1Micro-droplet Digital Polymerase Chain Reaction and Real-Time Quantitative Polymerase Chain Reaction Technologies Provide Highly Sensitive and Accurate Detection of Zika Virus显示文摘The establishment of highly sensitive diagnostic methods is critical in the early diagnosis and control of Zika virus(ZIKV)and in preventing serious neurological complications of ZIKV infection. In this study, we established micro-droplet digital polymerase chain reaction(ddPCR) and real-time quantitative PCR(RT-qPCR) protocols for the detection of ZIKV based on the amplification of the NS5 gene. For the ZIKV standard plasmid, the RT-qPCR results showed that the cycle threshold(Ct) value was linear from 10~1 to 10~8 copy/l L, with a standard curve R^2 of 0.999 and amplification efficiency of 92.203%;however, a concentration as low as 1 copy/l L could not be detected. In comparison with RT-qPCR, the dd PCR method resulted in a linear range of 10~1–10~4 copy/l L and was able to detect concentrations as low as 1 copy/l L. Thus, for detecting ZIKV from clinical samples, RT-qPCR is a better choice for high-concentration samples(above 10~1 copy/l L),while ddPCR has excellent accuracy and sensitivity for low-concentration samples. 2Chromosome-scale assembly of the Dendrobium chrysotoxum genome enhances the understanding of orchid evolution显示文摘As one of the largest families of angiosperms,the Orchidaceae family is diverse.Dendrobium represents the second largest genus of the Orchidaceae.However,an assembled high-quality genome of species in this genus is lacking.Here,we report a chromosome-scale reference genome of Dendrobium chrysotoxum,an important ornamental and medicinal orchid species.The assembled genome size of D.chrysotoxum was 1.37 Gb,with a contig N50 value of 1.54 Mb.Of the sequences,95.75%were anchored to 19 pseudochromosomes.There were 30,044 genes predicted in the D.chrysotoxum genome.Two whole-genome polyploidization events occurred in D.chrysotoxum.In terms of the second event,whole-genome duplication(WGD)was also found to have occurred in other Orchidaceae members,which diverged mainly via gene loss immediately after the WGD event occurred;the first duplication was found to have occurred in most monocots(tau event).We identified sugar transporter(SWEET)gene family expansion,which might be related to the abundant medicinal compounds and fleshy stems of D.chrysotoxum.MADS-box genes were identified in D.chrysotoxum,as well as members of TPS and Hsp90 gene families,which are associated with resistance,which may contribute to the adaptive evolution of orchids.We also investigated the interplay among carotenoid,ABA,and ethylene biosynthesis in D.chrysotoxum to elucidate the regulatory mechanisms of the short flowering period of orchids with yellow flowers.The reference D.chrysotoxum genome will provide important insights for further research on medicinal active ingredients and breeding and enhances the understanding of orchid evolution.Yongxia Zhang Guo-Qiang Zhang Diyang Zhang Xue-Die Liu Xin-Yu Xu Wei-Hong Sun Xia Yu Xiaoen Zhu Zhi-Wen Wang Xiang Zhao Wen-Ying Zhong Hongfeng Chen Wei-Lun Yin Tengbo Huang Shan-Ce Niu Zhong-Jian Liu 2021Horticulture Research2021,8,1:3

5An adaptive region growing algorithm for breast masses in mammograms显示文摘This study attempted to accurately segment the mammographic masses and distinguish malignant from benign tumors.An adaptive region growing algorithm with hybrid assessment function combined with maximum likelihood analysis and maximum gradient analysis was developed in this paper.In order to accommodate different situations of masses,the likelihood and the edge gradients of segmented masses were weighted adaptively by the use of information entropy.106 benign and 110 malignant tumors were included in this study.We found that the proposed algorithm obtained segmentation contour more accurately and delineated the tumor body as well as tumor peripheral regions covering typical mass boundaries and some spiculation patterns.Then the segmented results were evaluated by the classification accuracy.42 features including age,intensity,shape and texture were extracted from each segmented mass and support vector machine(SVM)was used as a classifier.The classification accuracy was evaluated using the area(A_(z))under the receiver operating characteristic(ROC)curve.It was found that the maximum likelihood analysis achieved an A_(z)value of 0.835,the maximum gradient analysis got an A_(z)value of 0.932 and the hybrid assessment function performed the best classification result where the value of A_(z)was 0.948.In addition,compared with traditional region growing algorithm,our proposed algorithm is more adaptive and provides a better performance for future works.Ying CAO Xin HAO Xiaoen ZHU Shunren XIA 2010Frontiers of Electrical and Electronic Engineering in China2010,5,2:1

13Tempospacial energy-saving effect-based diagnosis in large coal-fired power units:Energy-saving benchmark state显示文摘The energy-saving analytics of coal-fired power units in China is confronting new challenges especially with even more complicated system structure, higher working medium parameters, time-dependent varying operation conditions and boundaries such as load rate, coal quality, ambient temperature and humidity. Compared with the traditional optimization of specific operating parameters, the idea of the energy-consumption benchmark state was proposed. The equivalent specific fuel consumption(ESFC) analytics was introduced to determine the energy-consumption benchmark state, with the minimum ESFC under varying operation boundaries. Models for the energy-consumption benchmark state were established, and the endogenous additional specific consumption(ASFC) and exogenous ASFC were calculated. By comparing the benchmark state with the actual state, the energy-saving tempospacial effect can be quantified. As a case study, the energy consumption model of a 1000 MW ultra supercritical power unit was built. The results show that system energy consumption can be mainly reduced by improving the performance of turbine subsystem. This nearly doubles the resultant by improving the boiler system. The energy saving effect of each component increases with the decrease of load and has a greater influence under a lower load rate. The heat and mass transfer process takes priority in energy saving diagnosis of related components and processes. 14simple and effective aerosol pathogen disinfection test for a flowing air disinfector显示文摘Aerosol transmission is an important disease transmission route and has been especially pertinent to hospital and biosafety laboratories during the SARS-CoV-2 pandemic.The thermal resistance of airborne SARS-CoV-2 is lower than that of Bacillus subtilis spores,which are often used to test the effectiveness of SARS-CoV-2 and other pathogen disinfection methods.Herein,we propose a new method to test the disinfection ability of a flowing air disinfector(a digital electromagnetic induction air heater)using B.subtilis spores.The study provides an alternative air disinfection test method.The new test system combined an aerosol generator and a respiratory filter designed in-house and could effectively recover spores on the filter membrane at the air outlet after passing through the flowing air disinfector.The total number of bacterial spores used in the test was within the range of 5×10^(5)–5×10^(6)colony-forming units(CFUs)specified in the technical standard for disinfection.The calculation was based on the calculation method in Air Disinfection Effect Appraisal Test in Technical Standard for Disinfection(2002 Edition).At an air speed of 3.5 m/s,we used a digital electromagnetic induction air heater to disinfect flowing air containing 4.100×10^(6)CFUs of B.subtilis spores and determined that the minimum disinfection temperature was 350℃for a killing rate of 99.99%.At 400℃,additional experiments using higher spore concentrations(4.700×10^(6)±1.871×10^(5)CFU)and a higher airspeed(4 m/s)showed that the killing rate remained>99.99%.B.subtilis spores,as a biological indicator for testing the efficiency of dry-heat sterilization,were killed by the high temperatures used in this system.The proposed method used to test the flowing air disinfector is simple,stable,and effective.This study provides a reference for the development of test systems that can assess the disinfection ability of flowing air disinfectors.Xuling Liu Zhiran Qin Linqing Wang Xiaoting Xie Yifang Fu Jianhai Yu Zuxin Liang Xiaoen He Jingshu Li Hong Dai Jinxiu Yao Qinghua Wu Weiwei Xiao Li Zhu Chengsong Wan Bao Zhang Wei Zhao 2023Journal of Biosafety and Biosecurity2023,5,1:0
