1Paleoenvironmental implications of Holocene long-chain n-alkanes on the northern Bering Sea Slope显示文摘The records of high-resolution terrestrial biological markers(biomarkers) from Core B2-9 from the northern Bering Sea Slope over the last 9.6 ka BP were presented in the study.Variations in input of terrestrial long-chain nalkanes(referred to as n-alkanes) and vegetation structure in their source regions were investigated.The results show that the nC_(27) is the main carbon peak and has the greatest contribution rate of the total n-alkane content;this might be related to the abundance of woody plants and their spatial distribution in the source region.nC_(23) is another n-alkane having a relatively high content;this was mainly derived from submerged plants widespread along the coastal areas in the northern hemisphere.Total n-alkane content dropped quickly at ca.7.8 ka BP,ca.6.7 ka BP and ca.5.4 ka BP,and was followed by four relatively stable stages mostly controlled by sea-level rise,climate change and vegetation distribution in the source region.Variation in carbon preference index(CPI)indicates that the n-alkanes primarily originated from higher land plants,and the average chain length(ACL) and nC_(31)/nC_(27) ratio reveal the relatively stable presence of woody/herbaceous plants during the Holocene,and dominate woody plants in most of the time.Simultaneous variation in total n-alkane content,nC_(27) content and its contribution,CPI,ACL and nC_(31)/nC_(27) ratio over several short periods suggest that the growth rate of the woody plant n-alkane contribution was lower than that of herbaceous plants and fossil n-alkanes under the particular climatic conditions of the source region.ZHANG Haifeng WANG Rujian XIAO Wenshen 2017Acta Oceanologica Sinica2017,36,8:2
2Anti-Seizure and Neuronal Protective Effects of Irisin in Kainic Acid-Induced Chronic Epilepsy Model with Spontaneous Seizures显示文摘An increased level of reactive oxygen species is a key factor in neuronal apoptosis and epileptic seizures.Irisin reportedly attenuates the apoptosis and injury induced by oxidative stress.Therefore,we evaluated the effects of exogenous irisin in a kainic acid(KA)-induced chronic spontaneous epilepsy rat model.The results indicated that exogenous irisin significantly attenuated the KA-induced neuronal injury,learning and memory defects,and seizures.Irisin treatment also increased the levels of brain-derived neurotrophic factor(BDNF)and uncoupling protein 2(UCP2),which were initially reduced following KA administration.Furthermore,the specific inhibitor of UCP2(genipin)was administered to evaluate the possible protective mechanism of irisin.The reduced apoptosis,neurodegeneration,and spontaneous seizures in rats treated with irisin were significantly reversed by genipin administration.Our findings indicated that neuronal injury in KA-induced chronic epilepsy might be related to reduced levels of BDNF and UCP2.Moreover,our results confirmed the inhibition of neuronal injury and epileptic seizures by exogenous irisin.The protective effects of irisin may be mediated through the BDNF-mediated UCP2 level.Our results thus highlight irisin as a valuable therapeutic strategy against neuronal injury and epileptic seizures.Jie Yu Yao Cheng Yaru Cui Yujie Zhai Wenshen Zhang Mengdi Zhang Wenyu Xin Jia Liang Xiaohong Pan Qiaoyun Wang Hongliu Sun 2022Neuroscience Bulletin2022,38,11:1
3Ice rafting history and paleoceanographic reconstructions of Core 08P23 from southern Chukchi Plateau, western Arctic Ocean since Marine Isotope Stage 3显示文摘Multiproxy investigations have been performed on Core 08P23 collected from the Chukchi Plateau, the western Arctic Ocean, during the Third Chinese National Arctic Expedition. The core was dated back to Marine Isotope Stage(MIS) 3 by a combination of Accelerator Mass Spectrometric(AMS) carbon-14 dating and regional core correlation. A total of five prominent ice-rafted detritus(IRD) events were recognized in MIS 2 and MIS 3. The IRD sources in MIS 3 are originated from vast carbonate rock outcrops of the Canadian Arctic Archipelago and clastic quartz in MIS 2 may have a Eurasian origin. Most δ18O and δ13C values of Neogloboquadrina pachyderma(sinistral)(Nps) in Core 08P23 are lighter than the average values of surface sediments. The lighter δ18O and δ13C values of Nps in the two brown layers in MIS 1 and MIS 3 were resulted from meltwater events; and those in the gray layers in MIS 3 were caused by the enhanced sea ice formation. The δ18O values varied inversely with δ13C in MIS 2 indicate that the study area was covered by thick sea ice or ice sheet with low temperature and little meltwater, which prevented the biological productivity and sea-atmosphere exchange, as well as water mass ventilation. 7M2 macrophage-targeted iron oxide nanoparticles for magnetic resonance image-guided magnetic hyperthermia therapy显示文摘Tumor-associated macrophages(TAMs)play an important role in tumor development and progression.In particular,M2 TAMs can promote tumor growth by facilitating tumor progression and malignant behaviors.Selectively targeted elimination of M2 TAMs to inhibit tumor progression is of great significance for cancer treatment.Iron oxide nanoparticles based magnetic hyperthermia therapy(MHT)is a classical approach to destroy tumor tissue with deep penetration depth.In this study,we developed a typical M2 macrophage-targeted peptide(M2pep)functionalized superparamagnetic iron oxide nanoparticle(SPIO)for magnetic resonance imaging(MRI)-guided MHT in an orthotopic breast cancer mouse model.The obtained multifunctional SPIO-M2pep with a hydrodynamic diameter of 20 nm showed efficient targeting capability,high transverse relaxivity(149 mM^(-1) s^(-1))and satisfactory magnetic hyperthermia performance in vitro.In vivo studies demonstrated that the SPIO-M2pep based MRI can monitor the distribution of nanoparticles in tumor and indicate the suitable timing for MHT.The M2 macrophage-targeted MHT significantly reduced the tumor volume and the population of pro-tumoral M2 TAMs in tumor.In addition,the SPIO-M2pep based MHT can remodel the tumor immune microenvironment(TIME).The multifunctional SPIO-M2pep with M2 macrophage-targeting ability,high magnetic hyperthermia efficiency,MR imaging capability and effective role in remodeling the TIME hold great potential to improve clinical cancer therapy outcomes.Wenshen Wang Fenfen Li Shibo Li Yi Hu Mengran Xu Yuanyuan Zhang Muhammad Imran Khan Shaozhen Wang Min Wu Weiping Ding Bensheng Qiu 2021Journal of Materials Science & Technology2021,,22:0
8An electrochemical biosensor based on DNA“nano-bridge”for amplified detection of exosomal microRNAs显示文摘Exosomal miRNAs,as potential biomarkers in liquid biopsy for cancer early diagnosis,have aroused widespread concern.Herein,an electrochemical biosensor based on DNA“nano-bridge”was designed and applied to detect exosomal microRNA-21(miR-21)derived from breast cancer cells.In brief,the target miR-21 can specifically open the hairpin probe 1(HP1)labeled on the gold electrode(GE)surface through strand displacement reaction.Thus the exposed loop region of HP1 can act as an initiator sequence to activate the hybridization chain reaction(HCR)between two kinetically trapped hairpin probes:HP2 immobilized on the GE surface and biotin labeled HP3 in solution.Cascade HCR leads to the formation of DNA“nano-bridge”tethered to the GE surface with a great deal of“piers”.Upon addition of avidin-modified horseradish peroxidase(HRP),numerous HRP were bound to the formed“nano-bridge”through biotin-avidin interaction to arouse tremendous current signal.In theory,only a single miR-21 is able to trigger the continuous HCR between HP2 and HP3 until all of the HP2 are exhausted.Therefore the proposed biosensor achieved ultrahigh sensitivity toward miR-21 with the detection limit down to 168 amol/L,as well as little cross-hybridization even at the single-base-mismatched level.Successful attempts were also made in the detection of exosomal miR-21 obtained from the MCF-7 of breast cancer cell line.To our knowledge,this is the first attempt to built horizontal DNA nano-structure on the electrode surface for exosomal miRNAs detection.In a word,the high sensitivity,selectivity,low cost make the proposed method hold great potential application for early point-of-care(POC)diagnostics of cancer.Jing Zhang Meifeng Hou Guanyu Chen Huifang Mao Wenqian Chen Wenshen Wang Jinghua Chen 2021Chinese Chemical Letters2021,32,11:0
