1A complete sequence and comparative analysis of a SARS-associated virus(Isolate BJ01)显示文摘The genome sequence of the Severe Acute Respiratory Syndrome (SARS)-associated virus provides essential information for the identification of pathogen(s), exploration of etiology and evolution, interpretation of transmission and pathogenesis, development of diagnostics, prevention by future vaccination, and treatment by developing new drugs. We report the complete genome sequence and comparative analysis of an isolate (BJ01) of the coronavirus that has been recognized as a pathogen for SARS. The genome is 29725 nt in size and has 11 ORFs (Open Reading Frames). It is composed of a stable region encoding an RNA-dependent RNA polymerase (composed of 2 ORFs) and a variable region representing 4 CDSs (coding sequences) for viral structural genes (the S, E, M, N proteins) and 5 PUPs (putative uncharacterized proteins). Its gene order is identical to that of other known coronaviruses. The sequence alignment with all known RNA viruses places this virus as a member in the family of Coronaviridae. Thirty putative substitutions have been identified by comparative analysis of the 5 SARS- associated virus genome sequences in GenBank. Fifteen of them lead to possible amino acid changes (non-synonymous mutations) in the proteins. Three amino acid changes, with predicted alteration of physical and chemical features, have been detected in the S protein that is postulated to beinvolved in the immunoreactions between the virus and its host. Two amino acid changes have been detected in the Mprotein, which could be related to viral envelope formation. Phylogenetic analysis suggests the possibility of non-human origin of the SARS-associated viruses but provides noevidence that they are man-made. 2Controlling N-doping type in carbon to boost single-atom site Cu catalyzed transfer hydrogenation of quinoline显示文摘Single-atom site(SA)catalysts on N-doped carbon(CN)materials exhibit prominent performance for their active sites being M-Nx.Due to the commonly random doping behaviors of N species in these CN,it is a tough issue to finely regulate their doping types and clarify their effect on the catalytic property of such catalysts.Herein,we report that the N-doping type in CN can be dominated as pyrrolic-N and pyridinic-N respectively through compounding with different metal oxides.It is found that the proportion of distinct doped N species in CN depends on the acidity and basicity of compounded metal oxide host.Owing to the coordination by pyrrolic-N,the SA Cu catalyst displays an enhanced activity(two-fold)for transfer hydrogenation of quinoline to access the valuable molecule tetrahydroquinoline with a good selectivity(99%)under mild conditions.The higher electron density of SA Cu species induced by the predominate pyrrolic-N coordination benefits the hydrogen transfer process and reduces the energy barrier of the hydrogenation pathway,which accounts for the improved catalytic effeciency.Jian Zhang Caiyan Zheng Maolin Zhang Yajun Qiu Qi Xu Weng-Chon Cheong Wenxing Chen Lirong Zheng Lin Gu Zhengpeng Hu Dingsheng Wang Yadong Li 2020Nano Research2020,13,11:29
3Zircon U-Pb geochronology of the volcanic rocks from Fanchang-Ningwu volcanic basins in the Lower Yangtze region and its geological implications显示文摘The latest eruptions in two important Mesozoic volcanic basins of Fanchang and Ningwu located in the middle-lower reaches of the Yangtze River formed the bimodal volcanic rocks of the Kedoushan Formation and ultrapotassic volcanic rocks of the Niangniangshan Formation, respectively. The repre-sentative volcanic rocks of the two Formations were selected for LA-ICPMS zircon U-Pb dating. The results indicate that there exist a large amount of magmatic zircons as indicated by high Th/U ratios in these volcanic rocks. The weighted mean age of 21 analyses is 130.7±1.1 Ma for the Kedoushan Formation, and that of 20 analyses is 130.6±1.1 Ma for the Niangniangshan Formation. These U-Pb ages are interpreted to represent the formation times of the volcanic rocks. In combination with other known geochronological data for Mesozoic volcanic rocks from the Lower Yangtze region, it is proposed that the latest volcanic activations in the Jinniu, Luzong, Fanchang and Ningwu volcanic basins probably came to end prior to ca. 128 Ma. There is no significant time interval between the early and later volcanic activities in the Luzong and Ningwu basins, suggesting a short duration of volcanic activities and thus implying the onset of an extensional tectonic setting at about 130 Ma in the Lower Yangtze region. 6Prioritizing natural-selection signals from the deep-sequencing genomic data suggests multi-variant adaptation in Tibetan highlanders显示文摘Human genetic adaptation to high altitudes(>2500 m)has been extensively studied over the last few years,but few functional adaptive genetic variants have been identified,largely owing to the lack of deep-genome sequencing data available to previous studies.Here,w e build a list of putative adaptive variants,including 63 missense,7 loss-of-function;1,298 evolutionarily conserved variants and 509 expression quantitative traits loci.Notably,the top signal of selection is located in TMEM 247,a transmembrane protein-coding gene.The Tibetan version of TMEM 247 harbors one high-frequency(76.3%)missense variant,rsl 16983452(c.248C>T;p.Ala83Val);with the T allele derived from archaic ancestry and carried by>94%of Tibetans but absent or in low frequencies(<3%)in non-Tibetan populations.The rsl 16983452-T is strongly and positively correlated with altitude and significantly associated with reduced hemoglobin concentration(p=5.78×10^-5),red blood cell count(p=5.72×10^-7)and hematocrit(p=2.57×10^-6).In particular,TMEM247-rs 116983452 shows greater effect size and better predicts the phenotypic outcome than any E P A S1 variants in association with adaptive traits in Tibetans.Modeling the interaction between TM£M247-rsl 16983452 and EPAS1 variants indicates weak but statistically significant epistatic effects.Our results support that multiple variants may jointly deliver the fitness of the Tibetans on the plateau,where a complex model is needed to elucidate the adaptive evolution mechanism.Lian Deng Chao Zhang Kai Yuan Yang Gao Yuwen Pan Xueling Ge Yaoxi He Yuan Yuan Yan Lu Xiaoxi Zhang Hao Chen Haiyi Lou Xiaoji Wang Dongsheng Lu Jiaojiao Liu Lei Tian Qidi Feng Asifullah Khan Yajun Yang Zi-Bing Jin Jian Yang Fan Lu Jia Qu Longli Kang Bing Su Shuhua Xu 2019National Science Review2019,6,6:9
7First experiment of spectrometric observation of hydroxyl emission and rotational temperature in the mesopause in China显示文摘A SpectroMeter of Atmospheric RadiaTion (SMART) was developed and installed at the Xinglong station of the National Astronomical Observatories in Hebei province, China, which was supported by the Meridian Project [1]. The experimental tests of spectrometric observation of the hydroxyl emission and rotational temperature in China were conducted for the first time on the night of February 23, 2011 and the night of April 27, 2011, respectively. OH 6-2 band and OH 8-3 band spectra were measured and the rotational temperature was retrieved. Hourly average temperatures (186.82±6.40) K of OH 8-3 band and (178.07±6.73) K of OH 6-2 band were derived from the spectra observed on the night of February 23, 2011. Intensities and rotational temperature against local time were determined by the spectra measured in the whole night of April 27, 2011. The rotational temperature was consistent with the spatial average temperature of NRLMSISE00 empirical model at height 83-91 km and the average temperature of TIMED/SABER from April to May of seven years at height 83-91 km, with some discrepancies. 8Genome-wide variants of Eurasian facial shape differentiation and a prospective model of DNA based face prediction显示文摘It is a long-standing question as to which genes define the characteristic facial features among different ethnic groups. In this study, we use Uyghurs, an ancient admixed population to query the genetic bases why Europeans and Han Chinese look different. Facial traits were analyzed based on high-dense 3D facial images; numerous biometric spaces were examined for divergent facial features between European and Han Chinese, ranging from inter-landmark distances to dense shape geometrics. Genome-wide association studies(GWAS) were conducted on a discovery panel of Uyghurs. Six significant loci were identified, four of which, rs1868752, rs118078182, rs60159418 at or near UBASH3B, COL23A1, PCDH7 and rs17868256 were replicated in independent cohorts of Uyghurs or Southern Han Chinese. 9Sedimentary geochemistry and provenance of the Lower and Middle Devonian Laojunshan Formation,the North Qilian Orogenic Belt显示文摘The Laojunshan Formation is a suite of molasse formed during the rapid uplift of the North Qilian Orogenic Belt (NQOB). Forty-one samples of sandstone have been collected from the Sunan and Minle sections in the western sector and the Gulang and Jingyuan sections in the eastern sector of the NQOB belt. Geochemical analyses of those samples indicated: 1) The MgO+Fe2O3T and Al2O3/SiO2 values are higher, and K2O/Na2O ratios are lower in the western sector than those in the eastern sector. 2) All of them are depleted in Nb and Ta elements. The samples from the western sector are depleted in Rb element and enriched with Sc, Co, Ni, V, and Cr elements in the Upper Crust-normalized patterns. However, those from the eastern sector are depleted in Sr without enrichments of Sc, Co, Ni, V, and Cr. 3) All of the samples display a right-inclined REE pattern af- ter Chondrite-normalized REE pattern. But LaN/YbN and Eu/Eu* ratios of the samples from the western sector are lower than those of the samples from the eastern sector. These geochemical characteristics suggest the prominent input of mafic clast with minor granitic rocks into the Sunan area, felsic clast into the Gulang and Jingyuan area, and both mafic and felsic clast into the Minle area. The angular shapes of gravels imply that these ill-sorted sediments were deposited near their sources without recy- cling. Geochemical features above also demonstrated that no major chemical weathering occurred for the western provenance, but deposits in the eastern sector resulted from low or middle degree chemical weathering. Evidences combining tectonic discriminations and comparisons with potential provenances revealed that sediments in the Sunan area were derived mainly from the North Qilian Continental arc, whereas sediments in the Minle, Gulang, and Jingyuan areas were derived not only from the North Qilian Continental arc but also from the basement of the Middle Qilian block. 10The biological effect of recombinant humanized collagen on damaged skin induced by UV-photoaging:An in vivo study显示文摘The application of medical devices to repair skin damage is clinically accepted and natural polymer enjoys an important role in this field,such as collagen or hyaluronic acid,etc.However,the biosafety and efficacy of these implants are still challenged.In this study,a skin damage animal model was prepared by UV-photoaging and recombinant humanized type Ⅲ collagen(rhCol Ⅲ)was applied as a bioactive material to implant in vivo to study its biological effect,comparing with saline and uncrosslinked hyaluronic acid(HA).Animal skin conditions were non-invasively and dynamically monitored during the 8 weeks experiment.Histological observation,specific gene expression and other molecular biological methods were applied by the end of the animal experiment.The results indicated that rhCol Ⅲ could alleviate the skin photoaging caused by UV radiation,including reduce the thickening of epidermis and dermis,increase the secretion of Collagen Ⅰ(Col Ⅰ)and Collagen Ⅲ(Col Ⅲ)and remodel of extracellular matrix(ECM).Although the cell-material interaction and mechanism need more investigation,the effect of rhCol Ⅲ on damaged skin was discussed from influence on cells,reconstruction of ECM,and stimulus of small biological molecules based on current results.In conclusion,our findings provided rigorous biosafety information of rhCol Ⅲ and approved its potential in skin repair and regeneration.Although enormous efforts still need to be made to achieve successful translation from bench to clinic,the recombinant humanized collagen showed superiorities from both safety and efficacy aspects.Jing Wang He Qiu Yang Xu Yongli Gao Peijie Tan Rui Zhao Zhanhong Liu Yajun Tang Xiangdong Zhu Chongyun Bao Hang Wang Hai Lin Xingdong Zhang 2022Bioactive Materials2022,7,5:7
11Predicting the height of water-flow fractured zone during coal mining under the Xiaolangdi Reservoir显示文摘It is very important to determine the extent of the fractured zone through which water can flow before coal mining under the water bodies.This paper deals with methods to obtain information about overburden rock failure and the development of the fractured zone while coal mining in Xin'an Coal Mine.The risk of water inrush in this mine is great because 40%of the mining area is under the Xiaolangdi reservoir.Numerical simulations combined with geophysical methods were used in this paper to obtain the development law of the fractured zone under different mining conditions.The comprehensive geophysical method described in this paper has been demonstrated to accurately predict the height of the water-flow fractured zone.Results from the new model, which created from the results of numerical simulations and field measurements,were successfully used for making decisions in the Xin'an Coal Mine when mining under the Xiaolangdi Reservoir.Industrial scale experiments at the number 11201,14141 and 14191 working faces were safely carried out.These achievements provide a successful background for the evaluation and application of coal mining under large reservoirs.XU Zhimin SUN Yajun DONG Qinghong ZHANG Guowei LI Shi 2010Mining Science and Technology2010,20,3:6
12Creating a novel herbicide-tolerance OsALS allele using CRISPR/Cas9-mediated gene editing显示文摘Weeds and weedy rice plague commercial rice fields in many countries. Developingherbicide-tolerance rice is the most efficient strategy to control weed proliferation. CRISPR/Cas9-mediated gene editing, which generates small InDels and nucleotide substitutions atand around target sites using error-prone non-homologous end joining DNA repairing, hasbeen widely adopted for generation of novel crop germplasm with a wide range of geneticvariation in important agronomic traits. We created a novel herbicide-tolerance allele inrice by targeting the acetolactate synthase (OsALS) gene using CRISPR/Cas9-mediated geneediting. The novel allele (G628W) arose from a G-to-T transversion at position 1882 of OsALSand conferred a high level of herbicide tolerance. Transgene-free progeny carryinghomozygous G628W allele were identified and showed agronomic performance similar tothat of wild-type plants, suggesting that the G628W allele is a valuable resource fordeveloping elite rice varieties with strong herbicide tolerance. To promote use of the G628Wallele and to accelerate introgression and/or pyramiding of the G628W allele with other elitealleles, we developed a DNA marker for the G628W allele that accurately and robustlydistinguished homozygous from heterozygous segregants. 13A draft sequence of the rice (Oryza sativa ssp. indica) genome显示文摘The sequence of the rice genome holds fundamental information for its biology, including physiology, genetics, development, and evolution, as well as information on many beneficial phenotypes of economic significance. Using a 'whole genome shotgun' approach, we have pro-duced a draft rice genome sequence of Oryza sativa ssp. in-dica, the major crop rice subspecies in China and many other regions of Asia. The draft genome sequence is constructed from over 4.3 million successful sequencing traces with an accumulative total length of 2214.9 Mb. The initial assembly of the non-redundant sequences reached 409.76 Mb in length, based on 3.30 million successful sequencing traces with a total length of 1797.4 Mb from an indica variant cultivar 93-11, giving an estimated coverage of 95.29% of the rice genome with an average base accuracy of higher than 99%. 14Neutralization of interleukin-17A alleviates burn-induced intestinal barrier disruption via reducing pro-inflammatory cytokines in a mouse model显示文摘Background:The intestinal barrier integrity can be disrupted due to burn injury,which is responsible for local and systemic inflammatory responses.Anti-inflammation strategy is one of the proposed therapeutic approaches to control inflammatory cascade at an early stage.Interleukin-17A(IL-17A)plays a critical role in inflammatory diseases.However,the role of IL-17A in the progression of burn-induced intestinal inflammation is poorly understood.In this study,we aimed to investigate the effect of IL-17A and associated pro-inflammatory cytokines that were deeply involved in the pathogenesis of burn-induced intestinal inflammatory injury,and furthermore,we sought to determine the early source of IL-17A in the intestine.Methods:Mouse burn model was successfully established with infliction of 30%total body surface area scald burn.The histopathological manifestation,intestinal permeability,zonula occludens-1 expression,pro-inflammatory cytokines were determined with or without IL-17A-neutralization.Flow cytometry was used to detect the major source of IL-17A^(+)cells in the intestine.Results:Burn caused intestinal barrier damage,increase of intestinal permeability,alteration of zonula occludens-1 expressions,elevation of IL-17A,IL-6,IL-1βand tumor necrosis factor-α(TNF-α),whereas IL-17A neutralization dramatically alleviated burn-induced intestinal barrier disruption,maintained zonula occludens-1 expression,and noticeably,inhibited pro-inflammatory cytokines elevation.In addition,we observed that the proportion of intestinal IL-17A^(+)Vγ4^(+)T subtype cells(but not IL-17A^(+)Vγ1^(+)T subtype cells)were increased in burn group,and neutralization of IL-17A suppressed this increase.Conclusions:The main original findings of this study are intestinal mucosa barrier is disrupted after burn through affecting the expression of pro-inflammatory cytokines,and a protective role of IL-17A neutralization for intestinal mucosa barrier is determined.Furthermore,Vγ4^(+)T cells are identified as the major early producers of IL-17A that orchestrate an inflammatory response in the burn model.These data suggest that IL-17A blockage may provide a unique target for therapeutic intervention to treat intestinal insult after burn.Yajun Song Yang Li Ya Xiao Wengang Hu Xu Wang Pei Wang Xiaorong Zhang Jiacai Yang Yong Huang Weifeng He Chibing Huang 2019Burns & Trauma2019,7,1:5
15Regulation of mitochondrial NAD pool via NAD^+ transporter 2 is essential for matrix NADH homeostasis and ROS production in Arabidopsis显示文摘Reactive oxygen species(ROS) play a crucial role in numerous biological processes in plants, including development, responses to environmental stimuli, and programmed cell death(PCD). Deficiency in MOSAIC DEATH 1(MOD1), a plastid-localized enoyl-ACP reductase essential for de novo fatty acid biosynthesis in Arabidopsis thaliana, leads to the increased malate export from chloroplasts to mitochondria, and the subsequent accumulation of mitochondria-generated ROS and PCD. In this study, we report the identification and characterization of a mod1 suppressor, som592. SOM592 encodes mitochondrion-localized NAD^+ transporter 2(NDT2). We show that the mitochondrial NAD pool is elevated in the mod1 mutant. The som592 mutation fully suppressed mitochondrial NADH hyper-accumulation, ROS production, and PCD in the mod1 mutant, indicating a causal relationship between mitochondrial NAD accumulation and ROS/PCD phenotypes. We also show that in wild-type plants, the mitochondrial NAD+uptake is involved in the regulation of ROS production in response to continuous photoperiod. Elevation of the alternative respiration pathway can suppress ROS accumulation and PCD in mod1, but leads to growth restriction. 17Light-absorbing impurities on Keqikaer Glacier in western Tien Shan: concentrations and potential impact on albedo reduction显示文摘Light-absorbing impurities on glaciers are important factors that influence glacial surface albedo and accelerate glacier melt. In this study, the quantity of light-absorbing impurities on Keqikaer Glacier in western Tien Shan, Central Asia, was measured. We found that the average concentrations of black carbon was 2,180 ng/g, with a range from 250 ng/g to more than 10,000 ng/g. The average concentrations of organic carbon and mineral dust were 1,738 ng/g and 194 μg/g, respectively. Based on simulations performed with the Snow Ice Aerosol Radiative model simulations, black carbon and dust are responsible for approximately 64% and 9%, respectively, of the albedo reduction, and are associated with instantaneous radiative forcing of 323.18 W/m2(ranging from 142.16 to 619.25 W/m2) and 24.05 W/m2(ranging from 0.15 to69.77 W/m2), respectively. For different scenarios, the albedo and radiative forcing effect of black carbon is considerably greater than that of dust. The estimated radiative forcing at Keqikaer Glacier is higher than most similar values estimated by previous studies on the Tibetan Plateau, perhaps as a result of black carbon enrichment by melt scavenging. Light-absorbing impurities deposited on Keqikaer Glacier appear to mainly originate from central Asia, Siberia, western China(including the Taklimakan Desert) and parts of South Asia in summer, and from the Middle East and Central Asia in winter.A footprint analysis indicates that a large fraction(>60%) of the black carbon contributions on Keqikaer Glacier comes from anthropogenic sources. 18Phase-field simulation of dendritic solidification using a full threaded tree with adaptive meshing显示文摘Simulation of the microstructure evolution during solidifi cation is greatly benefi cial to the control of solidifi cation microstructures. A phase-fi eld method based on the full threaded tree(FTT) for the simulation of casting solidifi cation microstructure was proposed in this paper, and the structure of the full threaded tree and the mesh refi nement method was discussed. During dendritic growth in solidifi cation, the mesh for simulation is adaptively refi ned at the liquid-solid interface, and coarsened in other areas. The numerical results of a threedimension dendrite growth indicate that the phase-fi eld method based on FTT is suitable for microstructure simulation. Most importantly, the FTT method can increase the spatial and temporal resolutions beyond the limits imposed by the available hardware compared with the conventional uniform mesh. At the simulation time of 0.03 s in this study, the computer memory used for computation is no more than 10 MB with the FTT method, while it is about 50 MB with the uniform mesh method. In addition, the proposed FTT method is more effi cient in computation time when compared with the uniform mesh method. 19A 5D simulation method on post-earthquake repair process of buildings based on BIM显示文摘The post-earthquake repair process of buildings is one of the key components of building seismic resilience.Understanding the details of the repair process in advance will benefit the evaluation and decision making for the seismic resilience of buildings.To this end,a five-dimensional(5 D)simulation method on the post-earthquake repair process of buildings is proposed based on building information modeling(BIM),by which the repair activities can be presented in the way associated with a 3 D model,time(denoted as the fourth dimension),and the related costs(denoted as the fifth dimension).First,a decision-making model of repair schemes based on the interval index of the possibility degree is created to determine the optimal post-earthquake repair schemes for a damaged building.Subsequently,an algorithm based on separate flowrepetitive process is designed to automatically make the repair schedule on a BIM platform according to the determined repair schemes.Finally,an algorithm of automatic mapping between the components in BIM and their repair schedules is developed,so that a 5 D simulation of a post-earthquake repair process can be implemented.A six-story office building is selected as a case study,and the details of the post-earthquake repair process(e.g.,the costs of labor,material and equipment over the repair time)are presented by the proposed 5 D simulation method.The outcome of this study can be used to evaluate the seismic resilience of buildings and also to facilitate the decision making for seismically resilient buildings by providing the detailed guidance of the repair process.Xu Zhen Zhang Furong Jin Wei Wu Yingying Qi Mingzhu Yang Yajun 2020Earthquake Engineering and Engineering Vibration2020,19,3:4
