1Characterization of VOC emissions from construction machinery and river ships in the Pearl River Delta of China显示文摘Speciated characterization of Volatile Organic Compounds (VOCs),including oxygenated VOCs (OVOCs),from construction machinery and river ships in China is currently lacking.In this regard,we conducted field measurement on speciated VOC (including OVOC) emissions from six construction machinery and five river ships in the Pearl River Delta (PRD) region to identify VOC emission characteristics.We noticed that OVOC emissions from construction machinery and ships accounted for more than 50%of the total VOC emissions,followed by alkenes,aromatics and alkanes.Formaldehyde and acetaldehyde were the most emission species,accounting for 61.8%-83.2%of OVOCs.For construction machinery,the fuel-based emission factors of roller,grader and pile driver were 3.12,3.12 and 7.36 g/kg,respectively.With the rigorous restraint by the national emission standards,VOC emissions of construction machinery had decreased considerably,especially during stageⅢ.Ozone formation potential was also significantly reduced due to the significant decrease in emissions of OVOCs and alkenes with higher reactivity.For river ships,the fuel-based emission factors of cargo ships and speedboat were 1.46 and 0.44 g/kg,respectively.VOC emissions from construction machinery and river ships in Guangdong Province in 2017 were 8851.0 and 4361.0 ton,respectively.This study filled the knowledge gaps of reactive gas emissions from different kinds of non-road mobile sources over the PRD,and more importantly,highlighted the necessity in adding OVOC measurement to give a complete and accurate depiction of reactive gas emissions from non-road mobile sources.Richao Wang Zibing Yuan Junyu Zheng Cheng Li Zhijiong Huang Wenshi Li Yan Xie Yiran Wang Kaiyang Yu Lejun Duan 2020Journal of Environmental Sciences2020,32,10:5

5Gedunin Degrades Aggregates of Mutant Huntingtin Protein and Intranuclear Inclusions via the Proteasomal Pathway in Neurons and Fibroblasts from Patients with Huntington’s Disease显示文摘Huntington's disease(HD) is a deadly neurodegenerative disease with abnormal expansion of CAG repeats in the huntingtin gene. Mutant Huntingtin protein(m HTT) forms abnormal aggregates and intranuclear inclusions in specific neurons, resulting in cell death. Here,we tested the ability of a natural heat-shock protein 90 inhibitor, Gedunin, to degrade transfected m HTT in Neuro-2 a cells and endogenous m HTT aggregates and intranuclear inclusions in both fibroblasts from HD patients and neurons derived from induced pluripotent stem cells from patients. Our data showed that Gedunin treatment degraded transfected m HTT in Neuro-2 a cells, endogenous m HTT aggregates and intranuclear inclusions in fibroblasts from HD patients, and in neurons derived from induced pluripotent stem cells from patients in a dose-and time-dependent manner, and its activity depended on the proteasomal pathway rather than the autophagy route. 6Chaos Criteria Design Based on Modified Sign Functions with One or Three-Threshold显示文摘The complexity measures of chaotic or periodic signals are perpetual topics of interest to data scientists. This work adheres to the framework of the traditional 0-1 test for chaos and replaces sine and cosine functions by modified sign functions. The compressive mapping rules chosen are one-threshold of three-value or three-threshold of five-value. In new criteria for chaos in forms of the 3s plot and Ks metric compared with 0-1 test results, the periodic state of data features a short beeline instead of a big ring in the pq plot and signs the nearest zero mark, while the chaotic state signs a simple curve instead of a random-walking shape in the pq plot, and shows the nearest one mark. 7Imbalance between nitrogen and potassium fertilization influences potassium deficiency symptoms in winter oilseed rape (Brassica napus L.) leaves显示文摘Chlorosis at leaf margins is a typical symptom of potassium(K) deficiency, but inappropriate application of K with other nutrients often masks symptoms of K deficiency. A two-year field experiment was conducted to measure the interactive effects of N and K on leaf photosynthesis and dry matter accumulation and the resulting growth dilution effect on K concentration and leaf K deficiency symptoms. N application aggravated the imbalance of N and K nutrients and further exacerbated K deficiency symptoms under K limitation. Synergistic effects of N and K promoted plant growth, amplified the growth dilution effect, and reduced the critical K concentration in leaves. Using 90% of the maximum shoot biomass as a threshold,the critical K concentration was 0.72% at the recommended N(N180) fertilization level. The critical K concentration increased by 62.5% owing to the reduced biomass under insufficient N(N;) supply. In contrast, high N(N;) reduced the critical K concentration(0.64%), accelerating chlorophyll decomposition and exacerbating K deficiency symptoms. The basis of changing the critical K concentration by magnifying growth dilution effect was the functional synergistic effect of N and K on photosynthetic characteristics. Under insufficient N, the low maximum carboxylation rate(V;) limited the net photosynthetic rate(An) and necessitated more K to maintain high CO;transmission capacity, to improve the total conductance g;/V;ratio. High N supply increased gtotand V;possibly mitigating the effect of K reduction on photosynthesis. 9A numerical simulation of latent heating within Typhoon Molave显示文摘The weather research and forecasting(WRF) model is a new generation mesoscale numerical model with a fine grid resolution(2 km), making it ideal to simulate the macro-and micro-physical processes and latent heating within Typhoon Molave(2009). Simulations based on a single-moment, six-class microphysical scheme are shown to be reasonable, following verification of results for the typhoon track, wind intensity, precipitation pattern, as well as inner-core thermodynamic and dynamic structures. After calculating latent heating rate, it is concluded that the total latent heat is mainly derived from condensation below the zero degree isotherm, and from deposition above this isotherm. It is revealed that cloud microphysical processes related to graupel are the most important contributors to the total latent heat. Other important latent heat contributors in the simulated Typhoon Molave are condensation of cloud water, deposition of cloud ice, deposition of snow, initiation of cloud ice crystals, deposition of graupel, accretion of cloud water by graupel, evaporation of cloud water and rainwater,sublimation of snow, sublimation of graupel, melting of graupel, and sublimation of cloud ice. 11Sea-Salt Aerosol Effects on the Simulated Microphysics and Precipitation in a Tropical Cyclone显示文摘We investigate the effects of sea-salt aerosol(SSA) activated as cloud condensation nuclei on the microphysical processes, precipitation, and thermodynamics of a tropical cyclone(TC). The Weather Research and Forecasting model coupled with Chemistry(WRF-Chem) was used together with a parameterization of SSA production. Three simulations, with different levels of SSA emission(CTL, LOW, HIGH), were conducted. The simulation results show that SSA contributes to the processes of autoconversion of cloud water and accretion of cloud water by rain,thereby promoting rain formation. The latent heat release increases with SSA emission, slightly increasing horizontal wind speeds of the TC. The presence of SSA also regulates the thermodynamic structure and precipitation of the TC.In the HIGH simulation, higher latent heat release gives rise to stronger updrafts in the TC eyewall area, leading to enhanced precipitation. In the LOW simulation, due to decreased latent heat release, the temperature in the TC eye is lower, enhancing the downdrafts in the region; and because of conservation of mass, updrafts in the eyewall also strengthen slightly; as a result, precipitation in the LOW experiment is a little higher than that in the CTL experiment.Overall, the relationship between the precipitation rate and SSA emission is nonlinear.Baolin JIANG Wenshi LIN Fangzhou LI Junwen CHEN 2019Journal of Meteorological Research2019,33,1:1

17Effects of local and non-local closure PBL schemes on the simulation of Super Typhoon Mangkhut(2018)显示文摘With the convection-permitting simulation of Super Typhoon Mangkhut(2018)with a 3 km resolution for 10.5 days using mesoscale numerical model,Weather Research and Forecasting Model Version 4.1(WRFV4.1),the influences of local closure QNSE planetary boundary layer(PBL)scheme and non-local closure GFS planetary boundary layer scheme on super typhoon Mangkhut are mainly discussed.It is found that in terms of either track or intensity of typhoon,the local closure QNSE scheme is better than the non-local closure GFS scheme.Local and non-local closure PBL schemes have a large influence on both the intensity and the structure of typhoon.The maximum intensity difference of the simulated typhoon is 50 hPa.The intensity of typhoon is closely related to its variations in structure.In the rapid intensification stage,the typhoon simulated by the QNSE scheme has a larger friction velocity,stronger surface latent heat flux,sensible heat flux and vapor flux,related to a higher boundary height and stronger vertical mixing.The latent heat flux and sensible heat flux on the surface conveyed energy upward for the typhoon while the water vapor was transported upward through vertical mixing.While the water vapor condensed,the latent heat was released,which further warmed the typhoon eyewall,strengthening the convection.The stronger winds also intensified the vertical mixing and the warm-core structure,further strengthened the typhoon.The differences in surface layer schemes dominated the differences between the two simulations.Zixi RUAN Jiangnan LI Fangzhou LI Wenshi LIN 2022Frontiers of Earth Science2022,16,2:0
18Plasma HGF and OPN as Potential Biomarkers of Pulmonary Arterial Hypertension in Congenital Heart Disease显示文摘Objectives:Pulmonary arterial hypertension in congenital heart disease(PAH-CHD)is the most common type of PAH and increases morbidity and mortality in patients with CHD.Right heart catheterization(RHC)is the standard method to diagnose PAH.However,RHC is an invasive and complicated method with relatively high cost.Noninvasive,feasible,and cost-efficient methods are urgently needed.The objective of this study was to evaluate three potential biomarkers of PAH-CHD:Hepatocyte growth factor(HGF),osteopontin(OPN),and suppression of tumorigenicity 2(ST2).Methods:Plasma samples were collected from patients with CHD(n=46)and healthy individuals(n=22)and divided into four groups according to the severity of PAH.The levels of HGF,OPN,and ST2 were then analyzed using an enzyme-linked immunosorbent assay.Correlations between HGF,OPN,ST2,and clinical parameters of PAH-CHD were analyzed.Results:The plasma HGF levels in the moderate to the severe group were significantly higher than those in the mild group,nonPAH group,and healthy control group(p<0.05).Derived from a receiver operating characteristic(ROC)curve analysis,a cut-off value of 356.75 ng/ml for the HGF concentration was able to predict PAH-CHD with 53%sensitivity and 89%specificity.The HGF level was positively related to pulmonary arterial systolic pressure(PASP)(r=0.36,p<0.05)and pulmonary vascular resistance(PVR)(r=0.36,p<0.05).Plasma OPN levels in the mild group were significantly higher than other groups and positively correlated with the pulmonary-systemic shunt ratio(Qp/Qs)(r=0.33,p<0.05).There was no statistically significant between-group difference in plasma soluble ST2(sST2)levels.Conclusion:The plasma HGF level was elevated in PAH-CHD patients and can be used as a potential biomarker.The plasma OPN level was positively correlated with the Qp/Qs.Dongdong Zheng Chi Shen Wenshi Liu Wenjing Lv Xiaofei Li 2021Congenital Heart Disease2021,16,4:0
19Ancient dormant virus remnant ERVW-1 drives ferroptosis via degradation of GPX4 and SLC3A2 in schizophrenia显示文摘Human endogenous retroviruses(HERVs)are remnants of retroviral infections in human germline cells from millions of years ago.Among these,ERVW-1(also known as HERV-W-ENV,ERVWE1,or ENVW)encodes the envelope protein of the HERV-W family,which contributes to the pathophysiology of schizophrenia.Additionally,neuropathological studies have revealed cell death and disruption of iron homeostasis in the brains of individuals with schizophrenia.Here,our bioinformatics analysis showed that differentially expressed genes in the human prefrontal cortex RNA microarray dataset(GSE53987)were mainly related to ferroptosis and its associated pathways.Clinical data demonstrated significantly lower expression levels of ferroptosis-related genes,particularly Glutathione peroxidase 4(GPX4)and solute carrier family 3 member 2(SLC3A2),in schizophrenia patients compared to normal controls.Further in-depth analyses revealed a significant negative correlation between ERVW-1 expression and the levels of GPX4/SLC3A2 in schizophrenia.Studies indicated that ERVW-1 increased iron levels,malondialdehyde(MDA),and transferrin receptor protein 1(TFR1)expression while decreasing glutathione(GSH)levels and triggering the loss of mitochondrial membrane potential,suggesting that ERVW-1 can induce ferroptosis.Ongoing research has shown that ERVW-1 reduced the expression of GPX4 and SLC3A2 by inhibiting their promoter activities.Moreover,Ferrostatin-1(Fer-1),the ferroptosis inhibitor,reversed the iron accumulation and mitochondrial membrane potential loss,as well as restored the expressions of ferroptosis markers GSH,MDA,and TFR1 induced by ERVW-1.In conclusion,ERVW-1 could promote ferroptosis by downregulating the expression of GPX4 and SLC3A2,revealing a novel mechanism by which ERVW-1 contributes to neuronal cell death in schizophrenia.Dongyan Zhang Xiulin Wu Xing Xue Wenshi Li Ping Zhou Zhao Lv Kexin Zhao Fan Zhu 2024Virologica Sinica2024,39,1:0
20Drug repurposing screen identifies vidofludimus calcium and pyrazofurin as novel chemical entities for the development of hepatitis E interventions显示文摘Hepatitis E virus(HEV)infection can cause severe complications and high mortality,particularly in pregnant women,organ transplant recipients,individuals with pre-existing liver disease and immunosuppressed patients.However,there are still unmet needs for treating chronic HEV infections.Herein,we screened a best-in-class drug repurposing library consisting of 262 drugs/compounds.Upon screening,we identified vidofludimus calcium and pyrazofurin as novel anti-HEV entities.Vidofludimus calcium is the next-generation dihydroorotate dehydrogenase(DHODH)inhibitor in the phase 3 pipeline to treat autoimmune diseases or SARS-CoV-2 infection.Pyrazofurin selectively targets uridine monophosphate synthetase(UMPS).Their anti-HEV effects were further investigated in a range of cell culture models and human liver organoids models with wild type HEV strains and ribavirin treatment failure-associated HEV strains.Encouragingly,both drugs exhibited a sizeable therapeutic window against HEV.For instance,the IC50 value of vidofludimus calcium is 4.6–7.6-fold lower than the current therapeutic doses in patients.Mechanistically,their anti-HEV mode of action depends on the blockage of pyrimidine synthesis.Notably,two drugs robustly inhibited ribavirin treatment failure-associated HEV mutants(Y1320H,G1634R).Their combination with IFN-αresulted in synergistic antiviral activity.In conclusion,we identified vidofludimus calcium and pyrazofurin as potent candidates for the treatment of HEV infections.Based on their antiviral potency,and also the favorable safety profile identified in clinical studies,our study supports the initiation of clinical studies to repurpose these drugs for treating chronic hepatitis E.Hongbo Guo Dan Liu Kuan Liu Yao Hou Chunyang Li Qiudi Li Xiaohui Ding Monique M.A.Verstegen Jikai Zhang Lingli Wang Yibo Ding Renxian Tang Xiucheng Pan Kuiyang Zheng Luc J.Wvan der Laan Qiuwei Pan Wenshi Wang 2024Virologica Sinica2024,39,1:0
