| 1 | Efficacy and safety of a novel anti-HER2 therapeutic antibody RC48 in patients with HER2-overexpressing,locally advanced or metastatic gastric or gastroesophageal junction cancer:a single-arm phase II study显示文摘Background:Current treatment options for human epidermal growth factor receptor 2(HER2)-overexpressing gastric cancer at third-line have shown limited clinical benefit.Further,there is no specific treatment for HER2 immunohistochemistry(IHC)2+and fluorescence in-situ hybridization-negative patients.Here,we report the efficacy and safety of a novel anti-HER2 antibody RC48 for patients with HER2-overexpressing,advanced gastric or gastroesophageal junction cancer.Methods:Patients with HER2-overexpressing(IHC 2+or 3+),locally advanced or metastatic gastric or gastroesophageal junction cancer who were under at least second-line therapy were eligible and received RC482.5 mg/kg alone every 2 weeks.The primary endpoint was the objective response rate(ORR)assessed by an independent review committee.Secondary endpoints included progressionfree survival(PFS),overall survival(OS),duration of response,time to progression,disease control rate,and safety.Results:Of 179 patients screened,125 were eligible and received RC48 treatment.The ORR was 24.8%(95%confidence interval[CI]:17.5%-33.3%).The median PFS and OS were 4.1 months(95%CI:3.7-4.9 months)and 7.9 months(95%CI:6.7-9.9 months),respectively.The most frequently reported adverse events were decreased white blood cell count(53.6%),asthenia(53.6%),hair loss(53.6%),decreased neutrophil count(52.0%),anemia(49.6%),and increased aspartate aminotransferase level(43.2%).Serious adverse events(SAEs)occurred in 45(36.0%)patients,and RC48-related SAEs were mainly decreased neutrophil count(3.2%).Seven patients had adverse events that led to death were not RC48-related.Conclusions:RC48 showed promising activity with manageable safety,suggesting potential application in patients with HER2-overexpressing,advanced gastric or gastroesophageal junction cancer who have previously received at least two lines of chemotherapy. | Zhi Peng Tianshu Liu Jia Wei Airong Wang Yifu He Liuzhong Yang Xizhi Zhang Nanfeng Fan Suxia Luo Zhen Li Kangsheng Gu Jianwei Lu Jianming Xu Qingxia Fan Ruihua Xu Liangming Zhang Enxiao Li Yuping Sun Guohua Yu Chunmei Bai Yong Liu Jiangzheng Zeng Jieer Ying Xinjun Liang Nong Xu Chao Gao Yongqian Shu Dong Ma Guanghai Dai Shengmian Li Ting Deng Yuehong Cui Jianmin Fang Yi Ba Lin Shen | 2021 | Cancer Communications2021,41,11: | 29 |
| 2 | Effects of soil water and nitrogen availability on photosynthesis and water use efficiency of Robinia pseudoacacia seedlings显示文摘The efficient use of water and nitrogen (N) to promote growth and increase yield of fruit trees and crops is well studied.However,little is known about their effects on woody plants growing in arid and semiarid areas with limited water and N availability.To examine the effects of water and N supply on early growth and water use efficiency (WUE) of trees on dry soils,one-year-old seedlings of Robinia pseudoacacia were exposed to three soil water contents (non-limiting,medium drought,and severe drought) as well as to low and high N levels,for four months.Photosynthetic parameters,leaf instantaneous WUE (WUEi) and whole tree WUE (WUEb) were determined.Results showed that,independent of N levels,increasing soil water content enhanced the tree transpiration rate (Tr),stomatal conductance (Gs),intercellular CO2 concentration (Ci),maximum net assimilation rate (Amax),apparent quantum yield (AQY),the range of photosynthetically active radiation (PAR) due to both reduced light compensation point and enhanced light saturation point,and dark respiration rate (Rd),resulting in a higher net photosynthetic rate (Pn) and a significantly increased whole tree biomass.Consequently,WUEi and WUEb were reduced at low N,whereas WUE i was enhanced at high N levels.Irrespective of soil water availability,N supply enhanced Pn in association with an increase of Gs and Ci and a decrease of the stomatal limitation value (Ls),while Tr remained unchanged.Biomass and WUEi increased under non-limiting water conditions and medium drought,as well as WUEb under all water conditions;but under severe drought,WUEi and biomass were not affected by N application.In conclusion,increasing soil water availability improves photosynthetic capacity and biomass accumulation under low and high N levels,but its effects on WUE vary with soil N levels.N supply increased Pn and WUE,but under severe drought,N supply did not enhance WUEi and biomass. | Xiping Liu Yangyang Fan Junxia Long Ruifeng Wei Roger Kjelgren Chunmei Gong Jun Zhao | 2013 | Journal of Environmental Sciences2013,25,3: | 15 |

| 4 | Application of enriched stable isotope technique to the study of copper bioavailability in Daphnia magna显示文摘The biokinetics of Cu in Daphnia magna, including dissolved uptake, assimilation and efflux, has been determined using a gamma 67 Cu radiotracer methodology. However, this gamma emitting radioisotope is not readily available due to its very short half-life. In the present study, we employed a stable isotope tracer ( 65 Cu) to determine the Cu biokinetics and compared our results to those determined using 67 Cu. The dissolved uptake rate constant of 65 Cu was 3.36 L/(g·day), which is higher than that of 67 Cu (1.32 L/(g·day)). With increasing food concentrations from 2×10 4 to 1×10 5 cells/mL, the Cu assimilation efficiency (AE) decreased from 46% to 11%, compared to a decrease from 27% to 16% when determined using 67 Cu. The efflux of Cu from Daphnia magna was quantified following both dissolved and dietary uptake. The efflux of waterborne Cu was comparable to that of dietborne Cu and the efflux rate constant (0.32-0.52 day 1 ) was higher than that determined by 67 Cu (0.19-0.20 day 1 ). By considering different water properties and handling procedure between the two experiments, we believe that these differences are reasonable. | 5 | Mandible exosomal ssc-mir-133b regulates tooth development in miniature swine via endogenous apoptosis显示文摘Signal transduction between different organs is crucial in the normal development of the human body. As an important medium for signal communication, exosomes can transfer important information, such as microRNAs(miRNAs), from donors to receptors.MiRNAs are known to fine-tune a variety of biological processes, including maxillofacial development; however, the underlying mechanism remains largely unknown. In the present study, transient apoptosis was found to be due to the expression of a miniature swine maxillofacial-specific miRNA, ssc-mir-133b. Upregulation of ssc-mir-133b resulted in robust apoptosis in primary dental mesenchymal cells in the maxillofacial region. Cell leukemia myeloid 1(Mcl-1) was verified as the functional target, which triggered further downstream activation of endogenous mitochondria-related apoptotic processes during tooth development.More importantly, mandible exosomes were responsible for the initial apoptosis signal. An animal study demonstrated that ectopic expression of ssc-mir-133 b resulted in failed tooth formation after 12 weeks of subcutaneous transplantation in nude mice. | 7 | Inorganic nitrate alleviates the senescence-related decline in liver function显示文摘Senescence-related decline in liver function is a common complication in the elderly that can lead to impaired health in older individuals.Dietary nitrate supplements have physiological and therapeutic effects on organ function by nitrate(NO_3^-)-nitrite(NO_2^-)-nitric oxide(NO)pathway.However,the role of dietary nitrate on the senescence-related decline in liver function is unclear.The findings of the present study showed that nitrate levels in the serum and liver decreased,whereas the levels of alanine aminotransferase(ALT)and aspartate aminotransferase(AST)in serum increased in ageing mice.Consistently,cell senescence,decreased glycogen levels and increased lipid deposition were found in the liver of aged mice,both glucokinase(GCK)and phosphoenolpyruvate carboxykinase(PCK)were down-regulated(n=10).Daily nitrate intake(0.5 mmol L^(-1))restored nitrate levels,decreased ALT and AST levels,and prevented cell senescence and structural and glucose and lipid metabolism degeneration in liver tissue both in D-galactose(D-gal)-induced ageing mice(n=10)and in natural aged mice(n=10).In conclusion,the present study demonstrated that the reduction of nitrate levels was correlated with liver degeneration in ageing individuals and that dietary supplement of nitrate could restore the nitrate levels in serum and the liver and prevent ageing-related liver degeneration. | Haifeng Wang Lei Hu Le Li Xiaoshan Wu Zhipeng Fan Chunmei Zhang Jinsong Wang Jidong Jia Songlin Wang | 2018 | Science China(Life Sciences)2018,61,1: | 5 |

| 10 | Sensitivity of pollen factors in the climate transfer function显示文摘Constructing a mathematic transfer function of pollen and climatic factors is one of the most important approaches in the quantitative reconstruction of paleoclimate elements.In the function building,the key point is the response sensitivity to the climate change for different pollen families and categories. As an example in this paper,the pollen samples from the surface and stratum in Dajiuhu basin,Shennongjia are used to estimate the sensitivity of pollen factor-temperature in the transfer function by the EOF analysis,multiple regression and stepwise regression techniques.Thereafter,the selection methods of pollen factors are discussed and compared with other results from different researchers. The results show that in the pollen samples,the quantity of woody plants is larger than others,but the woody plants have relatively lower sensitivity and slow responses to the climate evolution and abrupt climate change.While the pollens from lower grade plants(herb and fern)have a relatively high sensi- tivity to the temperature change and fast response to the abrupt climate changes.Therefore,the pollen of herb and fern may significantly record the extreme events in the climate change.Also in different regions,the pollen samples have different sensitivity and optimum combination in the transfer functions.Final conclusion is that the stepwise regression is one of the best methods for transfer function building since it can obtain the maximum multiple correlation coefficients and optimum combination of sensitive pollen factors. | CHEN Xing ZHU Cheng MA ChunMei FAN Chao | 2008 | Chinese Science Bulletin2008,53,S1: | 3 |

Synergistic effects and mechanism显示文摘A novel ferruginous active absorbent, prepared by fly ash, industrial lime and the additive Fe(VI), was introduced for synchronous abatement of binary mixtures of SO2–NOx from simulated coal-fired flue gas. The synergistic action of various factors on the absorption of SO2 and NOx was investigated. The results show that a strong synergistic effect exists between Fe(VI) dose and reaction temperature for the desulfurization. It was observed that in the denitration process, the synergy of Fe(VI) dose and Ca/(S + N) had the most significant impact on the removal of NO, followed by the synergy of Fe(VI) and reaction temperature, and then the synergy of reaction temperature and flue gas humidity. A scanning electron microscope(SEM) and an accessory X-ray energy spectrometer(EDS)were used to observe the surface characteristics of the raw and spent absorbent as well as fly ash. A reaction mechanism was proposed based on chemical analysis of sulfur and nitrogen species concentrations in the spent absorbent. | 18 | The relationship between treatment-induced hypertension and efficacy of anlotinib in recurrent or metastatic esophageal squamous cell carcinoma显示文摘Objective:In this post-hoc analysis,we evaluated anlotinib treatment-induced hypertension as a potential predictive factor of efficacy in esophageal squamous cell carcinoma(ESCC)patients.Methods:A total of 109 patients enrolled in the anlotinib group in a phase 2 trial were included.The tumor response was assessed by computed tomography at week 3,week 6,and then every 6 weeks until progressive disease was observed.The primary endpoint of the study was progression free survival(PFS).The secondary endpoints included overall survival(OS)and objective response rate(ORR).Results:In all patients,the median PFS was 3.02 months[95%confidence interval(CI):2.63–3.65 months]and the OS was 6.11 months(95%CI:4.40–7.79 months).The ORR was 7.34%(95%CI:3.22%–13.95%).A total of 59(54%)patients were diagnosed with treatment-induced hypertension(Group A),and the remaining patients(n=50,46%)were in Group B.Baseline prognostic factors were similar between the 2 groups.Patients in Group A had a longer PFS and OS and higher ORR.When stratifying patients using a previously known history of hypertension,treatment-induced hypertension was a predictor only for patients without previous hypertension,who had longer PFS[hazard ratio(HR):0.40,95%CI:0.24–0.68]and OS(HR:0.37,95%CI:0.21–0.67).Conclusions:We showed,for the first time,a correlation between treatment-induced hypertension and better prognoses in recurrent or metastatic ESCC patients treated with anlotinib,without a previously known history of hypertension.Treatment-induced hypertension may be a simple and low cost predictor for anlotinib antitumor efficacy in these patients,which may also reflect the intended target inhibition. | Yan Song Juxiang Xiao Wentao Fang Ping Lu Qingxia Fan Yongqian Shu Jifeng Feng Shu Zhang Yi Ba Yang Zhao Ying Liu Chunmei Bai Yuxian Bai Yong Tang Jie He Jing Huang | 2021 | Cancer Biology & Medicine2021,18,2: | 1 |
