1m^6A Regulates Neurogenesis and Neuronal Development by Modulating Histone Methyltransferase Ezh2显示文摘N6-methyladenosine (m6A),catalyzed by the methyltransferase complex consisting of Mettl3 and Mettl14,is the most abundant RNA modification in mRNAs and participates in diverse biological processes. However,the roles and precise mechanisms of m6A modification in regulating neuronal development and adult neurogenesis remain unclear. Here,we examined the function of Mettl3,the key component of the complex,in neuronal development and adult neurogenesis of mice. We found that the depletion of Mettl3 significantly reduced m6A levels in adult neural stem cells (aNSCs) and inhibited the proliferation of aNSCs. Mettl3 depletion not only inhibited neu-ronal development and skewed the differentiation of aNSCs more toward glial lineage,but also affected the morphological maturation of newborn neurons in the adult brain. m6A immunoprecip-itation combined with deep sequencing (MeRIP-seq) revealed that m6A was predominantly enriched in transcripts related to neurogenesis and neuronal development. Mechanistically,m6A was present on the transcripts of histone methyltransferase Ezh2,and its reduction upon Mettl3 knockdown decreased both Ezh2 protein expression and consequent H3K27me3 levels. The defects of neurogenesis and neuronal development induced by Mettl3 depletion could be rescued by Ezh2 overexpression. 2Jointly Optimized Request Dispatching and Service Placement for MEC in LEO Network显示文摘Integrating Multi-access Edge Computing(MEC) in Low Earth Orbit(LEO) network is an important way to provide globally seamless low-delay service. In this paper, we consider the scenario that MEC platforms with computation and storage resource are deployed on LEO satellites, which is called 'LEO-MEC'. Service request dispatching decision is very important for resource utilization of the whole LEO-MEC system and Qo E of MEC users. Another important problem is service placement that is closely coupled with request dispatching. This paper models the joint service request dispatching and service placement problem as an optimization problem, which is a Mixed Integer Linear Programming(MILP). Our proposed mechanism solves this problem and uses the solved decision variables to dispatch requests and place services. 3Compositional dependence of microstructure and tribological properties of plasma sprayed Fe-based metallic glass coatings显示文摘Gas-atomized powders of three Fe-based glass-forming alloys were sprayed on mild steel substrates by atmospheric plasma spaying using the same spaying parameters. Microstructures, thermal stabilities and tribological properties of the sprayed coatings were analyzed. The coating performances showed a strong dependence on the intrinsic characters of the compositions, i.e., glass-forming ability (GFA) and supercooled liquid region (?Tx). The coatings tended to exhibit higher amorphous phase fraction for the composition with higher GFA and lower porosity for that with larger ?Tx. All the coatings exhibited superior wear resistance compared with the substrate. Higher wear resistance could be obtained in coatings with higher amorphous phase fraction, i.e. higher GFA of the composition. 4LIM domain proteins Pinch1/2 regulate chondrogenesis and bone mass in mice显示文摘The LIM domain-containing proteins Pinch1/2 regulate integrin activation and cell–extracellular matrix interaction and adhesion.Here,we report that deleting Pinch1 in limb mesenchymal stem cells(MSCs)and Pinch2 globally(double knockout;dKO)in mice causes severe chondrodysplasia,while single mutant mice do not display marked defects.Pinch deletion decreases chondrocyte proliferation,accelerates cell differentiation and disrupts column formation.Pinch loss drastically reduces Smad2/3 protein expression in proliferative zone(PZ)chondrocytes and increases Runx2 and Col10a1 expression in both PZ and hypertrophic zone(HZ)chondrocytes.Pinch loss increases sclerostin and Rankl expression in HZ chondrocytes,reduces bone formation,and increases bone resorption,leading to low bone mass.In vitro studies revealed that Pinch1 and Smad2/3 colocalize in the nuclei of chondrocytes.Through its C-terminal region,Pinch1 interacts with Smad2/3 proteins.Pinch loss increases Smad2/3 ubiquitination and degradation in primary bone marrow stromal cells(BMSCs).Pinch loss reduces TGF-β-induced Smad2/3 phosphorylation and nuclear localization in primary BMSCs.Interestingly,compared to those from single mutant mice,BMSCs from dKO mice express dramatically lower protein levels ofβ-catenin and Yap1/Taz and display reduced osteogenic but increased adipogenic differentiation capacity.Finally,ablating Pinch1 in chondrocytes and Pinch2 globally causes severe osteopenia with subtle limb shortening.Collectively,our findings demonstrate critical roles for Pinch1/2 and a functional redundancy of both factors in the control of chondrogenesis and bone mass through distinct mechanisms.Yiming Lei Xuekun Fu Pengyu Li Sixiong Lin Qinnan Yan Yumei Lai Xin Liu Yishu Wang Xiaochun Bai Chuanju Liu Di Chen Xuenong Zou Xu Cao Huiling Cao Guozhi Xiao 2020Bone Research2020,8,4:2

9Antioxidant effects of the aqueous extract of turmeric against hydrogen peroxide-induced oxidative stress in spotted seabass(Lateolabrax maculatus)显示文摘Intensive aquaculture-induced oxidative stress is detrimental to fish health and yield.Medicinal plants show promise as natural health boosters and antioxidants in the aquaculture industry.Therefore,this work investigated the effects of turmeric aqueous extract(TAE)on the growth performance,antioxidant status,and hydrogen peroxide(H2O2)-induced oxidative stress in spotted seabass(Lateolabrax maculatus).Fish were fed diets supplemented with 0(Con),2(TAE2),or 4(TAE4)g/kg TAE for eight weeks,then were injected with H2O2.The results showed that dietary supplementation of TAE did not affect fish growth,feed utilization,or body composition.TAE treatment increased liver antioxidant enzyme activities and decreased liver malondialdehyde content and serum levels of glutamate oxalate transaminase,glutamate pyruvate transaminase,and lactate dehydrogenase.Furthermore,the increases in mortality,liver malondialdehyde content,and serum biomarkers of liver injury in the H2O2-treated fish were inhibited as a consequence of the TAE treatment.In addition,TAE treatment activated the Nrf2/Keap1 pathway in the liver,supported by the up-regulated expression of nrf2,ho-1,and gclc,and down-regulated keap1 expression.Overall,dietary incorporation of TAE protected against H2O2-induced oxidative stress in spotted seabass probably by enhancing antioxidant capacity through the Nrf2/Keap1 pathway.Zhe Wang Xiuqin Wang Xueshan Li Kangle Lu Ling Wang Xuekun Ma Kai Song Chunxiao Zhang 2024Aquaculture and Fisheries2024,9,1:1

13Estimated HCFC-142b emissions in China: 2000–2050显示文摘1-Chloro-1,1-difluoroethane(HCFC-142b)was both ozone depleting substance under restriction of the Montreal Protocol on Substances that Deplete the Ozone Layer(Montreal Protocol)and potent greenhouse gas with high GWP.Controlling its emissions in China will contribute to both mitigating climate change and protecting ozone.A national emission inventory of HCFC-142b for China during 2000–2012 was established and projected to2050 based on the 2006 IPCC Guidelines for National Greenhouse Gas Inventories and the Montreal Protocol,showing that(i)in contrast to the downward trend revealed by existing researches,HCFC-142b emissions kept increasing from 0.1 kt/y in 2000 to the peak of 14.4 kt/y in2012,making China a crucial contributor to global HCFC-142b emissions and(ii)for future emission projections,a continuous increase from 14.9 kt/y in 2013 to 97.2 kt/y in2050 was anticipated under the business-as-usual(BAU)scenarios,while a reduction of about 90%of the projected BAU emissions would be obtained by fulfilling the Montreal Protocol,namely an accumulative mitigation of 1578kt HCFC-142b from 2013 to 2050,equal to 103 kt ODP and 3504 Tg CO2emissions.Emissions from each province in 2012 were also estimated to identify key emission areas.Among the 31 mainland provinces in China(Hong Kong,Macao,and Taiwan were not included),Jiangsu,Zhejiang,Shandong,and Guangdong had the highest emission ratesin 2012(2.06,1.85,1.52,and 1.04 kt/y,respectively);Zhejiang,Jiangsu,and Shanghai exhibit the strongest emission strength(0.83,0.59,and 0.54 t/km2,respectively),much higher than the average national level of 0.33t/km2.Jiarui Han Li Li Shenshen Su Jing Wu Xuekun Fang Shenglan Jia Jianbo Zhang Jianxin Hu 2014Chinese Science Bulletin2014,59,24:1

16Framework and development of data-driven physics based model with application in dimensional accuracy prediction in pocket milling显示文摘In the manufacturing of thin wall components for aerospace industry,apart from the side wall contour error,the Remaining Bottom Thickness Error(RBTE)for the thin-wall pocket component(e.g.rocket shell)is of the same importance but overlooked in current research.If the RBTE reduces by 30%,the weight reduction of the entire component will reach up to tens of kilograms while improving the dynamic balance performance of the large component.Current RBTE control requires the off-process measurement of limited discrete points on the component bottom to provide the reference value for compensation.This leads to incompleteness in the remaining bottom thickness control and redundant measurement in manufacturing.In this paper,the framework of data-driven physics based model is proposed and developed for the real-time prediction of critical quality for large components,which enables accurate prediction and compensation of RBTE value for the thin wall components.The physics based model considers the primary root cause,in terms of tool deflection and clamping stiffness induced Axial Material Removal Thickness(AMRT)variation,for the RBTE formation.And to incorporate the dynamic and inherent coupling of the complicated manufacturing system,the multi-feature fusion and machine learning algorithm,i.e.kernel Principal Component Analysis(kPCA)and kernel Support Vector Regression(kSVR),are incorporated with the physics based model.Therefore,the proposed data-driven physics based model combines both process mechanism and the system disturbance to achieve better prediction accuracy.The final verification experiment is implemented to validate the effectiveness of the proposed method for dimensional accuracy prediction in pocket milling,and the prediction accuracy of AMRT achieves 0.014 mm and 0.019 mm for straight and corner milling,respectively.Zhanying CHEN Liping WANG Jiabo ZHANG Guoqiang GUO Shuailei FU Chao WANG Xuekun LI 2021Chinese Journal of Aeronautics2021,34,6:1

18Global methyl halide emissions from biomass burning during 2003-2021显示文摘Methyl halides(CH3Cl,CH3Br,and CH3I)are ozone-depleting substances.Biomass burning(BB)is an important source of methyl halides.The temporal variations and global spatial distribution of BB methyl halide emissions are unclear.Thus,global methyl halide emissions from BB during 2003e2021 were estimated based on satellite data.A significant decreasing trend(p<0.01)in global methyl halide emissions from BB was found between 2003 and 2021,with CH3Cl emissions decreasing from 302 to 220 Gg yr^(-1),CH3Br emissions decreasing from 16.5 to 11.7 Gg yr^(-1),and CH3I emissions decreasing from 8.9 to 6.1 Gg yr^(-1).From a latitudinal perspective,the northern high-latitude region(60e90N)was the only latitude zone with significant increases in BB methyl halide emissions(p<0.01).Based on an analysis of the drivers of BB methyl halide emissions,emissions from cropland,grassland,and shrubland fires were more correlated with the burned area,while BB emissions from forest fires were more correlated with the emissions per unit burned area.The non-BB emissions of CH3Cl increased from 4749 Gg yr^(-1)in 2003 to 4882 Gg yr^(-1)in 2020,while those of CH3Br decreased from 136 Gg yr^(-1)in 2003 to 118 Gg yr^(-1)in 2020(global total CH3I emissions are not available).The finding indicates that global CH3Cl and CH3Br emissions from sources besides BB increased and decreased during 2003e2020.Based on our findings,not only searching for unknown sources is important,but also re-evaluating known sources is necessary for addressing methyl halide emissions.Xiaoyi Hu Di Chen Liting Hu Bowei Li Xinhe Li Xuekun Fang 2023Environmental Science and Ecotechnology2023,,2:0

20Gridded usage inventories of chlordane in China显示文摘Abstract Chlordane (1,2,4,5,6,7,8,8-octachloro-3a,4,7, 7a-tetra-hydro-4,7-methanoindane) is one of organochlor- ine pesticides (OCPs) which has been listed as one of the persistent organic pollutants (POPs) to be reduced and finally eliminated in the Stockholm Convention on Persistent Organic Pollutants, because of its great persistence, toxicity, bio-accumulation and long-range transport potential. It is critical to create a national chlordane usage inventories for China to compile chlordane emission inventories, which is helpful for carrying out risk assessments and other researches related to chlordane in China. The annual data of chlordane usage was calculated and modified in accordance with the reported annual production of chlordane which was caculated on the basis of the termite distribution, the data of chlordane usage rate and the annual new construction area (NCA). With the help of Geographic Information System, the usage data of this NCA were allocated to a grid system then, with a 1/4° longitude by 1/6°latitude resolution and a size for each grid cell of approximately 25 km by 25 km. Between 1988 and 2008, the total usage of chlordane in China was 2745 t, accounting for approximately 80% of the production in the same period. Zhejiang Province was the largest consumer of chlordane in China, whose usage adds up to 980 t, greatly exceeding other provinces/regions, followed by Jiangsu Province (534 t) and Sichuan Province (428 t). The region with the least usage of chlordane was Beijing. Provinces of Guizhou, Henan and Hebei did not use any chlordane, even though termites had occurred in these provinces. Gridded usage inventories showed that the intensive use of chlordane was concentrated in the southeast part of China, Yangtze River Delta and Pearl River Delta in particular.
