1A germline variant N375S in MET and gastric cancer susceptibility in a Chinese population显示文摘MET tyrosine kinase and its ligand,hepatocyte growth factor(HGF),play a pivotal role in the activties of tumor cells.A germline missense variant in exon 2 of the MET gene,N375S(rs33917957 A>G),may alter the binding affinity of MET for HGF and thus modify the risk of tumorigenesis.In this study,we performed a case-control study to assess the association between N375S and gastric cancer risk in 1,681 gastric cancer cases and 1,858 cancer-free controls.Logistic regression analysis was applied to estimate crude and adjusted odds ratios(ORs) and 95% confidence intervals(CIs) for the associations between genotypes and gastric cancer risk.We found that MET N375S variant genotypes(NS/SS) were associated with a significantly decreased risk of gastric cancer(OR = 0.78,95% CI = 0.63-0.96,P = 0.021) compared with the wildtype homozygote(NN).The finding indicates that this germline variant in MET may decrease gastric cancer susceptibility in Han Chinese.Yao Liu Qin Zhang Chuanli Ren Yanbing Drag Guangfu Jin Zhibin Hu Yaochu Xu Hongbing Shen 2012The Journal of Biomedical Research2012,26,5:2

6Tripodalsporormielones A-C,unprecedented cage-like polyketides with complex polyvdent bridged and fused ring systems显示文摘A chemical investigation on Sporormiella sp.led to the isolation and structural elucidation of tripodalsporormielones Ae C(1-3),a new class of polyketide possessing unprecedented cage-like skeletons with polyvdent bridged and fused ring systems.These polyketides with cage-like skeletons were characterized as a high non-protonated carbon-containing system,which resulted in few HMBC correlations observed and made the accurate structures hard to be obtained by NMR.Especially,some signals of non-protonated sp;carbons are weak and even unobservable in compound 1.In order to establish the structure of 1,the calculated NMR with DP4 evaluation was applied to determine the structure from the plausible structure candidates obtained from the detailed NMR analysis.Based on NMR experiments and calculated NMR,the structures of isolated compounds were established and confirmed by X-ray technology.Through chiral isolation,the optically pure enantiomers of 1 and 3 were obtained,and their absolute configurations were determined based on ECD quantum chemical calculation.Based on the isolated compounds and our previous work,1-3 would be derived from 3-methylorcinaldehyde,and their plausible biosynthetic mechanism was proposed.Furthermore,1 exhibited obvious short-term memory improvement activity on an Alzheimer’s disease fly model.Guodong Chen Bingxin Zhao Meijuan Huang Jia Tang Yanbing Li Liangdong Guo Rongrong He Dan Hu Xinsheng Yao Hao Gao 2021Acta Pharmaceutica Sinica B2021,11,11:1
7Development of a double antibodies sandwich ELISA for the detection of avian leukosis virus subgroup J based on monoclonal antibodies against gp85显示文摘Avian leukosis virus(ALV)belongs to the genus Alpharetrovirus of the family Retroviridae and can cause various types of neoplastic diseases,even mortality in infected chickens[1].ALV is categorized into seven subgroups,including A,B,C,D,E,J,and K,based on the pathogenicity and antibody neutralization characteristics[2].The subgroup E is endogenous and transmitted by Mendelian inheritance.The subgroups A,B,C,D,J,and K are exogenous and oncogenic,transmitted horizontally and vertically and caused different pathological lesions[3].Among them,subgroups A and B were most common in commercial poultry and subgroup J is the major cause for myeloid leukosis and nephromas in broilers.In 1980,ALV subgroup J(ALV-J)was first isolated and identified from infected broilers in Britain.It caused significant economic losses in the poultry industry because of increased myelocytomas and other tumors[4].In China,ALV-J was first detected in broilers in 1999 and,subsequently,spread to other chicken types.Compared to other ALV subgroups,ALV-J has been a serious issue in China poultry industry since 2007.Currently,there is no effective vaccine and drugs against ALV-J infection;thus,early detection and elimination of ALV-positive breeder chickens are key measures to block vertical and horizontal ALV-J transmission.Given the importance of ALV to the poultry industry,many ALV detection methods,including virus isolation,immunofluorescence assays,neutralization tests,reverse transcription polymerase chain reaction(PCR)or real-time fluorescent quantitative PCR-based molecular diagnosis ways,and enzyme-linked immunosorbent assays(ELISA)have been developed.Among them,ELISA is widely used for its efficiency,convenience,and sensitivity.Yun et al.[5]developed an antigen-capture ELISA for the detection of ALV based on highly conserved antigen p27.Yu et al.[6]exploited a colloidal gold test strip specific for the detection of ALV p27 antigen.These antigen capture-based serological detection methods share obviously simple,accurate,and rapid advantages in comparison with conventional time-consuming methods of indirect immunofluorescence assay or neutralization tests.However,it is difficult to identify different subgroups of ALV using p27-coated antibodies.Lili Cao Panpan Zhao He Ding Nan Wang Xue Zhang Shuxian Yuan Hang Dong Yanbing Guo Xinhua Yao Qinlei Yu Hongze Shao Pengtao Gong 2021Acta Biochimica et Biophysica Sinica2021,53,10:1
8From tuberculosis bedside to bench:UBE2B splicing as a potential biomarker and its regulatory mechanism显示文摘Alternative splicing(AS)is an important approach for pathogens and hosts to remodel transcriptome.However,tuberculosis(TB)-related AS has not been sufficiently explored.Here we presented the first landscape of TB-related AS by long-read sequencing,and screened four AS events(S100A8-intron1-retention intron,RPS20-exon1-alternaitve promoter,KIF13B-exon4-skipping exon(SE)and UBE2B-exon7-SE)as potential biomarkers in an in-house cohort-1.The validations in an in-house cohort-2(2274 samples)and public datasets(1557 samples)indicated that the latter three AS events are potential promising biomarkers for TB diagnosis,but not for TB progression and prognosis.The excellent performance of classifiers further underscored the diagnostic value of these three biomarkers.Subgroup analyses indicated that UBE2B-exon7-SE splicing was not affected by confounding factors and thus had relatively stable performance.The splicing of UBE2B-exon7-SE can be changed by heat-killed mycobacterium tuberculosis through inhibiting SRSF1 expression.After heat-killed mycobacterium tuberculosis stimulation,231 ubiquitination proteins in macrophages were differentially expressed,and most of them are apoptosis-related proteins.Taken together,we depicted a global TB-associated splicing profile,developed TB-related AS biomarkers,demonstrated an optimal application scope of target biomarkers and preliminarily elucidated mycobacterium tuberculosis-host interaction from the perspective of splicing,offering a novel insight into the pathophysiology of TB.Mengyuan Lyu Jian Zhou Yanbing Zhou Weelic Chong Wei Xu Hongli Lai Lu Niu Yang Hai Xiaojun Yao Sheng Gong Qinglan Wang Yi Chen Yili Wang Liyu Chen Zengwanggema Jiongjiong Zeng Chengdi Wang Binwu Ying 2023Signal Transduction and Targeted Therapy2023,8,3:0

10Red Blood Cell-Mimic Nanocatalyst Triggering Radical Storm to Augment Cancer Immunotherapy显示文摘Red blood cells(RBCs)have recently emerged as promosing candidates for cancer treatment in terms of relieving tumor hypoxia and inducing oxidative damage against cancer cells,but they are still far from satisfactory due to their limited oxygen transport and reactive oxygen species generation rate in tumor tissue.Herein,artificial RBCs(designated FTP@RBCM)with radical storm production ability were developed for oncotherapy through multidimensional reactivity pathways of Fe-protoporphyrin-based hybrid metal-organic frameworks(FTPs,as the core),including photodynamic/chemodynamic-like,catalase-like and glutathione peroxidase-like activities.Meanwhile,owing to the advantages of long circulation abilities of RBCs provided by their cell membranes(RBCMs),FTP with a surface coated with RBCMs(FTP@RBCM)could enormously accumulate at tumor site to achieve remarkably enhanced therapeutic efficiency.Intriguingly,this ROS-mediated dynamic therapy was demonstrated to induce acute local inflammation and high immunogenic cancer death,which evoked a systemic antitumor immune response when combined with the newly identified T cell immunoglobulin and mucin-containing molecule 3(Tim-3)checkpoint blockade,leading to not only effective elimination of primary tumors but also an abscopal effect of growth suppression of distant tumors.Therefore,such RBC-mimic nanocatalysts with multidimensional catalytic capacities might provide a promising new insight into synergistic cancer treatment.Jiong Li Sijia Wang Xinyi Lin Yanbing Cao Zhixiong Cai Jing Wang Zhenxi Zhang Xiaolong Liu Ming Wu Cuiping Yao 2022Nano-Micro Letters2022,14,4:0
