| 3 | mTERF5 Acts as a Transcriptional Pausing Factor to Positively Regulate Transcription of Chloroplast psbEFLJ显示文摘RNA polymerase transcriptional pausing represents a major checkpoint in transcription in bacteria and metazoans,but it is unknown whether this phenomenon occurs in plant organelles.Here,we report that transcriptional pausing occurs in chloroplasts.We found that mTERF5 specifically and positively regulates the transcription of chloroplast psbEFLJ in Arabidopsis thaliana that encodes four key subunits of photosystem II.We found that mTERF5 causes the plastid-encoded RNA polymerase(PEP)complex to pause at psbEFLJ by binding to the+30 to+51 region of double-stranded DNA.Moreover,we revealed that mTERF5 interacts with pTAC6,an essential subunit of the PEP complex,although pTAC6 is not involved in the transcriptional pausing at psbEFLJ.We showed that mTERF5 recruits additional pTAC6 to the transcriptionally paused region of psbEFLJ,and the recruited pTAC6 proteins could be assembled into the PEP complex to regulate psbEFLJ transcription.Taken together,our findings shed light on the role of transcriptional pausing in chloroplast transcription in plants. | Shunhua Ding Yi Zhang Zhi Hu Xiahe Huang Bohan Zhang Qingtao Lu Xiaogang Wen Yingchun Wang Congming Lu | 2019 | Molecular Plant2019,12,9: | 6 |
| 4 | Modulation of nitrate-induced phosphate response by the MYB transcription factor RLI1/HINGE1 in the nucleus显示文摘The coordinated utilization of nitrogen(N)and phosphorus(P)is vital for plants to maintain nutrient balance and achieve optimal growth.Previously,we revealed a mechanism by which nitrate induces genes for phosphate utilization;this mechanism depends on NRT1.1B-facilitated degradation of cytoplasmic SPX4,which in turn promotes cytoplasmic-nuclear shuttling of PHR2,the central transcription factor of phosphate signaling,and triggers the nitrate-induced phosphate response(NIPR)and N-P coordinated utilization in rice.In this study,we unveiled a fine-tuning mechanism of NIPR in the nucleus regulated by Highly Induced by Nitrate Gene 1(HINGE1,also known as RLI1),a MYB-transcription factor closely related to PHR2.RLI1/HINGE1,which is transcriptionally activated by PHR2 under nitrate induction,can directly activate the expression of phosphate starvation-induced genes.More importantly,RLI1/HINGE1 competes with PHR2 for binding to its repressor proteins in the nucleus(SPX proteins),and consequently releases PHR2 to further enhance phosphate response.Therefore,RLI1/HINGE1 amplifies the phosphate response in the nucleus downstream of the cytoplasmic SPX4-PHR2 cascade,thereby enabling fine-tuning of N-P balance when nitrate supply is sufficient. | Zhihua Zhang Zhao Li Wei Wang Zhimin Jiang Liping Guo Xiaohan Wang Yangwen Qian Xiahe Huang Yongqiang Liu Xiujie Liu Yahong Liu Aifu Li Yu Yan Junpeng Xie Shouyun Cao Stanislav Kopriva Legong Li Fanjiang Kong Baohui Liu Yingchun Wang Bin Hu Chengcai Chu | 2021 | Molecular Plant2021,14,3: | 4 |
| 5 | Microstructure induced galvanic corrosion evolution of SAC305 solder alloys in simulated marine atmosphere显示文摘Motivated by the increasing use of Sn-3.0 Ag-0.5 Cu(SAC305)solder in electronics worked in marine atmospheric environment and the uneven distribution of Ag3Sn and Cu6Sn5 intermetallic compounds(IMCs)inβ-Sn matrix,comb-like electrodes have been designed for in-situ EIS measurements to study the microstructure induced galvanic corrosion evolution of SAC305 solder in simulated marine atmosphere with high-temperature and high-humidity.Results indicate that in-situ EIS measurement by comb-like electrodes is an effective method for corrosion evolution behavior study of SAC305 solder.Besides,the galvanic effect between Ag3Sn IMCs andβ-Sn matrix can aggravate the corrosion of both as-received and furnace-cooled SAC305 solder as the exposure time proceeds in spite of the presence of corrosion product layer.Pitting corrosion can be preferentially found on furnace-cooled SAC305 with larger Ag3Sn grain size.Moreover,the generated inner stress during phases transformation process with Sn3O(OH)2Cl2 as an intermediate and the possible hydrogen evolution at local acidified sites are supposed to be responsible for the loose,porous,cracked,and non-adherent corrosion product layer.These findings clearly demonstrate the corrosion acceleration behavior and mechanism of SAC305 solder,and provide potential guidelines on maintenance of microelectronic devices for safe operation and longer in-service duration. | Mingna Wang Chuang Qiao Xiaolin Jiang Long Hao Xiahe Liu | 2020 | Journal of Materials Science & Technology2020,47,16: | 4 |
| 6 | Temperature and NaCl deposition dependent corrosion of SAC305 solder alloy in simulated marine atmosphere显示文摘The stable operation of electronic devices in marine atmospheric environment is affected by the corrosion deterioration of solder joints,and the effects by atmosphere temperature and chloride deposition are critical.In this work,NaCl deposition and temperature dependent corrosion of Pb-free SAC305 solder in simulated marine atmosphere has been investigated.The results indicate that higher NaCl deposition prolongs the surface wetting time and leads to the final thicker saturated electrolyte film for further corrosion.Higher temperature accelerates the evaporation and contributes to the final thinner saturated NaCl electrolyte film.Besides,the corrosion control process varies under the initially covered thicker NaCl electrolyte layer and under the final saturated much thinner NaCl electrolyte film as the evaporation proceeds.Moreover,the ready oxygen availability through the final thinner saturated NaCl electrolyte film facilitates the formation of corrosion product layer mainly of electrochemically stable SnO2,but higher temperature leads to the final corrosion product layer with smaller crystal size and large cracks.The findings clearly demonstrate the effects of NaCl deposition and temperature on corrosion evolution of SAC305 solder joints and are critical to the daily maintenance of electronic devices for longer service life in marine atmosphere. | Chuang Qiao Mingna Wang Long Hao Xiahe Liu Xiaolin Jiang Xizhong An Duanyang Li | 2021 | Journal of Materials Science & Technology2021,,16: | 3 |
| 7 | Controlling Effects of Tight Reservoir Micropore Structures on Seepage Ability: A Case Study of the Upper Paleozoic of the Eastern Ordos Basin, China显示文摘In this study, the types of micropores in a reservoir are analyzed using casting thin section(CTS) observation and scanning electron microscopy(SEM) experiments. The high-pressure mercury injection(HPMI) and constant-rate mercury injection(CRMI) experiments are performed to study the micropore structure of the reservoir. Nuclear magnetic resonance(NMR), gas-water relative seepage, and gas-water two-phase displacement studies are performed to examine the seepage ability and parameters of the reservoir, and further analyses are done to confirm the controlling effects of reservoir micropore structures on seepage ability. The experimental results show that Benxi, Taiyuan, Shanxi, and Shihezi formations in the study area are typical ultra-low porosity and ultra-low permeability reservoirs. Owing to compaction and later diagenetic transformation, they contain few primary pores. Secondary pores are the main pore types of reservoirs in the study area. Six main types of secondary pores are: intergranular dissolved pores, intragranular dissolved pores, lithic dissolved pores, intercrystalline dissolved pores, micropores, and microfracture. The results show that reservoirs with small pore-throat radius, medium displacement pressure, and large differences in pore-throat structures are present in the study area. The four types of micropore structures observed are: lower displacement pressure and fine pores with medium-fine throats, low displacement pressure and fine micropores with fine throats, medium displacement pressure and micropores with micro-fine throats, and high displacement pressure and micropores with micro throats. The micropore structure is complex, and the reservoir seepage ability is poor in the study areas. The movable fluid saturation, range of the gas-water two-phase seepage zone, and displacement types are the three parameters that well represent the reservoir seepage ability. According to the characteristic parameters of microscopic pore structure and seepage characteristics, the reservoirs in the study area are classified into four types(Ⅰ–Ⅳ), and types Ⅰ, Ⅱ, and Ⅲ are the main types observed. | 20 | An engineered platform for reconstituting functional multisubunit SCF E3 ligase in vitro显示文摘Multisubunit SKP1/Cullin1/F-box(SCF)E3 ligases play essential roles in regulating the stability of crucial regulatory factors and controlling growth and development in eukaryotes.Detecting E3 ligase activity in vitro is important forexploring the molecular mechanism of protein ubiquitination.However,in vitro ubiquitination assay systems for multisubunit E3 ligases remain difficult to achieve,especially in plants,mainly owing to difficulties in achieving active components of multisubunit E3 ligases with high purity and characterizing specific E2 and E3 pairs.In this study,we characterized components of the rice ScFDiwARF3(SCFDs)E3 ligase,screened the coordinated E2,and reconstituted active ScFD3 E3 ligase in vitro.We further engineered SCFD3 E3 ligase using a fused SKP1-Cullin1-RBX1(eSCR)protein and found that both the wild-type SCFD3 E3 ligase and the engineered SCFD3 E3 ligase catalyzed ubiquitination of the substrate D53,which is the key transcriptional repressor in strigolactone signaling.Finally,we replaced D3 with other F-box proteins from rice and humans and reconstituted active escF E3 ligases,including escFaID2,escFBxL1s,and escFcDC4 E3 ligases.Our work reconstitutes functional SCF E3 ligases in vitro and generates an engineered system with interchangeable F-box proteins,providing a powerful platform for studying the mechanisms of multisubunit SCF E3 ligases in eukaryotes. | Huihui Liu Simiao Liu Hong Yu Xiahe Huang Yingchun Wang Liang Jiang Xiangbing Meng Guifu Liu Mingjiang Chen Yanhui Jing Feifei Yui Bing Wang Jiayang Li | 2022 | Molecular Plant2022,15,8: | 1 |
