3p53 deficiency-induced Smad1 upregulation suppresses tumorigenesis and causes chemoresistance in colorectal cancers显示文摘The DNA damage response helps to maintain genome integrity,suppress tumorigenesis,and mediate the effects of radiotherapy and chemotherapy.Our previous studies have shown that Smad1 is upregulated and activated by Atm in DNA damage response,which can further bind to p53 and promote p53 stabilization.Herewe report another aspect of the interplay between p53 and Smad1.Comparison of rectal tumor against paired paraneoplastic specimens and analysis of>500 colorectal tumors revealed that Smad1 was upregulated in tumor samples,which was attributable to p53 defects.Using MEFs as a model,we found that knockdown of the elevated Smad1 in p532/2 MEFs promoted cell proliferation,E1A/Ras-induced cell transformation,and tumorigenesis.Mechanistic studies suggest that elevated Smad1 and momentary activation inhibit cell proliferation by upregulating p57Kip2 and enhancing Atm–Chk2 activation.Surprisingly,elevated Smad1 appears to have a negative effect on chemotherapy,as colorectal tumors,primary cancer cells,and cell lines with Smad1 knockdown all showed an increase in chemosensitivity,which could be attributable to elevated p57Kip2.These findings underscore the significance of Smad1–p53 interaction in tumor suppression and reveal an unexpected role for Smad1 in chemoresistance of colorectal cancers.Xinsen Ruan Qiao Zuo Hao Jia Jenny Chau Jinlin Lin Junping Ao Xuechun Xia Huijuan Liu Samy LHabib Chuangang Fu Baojie Li 2015Journal of Molecular Cell Biology2015,7,2:2

8Assessment of liver volume variation to evaluate liver function显示文摘In order to assess the value of liver volumetry in cirrhosis and acute liver failure(ALF)patients,we explored the correlation between hepatic volume and severity of the hepatic diseases.The clinical data of 48 cirrhosis patients with 60 normal controls and 39 ALF patients were collected.Computed tomography-derived liver volume(CTLV)and body surface area(BSA)of normal controls were calculated to get a regression formula for standard liver volume(SLV)and BSA.Then CTLV and SLV of all patients were calculated and grouped by Child-Turcotte-Pugh classification for cirrhosis patients and assigned according to prognosis of ALF patients for further comparison.It turned out that the mean liver volume of the control group was 1058±337 cm^(3).SLV was correlated with BSA according to the regression formula.The hepatic volume of cirrhosis patients in Child A,B level was not reduced,but in Child C level it was significantly reduced with the lowest liver volume index(CTLV/SLV).Likewise,in the death group of ALF patients,the volume index was significantly lower than that of the survival group.Based on volumetric study,we proposed an ROC(receiver operating characteristic)analysis to predict the prognosis of ALF patients that CTLV/SLV<83.9% indicates a poor prognosis.In conclusion,the CTLV/SLV ratio,which reflects liver volume variations,correlates well with the liver function and progression of cirrhosis and ALF.It is also a very useful marker for predicting the prognosis of ALF.Cong Tong Xinsen Xu Chang Liu Tianzheng Zhang Kai Qu 2012Frontiers of Medicine2012,6,4:1
9Pomegranate micro/nano hierarchical plasma structure for superior microwave absorption显示文摘Inspired by the pomegranate natural artful structure,pomegranate micro/nano hierarchical plasma configuration of Fe/Fe3C@graphitized carbon(FFC/pCL)was constructed based on the green sol-gel method and in-situ chemical vapor deposition(CVD)synthesis protocol.Pomegranate-like FFC/pCL successfully overcame the agglomeration phenomenon of magnetic nanoparticles with each seed of the pomegranate consisting of Fe/Fe_(3)C as cores and graphitized carbon layers as shells.The high-density arrangement of magnetic nanoparticles and the design of pomegranate-like heterostructures lead to enhanced plasmon resonance.Thus,the pomegranate-like FFC/pCL achieved a great electromagnetic wave(EMW)absorbing performance of 6.12 GHz wide band absorption at a low mass adding of only 16.7 wt.%.Such excellent EMW performance can be attributed to its unique pomegranate hierarchical plasma configuration with separated nanoscale iron cores,surface porous texture,and good carbon conductive network.This investigation provides a new paradigm for the development of magnetic/carbon based EMW absorbing materials by taking advantage of pomegranate hierarchical plasma configuration.Chunyan Ding Tao Wu Xinsen Hu Chengshuai Shao Zhipeng Xu Hui Fu Songsong Wu Guangwu Wen Xiaoxiao Huang 2022Nano Research2022,15,10:1
10Estimation of Probability of Damage Detection in Offshore Structural Inspection显示文摘- To characterize the uncertainty and fuzziness in offshore structural inspection, probability of detection (POD) must be determined. This paper presents the expressions for the POD of four different damage forms mainly existing in offshore structures. The fuzzy-set theory is applied to estimate human errors through the definition of inspection quality. Expressions of inspection quality are achieved. To verify the validity and correctness of the expressions, the data from an offshore platform field inspection of evaluation results of human errors affecting inspection quality are used to estimate the parameters of the POD. 13Femoral mechanical performance of precocial and altricial birds:a simulation study显示文摘Background:As the major load-bearing structures,bones exhibit various properties related to mechanical perfor-mance to adapt to different locomotor intensities.The habits and ontogenetic changes of locomotion in animals can,thus,be explored by assessing skeletal mechanical performance.Methods:In this study,we investigated the growing femoral mechanical performance in an ontogenetic series of Cabot’s Tragopans(Tragopan caboti)and Pigeons(Columba livia domestica).Micro-computed tomography-based finite element analysis was conducted to evaluate the stress,strain,and strain energy density(SED)of femora under axial and radial loading.Results:Femora deflected medio-laterally and dorso-ventrally under axial and radial loading,respectively.Femora deformed and tensed more severely under radial loading than axial loading.In adult individuals,Cabot’s Tragopans had lower strain and SED than pigeons.During ontogeny,the strain and SED of pigeons decreased sharply,while Cabot’s Tragopans showed moderately change.The structural properties of hatchling pigeons are more robust than those of hatchling Cabot’s Tragopans.Conclusions:Limb postures have dominant effect on skeletal deformation.The erect posture is preferred by large mammals and birds to achieve a high safety factor of bones during locomotion.Adult Cabot’s Tragopans have stronger femora than pigeons,reflecting a better bone adaption to the terrestrial locomotion of the studied pheas-ant species.Changes in strain and SED during growth reflect the marked difference in locomotor ability between precocial and altricial hatchlings.The femora of hatchling Cabot’s Tragopans were built with better energy efficiency than deformation resistance,enabling optimized mechanical performance.In contrast,although weak in mechani-cal function at the time of hatching,pigeon femora were suggested to be established with a more mature structural design as a prerequisite for rapid growth.These results will be helpful for studies regarding developmental patterns of fossil avian species.Xinsen Wei Zihui Zhang 2021Avian Research2021,12,2:1

15Model-based assessment of muscle forces and strain distribution in the femur of Cabot’s Tragopans(Tragopan caboti)显示文摘The hindlimbs play a crucial role in bird locomotion,making the biomechanical properties of the musculoskeletal system in these limbs a focal point for researchers studying avian behaviour.However,a comprehensive analysis of the mechanical performance within the long bones of hindlimbs during locomotion remains lacking.In the present study,the strain and deformation of the femur of Cabot’s Tragopans(Tragopan caboti)were estimated.We employed inverse simulation to calculate the force and moment of femoral muscles during mid-stance terrestrial locomotion and conducted finite element analysis to calculate femoral strain.Results showed that during mid-stance,the femur experiences combined deformation primarily characterized by torsion,bending,and compression.It emphasises the importance of considering the influence of varying loads on bone adaptation when investigating bone form-function relationships.Muscles were found to play a significant role in offsetting joint loads on the femur,subsequently reducing the deformation and overall strain on the bone.This reduction enhances femoral safety during locomotion,allowing birds to meet mechanical demands while maintaining a lightweight bone structure.Notably,the M.iliotrochantericus caudalis significantly reduces torsional deformation of the proximal femur,protecting the vulnerable femoral neck from high fracture risk induced by rotation load.Given that the femur torsion during terrestrial locomotion in birds is associated with changes in hindlimb posture due to their adaptation to flight,the characteristics of M.iliotrochantericus caudalis may provide insight into the locomotor evolution of theropods and the origin of avian flight.Xinsen Wei Zihui Zhang 2023Avian Research2023,14,4:0
16Boosting Kernel Search Optimizer with Slime Mould Foraging Behavior for Combined Economic Emission Dispatch Problems显示文摘Reducing pollutant emissions from electricity production in the power system positively impacts the control of greenhouse gas emissions.Boosting kernel search optimizer(BKSO)is introduced in this research to solve the combined economic emission dispatch(CEED)problem.Inspired by the foraging behavior in the slime mould algorithm(SMA),the kernel matrix of the kernel search optimizer(KSO)is intensified.The proposed BKSO is superior to the standard KSO in terms of exploitation ability,robustness,and convergence rate.The CEC2013 test function is used to assess the improved KSO's performance and compared to 11 well-known optimization algorithms.BKSO performs better in statistical results and convergence curves.At the same time,BKSO achieves better fuel costs and fewer pollution emissions by testing with four real CEED cases,and the Pareto solution obtained is also better than other MAs.Based on the experimental results,BKSO has better performance than other comparable MAs and can provide more economical,robust,and cleaner solutions to CEED problems.Ruyi Dong Lixun Sun Long Ma Ali Asghar Heidari Xinsen Zhou Huiling Chen 2023Journal of Bionic Engineering2023,20,6:0

19Reconstruction of the Hindlimb Locomotion of Confuciusornis (Aves) and Its Implication for the Origin of Avian Flight显示文摘As one of the most basal avian clades,the Confuciusornithids are ideal in revealing the early evolution of avianflight.Birds’hindlimbs are functionally diverse and contain a wealth of information about their behavior.The hindlimb of Confuciusornis,however,has only been studied in detail in terms of functional morphology,and quantitative studies that directly assess locomotor ability are relatively lacking.This has led to certain controver-sies on the behavior of Confuciusornis.This paper reviews the debates over the life habits and take-off ability of Confuciusornis,which are closely related to their hindlimb function.Several methodologies adopted engineering techniques,including the geometrical analysis of long bones,physiological reconstruction of muscles,kinematic and kinetic characteristics estimating,and appendage locomotor mechanism analysis,are recommended for esti-mating the hindlimb functions of Confuciusornis.Considering that the fossil bones are fragile,irregular in shape,and usually deformed,it is appropriate to apply computer numerical simulation techniques to such studies.A sufficient functional quantitative study will help clarify early bird locomotor behavior,which will provide clues and evidence for further exploration of the origin of birdflight and early bird movement.Xinsen Wei Ying Guo Yan Zhao 2023Molecular & Cellular Biomechanics2023,20,2:0
20Epidemiological study of a von Hippel-Lindau family in northwest China显示文摘von Hippel-Lindau(VHL)disease is a rare,inherited neoplastic disease characterized by hemangioblastomas(HBL)of the central nervous system(CNS),retinal angiomas,renal cell carcinomas(RCC),pancreatic endocrine tumors(PETs),pheochromocytomas,paragangliomas,and visceral cysts.We encountered a large VHL family in northwest China and conducted a systematic screening of the family members based on their epidemiological and clinical characteristics.A self-designed questionnaire was used to collect the general sociodemographic and health information of the family members.For the preliminary family screening,physical examination and abdomen B ultrasonography were performed.The suspected patients were subjected to cranial computerized tomography and fundus examination.The clinical data of the patients with confirmed VHL disease were collected from hospital records.A total of 63 lineal descendants in six generations were observed in the family(generations O,A,B,C,D,E),including 9 dead suspected cases(6 males,3 females)and 10 living cases(2 males,8 females).Among the 10 living cases,4,2,1,3,4,8,and 2 manifested HBLs of the CNS,PETs,RCC,pancreatic cysts,renal cysts,pheochromocytomas(4 hemi and 4 bilateral),and paragangliomas,respectively.Data showed that the morbidity of VHL disease in generation C was lower than that in generation B,but the age of onset was younger.This study is the first to report VHL disease in northwest China and VHL-associated PET cases in Chinese.Therefore,follow-up checkups of the family should be focused on younger generations.Proper family screening protocols should be followed for the treatment of patients with VHL disease.Jingyao Zhang Dapeng Wu Hong Ai Jigang Bai Shunbin Dong Qinling Yang Kai Qu Lei Zhou Xinsen Xu Chang Liu 2013Frontiers of Medicine2013,7,3:0
