1Role of contrast-enhanced ultrasonography in percutaneous radiofrequency ablation of liver metastases and efficacy evaluation显示文摘Objective: To retrospectively investigate the role of contrast-enhanced ultrasonography (CEUS) in percutaneous radiofrequency ablation (RFA) in patients with liver metastases and evaluate the therapeutic efficacy of RFA assisted by CEUS. Methods: From May 2004 to September 2010, 136 patients with 219 liver metastatic lesions received CEUS examination 1 h before RFA (CEUS group), and other 126 patients with 216 lesions without CEUS examination in the earlier period were served as a historical control group. The mean tumor size was 3.2 cm and the mean tumor number was 1.6 in the CEUS group, while 3.4 cm and 1.7 in the control group, respectively (P>0.05). The clinical characteristics, recurrence results and survival outcomes were compared between two groups. Results: In the CEUS group, two isoechoic tumors were not demonstrated on unenhanced ultrasonography (US), and 63 (47%) of 134 tumors examined with CEUS were 0.3 cm larger than with unenhanced US. Furthermore, in 18.4% of 136 patients, additional 1-3 tumors were detected on CEUS. The CEUS group showed higher early tumor necrosis and lower intrahepatic recurrence than the control group. The 3-year overall survival (OS) rate and the 3-year local recurrence-free survival (LRFS) rate in the CEUS group were 50.1% and 38.3%, in contrast to 25.3% and 19.3% in the control group, respectively (P=0.002 and P<0.001). 2Evaluation of contrast-enhanced ultrasound for diagnosis of dysplastic nodules with a focus of hepatocellular carcinoma in liver cirrhosis patients显示文摘Objective: To compare the enhancement features of dysplastic nodules with a focus of hepatocellular carcinoma(DN-HCC) versus HCC and regenerative nodules(RN) in cirrhotic patients.Methods: One hundred and ninety-three cirrhotic patients were enrolled in this study; they had 215 focal liver lesions, 1.0-3.5 cm in size, which were examined using contrast-enhanced ultrasound(CEUS) with Sono Vue?and diagnosed as HCC, RN or DN-HCC by biopsy. Samples were obtained using 18-gauge needles in the different enhanced areas. The enhancement features of DN-HCC, HCC and RN were evaluated.Results: There were 86 HCC lesions, 102 RN lesions, and 27 DN-HCC lesions diagnosed by biopsy. Of 86 HCC lesions, 87.2%(75/86) showed complete enhancement during the arterial phase, and 12.8%(11/86) had inhomogeneous enhancement, with no enhancement in the central area during the arterial phase; 100%(86/86) exhibited washout during the late phase. Of 102 RN lesions, 95.1%(97/102) had delayed or simultaneous enhancement during the arterial phase, and 4.9%(5/102) displayed slight enhancement during the arterial phase; 26.5%(27/102) exhibited washout and 73.5%(75/102) exhibited no washout during the late phase. In 27 DN-HCC lesions, only part of the lesions enhanced during the arterial phase and washed out during the late phase; the other areas had delayed or simultaneous enhancement during the arterial phase, and 29.6%(8/27) exhibited slight washout in the late phase. In 86 HCCs, the pathological feature was HCC in the enhanced area of 75 lesions, hepatocellular fatty degeneration in the slightly enhanced area of 7 lesions, and hepatocellular necrosis in the unenhanced area and HCC in the enhanced area of 4 lesions. In 102 RNs, the pathological diagnosis was hepatocyte proliferation with or without fatty degeneration. In 27 DN-HCCs, the pathological feature was HCC in the enhanced area and hepatocyte regeneration in the unenhanced area.Conclusions: CEUS is useful for the diagnosis of focal liver lesions in cirrhotic patients. CEUS can help determine the progression from RN to DN-HCC to HCC by analyzing the hemodynamics. 3Identification of mecciRNAs and their roles in the mitochondrial entry of proteins显示文摘Mammalian mitochondria have small genomes encoding very limited numbers of proteins.Over one thousand proteins and noncoding RNAs encoded by the nuclear genome must be imported from the cytosol into the mitochondria.Here,we report the identification of hundreds of circular RNAs(mecciRNAs)encoded by the mitochondrial genome.We provide both in vitro and in vivo evidence to show that mecciRNAs facilitate the mitochondrial entry of nuclear-encoded proteins by serving as molecular chaperones in the folding of imported proteins.Known components involved in mitochondrial protein and RNA importation,such as TOM40 and PNPASE,interact with mecciRNAs and regulate protein entry.The expression of mecciRNAs is regulated,and these transcripts are critical for the adaption of mitochondria to physiological conditions and diseases such as stresses and cancers by modulating mitochondrial protein importation.mecciRNAs and their associated physiological roles add categories and functions to the known eukaryotic circular RNAs and shed novel light on the communication between mitochondria and the nucleus.Xu Liu Xiaolin Wang Jingxin Li Shanshan Hu Yuqi Deng Hao Yin Xichen Bao Qiangfeng Cliff Zhang Geng Wang Baolong Wang Qinghua Shi Ge Shan 2020Science China(Life Sciences)2020,63,10:15
4Effect of contrast-enhanced ultrasound on differential diagnosis of intrahepatic cholangiocarcinoma and arterial phase enhanced hepatic inflammatory lesions显示文摘Objective: To investigate differential diagnosis between intrahepatic cholangiocarcinoma(ICC) and arterial phase enhanced hepatic inflammatory lesions in patients without liver cirrhosis using contrast-enhanced ultrasound(CEUS).Methods: ICC and hepatic inflammatory lesions cases with CEUS and pathological diagnosis between Sep 2013 and Oct 2016 were investigated retrospectively. Imaging features of conventional ultrasound and CEUS were analyzed. The parameters of time intensity curve(TIC), including the arrival time, peak intensity(PI) in the lesions, the starting time for washout, and the intensity difference at 3 min(ΔI_3) after contrast agent infection between the lesion and the liver parenchyma, were compared between ICC and hepatic inflammatory lesions.Results: Twenty-five ICC and fifteen inflammatory patients were included in this study. Seventeen ICC(68.0%)and two inflammatory cases(13.3%) showed bile duct dilatation on conventional ultrasound. Using CEUS, three ICC cases(12.0%) were misdiagnosed as inflammatory lesions and three inflammatory lesions(20.0%) as ICC; two ICC(8.0%) and one inflammatory case(6.7%) could not be made definite diagnosis. Washout started at 34.5±3.5 s and 61.5±12.9 s for ICC and inflammatory lesions respectively(P<0.001). The intensity difference between lesion and liver parenchyma at 3 min after contrast agent injection was 10.8±3.1 dB in ICC and 4.2±2.3 dB in inflammatory group(P<0.001). 5Exposure to polycyclic aromatic hydrocarbons and risk for premature ovarian failure and reproductive hormones imbalance显示文摘Polycyclic aromatic hydrocarbons(PAHs)are a typical class of persistent organic pollutants that is ubiquitous worldwide.Previous animal studies suggested that PAHs had adverse effects on female reproduction.However,the human data regarding relationship of PAHs exposure with women reproductive health,such as ovarian dysfunction,are scarce.In this case-control study,the associations of serum levels of PAHs with the risk of premature ovarian failure(POF)and reproductive hormones in Chinese women were investigated,with recruiting 157 POF patients and 217 healthy women.The serum levels of 12 types of PAHs,as well as reproductive hormones,including follicle-stimulating hormone,luteinizing hormone and anti-mullerian hormone,were determined.In the logistic regression models,most individual PAH congeners showed significantly positive correlations with the risk of POF(p<0.05),except for fluorine and pyrene.Benzo(a)pyrene(BaP),as the most carcinogenic PAH congener,was observed to be significantly positively associated with the risk of POF.After adjustment for age,body mass index,educational levels and household income,per one-unit increase in the log-transformed BaP concentration was significantly correlated with 2.191-fold increased risk of POF(OR=2.191,95%CI:1.634-2.938,p<0.05).To the best of our knowledge,this is the first study to report an association between internal exposure levels of PAHs and the increased risk of POF in women.Xiaoqing Ye Wuye Pan Chunming Li Xiaochen Ma Shanshan Yin Jianhong Zhou Jing Liu 2020Journal of Environmental Sciences2020,32,5:10

7Distribution and Provenance of Detrital Minerals in Southern Coast of Shandong Peninsula显示文摘Detrital minerals of 137 offshore and 22 river sediment samples collected from Qingdao coastal areas have been analyzed. Four mineral assemblage provinces can be classified by Q-mode cluster analysis. Factor analysis identifies two major factors that account for the total variability in most common minerals: 1) based on the relationship of quartz, hornblende, actinolite, micas, and authigenic pyrite, 41.55% of the variability is related to sediment sources; 2) based on the relationship of epidote, garnet, sphere, and ilmenite, 23.21% can be related to strong hydrodynamic conditions that control transport and sedimentation. By comparing mineral compositions of river waters in the study area, the following four mineral provenances can be identified. The Qingdao-Laoshan nearshore area has a quartz-feldspar-epidote-hornblende-limenite-limonite-sphene assemblage, which is largely attributed to relict sediment and coastal erosion. The Jimo-Haiyang nearshore area has a quartz-feldspar-hornblende-epidote-limonite-mica-actinolite assemblage, derived largely from the Wulong River and Rushan River, and is also affected by the Huanghe River, while the Qianliyan Island area in the deeper offshore area separated by a mud belt has a similar assemblage. The Haiyang-Rushan nearshore area has a quartz-feldspar-hornblende-epidote-micas-limonite assemblage, indicating multiple sources from the Rushan River, the Wulong River, the Huanghe River, and coastal erosion. 10An efficient VLSI architecture of speeded-up robust feature extraction for high resolution and high frame rate video显示文摘This paper proposes a VLSI architecture of the optimized Speeded-Up Robust Feature (SURF) algorithm. The SURF algorithm which is widely used in computer vision applications, locates interest points (IPoints) and extracts feature descriptors based on the surrounding gradients. In the proposed approach, the SURF algorithm is modified to make it more suitable for hardware implementation with little accuracy lost compared to OpenSURF, an open source software implementation based on OpenCV. The resource cost and throughput gain are balanced. Word Length Reduction (WLR) is adopted to compress the integral image and reduce the occupied memory resources. The orientations and the feature descriptors of the IPoints are calculated in a more efficient way, which significantly reduces memory accesses. Moreover, the operations are pipelined both in and among the proposed hardware modules. This VLSI architecture has been validated on FPGA (Xilinx Virtex-4 XC4VLX80), which is able to detect IPoints and extract SURF feature descriptors in a VGA (640 × 480) 64 fps video input with a 96 MHz working frequency while dissipating 1.278 W. 11Disentangling the factors that contribute to variation in forest biomass increments in the mid-subtropical forests of China显示文摘Mid-subtropical forests are the main vegetation type of global terrestrial biomes, and are critical for maintaining the global carbon balance. However, estimates of forest biomass increment in mid-subtropical forests remain highly uncertain. It is critically important to determine the relative importance of different biotic and abiotic factors between plants and soil, particularly with respect to their influence on plant regrowth. Consequently,it is necessary to quantitatively characterize the dynamicspatiotemporal distribution of forest carbon sinks at a regional scale. This study used a large, long-term dataset in a boosted regression tree(BRT) model to determine the major components that quantitatively control forest biomass increments in a mid-subtropical forested region(Wuyishan National Nature Reserve, China). Long-term,stand-level data were used to derive the forest biomass increment, with the BRT model being applied to quantify the relative contributions of various biotic and abiotic variables to forest biomass increment. Our data show that total biomass(t) increased from 4.62 9 106 to 5.30 9 106 t between 1988 and 2010, and that the mean biomass increased from 80.19 ± 0.39 t ha-1(mean ± standard error) to 94.33 ± 0.41 t ha-1in the study region. The major factors that controlled biomass(in decreasing order of importance) were the stand, topography, and soil. Stand density was initially the most important stand factor, while elevation was the most important topographic factor. Soil factors were important for forest biomass increment but have a much weaker influence compared to the other two controlling factors. 12Nb2C MXene-Functionalized Scaffolds Enables Osteosarcoma Phototherapy and Angiogenesis/Osteogenesis of Bone Defects显示文摘Early surgical resection and chemotherapy of bone cancer are commonly used in the treatment of bone tumor,but it is still highly challenging to prevent recurrence and fill the bone defect caused by the resection site.In this work,we report a rational integration of photonic-responsive two-dimensional(2D)ultrathin niobium carbide(Nb2C)MXene nanosheets(NSs)into the 3D-printed bone-mimetic scaffolds(NBGS)for osteosarcoma treatment.The integrated 2D Nb2C-MXene NSs feature specific photonic response in the second near-infrared(NIR-II)biowindow with high tissue-penetrating depth,making it highly efficient in killing bone cancer cells.Importantly,Nb-based species released by the biodegradation of Nb2C MXene can obviously promote the neogenesis and migration of blood vessels in the defect site,which can transport more oxygen,vitamins and energy around the bone defect for the reparative process,and gather more immune cells around the defect site to accelerate the degradation of NBGS.The degradation of NBGS provides sufficient space for the bone remodeling.Besides,calcium and phosphate released during the degradation of the scaffold can promote the mineralization of new bone tissue.The intrinsic multifunctionality of killing bone tumor cell and promoting angiogenesis and bone regeneration makes the engineered Nb2C MXeneintegrated composite scaffolds a distinctive implanting biomaterial on the efficient treatment of bone tumor.Junhui Yin Shanshan Pan Xiang Guo Youshui Gao Daoyu Zhu Qianhao Yang Junjie Gao Changqing Zhang Yu Chen 2021Nano-Micro Letters2021,13,2:5
13Silicon lithium-ion battery anode with enhanced performance: Multiple effects of silver nanoparticles显示文摘Silicon has been regarded as one of the most promising next generation lithium-ion battery anode. However, the poor cyclic stability of the Si based anode has severely limited its practical applications, which is even worse with high mass loading density(>1 mg cm^(-2)). A new concept has been developed to enhance the electrochemical performance of the Si nanoparticle anode. Silver nanoparticles are composited with the silicon nanoparticles in a facile way for the first time. It is found that the mechanical properties of the Si electrode have been significantly improved by the incorporation of the silver nanoparticles, leading to enhanced cyclic performance. With the Si/Ag mass ratio of 4:1, the reversible specific discharge capacity is retained as 1156 m A h g^(-1) after 100 cycles at 200 mA g^(-1), which is more than three times higher than that of the bare silicon(318 mA h g^(-1)). 15Cryo-EM structures of human m^(6)A writer complexes显示文摘N6-methyladenosine(m^(6)A)is the most abundant ribonucleotide modification among eukaryotic messenger RNAs.The m^(6)A“writer”consists of the catalytic subunit m^(6)A-METTL complex(MAC)and the regulatory subunit m^(6)A-METTL-associated complex(MACOM),the latter being essential for enzymatic activity.Here,we report the cryo-electron microscopy(cryo-EM)structures of MACOM at a 3.0-Åresolution,uncovering that WTAP and VIRMA form the core structure of MACOM and that ZC3H13 stretches the conformation by binding VIRMA.Furthermore,the 4.4-Åresolution cryo-EM map of the MACOM–MAC complex,combined with crosslinking mass spectrometry and GST pull-down analysis,elucidates a plausible model of the m^(6)A writer complex,in which MACOM binds to MAC mainly through WTAP and METTL3 interactions.In combination with in vitro RNA substrate binding and m^(6)A methyltransferase activity assays,our results illustrate the molecular basis of how MACOM assembles and interacts with MAC to form an active m^(6)A writer complex.Shichen Su Shanshan Li Ting Deng Minsong Gao Yue Yin Baixing Wu Chao Peng Jianzhao Liu Jinbiao Ma Kaiming Zhang 2022Cell Research2022,32,11:3

17Lipocalin-2-induced proliferative endoplasmic reticulum stress participates in Kawasaki disease-related pulmonary arterial abnormalities显示文摘Clinical cases have reported pulmonary arterial structural and functional abnormalities in patients with Kawasaki disease(KD);however,the underlying mechanisms are unclear.In this study,a KD rat model was established via the intraperitoneal injection of Lactobacillus casei cell wall extract(LCWE).The results showed that pulmonary arterial functional and structural abnormalities were observed in KD rats.Furthermore,proliferative endoplasmic reticulum stress(ER stress)was observed in the pulmonary arteries of KD rats.Notably,the level of lipocalin-2(Lcn 2),a trigger factor of inflammation,was remarkably elevated in the plasma and lung tissues of KD rats;increased Lcn 2 levels following LCWE stimulation may result from polymorphonuclear neutrophils(PMNs).Correspondingly,in cultured pulmonary artery smooth muscle cells(PASMCs),Lcn 2 markedly augmented the cleavage and nuclear localization of activating transcription factor-6(ATF6),upregulated the transcription of glucose regulated protein 78(GRP78)and neurite outgrowth inhibitor(NOGO),and promoted PASMCs proliferation.However,proapoptotic C/EBP homologous protein(CHOP)and caspase 12 levels were not elevated.Treatment with 4-phenyl butyric acid(4-PBA,a specific inhibitor of ER stress)inhibited PASMCs proliferation induced by Lcn 2 and attenuated pulmonary arterial abnormalities and right ventricular hypertrophy and reduced right ventricular systolic pressure in KD rats.In conclusion,Lcn 2 remarkably facilitates proliferative ER stress in PASMCs,which probably accounts for KD-related pulmonary arterial abnormalities.Zhaoling Shi Yue Yin Chen Li Hui Ding Nan Mu Yishi Wang Shanshan Jin Heng Ma Manling Liu Jie Zhou 2021Science China(Life Sciences)2021,64,6:1

19Lnc-RP5 Regulates the mi R-129-5p/Notch1/PFV Internal Promoter Axis to Promote the Expression of the Prototype Foamy Virus Transactivator Tas显示文摘Prototype foamy virus(PFV)is a unique retrovirus that infects animals and humans and does not cause clinical symptoms.Long noncoding RNAs(lncRNAs)are believed to exert multiple regulatory functions during viral infections.Previously,we utilized RNA sequencing(RNA-seq)to characterize and identify the lncRNA lnc-RP5-1086 D14.3.1-1:1(lnc-RP5),which is markedly decreased in PFV-infected cells.However,little is known about the function of lnc-RP5 during PFV infection.In this study,we identified lnc-RP5 as a regulator of the PFV transcriptional transactivator(Tas).Lnc-RP5 enhanced the activity of the PFV internal promoter(IP).The expression of PFV Tas was found to be promoted by lnc-RP5.Moreover,mi R-129-5 p was found to be involved in the lnc-RP5-mediated promotion of PFV IP activity,while the Notch1 protein suppressed the activity of PFV IP and the expression of Tas.Our results demonstrate that lnc-RP5 promotes the expression of PFV Tas through the miR-129-5 p/Notch1/PFV IP axis.This work provides evidence that host lnc RNAs can manipulate PFV replication by employing mi RNAs and proteins during an early viral infection.Shanshan Xu Liujun Chen Yinglian Tang Peipei Yuan Jun Yan Yingcheng Zheng Li Huang Zhi Li Yan Sun Song Han Jun Yin Qin Pan Biwen Peng Xiaohua He Wanhong Liu 2020Virologica Sinica2020,35,1:1
