| 2 | NLRP3 Deficiency Attenuates Secondary Degeneration of Visual Cortical Neurons Following Optic Nerve Injury显示文摘In the visual pathway, optic nerve(ON) injury may cause secondary degeneration of neurons in distal regions, such as the visual cortex. However, the role of the neuroinflammatory response in regulating secondary impairment in the visual cortex after ON injury remains unclear. The NOD-like receptor family pyrin domain containing 3(NLRP3) is an important regulator of neuroinflammation. In this study, we established a mouse model of unilateral ON crush(ONC) and showed that the expression of NLRP3 was significantly increased in the primary visual cortex(V1) as a response to ONC and that the NLRP3 inflammasome was activated in the contralateral V1 1 days–14 days after ONC. Ablation of the NLRP3 gene significantly decreased the trans-neuronal degeneration within 14 days. Visual electrophysiological function was improved in NLRP3-/- mice. | 6 | A novel multistage anoxic/aerobic process with sludge regeneration zone (R-MAO) for advanced nitrogen removal from domestic sewage显示文摘To achieve advanced nitrogen removal from actual municipal sewage,a novel multistage anoxic/aerobic process with sludge regeneration zone(R-MAO)was developed.The reactor was used to treat actual domestic sewage and the nitrogen removal capacity of the sludge regeneration zone(R zone)was investigated during the long-term operation.The best performance was obtained at the R zone’s Oxidation-Reduction Potential(ORP)of-50±30 mV and hydraulic residence times(HRT)of 1.2 hr.The average effluent COD,TN,NH_(4)^(+)-N and NO_(3)^(−)-N of the R-MAO process were 18.0±2.3,7.5±0.6,1.0±0.5 and 4.6±0.4 mg/L,respectively,with the corresponding removal efficiency of COD,TN and NH_(4)^(+)-N were 92.9%±1.0%,84.1%±1.5% and 97.5%±1.1%.Compared to the sole MAO system,the TN removal efficiency of the R-MAO increased by 10.1%.Besides,under the optimal conditions,the contribution of the R zone in the R-MAO that removal COD,TN,NH_(4)^(+)-N and NO_(3)^(−)-N were 0.36,0.15,0.032 and 0.82 g/day.High-throughput sequencing results showed that uncultured_bacterium_f_Burkholderiaceae(5.20%),OLB8(1.04%)and Ottowia(1.03%)played an important role in denitrification in the R zone.This study provided effective guidance for the design and operation of the R-MAO process in domestic sewage treatment. | Lin Cao Rong Sun Wenyi Dong Hongjie Wang Zhongyi Dai Xue Wang Jin Xie Hua Li | 2023 | Journal of Environmental Sciences2023,,2: | 1 |
| 7 | Aggregation-induced emission enhancement of yellow photoluminescent carbon dots for highly selective detection of environmental and intracellular copper(Ⅱ) ions显示文摘Carbon dots (CDs) are prepared through a simple one-step hydrothermal treatment of o-phenylendiamine (OPD) and show yellow photoluminescent (PL) emission under the ultraviolet excitation, which can be further enhanced by Cu2+ ions owing to Cu2+ ions induced aggregation of OPD-CDs through the coordination of Cu2+ with amino groups on the surface of OPD-CDs. The aggregation induced emission enhancement (AIEE) property enables it feasible to develop a simple, sensitive and selective method to detect environmental and intracellular copper (Ⅱ) ions. The limit of detection as lowas 0.28 μmol/L (3σ/k) and a dynamic range from 0.5 μmol/L to 40 μmol/L make it veryeasy to detect the copper content inwater samples, such as river closure reservoir. | 13 | CCGD-ESCC: A Comprehensive Database for Genetic Variants Associated with Esophageal Squamous Cell Carcinoma in Chinese Population显示文摘Esophageal squamous-cell carcinoma(ESCC) is one of the most lethal malignancies in the world and occurs at particularly higher frequency in China. While several genome-wide association studies(GWAS) of germline variants and whole-genome or whole-exome sequencing studies of somatic mutations in ESCC have been published, there is no comprehensive database publically available for this cancer. Here, we developed the Chinese Cancer Genomic Database-Esophageal Squamous Cell Carcinoma(CCGD-ESCC) database, which contains the associations of 69,593 single nucleotide polymorphisms(SNPs) with ESCC risk in 2022 cases and 2039 controls, survival time of 1006 ESCC patients(survival GWAS) and gene expression(expression quantitative trait loci,eQTL) in 94 ESCC patients. Moreover, this database also provides the associations between8833 somatic mutations and survival time in 675 ESCC patients. Our user-friendly database is a resource useful for biologists and oncologists not only in identifying the associations of genetic variants or somatic mutations with the development and progression of ESCC but also in studying the underlying mechanisms for tumorigenesis of the cancer. | 14 | Amphiphilic peptide dendrimer-based nanovehicles for safe and effective siRNA delivery显示文摘Small interfering RNA(siRNA)-based RNA interference has emerged as a promising therapeutic strategy for the treatment of a wide range of incurable diseases.However,the safe and effective delivery of siRNA therapeutics into the interior of target cells remains challenging.Here,we disclosed novel amphiphilic peptide dendrimers(AmPDs)that composed of hydrophobic two lipid-like alkyl chains and hydrophilic poly(lysine)dendrons with different generations(2C18-KK2 and 2C18-KK2K4)as nanovehicles for siRNA delivery.These AmPDs are able to self-assemble into supramolecular nanoassemblies that are capable of entrapping siRNA molecules into nanoparticles to protect siRNA from enzymatic degradation and promote efficient intracellular uptake without evident toxicity.Interestingly,by virtue of the optimal balance of hydrophobic lipid-like entity and hydrophilic poly(lysine)dendron generations,AmPD 2C18-KK2K4 bearing bigger hydrophilic dendron can package siRNA to form stable,but more ready to disassemble complexes,thereby resulting in more efficient siRNA releasing and better gene silencing effect in comparison with AmPD 2C18-KK2 bearing smaller dendron.Additional studies confirmed that 2Ci8-KK2K4 can capitalize on the advantages of lipid and peptide dendrimer vectors for effective siRNA delivery.Collectively,our AmPD-based nanocarriers indeed represent a safe and effective siRNA delivery system.Our findings also provide a new perspective on the modulation of selfassembly amphiphilic peptide dendrimers for the functional and adaptive delivery of siRNA therapeutics. | Chi Ma Dandan Zhu Yu Chen Yiwen Dong Wenyi Lin Ning Li Wenjie Zhang Xiaoxuan Liu | 2020 | Biophysics Reports2020,6,6: | 1 |

| 16 | A novel FGFR1 inhibitor CYY292 suppresses tumor progression,invasion,and metastasis of glioblastoma by inhibiting the Akt/GSK3β/snail signaling axis显示文摘Glioblastoma(GBM)is a malignant brain tumor that grows quickly,spreads widely,and is resistant to treatment.Fibroblast growth factor receptor(FGFR)1 is a receptor tyrosine kinase that regulates cellular processes,including proliferation,survival,migration,and dif-ferentiation.FGFR1 was predominantly expressed in GBM tissues,and FGFR1 expression was negatively correlated with overall survival.We rationally designed a novel small molecule CYY292,which exhibited a strong affinity for the FGFR1 protein in GBM cell lines in vitro.CYY292 also exerted an effect on the conserved Ser777 residue of FGFR1.CYY292 dose-depen-dently inhibited cell proliferation,epithelial-mesenchymal transition,stemness,invasion,and migration in vitro by specifically targeting the FGFR1/AKT/Snail pathways in GBM cells,and this effect was prevented by pharmacological inhibitors and critical gene knockdown.In vivo experiments revealed that CYY292 inhibited U87MG tumor growth more effectively than AZD4547.CYY292 also efficiently reduced GBM cell proliferation and increased survival in orthotopic GBM models.This study further elucidates the function of FGFR1 in the GBM and reveals the effect of CYY292,which targets FGFR1,on downstream signaling pathways directly reducing GBM cell growth,invasion,and metastasis and thus impairing the recruitment,activation,and function of immune cells. | Yanran Bi Ruiling Zheng Jiahao Hu Ruiqing Shi Junfeng Shi Yutao Wang Peng Wang Wenyi Jiang Gyudong Kim Zhiguo Liu Xiaokun Li Li Lin | 2024 | Genes & Diseases2024,11,1: | 0 |
| 17 | Highly crystalline core dominated the catalytic performance of carbon dot for cyclohexane to adipic acid reaction显示文摘Carbon dots(CDs)have uniquely structural,physicochemical and photochemical properties,suggesting a promising platform for catalysis applications.The in-depth understanding of the structure-activity relationship in the CDs-based catalyst system needs to know the effect of the crystalline core on their catalytic performance.The efficient catalytic oxidation of cyclohexane is an urgent challenge in current chemical industry,in which,adipic acid(AA)plays an important role in industry for synthesis of nylon6 and nylon-66.Here,we fabricated the pristine CDs by electrochemical etching graphite rod method and derived CDs with high crystalline core(CD-600,CD-800,and CD-1100)through a thermal treatment method in tube furnace.Furthermore,these CDs performed an outstanding catalytic performance for one-step synthesis of AA from cyclohexane.With the help of machine learning(ML),the deep correlations between features(structures of CDs,catalytic conditions)and catalytic performances were investigated by XGBoost(XGB)model.Then under the optimization and prediction of XGB,it was found that high crystalline core preceded the other features and CD-1100 could get the best conversion of 30.696%and selectivity to AA of 92.52%at reaction conditions of 130℃,1.5 MPa,and 10 h.This work pioneered the application of ML in industrial issues and demonstrated a comprehensive understanding on CDs as catalyst to realize one-step synthesis of AA. | Xiao Wang Wenyi Bian Tianyang Zhang Yajie Zhao Mingwang Shao Haiping Lin Yang Liu Hui Huang Zhenhui Kang | 2022 | Nano Research2022,15,8: | 0 |

| 19 | Synthetic and immunological studies on the OCT4 immunodominant motif antigen-based anti-cancer vaccine显示文摘Objective:Cancer stem cell is one of the important causes of tumorigenesis as well as a drug target in the treatment of malignant tumor.However,at present,there is no immune vaccine targeting these cells.Octamer-binding transcription factor 4(OCT4),a marker of embryonic stem cells and germ cells,often highly expresses in the early stages of tumorigenesis and is therefore a good candidate for cancer vaccine development.Methods:To identify the optimal carrier and adjuvant combination,we chemically synthesized and linked three different OCT4 epitope antigens to a carrier protein,keyhole limpet hemocyanin(KLH),combined with Toll-like receptor 9 agonist(TLR9).Results:Immunization with OCT4-3+TLR9 produced the strongest immune response in mice.In prevention assays,significant tumor growth inhibition was achieved in BABL/c mice treated with OCT4-3+TLR9(P<0.01).Importantly,the results showed that cytotoxic T lymphocyte activity and the inhibition of tumor growth were enhanced in mice immunized with OCT4-3 combined with TLR9.Meanwhile,multiple cytokines[such as interferon(IFN)-γ(P<0.05),interleukin(IL)-12(P<0.05),IL-2(P<0.01),and IL-6(P<0.05)]promoting cellular immune responses were shown to be greatly enhanced in mice immunized with OCT4-3+TLR9.Moreover,we considered safety considerations in terms of the composition of the vaccines to help facilitate the development of effective next-generation vaccines.Conclusions:Collectively,these experiments demonstrated that combination therapy with TLR9 agonist induced a tumor-specific adaptive immune response,leading to the suppression of primary tumor growth in testis embryonic carcinoma. | Tingting Chen Kan Liu Jiangyao Xu Tianying Zhan Maixian Liu Li Li Zhiwen Yang Shuping Yuan Wenyi Zou Guimiao Lin Dennis ACarson Christina CNWu Xiaomei Wang | 2020 | Cancer Biology & Medicine2020,17,1: | 0 |
