1A novel mechanism of inhibiting in-stent restenosis with arsenic trioxide drug-eluting stent: Enhancing contractile phenotype of vascular smooth muscle cells via YAP pathway显示文摘Objective:Arsenic trioxide(ATO or As2O3)has beneficial effects on suppressing neointimal hyperplasia and restenosis,but the mechanism is still unclear.The goal of this study is to further understand the mechanism of ATO's inhibitory effect on vascular smooth muscle cells(VSMCs).Methods and results:Through in vitro cell culture and in vivo stent implanting into the carotid arteries of rabbit,a synthetic-to-contractile phenotypic transition was induced and the proliferation of VSMCs was inhibited by ATO.F-actin filaments were clustered and the elasticity modulus was increased within the phenotypic modulation of VSMCs induced by ATO in vitro.Meanwhile,Yes-associated protein(YAP)nuclear translocation was inhibited by ATO both in vivo and in vitro.It was found that ROCK inhibitor or YAP inactivator could partially mask the phenotype modulation of ATO on VSMCs.Conclusions:The interaction of YAP with the ROCK pathway through ATO seems to mediate the contractile phenotype of VSMCs.This provides an indication of the clinical therapeutic mechanism for the beneficial bioactive effect of ATO-drug eluting stent(AES)on in-stent restenosis(ISR).Yinping Zhao Guangchao Zang Tieying Yin Xiaoyi Ma Lifeng Zhou Lingjuan Wu Richard Daniel Yunbing Wang Juhui Qiu Guixue Wang 2021Bioactive Materials2021,6,2:13
2A cabazitaxel liposome for increased solubility, enhanced antitumor effect and reduced systemic toxicity显示文摘The potential side effects of cabazitaxel(CBZ)in the field of cancer treatment have become a great limitation to its further clinical application.Liposomal delivery is a well-established approach to increase the therapeutic index of hydrophobic drugs.In this study,a PEGmodified liposome was developed for efficiently encapsulating CBZ,thus enhancing its specific tumor inhibition effect and reducing the systemic toxicity.It was found that the loading efficiency of CBZ into the liposome could be improved with the increase of lipophilic materials,as it could be over 80%under the weight ratio of 20:1(total lipid:CBZ).The diameter of CBZ loaded liposome(CBZ@Lipo)was^100 nm.And the liposome suspending in aqueous medium was stable at 4°C for at least one month,according to the change of its size distribution.The killing ability of CBZ@Lipo to cancer cells was significantly lower comparing to that of CBZ solution,which could be attributed to the slow release of CBZ from the liposomes.However,CBZ@Lipo could induce an obvious apoptosis of the cancer cells at low concentration.Furthermore,CBZ@Lipo exhibited an expressively enhanced tumor growth inhibition effect comparing to CBZ solution.More importantly,CBZ@Lipo showed an obviously higher biosafety proved by lower hemolysis probability,stable body weight of mice during the whole experiment and no obvious lesion in histology analysis.Our work provided a useful reference of the formulation of CBZ,which had potential for greater clinical application.Xiaoyi Yin Lihua Luo Wei Li Jie Yang Chunqi Zhu Mengshi Jiang Bing qin Xiaoling Yuan Hang Yin Yichao Lu Yongzhong Du Dawei Chen Jian You 2019Asian Journal of Pharmaceutical Sciences2019,14,6:3

4Extending generalized unsupervised manifold alignment显示文摘Building connections between different data sets is a fundamental task in machine learning and related application community. With proper manifold alignment, the correspondences between data sets will assist us with comprehensive study of data processes and analyses. Despite the several progresses in semi-supervised and unsupervised scenarios, potent manifold alignment methods in generalized and realistic circumstances remain in absence. Besides, theretofore unsupervised algorithms seldom prove themselves mathematically. In this paper, we devise an efficient method to properly solve the unsupervised manifold alignment problem and denominate it as extending generalized unsupervised manifold alignment(EGUMA)method. More specifically, an explicit relaxed integer programming method is adopted to solve the unsupervised manifold alignment problem, which reconciles three factors covering the updated local structure matching, the the feature comparability and geometric preservation. An additional effort is retained on extending the Frank Wolfe algorithm to tacking our optimization problem. Besides our previous endeavors we adopt a new strategy for neighborhood discovery in the manifolds. The main advantages over previous methods accommodate(1) simultaneous alignment and discovery of manifolds;(2) complete unsupervised learning structure without any prerequisite correspondence;(3) more concise local geometry for the embedding space;(4) efficient alternative optimization;(5) strict mathematical analysis on the convergence and efficiency issues. 5Insights into the proteomic profile of newly harvested corn and metagenomic analysis of the broiler intestinal microbiota显示文摘Background:The use of newly harvested corn in feed causes wet droppings in broilers and increased feed cost which was termed as“new season grain problem”.The present study was conducted to evaluate the proteomic profile of newly harvested corn and the subsequent influence on intestinal microbiol community for broiler chickens.Methods:Newly harvested corn stored for either half a month(HM)or two months(TM)was used,and the pasting properties,total soluble sugars,and proteomic analysis technology was used to explore the influence of storage on natural aging corn properties.Additionally,seventy-two 7-day-old Ross 308 male broiler chicken were fed with different stored corn.Apparent metabolizable energy(AME),digesta viscosity,intestinal morphology and microbiota were examined to explore the influence of feed corn storage on broiler chickens.Results:Pasting properties in the TM corn exhibited decreased viscoelastic properties.Proteomic studies found a total of 26 proteins that were differentially expressed between the two treatment groups.Proteins involved in starch and polysaccharides biosynthesis were upregulated in TM compared with HM.Chickens fed on TM diet had higher relative energy utilization compared to the HM birds.With increased corn storage,the relative digesta viscosity decreased significantly(P≤0.05).The total number of goblet cells and lymphocytes was lower in chickens fed the TM diet.The microbiota data showed that the TM chickens had decreased abundance of diarrheal bacteria such as Hungatella hathewayi and Bacteroides fragilis,and increased butyrate-producing bacteria such as Alistipes compared to the HM chickens.Conclusions:Storage of newly harvested corn induced the synthetic reaction of large molecules and changed the solubility of starch and protein with increasing soluble sugars and decreasing pasting properties that may improve the fermentation of intestinal microbiota,improve the energy utilization and protect gut health without the risk of diarrhea.Dafei Yin Youli Wang Liqun Wang Yuqin Wu Xiaoyi Bian Samuel E.Aggrey Jianmin Yuan 2022Journal of Animal Science and Biotechnology2022,13,3:0

7Commensal microbiota and host metabolic divergence are associated with the adaptation of Diploderma vela to spatially heterogeneous environments显示文摘Heterogeneous environment adaptation is critical to understand the species evolution and response to climate change.However,how narrow-range species adapt to micro-geographic heterogeneity has been overlooked,and there is a lack of insights from metabolism and commensal microbiota.Here,we studied the environmental adaptation for 3 geographic populations(>40 km apart)of Diploderma vela,a lizard endemic to dry-hot valleys of the Hengduan Mountain Region.The climatic boundary caused a cooler,droughtier,and barren environment for northernmost population(RM)than the middle(QZK)and southernmost populations(FS).Correspondingly,significant divergences in liver and muscle metabolism and commensal microbiota were detected between RM and QZK or FS individuals,but not between QZK and FS individuals.Phospholipid composition,coenzyme level(i.e.pyridoxal and NAD^(+)),and cholesterol metabolism(e.g.androgen and estriol synthesis)constituted the major metabolic difference between RM and QZK/FS groups.FS and QZK individuals kept abundant Proteobacteria and antifungal strains,while RM individuals maintained more Firmicutes and Bacteroidota.Strong associations existed between varied host metabolite and gut microbes.How were these interpopulation variations associated to the environment adaptation were discussed.These results provided some novel insights into the environmental adaptation and implicated the consequence of climate change on narrow-range species.Wei ZHU Xiudong SHI Yin QI Xiaoyi WANG Liming CHANG Chunlin ZHAO Lifeng ZHU Jianping JIANG 2022Integrative Zoology2022,17,3:0

9Systemic and single cell level responses to 1 nm size biomaterials demonstrate distinct biological effects revealed by multi-omics atlas显示文摘Although ultra-small nanoclusters(USNCs,<2 nm)have immense application capabilities in biomedicine,the investigation on body-wide organ responses towards USNCs is scant.Here,applying a novel strategy of single-cell mass cytometry combined with Nano Genome Atlas of multi-tissues,we systematically evaluate the interactions between the host and calcium phosphate(CaP)USNCs at the organism level.Combining single-cell mass cytometry,and magnetic luminex assay results,we identify dynamic immune responses to CaP USNCs at the single cell resolution.The innate immune is initially activated and followed by adaptive immune activation,as evidenced by dynamic immune cells proportions.Furthermore,using Nano Genome Atlas of multi-tissues,we uncover CaP USNCs induce stronger activation of the immune responses in the cartilage and subchondral bone among the five local tissues while promote metabolic activities in the liver and kidney.Moreover,based on the immunological response profiles,histological evaluation of major organs and local tissue,and a body-wide transcriptomics,we demonstrate that CaP USNCs are not more hazardous than the Food and Drug Administration-approved CaP nanoparticles after 14 days of injection.Our findings provide valuable information on the future clinical applications of USNCs and introduce an innovative strategy to decipher the whole body response to implants.Tao Zhang Tingyun Lei Ruojin Yan Bo Zhou Chunmei Fan Yanyan Zhao Shasha Yao Haihua Pan Yangwu Chen Bingbing Wu Yuwei Yang Lijuan Hu Shen Gu Xiaoyi Chen Fangyuan Bao Yu Li Hanqi Xie Ruikang Tang Xiao Chen Zi Yin 2022Bioactive Materials2022,7,12:0
10A novel mechanism of PHB2-mediated mitophagy participating in the development of Parkinson's disease显示文摘Endoplasmic reticulum stress and mitochondrial dysfunction play important roles in Parkinson s disease,but the regulato ry mechanism remains elusive.Prohibitin-2(PHB2)is a newly discove red autophagy receptor in the mitochondrial inner membrane,and its role in Parkinson’s disease remains unclear.Protein kinase R(PKR)-like endoplasmic reticulum kinase(PERK)is a factor that regulates cell fate during endoplasmic reticulum stress.Parkin is regulated by PERK and is a target of the unfolded protein response.It is unclear whether PERK regulates PHB2-mediated mitophagy thro ugh Parkin.In this study,we established a 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine(MPTP)-induced mouse model of Parkinson’s disease.We used adeno-associated virus to knockdown PHB2 expression.Our res ults showed that loss of dopaminergic neurons and motor deficits were aggravated in the MPTP-induced mouse model of Parkinson’s disease.Ove rexpression of PHB2 inhibited these abnormalities.We also established a 1-methyl-4-phenylpyridine(MPP+)-induced SH-SY5Y cell model of Parkinson’s disease.We found that ove rexpression of Parkin increased co-localization of PHB2 and microtubule-associated protein 1 light chain 3,and promoted mitophagy.In addition,MPP+regulated Parkin involvement in PHB2-mediated mitophagy through phosphorylation of PERK.These findings suggest that PHB2 participates in the development of Parkinson’s disease by intera cting with endoplasmic reticulum stress and Parkin.Yongjiang Zhang Shiyi Yin Run Song Xiaoyi Lai Mengmeng Shen Jiannan Wu Junqiang Yan 2024Neural Regeneration Research2024,19,8:0

12Survival outcomes of conversion surgery for metastatic pancreatic ductal adenocarcinoma after neoadjuvant therapy显示文摘Objective:To evaluate the survival outcomes of patients who underwent conversion surgery for metastatic pancreatic ductal adenocarcinoma(mPDAC)after neoadjuvant therapy(NAT)and to identify potential candidates that may benefit from this treat-ment strategy.Background:The role and eligibility population of conversion surgery for mPDAC remains controversial in the era of NAT.Methods:A consecutive cohort of patients diagnosed with mPDAC and treated with NAT followed by conversion surgery be-tween 2019 and 2021 were confirmed from a prospective database maintained by the Department of Pancreatic Hepatobiliary Surgery of Changhai Hospital.In accordance with residual metastases and technical resectability after NAT,patients were classi-fied as the complete pathological response of metastases(ypM0)resection group,residual metastases(ypM1)resection group,and exploration group.Median overall survival(mOS)was calculated using the Kaplan-Meier method,uni-and multivariable cox regression was performed to identify clinicopathological predictors of OS.Results:A total of 244 patients with mPDAC were identified from the prospective database,with 19(7.8%)patients who un-derwent ypM0 resection,22(9.0%)underwent ypM1 resection,and 23(9.4%)underwent explorative laparotomy.The mOS was 32.6 months for ypM0 resected patients,15.1 months for ypM1 resected patients,and 13.4 months for those who underwent explorative laparotomy(P<.001).Univariable and multivariable Cox regression analyses confirmed that ypM0 resection,normal-ization of preoperative CA19-9 levels,and continued adjuvant therapy were independent prognostic factors of conversion surgery for mPDAC after NAT.Subgroup analyses revealed that oligometastases and continued adjuvant therapy were associated with improved prognosis in the ypM1 resection group.Conclusion:In patients with mPDAC who underwent NAT followed by conversion surgery,the complete pathological response of metastases,normalization of preoperative CA19-9 levels,and continued adjuvant therapy were independent risk factors for prognosis.Patients with residual oligometastases after treatment were expected to prolong survival through resection.These patients may benefit from conversion surgery and should be potential candidates for this treatment strategy.Lingyu Zhu Suizhi Gao Xinqian Wu Bo Li Xiaohan Shi Xiaoyi Yin Huan Wang Meilong Shi Penghao Li Yikai Li Chaoliang Zhong Chuanqi Teng Jiawei Han Yiwei Ren Jian Wang Zhendong Fua Xinyu Liu Kailian Zheng Shiwei Guo Gang Jin 2023Journal of Pancreatology2023,6,3:0

14Ce-Ti catalysts modified with copper and vanadium to effectively remove slip NH_(3) and NO_x from coal-fired plants显示文摘In this work,V/Ce-Ti catalysts were modified with different kinds of transition metals(Cu,Fe,Co,Mn) by sol-gel and impregnation methods.The NH_(3) oxidation performance of them was tested to select the most active catalyst in NH_(3)-selective catalytic oxidation(NH_(3)-SCO).The effect of NO,SO_(2) and H_(2)O was also investigated.The experimental results indicate that 1% Cu-V/Ce-Ti catalyst exhibits the most significant ability to remove slip ammonia discharged from coal-fired plants and its NH_(3) conversion efficiency reaches90% at 300℃.In additio n,97% NO_x can be removed when NO is introduced in the gas.Cu-V/Ce-Ti catalyst also obtains good resistance to H_(2)O and SO_(2).Based on the characterization experiment,the introduced Cu and V are highly dispersed on Ce-Ti catalyst and they can increase the redox properties and the number of acidic sites.Besides,the redox cycles among Cu,V and Ce species on Cu-V/Ce-Ti catalyst surface are conducive to generating more active oxygen and promoting the oxidation capacity of the catalyst.Wenjie Liu Yifei Long Shinian Liu Yongyan Zhou Xin Tong Yajie Yin Xiaoyi Li Kang Hu Jiangjun Hu 2023Journal of Rare Earths2023,41,7:0
