| 1 | Natural Variation in CCD4 Promoter Underpins Species-Specific Evolution of Red Coloration in Citrus Peel显示文摘Carotenoids and apocarotenoids act as phytohormones and volatile precursors that influence plant development and confer aesthetic and nutritional value critical to consumer preference.Citrus fruits display considerable natural variation in carotenoid and apocarotenoid pigments.In this study,using an integrated genetic approach we revealed that a 5;c/s-regulatory change at CCD4b encoding CAROTENOID CLEAVAGE DIOXYGENASE 4b is a major genetic determinant of natural variation in C3 0 apocarotenoids responsible for red coloration of citrus peel.Functional analyses demonstrated that in addition the known role in synthesizing 3-citraurin,CCD4b is also responsible for the production of another important C3 0 apocarotenoid pigment,p-citraurinene.Furthermore,analyses of the CCD4b promoter and transcripts from various citrus germplasm accessions established a tight correlation between the presence of a putative 5'c/s-regulatory enhancer within an MITE transposon and the enhanced allelic expression of CCD4b in C3 0 apocarotenoid-rich red-peeled accessions.Phylogenetic analysis provided further evidence that functional diversification of CCD4b and naturally occurring variation of the CCD4b promoter resulted in the stepwise evolution of red peels in mandarins and their hybrids.Taken together,our findings provide new insights into the genetic and evolutionary basis of apocarotenoid diversity in plants,and would facilitate breeding efforts that aim to improve the nutritional and aesthetic value of citrus and perhaps other fruit crops. | Xiongjie Zheng Kaijie Zhu Quan Sun Weiyi Zhang Xia Wang Hongbo Cao Meilian Tan Zongzhou Xie Yunliu Zeng Junli Ye Lijun Chai Qiang Xu Zhiyong Pan Shunyuan Xiao Paul D.Fraser Xiuxin Deng | 2019 | Molecular Plant2019,12,9: | 18 |

| 4 | Genome-wide Characterization of cis-acting Elements in the Promoters of Key Carotenoid Pathway Genes from the Main Species of Genus Citrus显示文摘Carotenoids are indispensable for both human health and plant survival.Citrus,is one of the fruit crops richest in carotenoid compounds,with approximately 115 kinds of carotenoids;tremendous diversity in carotenoids composition and concentration exists among various species,showing different colors from nearly white to crimson.The carotenoid biosynthetic pathway and the key carotenogenic genes have been identified in citrus;however,the underlying regulatory mechanisms remain unclear.In this study,among the main species of genus Citrus(primitive,wild,and cultivated),we detected carotenoids in flavedo using High-Performance Liquid Chromatography,and analyzed variations in cis-acting elements in the promoters of key carotenoid pathway genes.Intriguingly,both carotenoid composition and content were generally increased during the evolution of citrus,and the corresponding variations in the promoters were identified,including the gain or loss of critical environmental stress-responsive elements and hormone-responsive elements,which are closely associated with carotenoid enhancement.In addition,pummelo has the most heat-responsive elements,but the Mangshan mandarin does not have this element in the promoters of PSY,which is highly related to their geographical origin and indicate that temperature is a critical environmental signal influencing carotenoid accumulation.Moreover,the abscisic acid-responsive motif was rich in almost all the seven species,but the ethylene-responsive motif was deficient,which demystified the unique phytohormone regulation mechanism of carotenoid accumulation in citrus.Overall,our study provides new insights into the molecular regulatory mechanism of carotenoid enhancement in the evolution of citrus,which can facilitate breeding and cultivation efforts to improve the nutritional quality and esthetic value in citrus and hopefully other fruit crops. | Kaijie Zhu Qingjiang Wu Yue Huang Junli Ye Qiang Xu Xiuxin Deng | 2020 | Horticultural Plant Journal2020,6,6: | 8 |

| 6 | Systematic review and meta analysis of randomized controlled trials on Tianmagouteng decoction in treatment of primary hypertension with liverYang hyperactivity syndrome显示文摘OBJECTIVE: To assess the efficacy of Tianma-gouteng decoction in the treatment of primary hypertension with liver yang hyperactivity syndrome. METHODS: The databases of MEDLINE (1950-2012), EMbase (1980-2012), China biology medicine (CBM) (1994-2012), China national knowledge infrastructure (CNKI) (1989-2012), Weipu (VIP) (1995-2012) and Wanfang (1989-2012) were searched and we performed manual searches for conference proceedings to select randomized controlled trials (RCTs) on Tianmagouteng decoction for the treatment of primary hypertension with liver Yang hyperactivity syndrome. Studies were enrolled according to the study-selecting criteria. The methodological quality was evaluated, data were extracted and meta-analyses were conducted with RevMan 4.2 software. RESULTS: Nine RCTs were selected involving 784 patients with no significant difference in decrease in systolic blood pressure (SBP) [OR=1.02, 95% CI (-0.24, 2.29), P=0.10] and diastolic blood pressure(DBP) [OR=0.56, 95% CI (-0.10,1.23), P=0.11] between the Tianmagouteng group and the control group. While there was a significantly larger increase in serum superoxide dismutase (SOD)[OR= 6.26, 95% CI (1.27,11.66), P=0. 01] in the Tianma-gouteng group than that in the control group CONCLUSION: Tianmagouteng decoction can decrease both systolic and diastolic blood pressure and greatly increase serum SOD. | 7 | Altered expression of mitochondrial related genes in the native Tibetan placents by mitochondrial cDNA array analysis显示文摘Objective: To explore the mechanism of native Tibetan fetuses adaptation to hypoxia, we tried to find the different expression genes about mitochondrial function in the native Tibetan placents. Methods: In this study, the placents of native Tibetan and the high-altitude Han (ha-Han) were collected. After the total RNA extraction, the finally synthesized cDNAs were hybridized to mitochondrial array to find the altered expression genes between them. Then, the cytochrome c oxidase 17 (Cox17), dynactin 2 (DCTN2, also known as p50), and vascular endothelial growth factor receptor (VEGFR, also known as KDR) were chosen from the altered expression genes to further verify the array results using the SYBR Green real-time PCR. Because the altered expression genes (such as Cybb and Cox17) in the array results related to the activities of COXI and COXIV, the placental mitochondria activities of COXI and COXIV were measured to find their changes in the hypoxia. Results: By a standard of ≥1.5 or ≤0.67, there were 24 different expressed genes between the native Tibetan and the ha-Han placents, including 3 up-regulated genes and 21 down-regulated genes. These genes were related to energy metabolism, signal transduction, cell proliferation, electron transport, cell adhesion, nucleotide-excision repair. The array results of Cox17, DCTN2 and KDR were further verified by the real-time RT-PCR. Through the mitochondria respiration measurements, the activity of COXI in the native Tibetan placents were higher than that of ha-Han, there was no difference in COXIV activity between them. | 8 | Difference of a citrus late-ripening mutant (Citrus sinensis) from its parental line in sugar and acid metabolism at the fruit ripening stage显示文摘‘Fengjiewancheng’(FW) (Citrus sinensis), a bud sport of‘Fengjie 72-1’navel orange (FJ), ripens one month later than its parental line. Differences in sugar and acid content and the transcript level of su-crose- and citric-metabolic enzymes for the two cultivars were investigated during fruit ripening. Re-sults showed that both sugar and acid metabolisms of the mutant were affected by the mutation. In the pulp of FW, sugar content was significantly lower than that in FJ before 227 DAF (days after flowering) and higher at 263 DAF; the mutant’s gene expression of one isoform of citrus sucrose synthase (CitSS1) was delayed, and its gene expression of citrus acid invertase (CitAI) was stronger than that in its pa-rental cultivars at 207 and 263 DAF. In the peel, only the sucrose content in FW was significantly lower than those in FJ at the early periods of fruit ripening (165 and 187 DAF); however the transcripts of the sucrose-cleaving enzymes in the mutant were higher than those in FJ at different ripening points. As regards acid accumulation in the two cultivars, it was observed that in the pulp of the mutant, the malic acid content was significantly lower than that in its parental cultivars from 187 to 263 DAF, and in the peel, remarkably higher during the whole fruit ripening period. The citric acid content in both the pulp and the peel of FW was higher than that in those of FJ during the early ripening period and lower during the late ripening period, which were correspondingly associated in part with the higher transcript level of citrus mitochondrial citrate synthase (CitCS) and with lower or undetectable transcript level of citrus cytosolic aconitase (CitAC). | 17 | Genetic diversity and relatedness analysis of nine wild species of tree peony based on simple sequence repeats markers显示文摘Tree peony has nine wild species,but the evolutionary relationship of them is still unclear.Here,a total of 274 specimens from 22 natural populations of nine wild species were collected,and their genetic diversity and similarity was analyzed based on Simple Sequence Repeats(SSR)molecular markers.A total of 106 alleles were generated based on 20 primers and with an average of 5.3 alleles per primer.Shannon’s information index(I)ranged from 0.6333 to 1.7842,and the average of Nei’s genetic diversity coefficient(H)was 0.5771.Polymorphism Information Content(PIC)value varied from 0.29 to 0.77,ten of these primers had high polymorphism(PIC≥0.50).All the above genetic parameters of primers reflect more rich genetic diversity information compared with other researches using SSR molecular markers to study the genetic diversity of tree peony wild species.At the population level,the lowest and highest degree of genetic diversity occurred in Paeonia ludlowii-P1 and P.delavayi-P3 population,respectively.Whereas at species level,the genetic diversity of 9 wild peony species was as follows:P.lutea>P.delavayi>P.rockii>P.qiui>P.ostii>P.decomposita>P.potaninii>P.spontanea>P.ludlowii.Furthermore,cluster analysis at species level divided the nine wild tree peony species into two branches.In branch I,the closest phylogenetic relationship was found between P.ostii and P.rockii,followed by P.spontanea,P.qiui,and P.decomposita.In branch II,the closest relationship occurred between P.lutea and P.delavayi,followed by P.potaninii and P.ludlowii.Clustering results supported the division of tree peonies into two subsects(Delavayanae and Vaginatae),it also supported P.potaninii and P.ludlowii as independent species.The results provided novel insight into the genetic diversity and phylogenetic relationship of nine wild tree peony species.It will help formulate comprehensive protection measures of wild germplasm resources and select proper parents for distant hybridization in the future. | Yuqian Xue Rong Liu Jingqi Xue Shunli Wang Xiuxin Zhang | 2021 | Horticultural Plant Journal2021,7,6: | 1 |

| 19 | Pro-inflammatory effects of a litchi protein extract in murine RAW264.7 macrophages显示文摘It has been observed that the consumption of litchi often causes symptoms characterized by itching or sore throat,gum swelling,oral cavity ulcers and even fever and inflammation,which significantly impair the quality of life of a large population.Using the RAW264.7 cell line,a step-by-step strategy was used to screen for the components in litchi fruits that elicited adverse reactions.The adverse reaction fractions were identified by mass spectrometry and analyzed using the SMART program,and a sequence alignment of the homologous proteins was performed.MTT tests were used to determine the cytotoxicity of a litchi protein extract in RAW264.7 macrophages,and real-time PCR was applied to analyze the expression of inflammatory genes in the RAW264.7 cells treated with lipopolysaccharide or the litchi protein extract.The results showed that the litchi water-soluble protein extract could increase the production of the pro-inflammatory mediators IL-1β,iNOS and COX-2,and the anti-inflammatory mediator HO-1 in the RAW264.7 cell line.The 14-3-3-like proteins GF14 lambda,GF14 omega and GF14 upsilon were likely the candidate proteins that caused the adverse effects. | Xiaoli Wang Xiaorong Hu Huiqing Yan Zhaocheng Ma Xiuxin Deng | 2016 | Horticulture Research2016,3,1: | 1 |
