| 1 | Reproductive management through integration of PGD and MPS-based noninvasive prenatal screening/diagnosis for a family with GJB2-associated hearing impairment显示文摘A couple with a proband child of GJB2(encoding the gap junction protein connexin 26)-associated hearing impairment and a previous pregnancy miscarriage sought for a reproductive solution to bear a healthy child. Our study aimed to develop a customized preconception-to-neonate care trajectory to fulfill this clinical demand by integrating preimplantation genetic diagnosis(PGD), noninvasive prenatal testing(NIPT), and noninvasive prenatal diagnosis(NIPD) into the strategy. Auditory and genetic diagnosis of the proband child was carried out to identify the disease causative mutations. The couple then received in-vitro-fertilization treatment, and eight embryos were obtained for day 5 biopsy. PGD was performed by short-tandem-repeat linkage analysis and Sanger sequencing of GJB2 gene. Transfer of a GJB2 c.235del C heterozygous embryo resulted in a singleton pregnancy. At the 13 th week of gestation, genomic DNA(g DNA) from the trio family and cell-free DNA(cf DNA) from maternal plasma were obtained for assessment of fetal chromosomal aneuploidy and GJB2 mutations. NIPT and NIPD showed the absence of chromosomal aneuploidy and GJB2-associated disease in the fetus, which was later confirmed by invasive procedures and postnatal genetic/auditory diagnosis. | 2 | A 2-kb Mycovirus Converts a Pathogenic Fungus into a Beneficial Endophyte for Brassica Protection and Yield Enhancement显示文摘Mycoviruses are viruses that infect fungi,and hypovirulence-associated mycoviruses have the potential to control fungal diseases.However,it is unclear how mycovirus-mediated hypovirulent strains live and survive in the field,and no mycovirus has been applied for field crop protection.In this study,we found that a previously identified small DNA mycovirus(SsHADV-1)can convert its host,Sclerotinia sclerotiorum,from a typical necrotrophic pathogen to a beneficial endophytic fungus.SsHADV-1 downregulates the expression of key pathogenicity factor genes in S.sclerotiorum during infection.When growing in rapeseed,the SsHADV-1-infected strain DT-8 significantly regulates the expression of rapeseed genes involved in defense,hormone signaling,and circadian rhythm pathways.As a result,plant growth is promoted and disease resistance is enhanced.Field experiments showed that spraying DT-8 at the early flowering stage can reduce the disease severity of rapeseed stem rot by 67.6%and improve yield by 14.9%.Moreover,we discovered that SsHADV-1 could also infect other S.sclerotiorum strains on DT-8-inoculated plants and that DT-8 could be recovered from dead plants.These findings suggest that the mycoviruses may have the ability to shape the origin of endophytism.Our discoveries suggest that mycoviruses may influence the origin of endophytism and may also offer a novel strategy for disease control in which mycovirus-infected strains are used to improve crop health and release mycoviruses into the field. | Hongxiang Zhang Jiatao Xie Yanping Fu Jiasen Cheng Zheng Qu Zhenzhen Zhao Shufen Cheng Tao Chen Bo Li Qianqian Wang Xinqiang Liu Binnian Tian David B.Collinge Daohong Jiang | 2020 | Molecular Plant2020,13,10: | 9 |
| 3 | Sources and Accumulation Risk of Heavy Metal in Major Animal Manure显示文摘Feed and corresponding manure samples were collected from 27 scale pig farms and 7 scale chicken farms in Jiaxing City,Zhejiang Province,and their heavy metal contents were analyzed. The weighted average values of Cr,Cu,Zn,Pb and As in pig feed were 4. 6,123. 4,370. 7,0. 69 and 6. 9 mg/kg,respectively. However,referred to Safe Use Specification of Feed Additives and MOA Announcement No. 1224,the exceeding standards rates of Cr,Cu,Zn,Pb and As in pig feed were 5. 36%,62. 5%,19. 64%,3. 57% and 37. 5%,respectively. The sequences of heavy metal content in pig feed successively were piglet > growing pig > big pig > sow,but the exceeding standard rates of heavy metal in feed were big pig > sow > growing pig > piglet,and the exceeding standard rate of big pig feed was more than 95%. The exceeding standard rates of Cu,Zn and As in chicken feed were 46. 7%,11. 1% and 33. 3% respectively. Based on Organic Fertilizer( NY525-2012) and Sludge Agricultural Pollution Control Standard( GB4284-84),Cu,Zn and As in pig manure and chicken manure exceeded the standard; the exceeding standard rates of pig manure were 71. 9%,78. 9% and 42. 1%,while that of chicken manure were 12. 5%,25. 0% and 44. 4%,respectively. The exceeding standard rates of Cu and Zn in piglet manure reached 100%. There were very significant positive correlations in Cu,Zn and As between pig feed and corresponding manure,or Hg,Cu and As between chicken feed and corresponding manure,and the correlation coefficients r^2 were greater than 0. 8. | 5 | Development of Cu foam-based Ni catalyst for solar thermal reforming of methane with carbon dioxide显示文摘Using solar energy to produce syngas via the endothermic reforming of methane has been extensively investigated at the laboratory- and pilot plant-scales as a promising method of storing solar energy. One of the challenges to scaling up this process in a tubular reformer is to improve the reactor's performance, which is limited by mass and heat transfer issues. High thermal conductivity Cu foam was therefore used as a substrate to improve the catalyst's thermal conductivity during solar reforming. We also developed a method to coat the foam with the catalytically active component NiMg_3AlO_x. The Cu foam-based NiMg_3AlO_x performs better than catalysts supported on SiSiC foam, which is currently used as a substrate for solar-reforming catalysts, at high gas hourly space velocity(≥400,000 mL/(g·h)) or at low reaction temperatures(≤ 720 °C). | 10 | SEM in-situ investigation on fatigue cracking behavior of P/M Rene95 alloy with surface inclusions显示文摘The low-cycle fatigue behavior of powder metallurgy Rene95 alloy containing surface inclusions was investigated by in-situ observation with scanning electron microscopy (SEM). The process of fatigue crack initiation and early stage of propagation behavior indicates that fatigue crack mainly occurs at the interface between the inclusion and the matrix. The effect of inclusion on the fatigue crack initiation and the early stage of crack growth was very obvious. The fatigue crack growth path in the matrix is similar to the shape of inclusion made on the basis of fatigue fracture image analysis. The empiric relation between the surface and inside crack growth length, near a surface inclusion, can be expressed. Therefore, the fatigue crack growth rate or life of P/M Rene95 alloy including the inclusions can be evaluated on the basis of the measurable surface crack length parameter. | 17 | Scriptaid affects histone acetylation and the expression of development-related genes at different stages of porcine somatic cell nuclear transfer embryo during early development显示文摘Although the somatic cell nuclear transfer(SCNT) technique has been used extensively for cloning and generating transgenic pigs,the cloning efficiency is still very low.It has been proposed that the low efficiency of this technique is the result of incomplete epigenetic reprogramming and abnormal gene expression during early embryonic development.In this study,we investigate the effect of Scriptaid,a low-toxicity histone deacetylase inhibitor,on the developmental competence of porcine SCNT embryos.We found that treating SCNT embryos with 500 nmol/L Scriptaid for 15 h after activation significantly enhanced the blastocyst formation rate(27.7%) compared with the untreated group(control)(12.2%,P<0.05).Using an immunofluorescence technique to measure the average fluorescence intensity,we also found that treating SCNT embryos with Scriptaid increased the level of histone acetylation on histone H3 at lysine 14(acH3K14).Furthermore,treating embryos with Scriptaid increased the expression level of three genes that play important roles during embryonic development(Oct4,Klf4 at the blastocyst stage and Nanog at the 4-cell stage).Moreover,the expression level of the apoptosis-related gene Caspase-3 was significantly lower in the Scriptaid-treated SCNT embryos compared with the control SCNT embryos at the 4-cell and blastocyst stages.In conclusion,these results indicate that Scriptaid treatment improves the development and nuclear reprogramming of porcine SCNT embryos. | ZHOU Yan HUANG YongYe XIE WanHua SONG Qi JI Yuan ZHANG YanPing OUYANG HongSheng LAI LiangXue PANG DaXin TANG XiaoChun | 2013 | Chinese Science Bulletin2013,58,18: | 1 |
