2Incorporation of silica nanoparticles to PLGA electrospun fibers for osteogenic differentiation of human osteoblast-like cells显示文摘The development of bone tissue engineering scaffolds still remains a challenging field,although various biomaterials have been developed for this purpose.Electrospinning is a promising approach to fabricate nanofibers with an interconnected porous structure,which can support cell adhesion,guide cell proliferation and regulate cell differentiation.The aim of this study is to fabricate composite fibers composed of poly(lactic-co-glycolic acid)(PLGA)and silica nanoparticles(NPs)via electrospinning and investigate the effect of PLGA/SiO_(2)composite fibers on the cellular response of osteoblast-like cells(SaOS-2 cells).SEM and EDX analysis showed that silica NPs were homogenously dispersed in the composite fibers.The mechanical behavior of the fibers showed that silica NPs acted as reinforcements at concentrations of 2.5 and 5 mg/ml.The incorporation of silica NPs led to enhancement of cell attachment and spreading on PLGA/SiO_(2)composite fibers.SaOS-2 cells cultured on PLGA/SiO_(2)composite fibers exhibited increased alkaline phosphatase activity,collagen secretion and bone nodules formation.The bone nodules formation of SaOS-2 cells increased along with the amount of incorporated silica NPs.The present findings indicate that PLGA/SiO_(2)composite fibers can stimulate osteogenic differentiation of SaOS-2 cells and may be a promising candidate scaffold for bone tissue engineering.Xing Yang Yuanyuan Li Xujie Liu Qianli Huang Ranran Zhang Qingling Feng 2018Regenerative Biomaterials2018,5,4:4
3Targeting a novel inducible GPX4 alternative isoform to alleviate ferroptosis and treat metabolic-associated fatty liver disease显示文摘Metabolic-associated fatty liver disease(MAFLD),which is previously known as non-alcoholic fatty liver disease(NAFLD),represents a major health concern worldwide with limited therapy.Here,we provide evidence that ferroptosis,a novel form of regulated cell death characterized by iron-driven lipid peroxidation,was comprehensively activated in liver tissues from MAFLD patients.The canonical-GPX4(cGPX4),which is the most important negative controller of ferroptosis,is downregulated at protein but not mRNA level.Interestingly,a non-canonical GPX4 transcript-variant is induced(inducible-GPX4,iGPX4)in MAFLD condition.The high fat-fructose/sucrose diet(HFFD)and methionine/choline-deficient diet(MCD)-induced MAFLD pathologies,including hepatocellular ballooning,steatohepatitis andfibrosis,were attenuated and aggravated,respectively,in cGPX4-and iGPX4-knockin mice.cGPX4 and iGPX4 isoforms also displayed opposing effects on oxidative stress and ferroptosis in hepatocytes.Knockdown of iGPX4 by siRNA alleviated lipid stress,ferroptosis and cell injury.Mechanistically,the triggered iGPX4 interacts with cGPX4 to facilitate the transformation of cGPX4 from enzymatic-active monomer to enzymatic-inactive oligomers upon lipid stress,and thus promotes ferroptosis.Co-immunoprecipitation and nano LC–MS/MS analyses confirmed the interaction between iGPX4 and cGPX4.Our results reveal a detrimental role of non-canonical GPX4 isoform in ferroptosis,and indicate selectively targeting iGPX4 may be a promising therapeutic strategy for MAFLD.Jie Tong Dongjie Li Hongbo Meng Diyang Sun Xiuting Lan Min Ni Jiawei Ma Feiyan Zeng Sijia Sun Jiangtao Fu Guoqiang Li Qingxin Ji Guoyan Zhang Qirui Shen Yuanyuan Wang Jiahui Zhu Yi Zhao Xujie Wang Yi Liu Shenxi Ouyang Chunquan Sheng Fuming Shen Pei Wang 2022Acta Pharmaceutica Sinica B2022,12,9:3
4Calcium carbonate nanoparticles promote osteogenesis compared to adipogenesis in human bone-marrow mesenchymal stem cells显示文摘Rhombohedron-like and fusiform calcium carbonate nanoparticles were fabricated using a new method. Their geometry was controlled by varying the mixing speed and ratio of ethanol versus water in reaction system. The calcium carbonate nanoparticles(CCNPs) have slight effect on viability of human bone-marrow mesenchymal stem cells(hBMSCs) with dose-dependent and shape-dependent, but they can significantly promote osteogenic differentiation of hBMSCs in vitro by 10–37% increase of alkaline phosphatase(ALP) activity, 9–36% growth of collagen secretion and 1.13–1.83 folds upregulation of osteogenesis-related genes, even at lower dose ranges(5–20 μg/ml). The efficacity of promoting osteogenesis depends on the shape and dose of CCNPs. Furthermore,adipogenesis was inhibited by less accumulation of lipid droplets, lower triglyceride(TG) secretion and downregulation of adipogenesis-related genes. 6A simple daily dynamic feeding regimen for reducing phosphorus consumption and excretion in laying hens显示文摘Phosphorus metabolism in laying hens is a highly dynamic process over the course of the 24 h egg-laying cycle.Adjusting the phosphorus feeding regimen according to the daily egg-laying cycle may help to improve phosphorus utilization efficiency.Hy-Line Brown layers(n=120;70 wk old)were offered 4different phosphorus daily regimens:(1)RR,fed regular phosphorus at both 09:00 and 17:00;(2)RL,fed regular phosphorus at 09:00 and low phosphorus at 17:00;(3)LR,fed low phosphorus at 09:00 and regular phosphorus at 17:00;(4)LL,fed low phosphorus at both 09:00 and 17:00.The regular and low phosphorus diets contained 0.32% and 0.14% non-phytate phosphorus,respectively.The feeding trial lasted for 12 wk.As a result,layers on the RL regimen had decreased laying rate(P<0.05;5 to 8,9 to 12,and 1 to 12 wk)when compared to all other regimens.Layers on the LL regimen had decreased eggshell thickness and specific gravity(P<0.05;wk 8)when compared to all other regimens,and had decreased egg shell strength(P<0.05;wk 8)when compared to RL and LR regimens.When compared to the RR regimen(a common practice in the industry),layers on the LR regimen had:(1)identical laying performance and egg quality(P>0.05);(2)decreased phosphorus excretion(P<0.05)during the period of09:00 to 17:00;(3)increased jejunal calbindin D28k protein expression(P<0.05)2 h after feeding in the morning;(4)decreased serum fibroblast growth factor 23 and calcitriol levels(P<0.05),decreased jejunal type Ⅲ sodium-phosphate cotransporter 2 gene and protein expression(P<0.05),and decreased renal type Ⅲ sodium-phosphate cotransporter 1 protein expression(P<0.05),2 h after feeding in the afternoon.In summary,when dietary phosphorus was supplemented in accordance with daily serum phosphorus rhythms(i.e.,the LR regimen),laying performance and egg quality were well supported whilst significantly decreasing phosphorus consumption and excretion.Thus,serum phosphorus rhythms will need to be carefully maintained when developing dietary phosphorus-reduction strategies in laying hens.Xujie Liao Jiakun Yan Jionghao Chen Zhenyu Huang Tianshuai Xiao Changqing Li Chong Pan Xin Yang Yanli Liu Thomas D.Crenshaw Xiaojun Yang Zhouzheng Ren 2022Animal Nutrition2022,,4:2
7Role of eIF4E on epithelial-mesenchymal transition,invasion,and chemoresistance of prostate cancer cells显示文摘Dear Editor,Prostate cancer(PCa)is the most common cancer and second leading cause of cancer death for men in the United States[1].PCa with similar Gleason score has been reported to show substantial interpatient heterogeneity and differential prostate cancer-specific mortality rate[2].Such heterogeneity in PCa often results in different therapeutic responses among patients,including therapy resistance,therapeutic failure,relapse,and metastasis[3].Numerous oncogenes,such as eukaryotic translation initiation factor 4E(eIF4E),have been reported to be involved in epithelial-mesenchymal transition(EMT)and/or drug resistance in PCa[4].We previously demonstrated that eIF4E overexpression was involved in chemoresistance of triple-negative breast cancer and silencing eIF4E significantly inhibited cancer cell proliferation and sensitized cancer cells to chemotherapy in a patient-derived xenograft mouse model[5].In addition,eIF4E phosphorylation is known to stimulate the translation of matrix metalloproteinase 3(MMP3)and Snail mRNAs to promote EMT in PCa[6].Furthermore,the complexity and dynamic nature of EMT contributes to the heterogeneity of aggressive cancer cells[7].The precise role of eIF4E in EMT,invasion,and chemoresistance in PCa is still to be established with consideration of different subpopulations in order to develop precision medicine for PCa.In this work,we aimed to explore the role of eIF4E in EMT,invasion,and chemoresistance in PCa for establishing a promising new therapeutic strategy by regulating eIF4E expression using(1-aminoethyl)iminobis[Noleicylcysteinyl-1-aminoethyl)propionamide](ECO)/small interfering RNA(siRNA)nanoparticles previously developed in our lab[8-10]for PCa therapy in the context of tumor heterogeneity.Xujie Liu Amita M Vaidya Da Sun Yan Zhang Nadia Ayat Andrew Schilb Zheng-Rong Lu 2020Cancer Communications2020,40,2:2
8Pressure responses of halide perovskites with various compositions, dimensionalities, and morphologies显示文摘Metal halide perovskites(HPVs)have been greatly developed over the last decade,with various compositions,dimensionalities,and morphologies,leading to an emergence of high-performance photovoltaic and optoelectronic applications.Despite the tremendous progress made,challenges remain,which calls for a better understanding of the fundamental mechanisms.Pressure,a thermodynamic variable,provides a powerful tool to tune materials’structures and properties.In combination with in situ characterization methods,high-pressure research could provide a better fundamental understanding.In this review,we summarize the recent studies of the dramatic,pressure-induced changes that occur in HPVs,particularly the enhanced and emergent properties induced under high pressure and their structure-property relationships.We first introduce the characteristics of HPVs and the basic knowledge of high-pressure techniques,as well as in situ characterization methods.We then discuss the effects of pressure on HPVs with different compositions,dimensionalities,and morphologies,and underline their common features and anomalous behaviors.In the last section,we highlight the main challenges and provide suggestions for possible future research on high-pressure HPVs.Mei Li Tianbiao Liu Yonggang Wang Wenge Yang Xujie Lu 2020Matter and Radiation at Extremes2020,5,1:2
9Nanoparticles and their effects on differentiation of mesenchymal stem cells显示文摘Over the past decades,advancements in nanoscience and nanotechnology have resulted in numerous nanomedicine platforms.Various nanoparticles,which exhibit many unique properties,play increasingly important roles in the field of biomedicine to realize the potential of nanomedicine.Due to the capacity of self-renewal and multilineage mesenchymal differentiation,mesenchymal stem cells(MSCs)have been widely used in the area of regenerative medicine and in clinical applications due to their potential to differentiate into various lineages.There are several factors that impact the differentiation of MSCs into different lineages.Many types of biomaterials such as polymers,ceramics,and metals are commonly applied in tissue engineering and regenerative therapies,and they are continuously refined over time.In recent years,along with the rapid development of nanotechnology and nanomedicine,nanoparticles have been playing more and more important roles in the fields of biomedicine and bioengineering.The combined use of nanoparticles and MSCs in biomedicine requires greater knowledge of the effects of nanoparticles on MSCs.This review focuses on the effects of four inorganic or metallic nanoparticles(hydroxyapatite,silica,silver,and calcium carbonate),which are widely used as biomaterials,on the osteogenic and adipogenic differentiation of MSCs.In this review,the cytotoxicity of these four nanoparticles,their effects on osteogenic/adipogenic differentiation of MSCs and the signalling pathways or transcription factors involved are summarized.In addition,the chemical composition,size,shape,surface area,surface charge and surface chemistry of nanoparticles,have been reported to impact cellular behaviours.In this review,we particularly emphasize the influence of their size on cellular responses.We envision our review will provide a theoretical basis for the combined application of MSCs and nanoparticles in biomedicine.Xing Yang Yuanyuan Li Xujie Liu Wei He Qianli Huang Qingling Feng 2020Biomaterials Translational2020,1,1:1

12Pressure-induced robust emission in a zero-dimensional hybrid metal halide (C_(9)NH_(20))_(6)Pb_(3)Br_(12)显示文摘Zero-dimensional(0D)hybrid metal halides are under intensive investigation owing to their unique physical properties,such as the broadband emission from highly localized excitons that is promising for white-emitting lighting.However,fundamental understanding of emission variations and structure–property relationships is still limited.Here,by using pressure processing,we obtain robust exciton emission in 0D(C_(9)NH_(20))_(6)Pb_(3)Br_(12) at room temperature that can survive to 80 GPa,the recorded highest value among all the hybrid metal halides.In situ experimental characterization and first-principles calculations reveal that the pressure-induced emission is mainly caused by the largely suppressed phonon-assisted nonradiative pathway.Lattice compression leads to phonon hardening,which considerably weakens the exciton–phonon interaction and thus enhances the emission.The robust emission is attributed to the unique structure of separated spring-like[Pb_(3)Br_(12)]^(6−)trimers,which leads to the outstanding stability of the optically active inorganic units.Our findings not only reveal abnormally robust emission in a 0D metal halide,but also provide new insight into the design and optimization of local structures of trimers and oligomers in lowdimensional hybrid materials.Mengting Chen Songhao Guo Kejun Bu Sujin Lee Hui Luo Yiming Wang Bingyan Liu Zhipeng Yan Hongliang Dong Wenge Yang Biwu Ma Xujie Lü 2021Matter and Radiation at Extremes2021,6,5:1
