4Zircon SHRIMP U-Pb geochronology of the Neoproterozoic Chengjiang Formation in central Yunnan Province(SW China) and its geological significance显示文摘The Chengjiang Formation is the earliest continental clastic deposit after the Jinning Orogeny in central Yunnan Province,and therefore its accurate depositional age is significant for understanding the formation and evolution of the Neoproterozoic rift basins in southern China.However,hampered by accuracy of the dating technique,the existing age data for the Chengjiang Formation are not very reliable.A large number of magmatic zircons were obtained from the tuff interbed in the lower part of the Chengjiang Formation in Jinyang area and the bottom part of the Chengjiang Formation in Dongchuan area,central Yunnan Province,and high-precision SHRIMP U-Pb dating was carried out on these zircons.The results show that the weighted mean 206 Pb/238 U ages are 797.8±8.2 and 803.1±8.7 Ma respectively.In combination with related geological evidence,it has been demonstrated that the bottom boundary age of the Chengjiang Formation should be 800±5 Ma,whereas the top boundary age could be ca 725 Ma.By synthetically analyzing the latest age data for the relevant strata,it has been confirmed that the Chengjiang Formation should be correlated with the Suxiong Formation and the Kaijianqiao Formation in western Sichuan Province,the Liantuo Formation in the middle and lower Yangtze,the Hongchicun Formation and the Shangshu Formation in northern Zhejiang Province,the Puling Formation in southern Anhui Province,the Luokedong Formation and the Majianqiao Formation in northwestern Jiangxi Province,the Wuqiangxi Formation in northwestern Hunan Province,the Fanzhao Formation and the Qingshuijiang Formation in southeastern Guizhou Province,and the Sanmenjie Formation and the Gongdong Formation in northern Guangxi Province.Sedimentary cycle analysis shows that the sedimentary filling sequence of the Neoproterozoic rift basins in southern China can be divided into four cycles.Among them,Cycle II began at ca.800 Ma,accompanied by intensive tectonic-thermal events.The zircon U-Pb ages from the bottom of the Chengjiang Formation reported in this paper indicate that the Neoproterozoic Kangdian rift subbasin probably started to develop at ca.800 Ma and therefore missed Cycle I of the Neoproterozoic sedimentary filling sequence in southern China.JIANG XinSheng WANG Jian CUI XiaoZhuang ZHUO JieWen XIONG GuoQing LU JunZe LIU JianHui 2012Science China Earth Sciences2012,55,11:25
5Modified Da-chai-hu Decoction regulates the expression of occludin and NF-κB to alleviate organ injury in severe acute pancreatitis rats显示文摘Modified Da-chai-hu Decoction(MDD), a traditional Chinese medicinal formulation, which was empirically generated from Da-chai-hu decoction, has been utilized to treat severe acute pancreatitis(SAP) for decades. The aim of the present study was to explore its potential organprotective mechanism in SAP. In the present study, rat SAP model was induced by retrograde injection of 3.5% sodium taurocholate into the biliopancreatic duct, MDD(23.35 g/kg body weight, twelve times the clinical dose) were orally given at 2 h before and 10 h after injection. At 12 h after model induction, blood was taken from vena cava for analysis of amylase, diamine oxidase(DAO), pulmonary surfactant protein-A(SP-A), and C-reactive protein(CRP). Histopathological change of pancreas, ileum and lung was assayed by H&E staining, myeloperoxidase(MPO) activity were determinated using colorimetric assay, and the expressions of occludin and nuclear factor-κB(NF-κB) were detected by real-time RT-PCR and western blot, respectively. In addition, the tissue concentrations of tumor necrosis factor-α(TNF-α), interleukin-1β(IL-1β), and monocyte chemoattractant protein-1(MCP-1) were measured by enzyme-linked immunosorbent assay(ELISA). The results showed that in SAP rats, MDD significantly alleviated histopathological damage, depressed the MPO activity and the concentrations of TNF-α, IL-1β, and MCP-1 of pancreas, ileum and lung, and reduced the serum levels of amylase [(3283.4±585.5) U·L^(-1) vs(5626.4±795.1) U·L^(-1)], DAO[(1100.1±334.3) U·L^(-1) vs(1666.4±525.3) U·L^(-1)] and CRP [(7.6±1.2) μg·mL^(-1) vs(17.8±3.8) μg·mL^(-1)]. However, the serum SP-A concentration [(106.1±16.6) pg·mL^(-1) vs(90.1±14.9) pg·mL^(-1)] was elevated when treated SAP rats with MDD. Furthermore, MDD increased the occludin expression and reduced the NF-κB expression in pancreas, ileum and lung of SAP rats. Our findings suggested that MDD administration was an effective therapeutic approach for SAP treatment. 6The Chromosome-Level Genome Sequence of the Autotetraploid Alfalfa and Resequencing of Core Germplasms Provide Genomic Resources for Alfalfa Research显示文摘Alfalfa(Medicago sativa)is one of the most important forage crops in the world;however,its molecular genetics and breeding research are hindered due to the lack of a high-quality reference genome.Here,we report a de novo assembled 816-Mb high-quality,chromosome-level haploid genome sequence for‘Zhongmu No.1’alfalfa,a heterozygous autotetraploid.The contig N50 is 3.92 Mb,and 49165 genes are annotated in the genome.The alfalfa genome is estimated to have diverged from M.truncatula approximately 8 million years ago.Genomic population analysis of 162 alfalfa accessions revealed high genetic diversity,weak population structure,and extensive gene flow from wild to cultivated alfalfa.Genome-wide association studies identified many candidate genes associated with important agronomic traits.Furthermore,we showed that MsFTa2,a Flowering Locus T homolog,whose expression is upregulated in salt-resistant germplasms,may be associated with fall dormancy and salt resistance.Taken together,these genomic resources will facilitate alfalfa genetic research and agronomic improvement.Chen Shen Huilong Du Zhuo Chen Hongwei Lu Fugui Zhu Hong Chen Xiangzhao Meng Qianwen Liu Peng Liu Lihua Zheng Xiuxiu Li Jiangli Dong Chengzhi Liang Tao Wang 2020Molecular Plant2020,13,9:21
7An implantable rat liver tumor model for experimental transarterial chemoembolization therapy and its imaging features显示文摘AIM: To establish an ideal implantable rat liver tumor model for interventional therapy study and examine its angiographic signs and MRI, CT features before and after embolization. METHODS: Forty male Wistar rats were implanted with Walker256 tumor in the left lateral lobe of liver. Digital subtraction angiography (DSA) and transarterial chemoembolization were performed on day 14 after implantation. Native computer tomography (CT, n=8) and native magnetic resonance (MR,n=40) were performed between the day 8 and day 21 after implantation. The radiological morphological characteristics were correlated with histological findings.RESULTS: Successful implantation was achieved in all forty rats, which was confirmed by CT and MRI. MR allowed tumor visualization from day 8 while CT from day 11 after implantation. The tumors were hypodensity on CT, hypointense on MR T1-weighted and hyperintense on T2-weighted. The model closely resembled human hepatocardnoma in growth pattem and the lesions were rich in vasculature on angiography and got its filling mainly from the hepatic artery. Before therapy, tumor size was 211.9±48.7 mm3. No ascites, satellite liver nodules or lung metastasis were found. One week after therapy, tumor size was 963.6±214.8 mm3 in the control group and 356.5±78.4mm3 in TACE group. Ascites (4/40), satellite liver nodules (7/40) or lung metastasis (3/40) could be seen on day 21.CONCLUSION: Walker-256 tumor rat model is suitable for the interventional experiment. 8Influence of transarterial chemoembolization on angiogenesis and expression of vascular endothelial growth factor and basic fibroblast growth factor in rat with Walker-256 transplanted hepatoma:An experimental study显示文摘AIM: After transarterial chemoembolization (TACE), the residual cancer cells are under extensive hypoxic or even anoxic environment. Hypoxia can lead to adaptive responses.For example, angiogenesis will help these cells survive. In this study, we examined the effect of TACE on angiogenesis and expression of vascular endothelial growth factor (VEGF)and basic fibroblast growth factor (b-FGF) and to assess their relevance to Walker-256 transplanted hepatoma.METHODS: Male Wistar rats were inoculated with Walker256 tumor in the left lobe of liver. Angiography and transarterial chemoembolization were performed at d14 after transplantation. Sixty rats bearing walker-256 transplanted hepatoma were randomly divided into control group, arterial infusion group and TACE group. Each group consisted of twenty rats. Normal saline, 5-Fu, 5-Fu and lipiodol were infused through hepatic artery respectively. Two weeks after the infusion, staining of factor Ⅷ, VEGF and b-FGF was performed by immunohistochemistry method in routine paraffin-embeded sections. Microvessel density(MVD) was counted in endothelial cells with positive factor Ⅷ. Their expression levels were analyzed in conjunction with the pathologic features.RESULTS: While a smaller tumor volume was found in TACE group (F=37.818, P<0.001), no statistical differences between MVD and expression of VEGF and b-FGF were found among the 3 groups. MVD of the control group, chemotherapy group and chemoemoblization group was 80.84±24.24,83.05±20.29 and 85.20±23.91 (F=0.193, P=0.873),respectively. The positive expression of VEGF and b-FGF was 75 %, 75 % , 85 % (X2=0.449, P=0.799) and 30 %,25 %, 30 % (X2=0.141, P=0.922), respectively. 9High-resolution records of Holocene from the Shuangchi Maar Lake in Hainan Island显示文摘The results of sedimentary characteristics, grain-size, clay mineral, C/N and pollen analysis reveal the evolution history of the Shuangchi Maar Lake of Hainan Island as well as the short-term climate variations in tropical areas. It is testified that the Maar sediment system was sensitive in recording the rapid climate changes in the Holocene. These multidisciplinary results demonstrate that the thermal maximum in the Holocene climate lasted from 7200 to 2700 aBP, with several abrupt ephemeral vibrations occurring between 7250-6120 aBP and at around 4460, 3850 and 2700 aBP. In the late Holocene, particularly at ~2700 aBP, a distinct environmental change indicating an important climate transition in the tropical eastern Asia was recorded, probably responding to the enhancement of ENSO phenomena in the Pacific Ocean. 10Precise nanomedicine for intelligent therapy of cancer显示文摘Precise nanomedicine has been extensively explored for efficient cancer imaging and targeted cancer therapy, as evidenced by a few breakthroughs in their preclinical and clinical explorations. Here, we demonstrate the recent advances of intelligent cancer nanomedicine, and discuss the comprehensive understanding of their structure-function relationship for smart and efficient cancer nanomedicine including various imaging and therapeutic applications, as well as nanotoxicity. In particular, a few emerging strategies that have advanced cancer nanomedicine are also highlighted as the emerging focus such as tumor imprisonment, supramolecular chemotherapy, and DNA nanorobot. The challenge and outlook of some scientific and engineering issues are also discussed in future development. 11Stereotactic aspiration for hypertensive intracerebral haemorrhage in a Chinese population:a retrospective cohort study显示文摘Objective We aimed to compare the therapeutic effects of stereotactic aspiration and best medical management in patients who developed supratentorial hypertensive intracerebral haemorrhage(HICH)with a volume of haemorrhage between 20 and 40 mL.Methods The clinical data of 220 patients with supratentorial HICH with a volume between 20 and 40 mL were retrospectively analysed.Among them,142 received stereotactic aspiration surgery(stereotactic aspiration group)and 78 received best medical management(conservative group).All were followed up for 6 months.Multivariate logistic regression and Kaplan-Meier survival curves were used to compare the outcome between the two groups.results The rebleeding rate was lower in the group that had stereotactic aspiration when compared with the group with medical treatment(6[4.2%]vs 9[11.5%],χ2=4.364,p=0.037).After 6 months,although the mortality rate did not differ significantly between the two groups(8 cases[5.6%]vs 10 cases[12.8%],χ2=3.461,p=0.063),the rate of a favourable outcome was higher in the group who received stereotactic aspiration(χ2=15.870,p=0.000).Logistic regression identified that medical treatment(OR=1.64,p=0.000)was an independent risk factor for an unfavourable outcome.The Kaplan-Meier curves indicated that the median favourable outcome time in the stereotactic aspiration group was 59.5 days compared with that in the medically treated group(87.0 days).The log-rank test indicated that the prognosis at 6 months was better for those treated with stereotactic haematoma aspiration(χ2=29.866,p=0.000).However,the 6-month survival rate was similar between the two groups(χ2=3.253,p=0.068).Conclusions Stereotactic haematoma aspiration significantly improved the quality of life,although did not effectively reduce the rate of mortality.When selected appropriately,patients with HICH may benefit from this type of surgical intervention.Xuyang Zhang Shaolong Zhou Qiang Zhang Xudong Fu Yuehui Wu Jiasheng Liu Bo Liang Zhuo Yang Xinjun Wang 2019Stroke & Vascular Neurology2019,4,1:19

14Association of polymorphism of tumor necrosis factor-alpha gene promoter region with outcome of hepatitis B virus infection显示文摘AIM: To determine whether -238G/A and -857C/T polymorphisms of tumor necrosis factor-alpha (TNF-α), gene promoter and hepatitis B (HB) viral genotypes were associated with outcomes of HBV infection.METHODS: A total of 244 HBV self-limited infected subjects, 208 asymptomatic carriers, and 443 chronic HB patients were recruited to conduct a case-control study.TNF-α -238G/A and -857C/T gene promoter polymorphisms were examined by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP), and HBV genotypes were examined by nested PCR.RESULTS: The positive rate of HBV DNA in asymptomatic carrier group and chronic HB group was 46.6% and 49.9%,respectively. HBV genotype proportion among the asymptomatic carriers was 2.1% for genotype A, 25.8% for genotype B, 68.0% for genotype C, and 4.1% for genotype B+C mixed infection, and 0.9% for genotype A,21.7% for genotype B, 71.5% for genotype C, 5.9% for genotype B+C mixed infection in chronic HB group. There was no significant difference in genotype distribution between the asymptomatic carrier group and chronic HB group (x2 = 1.66, P = 0.647). The frequency of -238GG genotype in self-limited group was 95.1%, significantly higher than 90.7% in chronic HB group and 89.0% in asymptomatic carrier group (P = 0.041 and P = 0.016,respectively).The frequency of TNF-α-857 CC in chronic HB group was 79.7%, significantly higher than 64.4% in asymptomatic carrier group and 70.9% in self-limited group (P<0.001 and P = 0.023, respectively). 17Hawthorn Extract Alleviates Atherosclerosis through Regulating Inflammation and Apoptosis Related Factors: An Experimental Study显示文摘Objective: To determine the effects of hawthorn extract on serum lipid levels, pathological changes in aortic atherosclerosis plaque, inflammatory factors, and apoptosis-related protein and mRNA expression in apolipoprotein E gene knockout(ApoE^(-/-)) mice. Methods: Thirty-six ApoE^(-/-) mice were fed with a high-fat diet starting at the age of 8 weeks. Mice were randomly divided into 3 groups by a random number table including model group, hawthorn extract group, and simvastatin group, 12 mice in each group. Twelve 8-week-old C57BL/6 mice were fed a basic diet and served as control. The mice in the control and model groups were administered 0.2 mL saline daily, the mice in the hawthorn extract and simvastatin groups were administered with 50 mg/kg hawthorn extract or 5 mg/kg simvastatin daily for 16 weeks. After 16 weeks, plasma lipids including total cholesterol(TC), triglyceride(TG), low-density lipoprotein cholesterol(LDL-C) and high-density lipoprotein cholesterol(HDL-C) were determined by an enzymatic assay. Aortic atherosclerotic lesions were observed by light microscopy, scanning and transmission electron microscopy, respectively. Plasma levels of monocyte chemoattractant protein-1(MCP-1), interleukin-1β(IL-1β), adiponectin(APN), and hypersensitive C-reactive protein(hs-CRP) were measured by enzyme-linked immunosorbent assay(ELISA). Protein and mRNA expressions of Bax and Bcl-2 in the aorta were assessed by Western blotting and quantitative real-time polymerase chain reaction(qR T-PCR), respectively. Results: Compared to the control group, the plasma levels of TC, TG and LDL-C were significantly increased and HDL-C were significantly decreased in the model group(P<0.01). Compared to the model group, treatment with hawthorn extract significantly decreased the plasma levels of TC, TG, and LDL-C and increased the plasma level of HDL-C in ApoE^(-/-)mice(P<0.01). The levels of MCP-1, IL-1β, and hs-CRP in the model group were significantly increased and APN was significantly decreased compared with the control group(P<0.01). Compared to the model group, treatment with hawthorn extract decreased the levels of MCP-1, IL-1β, and hs-CRP and increased the APN level(P<0.01). Compared to the control group, the protein and mR NA expression of Bax in the model group were significantly increased and the expression of Bcl-2 was significantly decreased(P<0.01). Hawthorn extract also reduced the protein and mR NA expression of Bax and increased the Bcl-2 expression in the aorta(P<0.01). Conclusion: Hawthorn extract has anti-atherosclerosis and stabilizing unstable plaque effects. Therefore, it is imperative to develop a new method to accelerate the discovery and design of new materials. In recent years, material discovery and design methods using machine learning have attracted much attention from material experts and have made some progress. This review first outlines available materials database and material data analytics tools and then elaborates on the machine learning algorithms used in materials science. Next, the field of application of machine learning in materials science is summarized, focusing on the aspects of structure determination, performance prediction, fingerprint prediction, and new material discovery. Finally, the review points out the problems of data and machine learning in materials science and points to future research. 19Antioxidant and Anti-inflammatory Capacity of Ferulic Acid Released from Wheat Bran by Solid-state Fermentation of Aspergillus niger显示文摘Objective A strain of Aspergillus niger(A. niger), capable of releasing bound phenolic acids from wheat bran, was isolated. This strain was identified by gene sequence identification. The antioxidant and anti-inflammatory capacity of ferulic acid released from wheat bran by this A. niger strain(FA-WB) were evaluated. Methods Molecular identification techniques based on PCR analysis of specific genomic sequences were conducted; antioxidant ability was examined using oxygen radical absorbance capacity(ORAC), cellular antioxidant activity(CAA) assays, and erythrocyte hemolysis assays. RAW264.7 cells were used as a model to detect anti-inflammatory activity. Results The filamentous fungal isolate was identified to be A. niger. ORAC and CAA assay showed that FA-WB had better antioxidant activity than that of the ferulic acid standard. The erythrocyte hemolysis assay results suggested that FA-WB could attenuate AAPH-induced oxidative stress through inhibition of reactive oxy gen species(ROS) generation. FA-WB could significantly restore the AAPH-induced increase in intracellular antioxidant enzyme activities to normal levels as well as inhibit the intracellular malondialdehyde formation. TNF-?, IL-6, and NO levels indicated that FA-WB can inhibit the inflammation induced by lipopolysaccharide(LPS).
