| 1 | Near-infrared responsive 5-fluorouracil and indocyanine green loaded MPEG-PCL nanoparticle integrated with dissolvable microneedle for skin cancer therapy显示文摘The prevalence of skin cancer is rising along with the rapid population aging in recent years.Traditional therapies,such as surgical treatment,radiotherapy,chemotherapy,photodynamic therapy,and immunotherapy,may accompany serious side effects,limiting their clinical benefits.According to the biological characteristics of skin cancer,we have already established two kinds of synergetic systems of photothermal therapy(microneedle)and chemotherapy,containing gold nanorods(GNR).Although the microneedle system exhibited great potential for skin cancer treatment,the system could be still improved further.So,we designed a near-infrared lightresponsive 5-fluorouracil(5-Fu)and indocyanine green(ICG)loaded monomethoxy-poly(ethylene glycol)-polycaprolactone(MPEG-PCL)nanoparticle(5-Fu-ICG-MPEG-PCL),and then 5-Fu-ICG-MPEG-PCL was integrated with a hyaluronic acid dissolvable microneedle system(HA MN)to get 5-Fu-ICG-MPEG-PCL loaded HA MN for treating skin cancers,including human epidermoid cancer and melanoma.In this system,hyaluronic acid,the microneedle carrier,possesses good skin penetration ability and is approved by FDA as a pharmaceutical adjuvant;5-Fu is recommended by FDA for skin cancer treatment;ICG,a photothermal agent,possesses a strong photothermal ability and is approved by FDA for its use in the human body.We hypothesized that 5-Fu-ICG-MPEG-PCL could be delivered by the dissolvable microneedle through the skin,and the release behavior of the drug in the nanoparticle could be controlled by near-infrared light for achieving a single-dose cure of skin cancer,improving the cure rate of skin cancer and providing a new idea and possibility for the clinical treatment of skin cancer. | Ying Hao YuWen Chen XinLong He Fan Yang RuXia Han ChengLi Yang Wei Li ZhiYong Qian | 2020 | Bioactive Materials2020,5,3: | 17 |
| 2 | A group of SUVH methyl-DNA binding proteins regulate expression of the DNA demethylase ROS_1 in Arabidopsis显示文摘DNA methylation is typically regarded as a repressive epigenetic marker for gene expression.Genome-wide DNA methylation patterns in plants are dynamically regulated by the opposing activities of DNA methylation and demethylation reactions. In Arabidopsis, a DNA methylation monitoring sequence(MEMS) in the promoter of the DNA demethylase gene ROS_1 functions as a methylstat that senses these opposing activities and regulates genome DNA methylation levels by adjusting ROS_1 expression. How DNA methylation in the MEMS region promotes ROS_1 expression is not known. Here, we show that several Su(var)3-9 homologs(SUVHs) can sense DNA methylation levels at the MEMS region and function redundantly to promote ROS_1 expression. The SUVHs bind to the MEMS region, and the extent of binding is correlated with the methylation level of the MEMS.Mutations in the SUVHs lead to decreased ROS_1 expression, causing DNA hypermethylation at more than 1,000 genomic regions. | 4 | New normalized LMS adaptive filter with a variable regularization factor显示文摘A new normalized least mean square(NLMS) adaptive filter is first derived from a cost function, which incorporates the conventional one of the NLMS with a minimum-disturbance(MD)constraint. A variable regularization factor(RF) is then employed to control the contribution made by the MD constraint in the cost function. Analysis results show that the RF can be taken as a combination of the step size and regularization parameter in the conventional NLMS. This implies that these parameters can be jointly controlled by simply tuning the RF as the proposed algorithm does. It also demonstrates that the RF can accelerate the convergence rate of the proposed algorithm and its optimal value can be obtained by minimizing the squared noise-free posteriori error. A method for automatically determining the value of the RF is also presented, which is free of any prior knowledge of the noise. While simulation results verify the analytical ones, it is also illustrated that the performance of the proposed algorithm is superior to the state-of-art ones in both the steady-state misalignment and the convergence rate. A novel algorithm is proposed to solve some problems. | 7 | Mechanism and kinetics study on removal of Iron from phosphoric acid by cation exchange resin显示文摘Iron element is one of the main impurities in wet-process phosphoric acid and it has a significant impact on the subsequent phosphorus chemical products. This paper studied the feasibility of using Sinco-430 cation exchange resin for iron removal from phosphoric acid. The specific surface area and the total exchange capacity of resin were 8.91 m2·g-1 and 5.18 mmol·g-1, respectively. The sorption mechanism was determined by FTIR and XPS and the results indicated that iron was combined with-SO3 H in resin. The removal process was studied as a function of temperature, H3 PO4 content and mass ratio between resin and solution. The unit mass of resin to remove iron was 0.058 g·g-1 resin when the operating parameters were T = 50 ℃, H3 PO4 content = 27.61 wt%and S/L = 0.1, respectively. Kinetics study demonstrated that pseudo-second-order reaction model fits this study best and the calculated activation energy of overall reaction is 29.10 kJ·mol-1. | 11 | Biomechanical properties of acellular sciatic nerves treated with a modified chemical method显示文摘Nerve grafts are able to adapt to surrounding biomechanical environments if the nerve graft itself exhibits appropriate biomechanical properties (load, elastic modulus, etc.). The present study was designed to determine the differences in biomechanical properties between fresh and chemically acellularized sciatic nerve grafts. Two different chemical methods were used to establish acellular nerve grafts. The nerve was chemically extracted in the Sondell method with a combination of Triton X-100 (nonionic detergent) and sodium deoxycholate (anionic detergent), and in the modified method with a combination of Triton X-200 (anionic detergent), sulfobetaine-10 (SB-10, amphoteric detergents), and sulfobetaine-16 (SB-16, amphoteric detergents). Following acellularization, hematoxylin-eosin staining and scanning electron microscopy demonstrated that the effect of acellularization via the modified method was similar to the traditional Sondell method. However, effects of demyelination and nerve fiber tube integrity were superior to the traditional Sondell method. Biomechanical testing showed that peripheral nerve graft treated using the chemical method resulted in decreased biomechanical properties (ultimate load, ultimate stress, ultimate strain, and mechanical work to fracture) compared with fresh nerves, but the differences had no statistical significance (P > 0.05). These results demonstrated no significant effect on biomechanical properties of nerves treated using the chemical method.
