| 1 | Essential roles of exosome and circRNA_101093 on ferroptosis desensitization in lung adenocarcinoma显示文摘Background:Resistance to ferroptosis,a regulated cell death caused by irondependent excessive accumulation of lipid peroxides,has recently been linked to lung adenocarcinoma(LUAD).Intracellular antioxidant systems are required for protection against ferroptosis.The purpose of the present studywas to investigate whether and how extracellular system desensitizes LUAD cells to ferroptosis.Methods:Established human lung fibroblasts MRC-5,WI38,and human LUAD H1650,PC9,H1975,H358,A549,and H1299 cell lines,tumor and matched normal adjacent tissues of LUAD,and plasma from healthy individuals and LUAD patients were used in this study.Immunohistochemistry and immunoblotting were used to analyze protein expression,and quantitative reverse transcription-PCR was used to analyze mRNA expression.Cell viability,cell death,and the lipid reactive oxygen species generationwere measured to evaluate the responses to ferroptosis.Exosomes were observed using transmission electron microscope.The localization of arachidonic acid(AA)was detected using click chemistry labeling followed by confocal microscopy.Interactions between RNAs and proteins were detected using RNA pull-down,RNA immunoprecipitation and photoactivatable ribonucleoside-enhanced crosslinking and immunoprecipitation methods.Proteomic analysis was used to investigate RNA-regulated proteins,and metabolomic analysis was performed to analyze metabolites.Cellderived xenograft,patient-derived xenograft,cell-implanted intrapulmonary LUAD mouse models and plasma/tissue specimens from LUAD patients were used to validate the molecular mechanism.Results:Plasma exosome from LUAD patients specifically reduced lipid peroxidation and desensitized LUAD cells to ferroptosis.A potential explanation is that exosomal circRNA_101093(cir93)maintained an elevation in intracellular cir93 in LUAD to modulate AA,a poly-unsaturated fatty acid critical for ferroptosisassociated increased peroxidation in the plasma membrane.Mechanistically,cir93 interacted with and increased fatty acid-binding protein 3(FABP3),which transported AA and facilitated its reaction with taurine.Thus,global AA was reduced,whereas N-arachidonoyl taurine(NAT,the product of AA and taurine)was induced.Notably,the role of NAT in suppressing AA incorporation into the plasma membrane was also revealed.In pre-clinical in vivo models,reducing exosome improved ferroptosis-based treatment.Conclusion:Exosome and cir93 are essential for desensitizing LUAD cells to ferroptosis,and blocking exosome may be helpful for future LUAD treatment. | Xiao Zhang Yunhua Xu Lifang Ma Keke Yu Yongjie Niu Xin Xu Yi Shi Susu Guo Xiangfei Xue Yikun Wang Shiyu Qiu Jiangtao Cui Hong Wang Xiaoting Tian Yayou Miao Fanyu Meng Yongxia Qiao Yongchun Yu Jiayi Wang | 2022 | Cancer Communications2022,42,4: | 10 |

| 3 | Serum semaphorin 4C as a diagnostic biomarker in breast cancer:A multicenter retrospective study显示文摘Background:To date,there is no approved blood-based biomarker for breast cancer detection.Herein,we aimed to assess semaphorin 4C(SEMA4C),a pivotal protein involved in breast cancer progression,as a serum diagnostic biomarker.Methods:We included 6,213 consecutive inpatients from Tongji Hospital,Qilu Hospital,and Hubei Cancer Hospital.Training cohort and two validation cohorts were introduced for diagnostic exploration and validation.A pan-cancer cohort was used to independently explore the diagnostic potential of SEMA4C among solid tumors.Breast cancer patients who underwent mass excision prior to modified radical mastectomy were also analyzed.We hypothesized that increased pretreatment serum SEMA4C levels,measured using optimized in-house enzymelinked immunosorbent assay kits,could detect breast cancer.The endpoints were diagnostic performance,including area under the receiver operating characteristic curve(AUC),sensitivity,and specificity.Post-surgery pathological diagnosis was the reference standard and breast cancer staging followed the TNM classification.There was no restriction on disease stage for eligibilities.Results:We included 2667 inpatients with breast lesions,2378 patients with other solid tumors,and 1168 healthy participants.Specifically,118 patients with breast cancer were diagnosed with stage 0(5.71%),620 with stage I(30.00%),966 with stage II(46.73%),217 with stage III(10.50%),and 8 with stage IV(0.39%).Patients with breast cancer had significantly higher serum SEMA4C levels than benign breast tumor patients and normal controls(P<0.001).Elevated serum SEMA4C levels had AUC of 0.920(95%confidence interval[CI]:0.900–0.941)and 0.932(95%CI:0.911–0.953)for breast cancer detection in the two validation cohorts.The AUCs for detecting early-stage breast cancer(n=366)and ductal carcinoma in situ(n=85)were 0.931(95%CI:0.916–0.946)and 0.879(95%CI:0.832–0.925),respectively.Serum SEMA4C levels significantly decreased after surgery,and the reduction was more striking after modified radical mastectomy,compared with mass excision(P<0.001).The positive rate of enhanced serum SEMA4C levels was 84.77%for breast cancer and below 20.75%for the other 14 solid tumors.Conclusions:Serum SEMA4C demonstrated promising potential as a candidate biomarker for breast cancer diagnosis.However,validation in prospective settings and by other study groups is warranted. | Ya Wang Long Qiao Jie Yang Xiong Li Yaqi Duan Jiahao Liu Shaoqi Chen Huayi Li Dan Liu Tian Fang Jingjing Ma Xiaoting Li Fei Ye Junxiang Wan Juncheng Wei Qin Xu Ensong Guo Ping Jin Mingfu Wu Lin Zhang Yun Xia Yaqun Wu Jun Shao Yaojun Feng Qing Zhang Zongyuan Yang Gang Chen Qinghua Zhang Xingrui Li Shixuan Wang Junbo Hu Xiaoyun Wang Mona P.Tan Kazuaki Takabe Beihua Kong Qifeng Yang Ding Ma Qinglei Gao | 2021 | Cancer Communications2021,41,12: | 3 |

| 12 | Targeting Kindlin-2 in adipocytes increases bone mass through inhibiting FAS/PPARγ/FABP4 signaling in mice显示文摘Osteoporosis(OP)is a systemic skeletal disease that primarily affects the elderly population,which greatly increases the risk of fractures.Here we report that Kindlin-2 expression in adipose tissue increases during aging and high-fat diet fed and is accompanied by decreased bone mass.Kindlin-2 specific deletion(K2KO)controlled by Adipoq-Cre mice or adipose tissue-targeting AAV(AAV-Rec2-CasRx-sgK2)significantly increases bone mass.Mechanistically,Kindlin-2 promotes peroxisome proliferator-activated receptor gamma(PPARγ)activation and downstream fatty acid binding protein 4(FABP4)expression through stabilizing fatty acid synthase(FAS),and increased FABP4 inhibits insulin expression and decreases bone mass.Kindlin-2 inhibition results in accelerated FAS degradation,decreased PPARγactivation and FABP4 expression,and therefore increased insulin expression and bone mass.Interestingly,we find that FABP4 is increased while insulin is decreased in serum of OP patients.Increased FABP4 expression through PPARγactivation by rosiglitazone reverses the high bone mass phenotype of K2KO mice.Inhibition of FAS by C75 phenocopies the high bone mass phenotype of K2KO mice.Collectively,our study establishes a novel Kindlin-2/FAS/PPARγ/FABP4/insulin axis in adipose tissue modulating bone mass and strongly indicates that FAS and Kindlin-2 are new potential targets and C75 or AAV-Rec2-CasRx-sgK2 treatment are potential strategies for OP treatment. | Wanze Tang Zhen Ding Huanqing Gao Qinnan Yan Jingping Liu Yingying Han Xiaoting Hou Zhengwei Liu Litong Chen Dazhi Yang Guixing Ma Huiling Cao | 2023 | Acta Pharmaceutica Sinica B2023,13,11: | 0 |
| 13 | 核素骨显像在多发性骨髓瘤疗效评价中的应用(英文)显示文摘Objective:To investigate the value of radionuclide whole-body bone imaging on monitoring chemotherapeutic effects for multiple myeloma(MM).Methods:Sixty patients were included.Twenty nine cases received CTD(thalidomide 100-200 mg/d;cyclophosphamide 200-300 mg/m2/d,1-4 days,every 4 weeks;and dexamethasone 20-40 mg/d,1-4 days,every 4 weeks);Thirty cases received VAD(vincristine 0.4 mg/d,1-4 days,every 4 weeks;adriamycin 10 mg/d,1-4 days,every 4 weeks;dexamethasone 40 mg/d,1-4 days,every 4 weeks).Radionuclide bone imagings were performed in all patients before chemotherapy,six months,twelve months and eighteen months after chemotherapy.The correlation of chemothera-peutic effects between CTD and VAD were analyzed.Results:One hundred and seventy nine bone lesions were visualized by bone scintigraphy before CTD treatment.Eighteen months after CTD chemotherapy,it was observed by bone scintigraphy that 39/179(21.78%) lesions disappeared,112/179(62.57%) improved,and 28/179(15.64%) had no change.One hundred and ninety one bone lesions were showed by bone imaging before VAD treatment,36/191(18.84%) lesions disappeared,eighteen months after chemotherapy,103/191(53.92%) improved,and 52/191(27.22%) had no change.The significant difference was observed in locations of MM induced bone lesions treated with CTD(H = 8.23,P < 0.05) and VAD(H = 11.18,P < 0.05).A significant chemotherapeutic sensitivity in detecting MM induced lesions in ribs was found compared with other bone lesions.The chemotherapeutic effect of CTD was statistically significant than that of VAD(U = 2.17,P < 0.05).Conclusion:Radionuclide whole-body bone imaging has great value in monitoring chemotherapeutic effects for MM. | Yongli Bai Shuyao Zuo Chao Ma Xiaoting Su | 2008 | The Chinese-German Journal of Clinical Oncology2008,7,10: | 0 |

| 15 | Intelligent reflecting surface-assisted federated learning in multi-platoon collaborative networks显示文摘Inspired by mobile edge computing(MEC),edge learning has gained a momentum by directly performing model training at network edge without sending massive data to a centralized data center.However,the quality of model training will be affected by the limited communication and computing resources of network edge.In this paper,how to improve the training performance of a federated learning system aided by intelligent reflecting surface(IRS)over vehicle platooning networks is studied,where multiple platoons train a shared federated learning model.Multi-platoon cooperation can alleviate the pressure of data processing caused by the limited computing resources of single platoon.Meanwhile,IRS can enhance the inter-platoon communication in a cost-effective and energy-efficient manner.Firstly,the federated learning optimization problem of maximizing the learning accuracy is formulated by jointing platoon scheduling,bandwidth allocation and phase shifts at the IRS to maximize the number of scheduled platoon.Specif-ically,in the proposed learning architecture each platoon updates the learning model with its own data and uploads it to the global model through IRS-based wireless networks.Then,a method based on sequential optimization algorithm(SOA)and a group-based optimization method are analyzed for single IRS aided and large-scale IRS aided commu-nication,respectively.Finally,a platoon scheduling scheme is designed based on the communication reliability and computing reliability of platoons.Simulation results demonstrate that large-scale IRS assisted communication can effectively improve the reliability of multi-user communication networks.The scheduling scheme based on learning reliability balances the communication performance and computing performance of platoons. | Xiaoting Ma Junhui Zhao Jieyu Liao Ziyang Zhang | 2023 | Digital Communications and Networks2023,9,3: | 0 |

| 19 | 沉默COX-2抑制A549细胞的恶性增殖(英文)显示文摘Objective: The aim of this study was to explore the effects on malignant proliferation of A549 cell by silencing cy-clooxygenase (COX)-2. Methods: In the present study, we constructed three siRNA vectors producing small interference RNA. The siRNA vectors and the vacant vectors were transfected into A549 cell with lipofectamine respectively and the transfected cell strains were constructed. The change of COX-2 expression levels was examined by Western blot and RT-PCR. The effects on the proliferation of lung cancer cells were studied by cell growth curve, clonogenic assay and xenograft assays. Results: The siRNA expression vectors produced marked effects in A549 cell but the inhibited effects were different. The effect of psi-10 was best and the mRNA and protein levels of COX-2 reduced 61.2% and 56.2% respectively in A549-si10 cell in contrast to the control. The growth of A549 cell slowed and the colony formation rate reduced after silencing COX-2. In xenograft assays, the growth speeds of tumor became slow and the numbers of tumor reduced after silencing COX-2. | 20 | Hybrid multibody system method for the dynamic analysis of an ultra‐precision fly‐cutting machine tool显示文摘The dynamics of an ultra‐precision machine tool determines the precision of the machined surface.This study aims to propose an effective method to model and analyze the dynamics of an ultra‐precision fly‐cutting machine tool.First,the dynamic model of the machine tool considering the deformations of the cutter head and the lathe head is developed.Then,the mechanical elements are classified into M subsystems and F subsystems according to their properties and connections.The M‐subsystem equations are formulated using the transfer matrix method for multibody systems(MSTMM),and the F‐subsystem equations are analyzed using the finite element method and the Craig-Bampton reduction method.Furthermore,all the subsystems are assembled by combining the restriction equations at connection points among the subsystems to obtain the overall transfer equation of the machine tool system.Finally,the vibration characteristics of the machine tool are evaluated numerically and are validated experimentally.The proposed modeling and analysis method preserves the advantages of the MSTMM,such as high computational efficiency,low computational load,systematic reduction of the overall transfer equation,and generalization of its computational capability to general flexible‐body elements.In addition,this study provides theoretical insights and guidance for the design of ultra‐precision machine tools. | Hanjing Lu Xiaoting Rui Ziyao Ma Yuanyuan Ding Yiheng Chen Yu Chang Xuping Zhang | 2022 | International Journal of Mechanical System Dynamics2022,2,3: | 0 |
