipitation particles in the form of(Nb,V,Ti)C with 35-50 nm grain size. The latter has a better precipitation strengthening effect, and the calculated PTT curve shows a typical 'C' shape. When the deformed storage energy is 3 820 J·mol^-1, the fastest precipitation temperature of calculated PTT curve is 925℃, and the calculated result is essentially consistent with experimental values.
