1Integrative lipidomic and transcriptomic study unravels the therapeutic effects of saikosaponins A and D on non-alcoholic fatty liver disease显示文摘Nonalcoholic fatty liver disease(NAFLD)has become one of the most prominent causes of chronic liver diseases and malignancies.However,few therapy has been approved.Radix Bupleuri(RB)is the most frequently used herbal medicine for the treatment of liver diseases.In the current study,we aim to systemically evaluate the therapeutic effects of saikosaponin A(SSa)and saikosaponin D(SSd),the major bioactive monomers in RB,against NAFLD and to investigate the underlying mechanisms.Our results demonstrated that both SSa and SSd improved diet-induced NAFLD.Integrative lipidomic and transcriptomic analysis revealed that SSa and SSd modulated glycerolipid metabolism by regulating related genes,like Lipe and Lipg.SSd profoundly suppressed the fatty acid biosynthesis by downregulating Fasn and Acaca expression and promoted fatty acid degradation by inducing Acox1 and Cpt1 a expression.Bioinformatic analysis further predicted the implication of master transcription factors,including peroxisome proliferator-activated receptor alpha(PPARα),in the protective effects of SSa and SSd.These results were further confirmed in vitro in mouse primary hepatocytes.In summary,our study uncoded the complicated mechanisms underlying the promising anti-steatosis activities of saikosaponins(SSs),and provided critical evidence inspiring the discovery of innovative therapies based on SSa and SSd for the treatment of NAFLD and related complications.Xiaojiaoyang Li Junde Ge Yajing Li Yajie Cai Qi Zheng Nana Huang Yiqing Gu Qi Han Yunqian Li Rong Sun Runping Liu 2021Acta Pharmaceutica Sinica B2021,11,11:15
2The Optimization of Ultrasonic Extraction Process of Total Flavonoids from Rhodiola Crenulata by Response Surface Method显示文摘[Objectives]To study the optimal extraction process of total flavonoids in Rhodiola rosea L.,and further facilitate the development and utilization of Rhodiola rosea L. [Methods]With Rhodiola crenulata as raw material,ethanol as extractant,ultrasonic extraction as the extraction method,the single factor method was first used for preliminary investigation of effect of ethanol volume fraction,solid-liquid ratio,extraction temperature and extraction time on the flavonoids extraction rate,and then the Box-Behnken response surface method was used to optimize the extraction process of total flavonoids in R. crenulata. [Results] The optimal extraction conditions: ethanol concentration of 72%;solid-liquid ratio of 1∶ 43; extraction temperature of 66℃; extraction time of 50 min. Under these conditions,the extraction rate of total flavonoids from R. crenulata was 2. 591%. 3Nuclear Aurora kinase A triggers programmed death-ligand 1-mediated immune suppression by activating MYC transcription in triple-negative breast cancer显示文摘Background:Increasing studies have reported that oncogenes regulate components of the immune system,suggesting that this is a mechanism for tumorigenesis.Aurora kinase A(AURKA),a serine/threonine kinase,is involved in cell mitosis and is essential for tumor cell proliferation,metastasis,and drug resistance.However,the mechanism by which AURKA is involved in immune response regulation is unclear.Therefore,this study aimed to investigate the role of AURKA in immune regulation in triple-negative breast cancer(TNBC).Methods:Peripheral blood mononuclear cells(PBMCs)were co-cultured with TNBC cells.The xCELLigence Real-Time Cell Analyzer-MP system was used to detect the killing efficiency of immune cells on TNBC cells.The expression of immune effector molecules was tested by quantitative real-time polymerase chain reaction(qRT-PCR)to evaluate immune function.Furthermore,to validate AURKA-regulated immune response in vivo,4T1 murine breast cancer cell line with AURKA overexpression or downregulation was engrafted into BALB/c mice.The distribution and proportion of immune cells in tumors were further evaluated by immunohistochemistry and flow cytometry.Results:Downregulation of AURKA in TNBC cells increased immune response by activating CD8^(+)T cell proliferation and activity.Nuclear rather than cytoplasmic AURKA-derived programmed death-ligand 1(PD-L1)expression was independent of its kinase activity.Mechanistic investigations showed that nuclear AURKA increased PD-L1 expression via an MYC-dependent pathway.PD-L1 overexpression mostly reversed AURKA silencing-induced expression of immune effector molecules,including interleukin-(IL-2),interferon-γ(IFN-γ),and perforin.Moreover,AURKA expression was negatively correlated with the enrichment and activity of tumor-infiltrating CD8^(+)T cells in 4T1 engrafted BALB/c mouse model.Conclusions:Nuclear AURKA elevated PD-L1 expression via an MYCdependent pathway and contributed to immune evasion in TNBC.Therapies targeting nuclear AURKA may restore immune responses against tumors.Shulan Sun Wei Zhou Xiaoxi Li Fei Peng Min Yan Yajing Zhan Fan An Xiaoyan Li Yunyong Liu Quentin Liu Haozhe Piao 2021Cancer Communications2021,41,9:2
4Recombinant human erythropoietin accelerated the proliferation of non-small cell lung cancer cell lines and reduced the expression of VEGF,HIF-1α,and PD-L1 under a simulated hypoxic environment in vitro显示文摘Background:As erythropoietin(EPO)has been used to treat anemia in cancer patients,negative controversy has continued.Unfortunately,its effects on non-small-cell lung carcinoma(NSCLC)cell lines are uncertain and the phenomenon of inducing immune escape of tumor cells remains to be explored.This study aimed to provide an important basis for the application of exogenous EPO in the treatment of tumor-associated anemia.Methods:Cells were cultured in 1%O2,5%CO_(2),and 94%N2 to simulate a hypoxic environment of the tumor.A549 cell line(lower expression EPOR)and NCI-H838 cell line(higher expression EPOR)were treated with 2 and 8 U/ml recombinant human EPO(rhEPO).CCK-8 method was used to determine the logarithmic growth phase of the cells and to detect cell proliferation.The expression levels of VEGF,HIF-1α,and PD-L1 were determined by western blot.One-way ANOVA was used for statistical analysis between groups,withp<0.05 indicating a significant difference.Results:Hypoxia itself could decrease the survival rate of NSCLC cells.Under the hypoxic condition,rhEPO induced tumor cells proliferation,especially in the NCI-H838 cell line,where 2 U/ml rhEPO increased the total number of surviving cells(Hypoxia+rhEPO 2 U/ml vs.Hypoxia,p<0.05).Western blot analysis showed that hypoxia upregulated the expression of VEGF,HIF-1α,and PD-L1 in NSCLC cell lines(Normoxia vs.Hypoxia,p<0.05),but may not be dependent on the expression levels of EPOR.RhEPO decreased the expression levels of VEGF and HIF-1α.In the A549 cell line,it depended on the concentration of rhEPO and was particularly obvious in HIF-1α(Hypoxia vs.Hypoxia+rhEPO 2 U/ml vs.Hypoxia+rhEPO 8 U/ml,p<0.05).A low concentration of rhEPO may not reduce VEGF expression.In the NCI-H838 cell line,the effect of rhEPO on VEGF was more obvious,but it may be independent of rhEPO concentrations.The downregulation of PD-L1 expression by rhEPO was only presented in the A549 cell line and required higher rhEPO concentrations(Hypoxia+rhEPO 8 U/ml vs.Hypoxia&Hypoxia+rhEPO 2 U/ml,p<0.05).Conclusion:The effect of prolonged high concentrations of rhEPO under hypoxic conditions resulted in accelerated cells proliferation of non-small-cell lung cancer and was independent of EPOR expression levels on the cell lines surface.Hypoxia resulted in increased expression of VEGF,HIF-1α,and PD-L1 on the NSCLC cell lines.Under normoxic conditions,rhEPO did not affect the expression of VEGF,HIF-1α,and PD-L1;but under hypoxic conditions,the application of rhEPO reduced the expression of VEGF,HIF-1α,and PD-L1,producing an impact on the biological behavior of tumor cells.Yajing Zhang Yangchun Feng Xiaojie Sun 2022Chronic Diseases and Translational Medicine2022,8,2:2

6Trans-cinnamaldehyde promotes nitric oxide release via the protein kinase-B/v-Akt murine thymoma viral oncogene-endothelial nitric oxide synthase pathway to alleviate hypertension in SHR.Cg-Lepr^(cp)/NDmcr rats显示文摘OBJECTIVE:To evaluate whether endothelial dysfunction and hypertension are prevented by trans-cinnamaldehyde(t CA) through the activation of endothelial nitric oxide synthase(e NOS).METHODS:Human umbilical vein endothelial cells(HUVECs) were cultured in vitro and stimulated with t CA to determine cell viability using the methyl thiazolyl tetrazolium assay.The effect of t CA on nitric oxide(NO) production was determined by diaminofluorescein-dyes in the absence or presence of inhibitors of e NOS,AMPK,PKA,and AKT.The effect of t CA on blood pressure was determined by the tail-cuff method in obesity spontaneous hypertension(SHR.Cg-Leprcp/NDmcr) rats.The phosphorylation of e NOS and protein expression of the insulin-signaling pathway(Ins R-IRS1-PI3 K-AKT)were measured by western blot.RESULTS:t CA at concentrations less than 100 μM did not affect cell viability in cultured HUVECs.Stimulation with t CA promoted NO release in a time-dependent manner compared with the control group.t CA-treated HUVECs also significantly increased AKT-Ser473 and e NOS-Ser1177 phosphorylation.In SHR-CP rats,treatment with t CA at a dose of 40 mg/kg/day for 6 weeks markedly reduced the systolic blood pressure and diastolic blood pressure,increased the phosphorylation of AKT and e NOS,and increased urinary nitric oxidation.CONCLUSION:t CA attenuated endothelial dysfunction and reduced blood pressure in SHR-CP rats.The underlying mechanisms may involve the increase in AKT and e NOS phosphorylation and the release of e NOS-derived NO.Zhang Lu Wu Lili Liu Ximing Yoshitomi Hisae Ikeda Katsumi Negishi Hiroko Pan Yajing Sun Wen Qin Lingling Li Juan-E Xu Tunhai Liu Tonghua Gao Ming 2018Journal of Traditional Chinese Medicine2018,38,4:2
7Knowledge-aware path: interpretable graph reasoning in proactive dialogue generation显示文摘Proactive dialogue generates dialogue utterance based on a conversation goal and a given knowledge graph(KG). Existing methods combine knowledge of each turn of dialogue with knowledge triples by hidden variables, resulting in the interpretability of generation results is relatively poor. An interpretable knowledge-aware path(KAP) model was proposed for knowledge reasoning in proactive dialogue generation.KAP model can transform explicit and implicit knowledge of each turn of dialogue into corresponding dialogue state matrix, thus forming the KAP for dialogue history. Based on KAP, the next turn of dialogue state vector can be infered from both the topology and semantic of KG. This vector can indicate knowledge distribution of next sentence, so it enhances the accuracy and interpretability of dialogue generation. 8Dimensional Gradient Structure of CoSe2@CNTs-MXene Anode Assisted by Ether for High-Capacity,Stable Sodium Storage显示文摘Recently,abundant resources,low-cost sodium-ion batteries are deemed to the new-generation battery in the field of largescale energy storage.Nevertheless,poor active reaction dynamics,dissolution of intermediates and electrolyte matching problems are significant challenges that need to be solved.Herein,dimensional gradient structure of sheet-tube-dots is constructed with CoSe2@CNTs-MXene.Gradient structure is conducive to fast migration of electrons and ions with the association of ether electrolyte.For half-cell,CoSe2@CNTs-MXene exhibits high initial coulomb efficiency(81.7%)and excellent cycling performance(400 mAh g^-1 cycling for 200 times in 2 Ag^−1).Phase transformation pathway from crystalline CoSe2-Na2Se with Co and then amorphous CoSe2 in the discharge/charge process is also explored by in situ X-ray diffraction.Density functional theory study discloses the CoSe2@CNTs-MXene in ether electrolyte system which contributes to stable sodium storage performance owing to the strong adsorption force from hierarchical structure and weak interaction between electrolyte and electrode interface.For full cell,CoSe2@CNTs-MXene//Na3V2(PO4)3/C full battery can also afford a competitively reversible capacity of 280 mAh g^−1 over 50 cycles.Concisely,profiting from dimensional gradient structure and matched electrolyte of CoSe2@CNTs-MXene hold great application potential for stable sodium storage.Enze Xu Pengcheng Li Junjie Quan Hanwen Zhu Li Wang Yajing Chang Zhenjie Sun Lei Chen Dabin Yu Yang Jiang 2021Nano-Micro Letters2021,13,2:1

12Distinct functions of Trio GEF domains in axon outgrowth of cerebellar granule neurons显示文摘As a critical guanine nucleotide exchange factor(GEF) regulating neurite outgrowth, Trio coordinates multiple processes of cytoskeletal dynamics through activating Rac1, Cdc42 and RhoA small GTPases by two GEF domains, but the in vivo roles of these GEF domains and corresponding downstream effectors have not been determined yet. We established multiple lines of knockout mice and assessed the respective roles of Trio GEF domains and Rac1 in axon outgrowth. Knockout of total Trio in cerebellar granule neurons(CGNs) led to an impaired F-actin rearrangement of growth cone and hence a retarded neurite outgrowth. Such a retardation was reproduced by inhibition of GEF1 domain or knockdown of Cdc42 and restored apparently by introduction of active Cdc42. As Rac1 deficiency did not affect the neurite outgrowth of CGNs, we suggested that Trio GEF1-mediated Cdc42 activation was required for neurite outgrowth. We established a GEF2-knockout line with deletion of all Trio isoforms except a cerebella-specific Trio8, a short isoform of Trio without GEF2 domain, and used this line as a GEF2-deficient animal model. The GEF2-deficient CGNs had a normal neurite outgrowth but abolished Netrin-1-promoted growth, without affecting Netrin-1 induced Rac1 activation. We thus suggested that Trio GEF1-mediated Cdc42 activation rather than Rac1 activation drives the F-actin dynamics necessary for neurite outgrowth, while GEF2 functions in Netrin-1-promoted neurite elongation. 13A bacterial artificial chromosome transgenic mouse model for visualization of neurite growth显示文摘Class III β-tubulin(Tubb3) is a component of the microtubules in neurons and contributes to microtubule dynamics that are required for axon outgrowth and guidance during neuronal development. We here report a novel bacterial artificial chromosome(BAC) transgenic mouse line that expresses Class III β-tubulin fused to m Cherry, an improved monomeric red fluorescent protein, for the visualization of microtubules during neuronal development. A BAC containing Tubb3 gene was modified by insertion of m Cherry complementary DNA downstream of Tubb3 coding sequence via homologous recombination. m Cherry fusion protein was expressed in the nervous system and testis of the transgenic animal, and the fluorescent signal was observed in the neurons that located in the olfactory bulb, cerebral cortex, hippocampal formation, cerebellum, as well as the retina. Besides, Tubb3-m Cherry fusion protein mainly distributed in neurites and colocalized with endogenous Class III β-tubulin. The fusion protein labels Purkinje cell dendrites during cerebellar circuit formation. 17Novel machine learning framework for thermal conductivity prediction by crystal graph convolution embedded ensemble显示文摘With the advent of the big data era,artificial intelligence technology has penetrated and deeply affected our daily life.In addition,data-based machine learning algorithms have been applied to physics,chemistry,material science,and other basic science fields.However,the scarcity of data sets is known as the main obstacle to its development.Mining effective information from the limited data samples and building an appropriate machine learning algorithms framework are the major breakthroughs.For solid materials,the intrinsic properties are closely related to their atomic composition and relative positions,namely crystal structures.Here,inspired by the emerging of graph convolution neural network and material crystal graph,we proposed an integrated algorithms framework embedded crystal graph to train and predict the lattice thermal conductivities of crystal materials.This machine learning algorithms framework showed superior learning and generalization ability.In addition,not only in predicting thermal conductivities,but our framework also has great performance in predicting other phonon or electron-related properties.This strategy provided a new approach in the design of machine learning framework,which indicated the great potential for the application of machine learning in material science.Yajing Sun Wenping Hu 2022SmartMat2022,3,3:1
18Efficient gene transfection of suspension cells by highly branched poly(β-amino ester)显示文摘Suspension cells play a crucial role in many biological processes. However, compared to adherent cells, it is particularly challenging to introduce exogenous genes into suspension cells to regulate their biological functions with non-viral gene vectors, mainly due to the low cellular uptake and endosomal escape of polyplexes. Herein, to improve the interactions of polyplexes with cellular membranes, we design and synthesize highly branched poly(β-amino ester)(HPAE) via an “A2 + B4 + C2” Michael addition strategy.Results show that branching significantly increases DNA condensation of HPAE, cellular uptake and endosomal escape of HPAE/DNA polyplexes. In mast cells(MCs), HPAE exhibits up to 80-fold higher gene transfection efficiency compared to the corresponding linear poly(β-amino ester)(LPAE) and the leading commercial gene transfection reagents PEI25k, jetPEI, and Lipofectamine 3000, without causing obvious cytotoxicity.
