1Humoral immune response to circulating SARS-CoV-2 variants elicited by inactivated and RBD-subunit vaccines显示文摘SARS-CoV-2 variants could induce immune escape by mutations on the receptor-binding domain(RBD)and N-terminal domain(NTD).Here we report the humoral immune response to circulating SARS-CoV-2 variants,such as 501Y.V2(B.1.351),of the plasma and neutralizing antibodies(NAbs)elicited by CoronaVac(inactivated vaccine),ZF2001(RBD-subunit vaccine)and natural infection.Among 86 potent NAbs identified by high-throughput single-cell VDJ sequencing of peripheral blood mononuclear cells from vaccinees and convalescents,near half anti-RBD NAbs showed major neutralization reductions against the K417N/E484K/N501Y mutation combination,with E484K being the dominant cause.VH3-53/VH3-66 recurrent antibodies respond differently to RBD variants,and K417N compromises the majority of neutralizing activity through reduced polar contacts with complementarity determining regions.In contrast,the 242–244 deletion(242–244Δ)would abolish most neutralization activity of anti-NTD NAbs by interrupting the conformation of NTD antigenic supersite,indicating a much less diversity of anti-NTD NAbs than anti-RBD NAbs.Plasma of convalescents and CoronaVac vaccinees displayed comparable neutralization reductions against pseudo-and authentic 501Y.V2 variants,mainly caused by E484K/N501Y and 242–244Δ,with the effects being additive.Importantly,RBD-subunit vaccinees exhibit markedly higher tolerance to 501Y.V2 than convalescents,since the elicited anti-RBD NAbs display a high diversity and are unaffected by NTD mutations.Moreover,an extended gap between the third and second doses of ZF2001 leads to better neutralizing activity and tolerance to 501Y.V2 than the standard three-dose administration.Together,these results suggest that the deployment of RBD-vaccines,through a third-dose boost,may be ideal for combating SARS-CoV-2 variants when necessary,especially for those carrying mutations that disrupt the NTD supersite.Yunlong Cao Ayijiang Yisimayi Yali Bai Weijin Huang Xiaofeng Li Zhiying Zhang Tianjiao Yuan Ran An Jing Wang Tianhe Xiao Shuo Du Wenping Ma Liyang Song Yongzheng Li Xiang Li Weiliang Song Jiajing Wu Shuo Liu Xuemei Li Yonghong Zhang Bin Su Xianghua Guo Yangyang Wei Chuanping Gao Nana Zhang Yifei Zhang Yang Dou Xiaoyu Xu Rui Shi Bai Lu Ronghua Jin Yingmin Ma Chengfeng Qin Youchun Wang Yingmei Feng Junyu Xiao Xiaoliang Sunney Xie 2021Cell Research2021,31,7:12
2Reproductive management through integration of PGD and MPS-based noninvasive prenatal screening/diagnosis for a family with GJB2-associated hearing impairment显示文摘A couple with a proband child of GJB2(encoding the gap junction protein connexin 26)-associated hearing impairment and a previous pregnancy miscarriage sought for a reproductive solution to bear a healthy child. Our study aimed to develop a customized preconception-to-neonate care trajectory to fulfill this clinical demand by integrating preimplantation genetic diagnosis(PGD), noninvasive prenatal testing(NIPT), and noninvasive prenatal diagnosis(NIPD) into the strategy. Auditory and genetic diagnosis of the proband child was carried out to identify the disease causative mutations. The couple then received in-vitro-fertilization treatment, and eight embryos were obtained for day 5 biopsy. PGD was performed by short-tandem-repeat linkage analysis and Sanger sequencing of GJB2 gene. Transfer of a GJB2 c.235del C heterozygous embryo resulted in a singleton pregnancy. At the 13 th week of gestation, genomic DNA(g DNA) from the trio family and cell-free DNA(cf DNA) from maternal plasma were obtained for assessment of fetal chromosomal aneuploidy and GJB2 mutations. NIPT and NIPD showed the absence of chromosomal aneuploidy and GJB2-associated disease in the fetus, which was later confirmed by invasive procedures and postnatal genetic/auditory diagnosis. 3Characterization of the essential role of bone morphogenetic protein 9 (BMP9) in osteogenic differentiation of mesenchymal stem cells (MSCs) through RNA interference显示文摘Mesenchymal stem cells(MSCs)are multipotent stem cells and capable of differentiating into multiple cell types including osteoblastic,chondrogenic and adipogenic lineages.We previously identified BMP9 as one of the most potent BMPs that induce osteoblastic differentiation of MSCs although exact molecular mechanism through which BMP9 regulates osteogenic differentiation remains to be fully understood.Here,we seek to develop a recombinant adenovirus system to optimally silence mouse BMP9 and then characterize the important role of BMP9 in osteogenic differentiation of MSCs.Using two different siRNA bioinformatic prediction programs,we design five siRNAs targeting mouse BMP9(or simB9),which are expressed under the control of the converging H1 and U6 promoters in recombinant adenovirus vectors.We demonstrate that two of the five siRNAs,simB9-4 and simB9-7,exhibit the highest efficiency on silencing exogenous mouse BMP9 in MSCs.Furthermore,simB9-4 and simB9-7 act synergistically in inhibiting BMP9-induced expression of osteogenic markers,matrix mineralization and ectopic bone formation from MSCs.Thus,our findings demonstrate the important role of BMP9 in osteogenic differentiation of MSCs.The characterized simB9 siRNAs may be used as an important tool to investigate the molecular mechanism behind BMP9 osteogenic signaling.Our results also indicate that recombinant adenovirus-mediated expression of siRNAs is efficient and sustained,and thus may be used as an effective delivery vehicle of siRNA therapeutics.Shujuan Yan Ruyi Zhang Ke Wu Jing Cui Shifeng Huang Xiaojuan Ji Liping An Chengfu Yuan Cheng Gong Linghuan Zhang Wei Liu Yixiao Feng Bo Zhang Zhengyu Dai Yi Shen Xi Wang Wenping Luo Bo Liu Rex C.Haydon Michael J.Lee Russell R.Reid Jennifer Moriatis Wolf Qiong Shi Hue H.Luu Tong-Chuan He Yaguang Weng 2018Genes & Diseases2018,5,2:8
4Aerodynamic performance enhancement forflapping airfoils by co-flow jet显示文摘Introducing active flow control into the design of flapping wing is an effective way to enhance its aerodynamic performance.In this paper,a novel active flow control technology called Co-Flow Jet(CFJ)is applied to flapping airfoils.The effect of CFJ on aerodynamic performance of flapping airfoils at low Reynolds number is numerically investigated using Unsteady Reynolds Averaged Navier-Stokes(URANS)simulation with Spalart-Allmaras(SA)turbulence model.Numerical methods are validated by a NACA6415-based CFJ airfoil case and a S809 pitching airfoil case.Then NACA6415 baseline airfoil and NACA6415-based CFJ airfoil with jet-off and jet-on are simulated in flapping motion,with Reynolds number 70,000 and reduced frequency 0.2.As a result,CFJ airfoils with jet-on generally have better lift and thrust characteristics than baseline airfoils and jet-off airfoil when Cμgreater than 0.04,which results from the CFJ effect of reducing flow separation by injecting high-energy fluid into boundary layer.Besides,typical kinematic and geometric parameters,including the reduced frequency and the positions of the suction and injection slot,are systematically studied to figure out their influence on aerodynamic performance of the CFJ airfoil.And a variable Cμjet control strategy is proposed to further improve effective propulsive efficiency.Compared with using constant Cμ,an increase of effective propulsive efficiency by22.6%has been achieved by using prescribed variable CμNACA6415-based CFJ airfoil at frequency 0.2.This study may provide some guidance to performance enhancement for Flapping wing Micro Air Vehicles(FMAV).Tao WU Bifeng SONG Wenping SONG Wenqing YANG Zhonghua HAN 2020Chinese Journal of Aeronautics2020,33,10:7
5A transition prediction method for flow over airfoils based on high-order dynamic mode decomposition显示文摘This article presents a novel approach for predicting transition locations over airfoils,which are used to activate turbulence model in a Reynolds-averaged Navier-Stokes flow solver.This approach combines Dynamic Mode Decomposition(DMD)with e^Ncriterion.The core idea is to use a spatial DMD analysis to extract the modes of unstable perturbations from a steady flowfield and substitute the local Linear Stability Theory(LST)analysis to quantify the spatial growth of Tollmien–Schlichting(TS)waves.Transition is assumed to take place at the stream-wise location where the most amplified mode’s N-factor reaches a prescribed threshold and a turbulence model is activated thereafter.To improve robustness,the high-order version of DMD technique(known as HODMD)is employed.A theoretical derivation is conducted to interpret how a spatial highorder DMD analysis can extract the growth rate of the unsteady perturbations.The new method is validated by transition predictions of flows over a low-speed Natural-Laminar-Flow(NLF)airfoil NLF0416 at various angles of attack and a transonic NLF airfoil NPU-LSC-72613.The transition locations predicted by our HODMD/e^Nmethod agree well with experimental data and compare favorably to those obtained by some existing methods■.It is shown that the proposed method is able to predict transition locations for flows over different types of airfoils and offers the potential for application to 3D wings as well as more complex configurations.Mengmeng WU Zhonghua HAN Han NIE Wenping SONG Soledad Le CLAINCHE Esteban FERRER 2019Chinese Journal of Aeronautics2019,32,11:7

7Nonselective etching of GaN/AlGaN heterostructures by Cl2/Ar/BCl3 inductively coupled plasmas显示文摘A systematic study of the nonselective and smooth etching of GaN/AIGaN het-erostructures was performed using C12/Ar/BCI3 inductively coupled plasmas (ICR).Nonselective etching can be realized by adjusting the BCI3 ratio in the Cl2/Ar/BCI3 mixture (20%—60%), increasing the ICR power and dc bias, and decreasing the chamber pressure. Surface morphology of the etched heterostructures strongly depends on the gas chemistry and the chamber pressure. Specifically, with the addition of 20% BCI3 to Cl2/Ar (4:1) gas mixture, nonselective etching of GaN/Al0.28Ga0.72N heterostructures at high etch rate is maintained and the surface root-mean-square (rms) roughness is reduced from 10.622 to 0.495 nm, which is smoother than the as-grown sample. 9Establishment and functional characterization of the reversibly immortalized mouse glomerular podocytes(imPODs)显示文摘Glomerular podocytes are highly specialized epithelial cells and play an essential role in establishing the selective permeability of the glomerular filtration barrier of kidney.Maintaining the viability and structural integrity of podocytes is critical to the clinical management of glomerular diseases,which requires a thorough understanding of podocyte cell biology.As mature podocytes lose proliferative capacity,a conditionally SV40 mutant tsA58-immortalized mouse podocyte line(designated as tsPC)was established from the Immortomouse over 20 years ago.However,the utility of the tsPC cells is hampered by the practical inconvenience of culturing these cells.In this study,we establish a user-friendly and reversibly-immortalized mouse podocyte line(designated as imPOD),on the basis of the tsPC cells by stably expressing the wildtype SV40 T-antigen,which is flanked with FRT sites.We show the imPOD cells exhibit long-term high proliferative activity,which can be effectively reversed by FLP recombinase.The imPOD cells express most podocyte-related markers,including WT-1,Nephrin,Tubulin and Vinculin,but not differentiation marker Synaptopodin.The imPOD cells do not form tumor-like masses in vivo.We further demonstrate that TGFb1 induces a podocyte injury-like response in the FLP-reverted imPOD cells by suppressing the expression of slit diaphragm-associated proteins P-Cadherin and ZO-1 and upregulating the expression of mesenchymal markers,a-SMA,Vimentin and Nestin,as well as fibrogenic factors CTGF and Col1a1.Collectively,our results strongly demonstrate that the newly engineered im-POD cells should be a valuable tool to study podocyte biology both under normal and under pathological conditions.Xinyi Yu Liqun Chen Ke Wu Shujuan Yan Ruyi Zhang Chen Zhao Zongyue Zeng Yi Shu Shifeng Huang Jiayan Lei Xiaojuan Ji Chengfu Yuan Linghuan Zhang Yixiao Feng Wei Liu Bo Huang Bo Zhang Wenping Luo Xi Wang Bo Liu Rex C.Haydon Hue H.Luu Tong-Chuan He Hua Gan 2018Genes & Diseases2018,5,2:5

12Local delivery of arsenic trioxide nanoparticles for hepatocellular carcinoma treatment显示文摘Hepatocellular carcinoma(HCC)is a malignancy with a poor prognosis.Surgery combined with chemotherapy has been recommended as a curative regimen for HCC.Nevertheless,the anticancer mechanisms of chemicals in hepatocellular carcinoma remain unclear.Pyroptosis is a type of programmed necrosis,and its mechanism in hepatocellular carcinoma is poorly understood.The efficacy and mechanism of arsenic trioxide nanoparticles in the treatment of HCC were explored in this research.Arsenic trioxide alone and arsenic trioxide nanoparticles were conveniently administered to mice intratumorally using a needle.Compared with As_(2)O_(3),As_(2)O_(3)nanoparticles(As_(2)O_(3)-NPs)showed better inhibition,promoted greater LDH release,and induced cell morphology indicative of pyroptosis in vitro.Compared with the free drug,As_(2)O_(3)-NPs increased GSDME-N expression and decreased Dnmt3a,Dnmt3b,and Dnmt1 expression in Huh7 cells.In vivo,As_(2)O_(3)-NPs induced a significant decrease in the expression of Dnmt3a,Dnmt3b and Dnmt1,but significantly upregulated the expression of GSDME-N(gasdermin E(GSDME)was originally found to be related to deafness;recently,it has been defined as a gasdermin family member associated with pyroptosis).As_(2)O_(3)-NPs inhibited tumor growth more strongly than As_(2)O_(3)or control,a finding likely attributed to the downregulation of PCNA and DNMT-related proteins and the upregulation of GSDME-N.Jian Hu Yi Dong Li Ding Yang Dong Zhihua Wu Wenping Wang Ming Shen Yourong Duan 2019Signal Transduction and Targeted Therapy2019,4,1:2
13Molecular dynamics simulation of rhenium effects on creep behavior of Ni-based single crystal superalloys显示文摘The tensile creep behavior of Ni-based single crystal superalloys was investigated by molecular dynamics simulations.The effects of rhenium(Re)on the creep behavior and microstructural evolution mechanism during nanoscale creep tests were discussed.The results indicate that the addition of Re can effectively enhance the creep resistance of the alloys.The reason is that Re plays an important role in the evolution of microstructure at each stage of the creep.Firstly,Re has a pinning effect to delay the failure of the dislocation network at the primary creep stage.And then,the addition of Re decreases dislocation density and increases steady creep time at the steady-state creep.Finally,Re segregates at the dislocations in theγ0 precipitates phase and prevents the dislocation from cutting into theγ0 phase by dragging dislocations at the tertiary creep stage,which enhances effectively the creep resistance of superalloys.These mechanisms help to further understand the creep behavior and derive an atomistic description of the Re effects on creep behavior and creep mechanisms in Ni-based single crystal superalloys.Wenping Wu Bin Chen Hongfei Shen Zijun Ding 2022Progress in Natural Science:Materials International2022,32,2:2
14A branched dihydrophenazine-based polymer as a cathode material to achieve dual-ion batteries with high energy and power density显示文摘Organic electrode materials have exhibited good electrochemical performance in batteries,but their voltages and rate capabilities still require improvement to meet the increasing demand for batteries with high energy and power density.Herein,we design and synthesize a branched dihydrophenazine-based polymer(p-TPPZ)as a cathode material for dual-ion batteries(DIBs)through delicate molecular design.Compared with the linear dihydrophenazine-based polymer(p-DPPZ,with a theoretical capacity of 209 mAh g^(−1)),p-TPPZ possessed a higher theoretical capacity of 233 mAh g^(−1)and lower highest occupied molecular orbital energy levels,which resulted in a high actual capacity(169.3 mAh g^(−1)at 0.5 C),an average discharge voltage of 3.65 V(vs.Li+/Li)and a high energy density(618.2 Wh kg^(−1),based on the cathode materials).The branched structure of p-TPPZ led to a larger specific surface area than that of p-DPPZ,which was beneficial for the electrolyte infiltration and fast ionic transport,contributing to the high power density.Due to the fast reaction kinetics,even at a power density of 23,725 W kg^(−1)(40 C),the energy density still reached 474.5 Wh kg^(−1).We also made a detailed investigation of the p-TPPZ cathode’s charge storage mechanism.This work will stimulate the further molecular design to develop organic batteries with both high energy and power density.Shuaifei Xu Huichao Dai Shaolong Zhu Yanchao Wu Mingxuan Sun Yuan Chen Kun Fan Chenyang Zhang Chengliang Wang Wenping Hu 2021eScience2021,1,1:2
15Crystal structure of a secreted lipase from Gibberella zeae reveals a novel “double-lock” mechanism显示文摘Fusarium graminearum(sexual stage:Gibberella zeae)is the causative agent of Fusarium Head Blight(FHB),which is one of the most destructive plant disease of cereals,accounting for high grain yield losses,especially for wheat and maize.Like other fungal pathogens,several extracellular enzymes secreted by G.zeae are known to be involved in host infection.Among these secreted lipases,G.zeae lipase(GZEL),which is encoded by the FGL1 gene,was demonstrated to be crucial to G.zeae pathogenicity.However,the precise mechanism of GZEL remains unclear due to a lack of detailed structural information.In this study,we report the crystal structure of GZEL at the atomic level.The structure of GZEL displays distinct structural differences compared to reported homologues and indicates a unique“double lock”enzymatic mechanism.To gain insight into substrate/inhibitor recognition,we proposed a model of GZEL in complex with substrate and the lipase inhibitor ebelactone B(based on the reported structures of GZEL homologues),which defines possible substrate binding sites within the catalytic cleft and suggests an“anti sn-l”binding mode.These results pave the way to elucidating the mechanism of GZEL and thus provide clues for the design of anti-FHB inhibitors.Zhiyong Lou Ming Li Yuna Sun Ye Liu Zheng Liu Wenping Wu Zihe Rao 2010Protein & Cell2010,1,8:2

17Lightweight Design of Spaceborne CFRP Rigid Antenna Reflector显示文摘In order to overcome the shortcoming of space-borne rigid antenna reflector made of carbon fiber reinforced plastic(CFRP)skins with aluminum honeycomb sandwich(SAHS)structure,a new type of full CFRP skin plus rib(SPR)structure ring-focused parabolic surface antenna reflector with the size of 2.5 m×1.9 m is designed.Under the condition that the original caliber,surface type,and interface remain unchanged,the main influence factors are designed and controlled.First,from the perspective of high stiffness,lightweight,and easy to form,a finite element simulation software is used to analyze and optimize the layout of the rib,the cross-sectional shape of the rib,the size of the rib,and the matching of the size and the coefficients of thermal expansion(CTEs)of the rib and the skin.Second,two structures are prepared by the autoclave molding process.Third,the weight and the surface precision root mean square(RMS)value are measured.The results show that the fundamental frequency of the SPR structure is 142.2 Hz,which is 3.5 Hz higher;the number of the new structural parts is reduced by 40%,and the forming process is greatly simplified.The total weight of the new structure is 11.9 kg,lighter 42.5%,indicating that the weight loss is obvious.The RMS value is 0.15 mm,which is slightly lower 0.01 mm but satisfies the accuracy requirement not greater than 0.3 mm.It is proved that the SPR structure reflector is a superior structure of the lightweight spaceborne antenna reflector.ZHOU Tao LIANG Yanmin WU Wenping WU Xinrui YE Zhoujun DONG Bin 2022上海航天(中英文)2022,39,1:1

20Anamorphic chaetosphaeriaceous fungi from China显示文摘Chaetosphaeriaceae is one of the largest families in Sordariomycetes with its members commonly found on decaying leaf,fruit,branch,bark and wood in both terrestrial and submerged environment in nature.This paper reports our research result of diversity,taxonomy and phylogeny of anamorphic Chaetosphaeriaceae in China,which is based on a systematic study with an integrated approach of morphological observation and phylogenetic analysis for a large collection(>1300 herbarium specimens and 1100 living strains).The family Chaetosphaeriaceae is expanded to accommodate 89 accepted genera,including 22 new genera and 10 newly assigned genera.Most of these genera(except for Chaetosphaeria and several other relatively large genera)are delimitated as monophyletic genera with well-defined diagnostic characters in morphology.The phylogenetic connection of non-phialidic Sporidesmium-like fungi is further confirmed and expanded to 10 different genera.The polyphyletic Codinaea/Dictyochaeta/Tainosphaeria complex is further resolved with a taxonomic framework of 28 monophyletic genera by redelimitation of Codinaea and Dictyochaeta with narrower concept,acceptance of the 16 established genera,and finally introduction of 10 new genera.Chloridium is phylogenetically redefined as monophyletic genus with narrower concept as typified by the type species,but a systematic review in both generic and species level is still needed.For biodiversity of chaetosphaeriaceous fungi,a total of 369 species in 76 genera,including 119 new species,47 new combinations,and one new name,are documented.The identification keys are provided for most genera,especially the large genera such as Codinaea s.str.,Codinaeella,Stilbochaeta,Cryptophiale,Thozetella,Dinemasporium and Pseudolachnella.In addition,ten known species were excluded from the family and reclassified.Systematic revision of several relatively large polyphyletic genera should be conducted in future studies,including Bahusutrabeeja,Ellisembia,Stanjehughesia,Cacumisporium,Chaetosphaeria,Chloridium,Craspedodidymum,Cryptophiale,Cryptophialoidea,Dictyochaetopsis,Minimidochium,and many published species of Codinaea and Dictyochaeta.Wenping Wu Yongzhao Diao 2022Fungal Diversity2022,,5:1
