1Prioritizing natural-selection signals from the deep-sequencing genomic data suggests multi-variant adaptation in Tibetan highlanders显示文摘Human genetic adaptation to high altitudes(>2500 m)has been extensively studied over the last few years,but few functional adaptive genetic variants have been identified,largely owing to the lack of deep-genome sequencing data available to previous studies.Here,w e build a list of putative adaptive variants,including 63 missense,7 loss-of-function;1,298 evolutionarily conserved variants and 509 expression quantitative traits loci.Notably,the top signal of selection is located in TMEM 247,a transmembrane protein-coding gene.The Tibetan version of TMEM 247 harbors one high-frequency(76.3%)missense variant,rsl 16983452(c.248C>T;p.Ala83Val);with the T allele derived from archaic ancestry and carried by>94%of Tibetans but absent or in low frequencies(<3%)in non-Tibetan populations.The rsl 16983452-T is strongly and positively correlated with altitude and significantly associated with reduced hemoglobin concentration(p=5.78×10^-5),red blood cell count(p=5.72×10^-7)and hematocrit(p=2.57×10^-6).In particular,TMEM247-rs 116983452 shows greater effect size and better predicts the phenotypic outcome than any E P A S1 variants in association with adaptive traits in Tibetans.Modeling the interaction between TM£M247-rsl 16983452 and EPAS1 variants indicates weak but statistically significant epistatic effects.Our results support that multiple variants may jointly deliver the fitness of the Tibetans on the plateau,where a complex model is needed to elucidate the adaptive evolution mechanism.Lian Deng Chao Zhang Kai Yuan Yang Gao Yuwen Pan Xueling Ge Yaoxi He Yuan Yuan Yan Lu Xiaoxi Zhang Hao Chen Haiyi Lou Xiaoji Wang Dongsheng Lu Jiaojiao Liu Lei Tian Qidi Feng Asifullah Khan Yajun Yang Zi-Bing Jin Jian Yang Fan Lu Jia Qu Longli Kang Bing Su Shuhua Xu 2019National Science Review2019,6,6:9
2Vertically standing PtSe_2 film: a saturable absorber for a passively mode-locked Nd:LuVO_4 laser显示文摘The novel vertically standing Pt Se2 film on transparent quartz was prepared by selenization of platinum film deposited by the magnetron sputtering method, and an Nd:Lu VO4 passively mode-locked solid-state laser was realized by using the fabricated Pt Se2 film as a saturable absorber. The X-ray diffraction pattern and Raman spectrum of the film indicate its good crystallinity with a layered structure. The thickness of Pt Se2 film is measured to be 24 nm according to the cross-section height profile of the atomic force microscope image. Highresolution transmission electron microscopy images clearly demonstrate its vertically standing structure with an interlayer distance of 0.54 nm along the c-axis direction. The modulation depth(ΔT) and saturation fluence(Φs)of Pt Se2 film are measured to be 12.6% and 17.1 μJ∕cm2, respectively. The obtained mode-locked laser spectrum has a central wavelength of 1066.573 nm, with a 3 d B bandwidth of 0.106 nm. The transform limited pulse width of the mode-locked laser was calculated to be 15.8 ps. A maximum average output power of 180 m W with a working repetition rate of 61.3 MHz is obtained. 3Genome-wide variants of Eurasian facial shape differentiation and a prospective model of DNA based face prediction显示文摘It is a long-standing question as to which genes define the characteristic facial features among different ethnic groups. In this study, we use Uyghurs, an ancient admixed population to query the genetic bases why Europeans and Han Chinese look different. Facial traits were analyzed based on high-dense 3D facial images; numerous biometric spaces were examined for divergent facial features between European and Han Chinese, ranging from inter-landmark distances to dense shape geometrics. Genome-wide association studies(GWAS) were conducted on a discovery panel of Uyghurs. Six significant loci were identified, four of which, rs1868752, rs118078182, rs60159418 at or near UBASH3B, COL23A1, PCDH7 and rs17868256 were replicated in independent cohorts of Uyghurs or Southern Han Chinese. 4A compact fluorescence/circular dichroism dual-modality probe for detection, differentiation, and detoxification of multiple heavy metal ions via bond-cleavage cascade reactions显示文摘Selective detection of multiple analytes in a compact design with dual-modality and theranostic features presents great challenges. Herein, we wish to report a coumarin-thiazolidine masked D-penicillamine based dual-modality fluorescent probe COU-DPA-1 for selective detection, differentiation, and detoxification of multiple heavy metal ions(Ag^(+), Hg^(2+), Cu^(2+)). The probe shows divergent fluorescence(FL)/circular dichroism(CD) responses via divergent bond-cleavage cascade reactions(metal ion promoted C-S cleavage and hydrolysis at two distinctive cleavage sites): FL “turn-off” and CD “turn-on” for Ag+(no hydrolysis), FL “turn-on” and CD “turn-off” for Hg^(+)(imine hydrolysis), and FL “self-threshold ratiometric” and CD “turn-off” for excess Cu^(2+)(lactone and imine hydrolysis), providing the first example of a fluorescence/CD dual-modality probe for multiple species with complimentary responses. 6Effects of curcumin on hippocampal Bax and Bcl-2 expression and cognitive function of a rat model of Alzheimer's disease显示文摘We tested the effects of curcumin treatment on a rat model of Alzheimer's disease induced by beta-amlyoid (Aβ1-40) expression. We investigated alterations in the expression of the apoptosis-related genes Bax and Bcl-2 in the hippocampus, as well as changes in the spatial memory and cognitive function of the rats. Reverse transcription-polymerase chain reaction and immunohistochemistry results showed that Bax expression was remarkably decreased and Bcl-2 expression was increased in the rat Alzheimer's disease model after curcumin treatment. Morris water maze results showed that the average time of escape latency was shortened in the curcumin treated model rats. 7A pinacol boronate caged NIAD-4 derivative as a near-infrared fluorescent probe for fast and selective detection of hypochlorous acid显示文摘Hypochlorous acid(HOCI) is one of highly reactive oxygen species(ROS). It is involved in both immune defense against invading microbes and the progression of many diseases including cardiovascular disease and neurodegeneration disorders. It is generated from hydrogen peroxide(H_2O_2) and chloride ions in the presence of myeloperoxidase in activated neutrophils. To illustrate HOCl's biological functions,fluorescent probes, particularly those fluorescence emissions are in the near-infrared range, are highly needed for in vivo applications. Herein, we reported the design of a pinacol boronate caged near-infrared(NIR) fluorescent probe 1 derived from an Aβ binding fluorophore NIAD-4 for fast and selective detection of HOCl/ClO^-over other ROS. Upon exposure to HOCl/ClO^-, the pinacol boronate caging group of the probe 1 was quickly converted to electron-donating hydroxyl group, which increased intramolecular charge transfer(ICT) in the excited state and resulted in the red-shift and intensity enhancement of fluorescence emission. The probe bears several unique features:(1) It could be used as either a ratiomatic or turn-on fluorescent probe;(2) Reaction of the caging group boronate with HOCl is very fast and finishes within seconds, which provides the selectivity over H_2O_2;(3) The NIAD-4 fluorophore provides additional selectivity for detection of HOCI over peroxynitrite. 8氟比洛芬酯和曲马多静脉超前镇痛对乳腺癌患者淋巴细胞计数的影响(英文)显示文摘Objective: In this study, we evaluated the effect of preemptive analgesia of flurbiprofen axetil (FA) and tramadol on immune response in patients undergoing radical mastectomy. Methods: In this prospective randomized, double-blind, placebo-controlled study, 80 patients were randomly assigned to one of four groups (n = 20). The FA group (group A) patients received FA before tracheal intubation and at the end of surgery. The tramadol group (group B) patients received tramadol before tracheal intubation and at the end of surgery. The control group (group C) patients received saline before tracheal intubation and at the end of surgery. The combination group (group D) patients received FA and tramadol before tracheal intubation and at the end of surgery. Cluster of differentiation and lymphocytes were measured. Results: The CD3, CD4, CD8 and lymphocytes decreased postoperatively of all groups except of group A and D at 0.5 h postoperatively (P < 0.05). The CD3, CD4, CD8 and lymphocytes of group D were higher than those of group C at 0.5 h and 4 h postoperatively (P < 0.05). Conclusion: Preemptive analgesia using flurbiprofen axetil (FA) and tramadol may protect the immune system. The immune protective effect of FA may be better than that of tramadol. 9The Influence of Age on Blood Flow and Temperature of Acupuncture Points: A Study based on Laser Doppler Flowmetry and Infrared Thermography显示文摘Background:To date,there has been a great lack of investigation on the influence of age on blood flow and temperature of acupoints in specific regions.Objective:This study aimed to determine the association between different age categories and acupoint blood flow/temperature on the forearm.Methods:Acupoint blood flow and temperature were measured in healthy adults of different age categories using laser doppler flowmetry(LDF)and infrared thermography(IRT),respectively.A total of 60 eligible healthy volunteers were divided into the young group,mid age group and old age group.All groups received LDF and IRT examination.Shenmen(HT7),Shaohai(HT3),Taiyuan(LU9)and Chize(LU5)of the Heart and Lung meridians on the forearm were selected as 4 test acupoints.Results:Regarding blood flow of the 4 test acupoints,the PU of Taiyuan(LU9)in the old age group was sig-nificantly different compared with that of the young age group(P<0.05)and the mid age group(P<0.05),while there was no significant difference in PU of the other acupoints between 3 groups(all P>0.05).Simi-larly,regarding acupoint temperature of the 4 test sites,the temperature of Shaohai(HT3)in the old age group was significantly different compared with that of the mid age group(P<0.05),while there was no significant difference in the temperature of the other 3 acupoints between 3 age groups(all P>0.05).Conclusion:Age category tends to have notable influence on the blood flow and temperature in specific acupoints in the forearm.Therefore,particular concerns should be taken into consideration regarding the effect of age differences for future studies in this field.Nevertheless,further studies with a large sample size and more test acupoints are needed to further verify current findings.Yajun Zhang Xiaoyu Li Xiaofen He Yongliang Jiang Hantong Hu Jiali Lou Ruijie Ma Yi Liang Xiaomei Shao Jianqiao Fang 2022Clinical Complementary Medicine and Pharmacology2022,2,3:0
