| 1 | Niacin alters the ruminal microbial composition of cattle under high-concentrate condition显示文摘To understand the effects of niacin on the ruminal microbial ecology of cattle under high-concentrate diet condition, Illumina MiSeq sequencing technology was used. Three cattle with rumen cannula were used in a 3 x 3 Latin-square design trial. Three diets were fed to these cattle during 3 periods for 3 days, respectively: high-forage diet(HF; forage-to-concentrate ratio = 80:20), high-concentrate diet(HC;forage-to-concentrate ratio=20:80), and HC supplemented with 800 mg/kg niacin(HCN). Ruminal pH was measured before feeding and every 2 h after initiating feeding. Ruminal fluid was sampled at the end of each period for microbial DNA extraction. Overall, our findings revealed that subacute ruminal acidosis(SARA) was induced and the a-diversity of ruminal bacterial community decreased in the cattle of HC group. Adding niacin in HC could relieve the symptoms of SARA in the cattle but the ruminal pH value and the Shannon index of ruminal bacterial community of HCN group were still lower than those of HF group. Whatever the diet was, the ruminal bacterial community of cattle was dominated by Bacteroidetes,Firmicutes and Proteobacteria. High-concentrate diet significantly increased the abundance of Prevotella,and decreased the abundance of Paraprevotella, Sporobacter, Ruminococcus and Treponema than HF.Compared with HC, HCN had a trend to decrease the percentage of Prevotella, and to increase the abundance of Succiniclasticum, Acetivibrio and Treponema. Increasing concentrate ratio could decrease ruminal pH value, and change the ruminal microbial composition. | 2 | LncDACH1 promotes mitochondrial oxidative stress of cardiomyocytes by interacting with sirtuin3 and aggravates diabetic cardiomyopathy显示文摘Diabetic cardiomyopathy(DCM)is a common complication in diabetic patients.The molecular mechanisms of DCM remain to be fully elucidated.The intronic long noncoding RNA of DACH1(lnc DACH1)has been demonstrated to be closely associated with heart failure and cardiac regeneration.In this study,we investigated the role of lnc DACH1 in DCM and the underlying molecular mechanisms.The expression of lnc DACH1 was increased in DCM hearts and in high glucose-treated cardiomyocytes.Knockout of lnc DACH1 reduced mitochondrial oxidative stress,cell apoptosis,cardiac fibrosis and hypertrophy,and improved cardiac function in DCM mice.Overexpression of lnc DACH1 exacerbated mitochondria-derived reactive oxygen species(ROS)level and apoptosis,decreased activity of manganese superoxide dismutase(Mn-SOD);while silencing of lnc DACH1 attenuated ROS production,mitochondrial dysfunction,cell apoptosis,and increased the activity of Mn-SOD in cardiomyocytes treated with high glucose.Lnc DACH1 directly bound to sirtuin3(SIRT3)and facilitated its degradation by ubiquitination,therefore promoting mitochondrial oxidative injury and cell apoptosis in mouse hearts.In addition,SIRT3 silencing abrogated the protective effects of lnc DACH1 deficiency in cardiomyocytes.In summary,lnc DACH1 aggravates DCM by promoting mitochondrial oxidative stress and cell apoptosis via increasing ubiquitination-mediated SIRT3 degradation in mouse hearts.Inhibition of lnc DACH1 represents a novel therapeutic strategy for the intervention of diabetic cardiomyopathy. | Qi Zhang Danyang Li Xue Dong Xiaowen Zhang Junwu Liu Lili Peng Bo Meng Qi Hua Xinyu Pei Lu Zhao Xiaoxi Hu Yang Zhang Zhenwei Pan Yanjie Lu Baofeng Yang | 2022 | Science China(Life Sciences)2022,65,6: | 4 |
| 3 | Severity of early allograft dysfunction following donation after circulatory death liver transplantation:a multicentre study显示文摘Background:Early allograft dysfunction(EAD)is associated with decreased graft and patient survival rates.This study aimed to identify the severity of EAD and develop a predictive model for EAD after donation after circulatory death(DCD)liver transplantation(LT).Furthermore,the influence of operative time on EAD incidence was also evaluated.Methods:In this retrospective,multicentre cohort study,nomograms were established based on a single-centre training cohort(n=321)and validated in a 3-center validation cohort(n=501).Results:The incidence rate of EAD was 46.4%(149/321)in the training cohort and 40.5%(203/501)in the validation cohort.Of the 149 EAD patients in the training cohort,77 patients with either elevated alanine aminotransferase(ALT)or aspartate aminotransferase(AST)were classified as having EAD type A,and the rest of the EAD patients were classified as having EAD type B.Recipients with EAD type B had lower graft and patient survival rates than recipients with EAD type A(P=0.043 and 0.044,respectively).We further developed a nomogram to predict EAD(graft weight,cold ischemia time,donor age,model for end-stage liver disease(MELD)score)and another nomogram to predict EAD type B(graft weight,cold ischemia time,MELD score).The nomograms for the prediction of EAD and EAD type B had good discrimination[concordance index(C-index)=0.712(0.666-0.758),0.707(0.641-0.773)]and calibration[Hosmer-Lemeshow(HL)P=0.384,P=0.425]in the validation cohort.An increased operative time(>6 h)was associated with increased EAD and EAD type B incidence in the high-risk group(P=0.005,P=0.020,respectively).Conclusions:EAD type B was associated with decreased graft and patient survival rates.The novel nomograms effectively predicted the incidence of EAD and EAD type B in DCD LT patients. | Kun Wang Di Lu Yuhui Liu Wangyao Li Li Zhuang Zhenyu Ma Qinfen Xie Binhua Pan Yichao Wu Junli Chen Lidan Lin Xiaowen Feng Qiang Wei Xuyong Wei Haiyang Xie Zhengxin Wang Shusen Zheng Xiao Xu | 2021 | Hepatobiliary Surgery and Nutrition2021,10,1: | 4 |
| 4 | Impacts of different feeding modes on the gonadal development, total edible yield, and nutritional composition of male Chinese mitten crab (Eriocheir sinensis)显示文摘This study analyzed the tissue indices,proximate composition,fatty acid and amino acid composition of edible tissues of male E.sinensis under three feeding modes(formulated diets(FD),natural diets(ND),and traditional diets(TD)).The results showed that different feeding modes had no impacts on the overall gonadal development,as no significant difference was found in gonadsomatic index among three treatments.For edible yield,three modes all had similar levels of the meat yield,hepatosomatic index,and total edible yield.Although different feeding modes did not affect the individual edible yield and gonadal development,significant differences were detected in the nutritional composition of males from three modes.For proximate composition,significant differences were found in the total lipids of gonads and the carbohydrate of hepatopancreas and meat.For fatty acids,crabs from ND had a higher proportion of monounsaturated fatty acids.In gonads and hepatopancreas,FD had a higher proportion of long-chain polyunsaturated fatty acids.For amino acids,FD had higher amino acid contents,while ND had the lowest.Except tryptophan,the essential amino acids scores of the meat were all greater than 100.As there was no significant difference on meat quality and meat yields between three feeding modes,this study suggests that the formulated diet could replace the natural diet and traditional diet towards sustainable crab farming in future. | Jie Pan Renfu Wu Xugan Wu Xiaowen Long Xichang Wang Yongxu Cheng | 2020 | Aquaculture and Fisheries2020,5,6: | 3 |

| 6 | Tocilizumab in patients with moderate or severe COVID-19: a randomized, controlled, open-label, multicenter trial显示文摘Tocilizumab has been reported to attenuate the“cytokine storm”in COVID-19 patients.We attempted to verify the effectiveness and safety of tocilizumab therapy in COVID-19 and identify patients most likely to benefit from this treatment.We conducted a randomized,controlled,open-label multicenter trial among COVID-19 patients.The patients were randomly assigned in a 1:1 ratio to receive either tocilizumab in addition to standard care or standard care alone.The cure rate,changes of oxygen saturation and interference,and inflammation biomarkers were observed.Thirty-three patients were randomized to the tocilizumab group,and 32 patients to the control group.The cure rate in the tocilizumab group was higher than that in the control group,but the difference was not statistically significant(94.12%vs.87.10%,rate difference 95%CI−7.19%–21.23%,P=0.4133).The improvement in hypoxia for the tocilizumab group was higher from day 4 onward and statistically significant from day 12(P=0.0359).In moderate disease patients with bilateral pulmonary lesions,the hypoxia ameliorated earlier after tocilizumab treatment,and less patients(1/12,8.33%)needed an increase of inhaled oxygen concentration compared with the controls(4/6,66.67%;rate difference 95%CI−99.17%to−17.50%,P=0.0217).No severe adverse events occurred.More mild temporary adverse events were recorded in tocilizumab recipients(20/34,58.82%)than the controls(4/31,12.90%).Tocilizumab can improve hypoxia without unacceptable side effect profile and significant influences on the time virus load becomes negative.For patients with bilateral pulmonary lesions and elevated IL-6 levels,tocilizumab could be recommended to improve outcome. | Dongsheng Wang Binqing Fu Zhen Peng Dongliang Yang Mingfeng Han Min Li Yun Yang Tianjun Yang Liangye Sun Wei Li Wei Shi Xin Yao Yan Ma Fei Xu Xiaojing Wang Jun Chen Daqing Xia Yubei Sun Lin Dong Jumei Wang Xiaoyu Zhu Min Zhang Yonggang Zhou Aijun Pan Xiaowen Hu Xiaodong Mei Haiming Wei Xiaoling Xu | 2021 | Frontiers of Medicine2021,15,3: | 2 |

| 14 | Diverse Asgard archaea including the novel phylum Gerdarchaeota participate in organic matter degradation显示文摘Asgard is an archaeal superphylum that might hold the key to understand the origin of eukaryotes, but its diversity and ecological roles remain poorly understood. Here, we reconstructed 15 metagenomic-assembled genomes from coastal sediments covering most known Asgard archaea and a novel group, which is proposed as a new Asgard phylum named as the 'Gerdarchaeota'.Genomic analyses predict that Gerdarchaeota are facultative anaerobes in utilizing both organic and inorganic carbon. Unlike their closest relatives Heimdallarchaeota, Gerdarchaeota have genes encoding for cellulase and enzymes involved in the tetrahydromethanopterin-based Wood–Ljungdahl pathway. Transcriptomics showed that most of our identified Asgard archaea are capable of degrading organic matter, including peptides, amino acids and fatty acids, occupying ecological niches in different depths of layers of the sediments. | 15 | Automatic calculation of the magnetometer zero offset using the interplanetary magnetic field based on the Wang-Pan method显示文摘The space-borne fluxgate magnetometer(FGM)requires regular in-flight calibration to obtain its zero offset.Recently,Wang GQ and Pan ZH(2021a)developed a new method for the zero offset calibration based on the properties of Alfvén waves.They found that an optimal offset line(OOL)exists in the offset cube for a pure Alfvén wave and that the zero offset can be determined by the intersection of at least two nonparallel OOLs.Because no pure Alfvén waves exist in the interplanetary magnetic field,calculation of the zero offset relies on the selection of highly Alfvénic fluctuation events.Here,we propose an automatic procedure to find highly Alfvénic fluctuations in the solar wind and calculate the zero offset.This procedure includes three parts:(1)selecting potential Alfvénic fluctuation events,(2)obtaining the OOL,and(3)determining the zero offset.We tested our automatic procedure by applying it to the magnetic field data measured by the FGM onboard the Venus Express.The tests revealed that our automatic procedure was able to achieve results as good as those determined by the Davis-Smith method.One advantage of our procedure is that the selection criteria and the process for selecting the highly Alfvénic fluctuation events are simpler.Our automatic procedure could also be applied to find fluctuation events for the Davis-Smith method. | XiaoWen Hu GuoQiang Wang ZongHao Pan | 2022 | Earth and Planetary Physics2022,6,1: | 1 |
