1A novel method for rapid and reliable detection of complex vertebral malformation and bovine leukocyte adhesion deficiency in Holstein cattle显示文摘Background: Complex vertebral malformation (CVM) and bovine leukocyte adhesion deficiency (BLAD) are two autosomal recessive lethal genetic defects frequently occurring in Holstein cattle, identifiable by single nucleotide polymorphisms. The objective of this study is to develop a rapid and reliable genotyping assay to screen the active Holstein sires and determine the carrier frequency of CVM and BLAD in Chinese dairy cattle population. Results: We developed real-time PCR-based assays for discrimination of wild-type and defective alleles, so that carriers can be detected. Only one step was required after the DNA extraction from the sample and time consumption was about 2 hours. A total of 587 Chinese Holstein bulls were assayed, and fifty-six CVM-carriers and eight BLAD-carriers were identified, corresponding to heterozygote carrier frequencies of 9.54% and 1.36%, respectively. The pedigree analysis showed that most of the carriers could be traced back to the common ancestry, Osborndale Ivanhoe for BLAD and Pennstate Ivanhoe Star for CVM. Conclusions: These results demonstrate that real-time PCR is a simple, rapid and reliable assay for BLAD and CVM defective allele detection. 2A Recombinant Rabies Virus Expressing Fms-like Tyrosine Kinase 3 Ligand(Flt3L) Induces Enhanced Immunogenicity in Mice显示文摘Rabies is a zoonotic disease that still causes 59,000 human deaths each year,and rabies vaccine is the most effective way to control the disease.Our previous studies suggested that the maturation of DC plays an important role in enhancing the immunogenicity of rabies vaccine.Flt3L has been reported to own the ability to accelerate the DC maturation,therefore,in this study,a recombinant rabies virus expressing mouse Flt3L,designated as LBNSE-Flt3L,was constructed,and its immunogenicity was characterized.It was found that LBNSE-FU3L could enhance the maturation of DC both in vitro and in vivo,and significantly more TFH cells and Germinal Center B(GC B)cells were generated in mice immunized with LBNSE-FU3L than those immunized with the parent virus LBNSE.Consequently,expressing of Flt3L could elevate the level of virus-neutralizing antibodies(VNA)in immunized mice which provides a better protection from a lethal rabies virus challenge.Taken together,our study extends the potential of Flt3L as a good adjuvant to develop novel rabies vaccine by enhancing the VNA production through activating the DC—Tfh^GC B axis in immunized mice.Yachun Zhang Jie Yang Mingming Li Min Cui Zhen FFu Ling Zhao Ming Zhou 2019Virologica Sinica2019,34,6:3
3Identification of novel molecular markers of mastitis caused by Staphylococcus aureus using gene expression profiling in two consecutive generations of Chinese Holstein dairy cattle显示文摘Background: Mastitis in dairy cows caused by Staphylococcus aureus is a major problem hindering economic growth in dairy farms worldwide. It is difficult to prevent or eliminate due to its asymptomatic nature and long persistence of infection. Although transcriptomic responses of bovine mammary gland cells to pathogens that cause mastitis have been studied, the common responses of peripheral blood leukocytes to S. aureus infection across two consecutive generations of dairy cattle have not been investigated.Methods: In the current study, RNA-Seq was used to profile the transcriptomes of peripheral blood leukocytes sampled from S. aureus-infected mothers and their S. aureus-infected daughters, and also healthy non-infected mothers and their healthy daughters. Differential gene expression was evaluated as follows: 1) S. aureus-infected cows versus healthy non-infected cows(S vs. H, which include all the mothers and daughters), 2) S. aureus-infected mothers versus healthy non-infected mothers(SM vs. HM), and 3) S. aureus-infected daughters versus healthy noninfected daughters(SMD vs. HMD).Results: Analysis of all identified expressed genes in the four groups(SM, SMD, HM, and HMD) showed that EPOR,IL9, IFNL3, CCL26, IL26 were exclusively expressed in both the HM and HMD groups, and that they were significantly(P < 0.05) enriched for the cytokine-cytokine receptor interaction pathway. A total of 17, 13 and 10 differentially expressed genes(DEGs)(FDR Padj. < 0.1 and |FC| > 1.2) were detected in the three comparisons, respectively. DEGs with P < 0.05 and |FC| > 2 were used for functional enrichment analyses. For the S vs. H comparison, DEGs detected included CCL20, IL13 and MMP3, which are associated with the IL-17 signaling pathway. In the SM vs. HM and SMD vs. HMD comparisons, five(BLA-DQB, C1 R, C2, FCGR1 A, and KRT10) and six(BLA-DQB, C3 AR1, CFI, FCAR, FCGR3 A, and LOC10498484) genes, respectively, were involved in the S. aureus infection pathway.Conclusions: Our study provides insights into the transcriptomic responses of bovine peripheral blood leukocytes across two generations of cattle naturally infected with S. aureus. 4Simulation study on the efficiencies of MOET nucleus breeding schemes applying marker assisted selection in dairy cattle显示文摘Advantages of breeding schemes using genetic marker information and/or multiple ovulation and embryo transfer(MOET) technology over the traditional approach were extensively evaluated through simulation.Milk yield was the trait of interest and QTL was the genetic marker utilized.Eight dairy cattle breeding scenarios were considered,i.e.,traditional progeny testing breeding scheme(denoted as STANPT),GASPT scheme including a pre-selection of young bulls entering progeny testing based on their own QTL information,MOETPT scheme using MOET technology to generate young bulls and a selection of young bulls limited within the full-sib family,GAMOPT scheme adopting both QTL pre-selection and MOET technology,COMBPT scheme using a mixed linear model which considered QTL genotype instead of the BLUP model in GAMOPT,and three non-progeny testing schemes,i.e.the MOET,GAMO and COMB schemes,corresponding to MOETPT,GAMOPT and COMBPT with progeny testing being part of the system.Animals were selected based on their breeding value which was estimated under an animal model framework.Sequential selection over 17 years was performed in the simulations and 30 replicates were designed for each scenario.The influences of using QTL information and MOET technology on favorable QTL allele frequency,true breeding values,polygenetic breeding values and the accumulated genetic superiority were extensively evaluated,for five different populations including active sires,lactating cows,bull dams,bull sires,and young bulls.The results showed that the combined schemes significantly outperformed other approaches wherein accumulated true breeding value progressed.The difference between schemes exclusively using QTL information or MOET technology was not significant.The STANPT scheme was the least efficient among the 8 schemes.The schemes using MOET technology had a higher polygenetic response than others in the 17th year.The increases of frequency of the favorable QTL allele varied more greatly across the 3 male groups than in the lactating cows group.The accumulated genetic superiorities of the GASPT scheme,MOETPT scheme,GAMOPT scheme,COMBPT scheme,MOET scheme,GAMO scheme and COMB scheme over the STANPT scheme were 8.42%,3.59%,14.58%,18.54%,4.12%,14.12%,16.50% in active sires and 2.70%,5.00%,11.05%,12.78%,7.51%,17.12%,25.38% in lactating cows.LUO WeiZhen,WANG YaChun & ZHANG Yuan Department of Animal Genetics & Breeding,College of Animal Science and Technology,China Agricultural University,Beijing 100094,China 2009Science China(Life Sciences)2009,52,3:2
5Genomic and transcriptomic analyses enable the identification of important genes associated with subcutaneous fat deposition in Holstein cows显示文摘Subcutaneous fat deposition has many important roles in dairy cattle,including immunological defense and mechanical protection.The main objectives of this study are to identify key candidate genes regulating subcutaneous fat deposition in high-producing dairy cows by integrating genomic and transcriptomic datasets.A total of 1654 genotyped Holstein cows are used to perform a genome-wide association study(GWAS)aiming to identify genes associated with subcutaneous fat deposition.Subsequently,weighted gene co-expression network analyses(WGCNA)are conducted based on RNA-sequencing data of 34 cows and cow yield deviations of subcutaneous fat deposition.Lastly,differentially expressed(DE)m RNA,lnc RNA,and differentially alternative splicing genes are obtained for 12 Holstein cows with extreme and divergent phenotypes for subcutaneous fat deposition.Forty-six protein-coding genes are identified as candidate genes regulating subcutaneous fat deposition in Holstein cattle based on GWAS.Eleven overlapping genes are identified based on the analyses of DE genes and WGCNA.Furthermore,the candidate genes identified based on GWAS,WGCNA,and analyses of DE genes are significantly enriched for pathways involved in metabolism,oxidative phosphorylation,thermogenesis,fatty acid degradation,and glycolysis/gluconeogenesis pathways.Integrating all findings,the NID2,STARD3,UFC1,DEDD,PPP1R1B,and USP21 genes are considered to be the most important candidate genes influencing subcutaneous fat deposition traits in Holstein cows.This study provides novel insights into the regulation mechanism underlying fat deposition in high-producing dairy cows,which will be useful when designing management and breeding strategies.Hailiang Zhang Siyuan Mia Luiz F.Brito Lirong Hu Lei Wang Longgang Ma Qing Xu Gang Guo Ying Yu Yachun Wang 2023Journal of Genetics and Genomics2023,50,6:1

9Crude oil mobility and its controlling factors in tight sand reservoirs in northern Songliao Basin, East China显示文摘Taking tight oil in Gaotaizi and Fuyu oil layers of the Upper Cretaceous Qingshankou Formation in northern Songliao Basin as an example, based on analyses of nuclear magnetic resonance and high pressure mercury injection, experiment methods of supercritical carbon dioxide displacement and extraction are firstly employed to quantify crude oil mobility in tight sand reservoirs with different lithologies and oil contents. The results show that, under the conditions of simulating the Cretaceous Qingshankou Formation in the northern Songliao Basin at a temperature of 76-89 °C and a pressure of 35-42 MPa, the lower limit of the porosity of the movable oil is4.4%, and the lower limit of the permeability is 0.015′10-3 mm2. The lower limit of the average pore throat radius is 21 nm. On this basis,a classification standard for three types of tight sand reservoirs is proposed. Type I reservoirs are characterized by the movable fluid saturation larger than 40%, the movable oil ratio(ratio of movable oil to total oil) greater than 30% and the starting pressure gradient in the range of 0.3-0.6 MPa/m; Type II reservoirs are characterized by the movable fluid saturation in the range of 10%–40%, the movable oil ratio in the range of 5%–30% and the starting pressure gradient in the range of 0.6–1.0 MPa/m; Type III reservoirs are characterized by the movable fluid saturation less than 10% in general, the movable oil ratio less than 5%, and the starting pressure gradient greater than1.0 MPa/m. The fluid mobility in tight sand reservoirs is mainly affected by diagenesis and sedimentary environment. 10First Look of Surface Vegetation from the Advanced Geostationary Radiation Imager(AGRI) onboard Fengyun-4B显示文摘For many years, the status of surface vegetation has been monitored by using polar-orbiting satellite imagers such as Moderate Resolution Imaging Spectroradiometer(MODIS). However, limited availability of clear-sky samples makes the derived vegetation index dependent on multiple days of observations. High-frequency observations from the geostationary Fengyun(FY) satellites can significantly reduce the influence of clouds on the synthesis of terrestrial normalized difference vegetation index(NDVI). In this study, we derived the land surface vegetation index based on observational data from the Advanced Geostationary Radiation Imager(AGRI) onboard the FY-4B geostationary satellite. First, the AGRI reflectance of visible band and near-infrared band is corrected to the land surface reflectance by the 6S radiative transfer model. The bidirectional reflectance distribution function(BRDF) model is then used to normalize the AGRI surface reflectance at different observation angles and solar geometries, and an angle-independent reflectance is derived. The AGRI surface reflectance is further corrected to the MODIS levels according to the AGRI spectral response function(SRF). Finally, the daily AGRI data are used to synthesize the surface vegetation index. It is shown that the spatial distribution of NDVI images retrieved by single-day AGRI is consistent with that of 16-day MODIS data. 12Characteristics and main control factors of Ordovician shoal dolomite gas reservoir in Gucheng area,Tarim Basin,NW China显示文摘Based on seismic, drilling data and experimental analysis, the characteristics and main controlling factors of shoal dolomite gas reservoir in the third member of Ordovician Yingshan Formation of Gucheng area, Tarim basin were examined.The study shows that the dolomite gas reservoir in Gucheng area is lithologic gas reservoir controlled by shoal and fault jointly,and its formation is mainly attributed to the following factors:(1) The continuously developing paleotectonic structure has been in the direction of gas migration and accumulation;(2) The large area of medium-high energy grain bank deposited in gentle slope environment is the material basis for the formation of dolomite reservoir;(3) Atmospheric water leaching and dolomitization and fluid dissolution in fault zone are the key factors for the formation of high-quality dolomite reservoir;(4)The natural gas comes from cracking of the ancient oil reservoir and hydrocarbon generation of dispersed organic matter in source rocks, and the NNE-trending strike-slip fault is the dominant channel for vertical migration of natural gas;(5) Limestone cap rocks in the first and second members of Yingshan Formation provide direct sealing for the formation of gas reservoir there. 13An epi-allele of SMS causes Sanming dominant genic male sterility in rice显示文摘Male sterility is an important trait in rice for hybrid rice(Oryza sativa)breeding.However,the factors involved in dominant male sterility are largely unknown.Here,we identified a gene from Sanming dominant genic male sterile rice,named Sanming dominant male sterility(SMS),and reported that an epi-allele of this locus contributes to male sterility.Segregation analysis attributed dominant male sterility to a single locus,SMS,which we characterized using a male-sterile near isogenic line(NIL)of rice cultivar 93-11.The SMS locus was heterozygous in the male-sterile 93-11 NIL,containing an epi-allele identical to that in 93-11,and an epi-allele identical to that in rice cultivar Nipponbare,which we refer to as SMS9 and SMSN,respectively.SMS9is silent and hyper-methylated,whereas SMSNis expressed and hypo-methylated in the 93-11 NIL.Overexpressing SMSNled to male sterility.Mutations in SMS rescued the male sterility of the 93-11 NIL.Interestingly,we observed the duplication of SMSN in Nipponbare,but did not observe the duplication of SMS9in 93-11.Together,these findings suggest that the reduced methylation and enhanced expression of the SMSNepi-allele in the 93-11 NIL is responsible for its role in conferring dominant male sterility.Yachun Yang Cheng Zhang Hao Li Zeyuan Yang Zuntao Xu Dewei Tai Dahu Ni Pengcheng Wei Chengxin Yi Jianbo Yang Yong Ding 2023Science China(Life Sciences)2023,66,12:0

15Achieving 1.2 fm/Hz^(1/2)Displacement Sensitivity with Laser Interferometry in Two-Dimensional Nanomechanical Resonators:Pathways towards Quantum-Noise-Limited Measurement at Room Temperature显示文摘Laser interferometry is an important technique for ultrasensitive detection of motion and displacement.We push the limit of laser interferometry through noise optimization and device engineering.The contribution of noises other than shot noise is reduced from 92.6%to 62.4%,demonstrating the possibility towards shotnoise-limited measurement.Using noise thermometry,we quantify the laser heating effect and determine the range of laser power values for room-temperature measurements.With detailed analysis and optimization of signal transduction,we achieve 1.2 fm/Hz^(1/2)displacement measurement sensitivity at room temperature in twodimensional(2D)Ca Nb_(2)O_(6)nanomechanical resonators,the best value reported to date among all resonators based on 2D materials.Our work demonstrates a possible pathway towards quantum-noise-limited measurement at room temperature.Jiankai Zhu Luming Wang Jiaqi Wu Yachun Liang Fei Xiao Bo Xu Zejuan Zhang Xiulian Fan Yu Zhou Juan Xia Zenghui Wang 2023Chinese Physics Letters2023,40,3:0
