Breast cancer development and progression:Risk factors, cancer stem cells, signaling pathways, genomics, and molecular pathogenesis显示文摘As the most commonly occurring cancer in women worldwide, breast cancer poses a formidable public health challenge on a global scale.Breast cancer consists of a group of biologically and molecularly heterogeneous diseases originated from the breast. While the risk factors associated with this cancer varies with respect to other cancers, genetic predisposition, most notably mutations in BRCA1 or BRCA2 gene,is an important causative factor for this malignancy.Breast cancers can begin in different areas of the breast, such as the ducts, the lobules,or the tissue in between.Within the large group of diverse breast carcinomas, there are various denoted types of breast cancer based on their invasiveness relative to the primary tumor sites. It is important to distinguish between the various subtypes because they have different prognoses and treatment implications. As there are remarkable parallels between normal development and breast cancer progression at the molecular level,it has been postulated that breast cancer may be derived from mammary cancer stem cells.Normal breast development and mammary stem cells are regulated by several signaling pathways, such as estrogen receptors(ERs),HER2,and Wnt/b-catenin signaling pathways, which control stem cell proliferation, cell death, cell differentiation, and cell motility.Furthermore, emerging evidence indicates that epigenetic regulations and noncoding RNAs may play important roles in breast cancer development and may contribute to the heterogeneity and metastatic aspects of breast cancer, especially for triple-negative breast cancer. This review provides a comprehensive survey of the molecular, cellular and genetic aspects of breast cancer.Yixiao Feng Mia Spezia Shifeng Huang Chengfu Yuan Zongyue Zeng Linghuan Zhang Xiaojuan Ji Wei Liu Bo Huang Wenping Luo Bo Liu Yan Lei Scott Du Akhila Vuppalapati Hue H.Luu Rex C.Haydon Tong-Chuan He Guosheng Ren 2018Genes & Diseases2018,5,2:24
A long-term Global LAnd Surface Satellite(GLASS)data-set for environmental studies显示文摘Recently, five Global LAnd Surface Satellite(GLASS)products have been released:leaf area index(LAI),shortwave broadband albedo, longwave broadband emissivity, incident short radiation, and photosynthetically active radiation(PAR).The first three products cover the years 19822012(LAI)and 19812010(albedo and emissivity)at 15 km and 8-day resolutions, and the last two radiation products span the period 20082010 at 5 km and 3-h resolutions. These products have been evaluated and validated, and the preliminary results indicate that they are of higher quality and accuracy than the existing products. In particular, the first three products have much longer time series, and are therefore highly suitable for various environmental studies. This paper outlines the algorithms, product characteristics, preliminary validation results, potential applications and some examples of initial analysis of these products.Shunlin Liang Xiang Zhao Suhong Liu Wenping Yuan Xiao Cheng Zhiqiang Xiao Xiaotong Zhang Qiang Liu Jie Cheng Hairong Tang Yonghua Qu Yancheng Bo Ying Qu Huazhong Ren Kai Yu John Townshend 2013International Journal of Digital Earth2013,6,S01:16
Humoral immune response to circulating SARS-CoV-2 variants elicited by inactivated and RBD-subunit vaccines显示文摘SARS-CoV-2 variants could induce immune escape by mutations on the receptor-binding domain(RBD)and N-terminal domain(NTD).Here we report the humoral immune response to circulating SARS-CoV-2 variants, such as 501Y.V2(B.1.351),of the plasma and neutralizing antibodies(NAbs)elicited by CoronaVac(inactivated vaccine),ZF2001(RBD-subunit vaccine)and natural infection.Among 86 potent NAbs identified by high-throughput single-cell VDJ sequencing of peripheral blood mononuclear cells from vaccinees and convalescents, near half anti-RBD NAbs showed major neutralization reductions against the K417N/E484K/N501Y mutation combination, with E484K being the dominant cause.VH3-53/VH3-66 recurrent antibodies respond differently to RBD variants, and K417N compromises the majority of neutralizing activity through reduced polar contacts with complementarity determining regions. In contrast, the 242–244 deletion(242–244Δ)would abolish most neutralization activity of anti-NTD NAbs by interrupting the conformation of NTD antigenic supersite, indicating a much less diversity of anti-NTD NAbs than anti-RBD NAbs.Plasma of convalescents and CoronaVac vaccinees displayed comparable neutralization reductions against pseudo-and authentic 501Y.V2 variants, mainly caused by E484K/N501Y and 242–244Δ,with the effects being additive.Importantly,RBD-subunit vaccinees exhibit markedly higher tolerance to 501Y.V2 than convalescents, since the elicited anti-RBD NAbs display a high diversity and are unaffected by NTD mutations. Moreover,an extended gap between the third and second doses of ZF2001 leads to better neutralizing activity and tolerance to 501Y.V2 than the standard three-dose administration.Together, these results suggest that the deployment of RBD-vaccines,through a third-dose boost, may be ideal for combating SARS-CoV-2 variants when necessary, especially for those carrying mutations that disrupt the NTD supersite.Yunlong Cao Ayijiang Yisimayi Yali Bai Weijin Huang Xiaofeng Li Zhiying Zhang Tianjiao Yuan Ran An Jing Wang Tianhe Xiao Shuo Du Wenping Ma Liyang Song Yongzheng Li Xiang Li Weiliang Song Jiajing Wu Shuo Liu Xuemei Li Yonghong Zhang Bin Su Xianghua Guo Yangyang Wei Chuanping Gao Nana Zhang Yifei Zhang Yang Dou Xiaoyu Xu Rui Shi Bai Lu Ronghua Jin Yingmin Ma Chengfeng Qin Youchun Wang Yingmei Feng Junyu Xiao Xiaoliang Sunney Xie 2021Cell Research2021,31,7:12
Reproductive management through integration of PGD and MPS-based noninvasive prenatal screening/diagnosis for a family with GJB2-associated hearing impairment显示文摘A couple with a proband child of GJB2(encoding the gap junction protein connexin 26)-associated hearing impairment and a previous pregnancy miscarriage sought for a reproductive solution to bear a healthy child. Our study aimed to develop a customized preconception-to-neonate care trajectory to fulfill this clinical demand by integrating preimplantation genetic diagnosis(PGD), noninvasive prenatal testing(NIPT), and noninvasive prenatal diagnosis(NIPD) into the strategy. Auditory and genetic diagnosis of the proband child was carried out to identify the disease causative mutations. The couple then received in-vitro-fertilization treatment, and eight embryos were obtained for day 5 biopsy. PGD was performed by short-tandem-repeat linkage analysis and Sanger sequencing of GJB2 gene. Transfer of a GJB2 c.235del C heterozygous embryo resulted in a singleton pregnancy. At the 13 th week of gestation, genomic DNA(g DNA) from the trio family and cell-free DNA(cf DNA) from maternal plasma were obtained for assessment of fetal chromosomal aneuploidy and GJB2 mutations. NIPT and NIPD showed the absence of chromosomal aneuploidy and GJB2-associated disease in the fetus, which was later confirmed by invasive procedures and postnatal genetic/auditory diagnosis. This strategy successfully prevented the transmission of hearing impairment in the newborn, thus providing a valuable experience in reproductive management of similar cases and potentially other monogenic disorders.XIONG WenPing WANG DaYong GAO Yuan GAO Ya WANG HongYang GUAN Jing LAN Lan YAN JunHao ZONG Liang YUAN Yuan DONG Wei HUANG SeXin WU KeLiang WANG YaoShen WANG ZhiLi PENG HongMei LU YanPing XIE LinYi ZHAO Cui WANG Li ZHANG QiuJing GAO Yun LI Na YANG Ju YIN ZiFang HAN Bing WANG Wei CHEN Zi-Jiang WANG QiuJu 2015Science China(Life Sciences)2015,58,9:12

A Brief Introduction to BNU-HESM1.0 and Its Earth Surface Temperature Simulations显示文摘Integrated assessment models and coupled earth system models both have their limitations in understanding the interactions between human activity and the physical earth system. In this paper,a new human–earth system model,BNUHESM1.0,constructed by combining the economic and climate damage components of the Dynamic Integrated Model of Climate Change and Economy to the BNU-ESM model,is introduced. The ability of BNU-HESM1.0 in simulating the global CO2 concentration and surface temperature is also evaluated. We find that, compared to observation,BNU-HESM1.0underestimates the global CO2 concentration and its rising trend during 1965–2005,due to the uncertainty in the economic components. However, the surface temperature simulated by BNU-HESM1.0 is much closer to observation, resulting from the overestimates of surface temperature by the original BNU-ESM model. The uncertainty of BNU-ESM falls within the range of present earth system uncertainty,so it is the economic and climate damage component of BNU-HESM1.0 that needs to be improved through further study. However, the main purpose of this paper is to introduce a new approach to investigate the complex relationship between human activity and the earth system. It is hoped that it will inspire further ideas that prove valuable in guiding human activities appropriate for a sustainable future climate.YANG Shili DONG Wenjie CHOU Jieming FENG Jinming YAN Xiaodong WEI Zhigang YUAN Wenping GUO Yan TANG Yanli HU Jiacong 2015Advances in Atmospheric Sciences2015,32,12:8
Characterization of the essential role of bone morphogenetic protein 9 (BMP9) in osteogenic differentiation of mesenchymal stem cells (MSCs) through RNA interference显示文摘Mesenchymal stem cells(MSCs)are multipotent stem cells and capable of differentiating into multiple cell types including osteoblastic, chondrogenic and adipogenic lineages. We previously identified BMP9 as one of the most potent BMPs that induce osteoblastic differentiation of MSCs although exact molecular mechanism through which BMP9 regulates osteogenic differentiation remains to be fully understood. Here,we seek to develop a recombinant adenovirus system to optimally silence mouse BMP9 and then characterize the important role of BMP9 in osteogenic differentiation of MSCs.Using two different siRNA bioinformatic prediction programs,we design five siRNAs targeting mouse BMP9(or simB9),which are expressed under the control of the converging H1 and U6 promoters in recombinant adenovirus vectors. We demonstrate that two of the five siRNAs,simB9-4 and simB9-7,exhibit the highest efficiency on silencing exogenous mouse BMP9 in MSCs.Furthermore,simB9-4 and simB9-7 act synergistically in inhibiting BMP9-induced expression of osteogenic markers, matrix mineralization and ectopic bone formation from MSCs.Thus, our findings demonstrate the important role of BMP9 in osteogenic differentiation of MSCs.The characterized simB9 siRNAs may be used as an important tool to investigate the molecular mechanism behind BMP9 osteogenic signaling. Our results also indicate that recombinant adenovirus-mediated expression of siRNAs is efficient and sustained, and thus may be used as an effective delivery vehicle of siRNA therapeutics.Shujuan Yan Ruyi Zhang Ke Wu Jing Cui Shifeng Huang Xiaojuan Ji Liping An Chengfu Yuan Cheng Gong Linghuan Zhang Wei Liu Yixiao Feng Bo Zhang Zhengyu Dai Yi Shen Xi Wang Wenping Luo Bo Liu Rex C.Haydon Michael J.Lee Russell R.Reid Jennifer Moriatis Wolf Qiong Shi Hue H.Luu Tong-Chuan He Yaguang Weng 2018Genes & Diseases2018,5,2:8
Influence of the carbon cycle on the attribution of responsibility for climate change显示文摘The carbon cycle is one of the fundamental climate change issues. Its long-term evolution largely affects the amplitude and trend of human-induced climate change,as well as the formulation and implementation of emission reduction policy and technology for stabilizing the atmospheric CO2concentration.Two earth system models incorporating the global carbon cycle, the Community Earth System Model and the Beijing Normal University-Earth System Model, were used to investigate the effect of the carbon cycle on the attribution of the historical responsibility for climate change. The simulations show that when compared with the criterion based on cumulative emissions, the developed(developing)countries’responsibility is reduced(increased)by 6%–10%using atmospheric CO2concentration as the criterion. This discrepancy is attributed to the fact that the developed world contributed approximately61%–68%(61%–64%)to the change in global oceanic(terrestrial)carbon sequestration for the period from 1850 to2005,whereas the developing world contributed approximately 32%–49%(36%–39%).Under a developed world emissions scenario, the relatively larger uptake of global carbon sinks reduced the developed countries’responsibility for carbon emissions but increased their responsibility for global ocean acidification(68%).In addition, the large emissions from the developed world reduced the efficiency of the global carbon sinks, which may affect the long-term carbon sequestration and exacerbate global warming in the future. Therefore,it is necessary to further consider the interaction between carbon emissions and the carbon cycle when formulating emission reduction policy.Ting Wei Wenjie Dong Wenping Yuan Xiaodong Yan Yan Guo 2014Chinese Science Bulletin2014,59,19:6
Establishment and functional characterization of the reversibly immortalized mouse glomerular podocytes(imPODs)显示文摘Glomerular podocytes are highly specialized epithelial cells and play an essential role in establishing the selective permeability of the glomerular filtration barrier of kidney.Maintaining the viability and structural integrity of podocytes is critical to the clinical management of glomerular diseases, which requires a thorough understanding of podocyte cell biology. As mature podocytes lose proliferative capacity,a conditionally SV40 mutant tsA58-immortalized mouse podocyte line(designated as tsPC)was established from the Immortomouse over 20 years ago. However, the utility of the tsPC cells is hampered by the practical inconvenience of culturing these cells. In this study,we establish a user-friendly and reversibly-immortalized mouse podocyte line(designated as imPOD),on the basis of the tsPC cells by stably expressing the wildtype SV40 T-antigen,which is flanked with FRT sites. We show the imPOD cells exhibit long-term high proliferative activity, which can be effectively reversed by FLP recombinase. The imPOD cells express most podocyte-related markers, including WT-1,Nephrin,Tubulin and Vinculin, but not differentiation marker Synaptopodin. The imPOD cells do not form tumor-like masses in vivo.We further demonstrate that TGFb1 induces a podocyte injury-like response in the FLP-reverted imPOD cells by suppressing the expression of slit diaphragm-associated proteins P-Cadherin and ZO-1 and upregulating the expression of mesenchymal markers,a-SMA,Vimentin and Nestin,as well as fibrogenic factors CTGF and Col1a1.Collectively,our results strongly demonstrate that the newly engineered im-POD cells should be a valuable tool to study podocyte biology both under normal and under pathological conditions.Xinyi Yu Liqun Chen Ke Wu Shujuan Yan Ruyi Zhang Chen Zhao Zongyue Zeng Yi Shu Shifeng Huang Jiayan Lei Xiaojuan Ji Chengfu Yuan Linghuan Zhang Yixiao Feng Wei Liu Bo Huang Bo Zhang Wenping Luo Xi Wang Bo Liu Rex C.Haydon Hue H.Luu Tong-Chuan He Hua Gan 2018Genes & Diseases2018,5,2:5
Natural Climate Solutions for China:The Last Mile to Carbon Neutrality显示文摘“I call on all leaders worldwide to declare a State of Climate Emergency in their own countries until carbon neutrality is reached.”–António GUTERRES(United Nations Secretary General),12 December,2020 There is no shortcut to a carbon neutral society;solutions are urgently required from both energy&industrial sectors and global ecosystems. While the former is often held accountable and emphasized in terms of its emissions reduction capability, the latter(recently termed natural climate solutions)should also be assessed for potential and limitations by the scientific community, the public, and policy makers.Zhangcai QIN Xi DENG Bronson GRISCOM Yao HUANG Tingting LI Pete SMITH Wenping YUAN Wen ZHANG 2021Advances in Atmospheric Sciences2021,38,6:5
The development of a sensitive fluorescent protein-based transcript reporter for high throughput screening of negative modulators of lncRNAs显示文摘While the human genome is pervasively transcribed,<2%of the human genome is transcribed into protein-coding mRNAs,leaving most of the transcripts as noncoding RNAs,such as microRNAs and long-noncoding RNAs(lncRNAs),which are critical components of epigenetic regulation.lncRNAs are emerging as critical regulators of gene expression and genomic stability. However,it remains largely unknown about how lncRNAs are regulated. Here,we develop a highly sensitive and dynamic reporter that allows us to identify and/or monitor negative modulators of lncRNA transcript levels in a high throughput fashion.Specifically,we engineer a fluorescent fusion protein by fusing three copies of the PEST destruction domain of mouse ornithine decarboxylase(MODC)to the C-terminal end of the codon-optimized bilirubin-inducible fluorescent protein, designated as dBiFP,and show that the dBiFP protein is highly destabilized, compared with the commonly-used eGFP protein. We further demonstrate that the dBiFP signal is effectively down-regulated when the dBiFP and mouse lncRNA H19 chimeric transcript is silenced by mouse H19-specific siRNAs.Therefore, our results strongly suggest that the dBiFP fusion protein may serve as a sensitive and dynamic transcript reporter to monitor the inhibition of lncRNAs by microRNAs,synthetic regulatory RNA molecules,RNA binding proteins,and/or small molecule inhibitors so that novel and efficacious inhibitors targeting the epigenetic circuit can be discovered to treat human diseases such as cancer and other chronic disorders.Zongyue Zeng Bo Huang Shifeng Huang Ruyi Zhang Shujuan Yan Xinyi Yu Yi Shu Chen Zhao Jiayan Lei Wenwen Zhang Chao Yang Ke Wu Ying Wu Liping An Xiaojuan Ji Cheng Gong Chengfu Yuan Linghuan Zhang Wei Liu Yixiao Feng Bo Zhang Zhengyu Dai Yi Shen Xi Wang Wenping Luo Rex C.Haydon Hue H.Luu Lan Zhou Russell R.Reid Tong-Chuan He Xingye Wu 2018Genes & Diseases2018,5,1:4
Soil erosion affects variations of soil organic carbon and soil respiration along a slope in Northeast China显示文摘Background:Although soil erosion plays a key role in the carbon cycle,a holistic and mechanistic understanding of the soil erosion process within the cycle is still lacking. The aim of this study was therefore to improve our mechanistic understanding of soil organic carbon(SOC)and soil respiration dynamics through an experiment conducted in an eroding black soil farmland landscape in Northeast China. Results:The depositional profiles store 5.9 times more SOC than the eroding profiles and 3.3 times more SOC than the non-eroding profiles.A linear correlation between the SOC and 137Cs(Caesium-137)was observed in our study, suggesting that the SOC decreased with increased soil erosion.Furthermore, the fractions of intermediate C and the microaggregate C were lowest at the eroding position and highest at the depositional position. In the depositional topsoil, the input of labile materials plays a promotional role in soil respiration.Conversely,in the subsoil(i.e.,below 10 cm),the potential mineralization rates were lowest at the depositional position—due to effective stabilization by physical protection within soil microaggregates. The field results of soil surface respiration also suggest that the depositional topsoil SOC is prone to be mineralized and that SOC at this depositional context is stabilized at subsoil depth. In addition, the high water contents at the depositional position can limit the decomposition rates and stabilize the SOC at the same time. Conclusions:The findings from this study support that a majority of the SOC at footslope is stored within most of the soil profile(i.e.,below 10 cm)and submitted to long-term stabilization, and meanwhile support that the depositional profile emits more CO2 than the summit due to its high amount and quality of SOC.Tong Li Haicheng Zhang Xiaoyuan Wang Shulan Cheng Huajun Fang Gang Liu Wenping Yuan 2019Ecological Processes2019,8,1:4

A branched dihydrophenazine-based polymer as a cathode material to achieve dual-ion batteries with high energy and power density显示文摘Organic electrode materials have exhibited good electrochemical performance in batteries, but their voltages and rate capabilities still require improvement to meet the increasing demand for batteries with high energy and power density.Herein,we design and synthesize a branched dihydrophenazine-based polymer(p-TPPZ)as a cathode material for dual-ion batteries(DIBs)through delicate molecular design.Compared with the linear dihydrophenazine-based polymer(p-DPPZ,with a theoretical capacity of 209 mAh g^(−1)),p-TPPZ possessed a higher theoretical capacity of 233 mAh g^(−1)and lower highest occupied molecular orbital energy levels, which resulted in a high actual capacity(169.3 mAh g^(−1)at 0.5 C),an average discharge voltage of 3.65 V(vs.Li+/Li)and a high energy density(618.2 Wh kg^(−1),based on the cathode materials). The branched structure of p-TPPZ led to a larger specific surface area than that of p-DPPZ,which was beneficial for the electrolyte infiltration and fast ionic transport, contributing to the high power density.Due to the fast reaction kinetics, even at a power density of 23,725 W kg^(−1)(40 C),the energy density still reached 474.5 Wh kg^(−1).We also made a detailed investigation of the p-TPPZ cathode’s charge storage mechanism. This work will stimulate the further molecular design to develop organic batteries with both high energy and power density.Shuaifei Xu Huichao Dai Shaolong Zhu Yanchao Wu Mingxuan Sun Yuan Chen Kun Fan Chenyang Zhang Chengliang Wang Wenping Hu 2021eScience2021,1,1:2

Early embryonic transcriptomes of Zeugodacus tau provide insight into sex determination and differentiation genes显示文摘Zeugodacus tau(Walker)is an invasive pest. The sterile insect technique is an environment-friendly method for pest control.Understanding the mechanism of sex determination will contribute to improving efficiency of this technique. In this study,we identified the transformer(tra)gene in Z.tau.One female-specific and two male-specific isoforms of tra were found in Z.tau, and the male-specific splicing pattern of tra was found to occur 5 h after egg laying. We performed transcriptome sequencing at 1 h(E1),5 h(E5),and 9 h(E9)after egg laying and obtained high-quality transcriptome libraries of early embryo development. We identified 13297 and 11713 differentially expressed genes(DEGs)from E5 versus E1 and E9 versus E1 comparisons, respectively. To explore the potential functions of the DEGs during embryonic development,Gene Ontology,Clusters of Orthologous Groups of proteins, and Kyoto Encyclopedia of Genes and Genomes analyses were performed.Twenty-six genes potentially involved in sex determination or differentiation, including Maleness-on-the-Y(MoY),were identified in Z.tau.To verify the transcriptome results,15 genes were selected for quantitative real-time PCR validation. The results were consistent with the transcriptome sequencing results.Moreover,U2 small nuclear riboprotein auxiliary factor(U2AF-50),female lethal d(fl(2)d),and virilizer(vir)were highly expressed at E5,indicating that they may be related to the sex-specific splicing of tra.Further functional analysis is needed to confirm this speculation. Our data provide an insight into the mechanism underlying sex determination and differentiation in tephritid species.Peipei Liu Wenping Zheng Jiao Qiao Ziniu Li Zhurong Deng Yimin Yuan Hongyu Zhang 2022Insect Science2022,29,3:1
