1Diagnostic value of 5 serum biomarkers for hepatocellular carcinoma with different epidemiological backgrounds:A large-scale,retrospective study显示文摘Objective:Hepatocellular carcinoma(HCC)is a lethal global disease that requires an accurate diagnosis.We assessed the potential of 5 serum biomarkers(AFP,AFU,GGT-II,GPC3,and HGF)in the diagnosis of HCC.Methods:In this retrospective study,we measured the serum levels of each biomarker using ELISAs in 921 participants,including 298 patients with HCC,154 patients with chronic hepatitis(CH),122 patients with liver cirrhosis(LC),and 347 healthy controls from 3 hospitals.Patients negative for hepatitis B surface antigen and hepatitis C antibody(called'NBNC-HCC')and patients positive for the above indices(called'HBV-HCC and HCV-HCC')were enrolled.The selected diagnostic model was constructed using a training cohort(n=468),and a validation cohort(n=453)was used to validate our results.Receiver operating characteristic analysis was used to evaluate the diagnostic accuracy.Results:Theα-L-fucosidase(AFU)/α-fetoprotein(AFP)combination was best able to distinguish NBNC-HCC[area under the curve:0.986(95%confidence interval:0.958–0.997),sensitivity:92.6%,specificity:98.9%]from healthy controls in the test cohort.For screening populations at risk of developing HCC(CH and LC),the AFP/AFU combination improved the diagnostic specificity for early-stage HCC[area under the curve:0.776(0.712–0.831),sensitivity:52.5%,specificity:91.6%in the test group].In all-stage HBV-HCC and HCV-HCC,AFU was also the best candidate biomarker combined with AFP[area under the curve:0.835(0.784–0.877),sensitivity 69.1%,specificity:87.4%in the test group].All results were verified in the validation group.Conclusions:The AFP/AFU combination could be used to identify NBNC-HCC from healthy controls and hepatitis-related HCC from at-risk patients.Dongming Liu Yi Luo Lu Chen Liwei Chen Duo Zuo Yueguo Li Xiaofang Zhang Jing Wu Qing Xi Guangtao Li Lisha Qi Xiaofen Yue Xiehua Zhang Zhuoyu Sun Ning Zhang Tianqiang Song Wei Lu Hua Guo 2021Cancer Biology & Medicine2021,18,1:10
2CPI-17-mediated contraction of vascular smooth muscle is essential for the development of hypertension in obese mice显示文摘Several factors have been implicated in obesity-related hypertension, but the genesis of the hypertension is largely unknown. In this study, we found a significantly upregulated expression of CPI-17(C-kinasepotentiated protein phosphatase 1 inhibitor of 17 kDa) and protein kinase C(PKC) isoforms in the vascular smooth muscles of high-fat diet(HFD)-fed obese mice. The obese wild-type mice showed a significant elevation of blood pressure and enhanced calcium-sensitized contraction of vascular smooth muscles. However, the obese CPI-17-deficient mice showed a normotensive blood pressure, and the calcium-sensitized contraction was consistently reduced. In addition, the mutant muscle displayed an abolished responsive force to a PKC activator and a 30%-50% reduction in both the initial peak force and sustained force in response to various G protein-coupled receptor(GPCR) agonists. Our observations showed that CPI-17-mediated calcium sensitization is mediated through a GPCR/PKC/CPI-17/MLCP/RLC signaling pathway. 4Methionine oxidation and reduction of the ethylene signaling component MaEIL9 are involved in banana fruit ripening显示文摘The ethylene insensitive 3/ethylene insensitive3-like(EIN3/EIL)plays an indispensable role in fruit ripening.However,the regulatory mechanism that links post-translational modification of EIN3/EIL to fruit ripening is largely unknown.Here,we studied the expression of 13 MaE IL genes during banana fruit ripening,among which MaE IL9 displayed higher enhancement particularly in the ripening stage.Consistent with its transcript pattern,abundance of MaE IL9 protein gradually increased during the ripening process,with maximal enhancement in the ripening.DNA affinity purification(DAP)-seq analysis revealed that MaE IL9 directly targets a subset of genes related to fruit ripening,such as the starch hydrolytic genes MaA MY3D and MaB AM1.Stably overexpressing MaE IL9 in tomato fruit hastened fruit ripening,whereas transiently silencing this gene in banana fruit retarded the ripening process,supporting a positive role of MaEIL9 in fruit ripening.Moreover,oxidation of methionines(Met-129,Met-130,and Met-282)in MaEIL9 resulted in the loss of its DNA-binding capacity and transcriptional activation activity.Importantly,we identified MaEIL9 as a potential substrate protein of methionine sulfoxide reductase A MaMsrA4,and oxidation of Met-129,Met-130,and Met-282in MaEIL9 could be restored by MaMsrA4.Collectively,our findings reveal a novel regulatory network controlling banana fruit ripening,which involves MaMsrA4-mediated redox regulation of the ethylene signaling component MaEIL9.Lisha Zhu Lin Chen Chaojie Wu Wei Shan Danling Cai Zengxiang Lin Wei Wei Jianye Chen Wangjin Lu Jianfei Kuang 2023Journal of Integrative Plant Biology2023,65,1:1

6Distinct functions of Trio GEF domains in axon outgrowth of cerebellar granule neurons显示文摘As a critical guanine nucleotide exchange factor(GEF) regulating neurite outgrowth, Trio coordinates multiple processes of cytoskeletal dynamics through activating Rac1, Cdc42 and RhoA small GTPases by two GEF domains, but the in vivo roles of these GEF domains and corresponding downstream effectors have not been determined yet. We established multiple lines of knockout mice and assessed the respective roles of Trio GEF domains and Rac1 in axon outgrowth. Knockout of total Trio in cerebellar granule neurons(CGNs) led to an impaired F-actin rearrangement of growth cone and hence a retarded neurite outgrowth. Such a retardation was reproduced by inhibition of GEF1 domain or knockdown of Cdc42 and restored apparently by introduction of active Cdc42. As Rac1 deficiency did not affect the neurite outgrowth of CGNs, we suggested that Trio GEF1-mediated Cdc42 activation was required for neurite outgrowth. We established a GEF2-knockout line with deletion of all Trio isoforms except a cerebella-specific Trio8, a short isoform of Trio without GEF2 domain, and used this line as a GEF2-deficient animal model. The GEF2-deficient CGNs had a normal neurite outgrowth but abolished Netrin-1-promoted growth, without affecting Netrin-1 induced Rac1 activation. We thus suggested that Trio GEF1-mediated Cdc42 activation rather than Rac1 activation drives the F-actin dynamics necessary for neurite outgrowth, while GEF2 functions in Netrin-1-promoted neurite elongation. 12Construction of n-TiO_(2)/p-Ag_(2)O Junction on Carbon Fiber Cloth with Vis-NIR Photoresponse as a Filter-Membrane-Shaped Photocatalyst显示文摘The development of effective and reusable photocatalysts with broad-spectra activity has attracted attention.Herein,we have constructed n-TiO_(2)/p-Ag_(2)O junction on carbon fiber(CF)cloth as an efficient and recyclable photocatalyst.With CF cloth as the substrate,TiO_(2) nanorods(length:1-2μm)are prepared by a hydrothermal process,and the in-situ growth of Ag_(2)O nanoparticles(10-20 nm)is then realized by chemical bath deposition route.The flexible CF/TiO_(2)/Ag_(2)O cloth(area:4×4 cm^(2))shows a broad and strong photo-absorption(200-1000 nm).Under the illumination of visible-light(λ>400 nm),CF/TiO_(2)/Ag_(2)O cloth can efficiently eliminate 99.2%rhodamine B(RhB),99.4%acid orange 7(AO7),87.6%bisphenol A(BPA),and 89.5%hexavalent chromium(Cr^(6+))in 100 min,superior to CF/Ag_(2)O cloth(83.5%RhB,60.0%AO7,31.2%BPA and 41.8%Cr^(6+)).In particular,under the NIR-light illumination(980 nm laser),CF/TiO_(2)/Ag_(2)O cloth can remove 70.9%AO7 and 60.0%Cr^(6+) in 100 min,which are significantly higher than those by CF/Ag_(2)O cloth(19.8%AO7 and 18.9%Cr^(6+)).In addition,CF/TiO_(2)/Ag_(2)O cloth(diameter:10 cm),as a filter-membrane,can effectively wipe off 94.4%flowing RhB solution(rate:~1 L h^(−1))at 6th filtering/degrading grade.Thus,CF/TiO_(2)/Ag_(2)O cloth can be used as a Vis-NIR-responded filter-membrane-shaped photocatalyst with high-efficiency for purifying wastewater.Gumila Duoerkun Yan Zhang Zhun Shi Xiaofeng Shen Wei Cao Ting Liu Jianshe Liu Quanyuan Chen Lisha Zhang 2020Advanced Fiber Materials2020,2,1:0
13Nucleoporin 160 Regulates Flowering through Anchoring HOS1 for Destabilizing CO in Arabidopsis显示文摘Nuclear pore complexes(NPCs),which comprise multiple copies of nucleoporins(Nups),are large protein assemblies embedded in the nuclear envelope connecting the nucleus and cytoplasm.Although it has been known that Nups affect flowering in Arabidopsis,the underlying mechanisms are poorly understood.Here,we show that loss of function of Nucleoporin 160(Nup160)leads to increased abundance of CONSTANS(CO)protein and the resulting upregulation of FLOWERING LOCUS T(FT)specifically in the morning.We demonstrate that Nup160 regulates CO protein stability through affecting NPC localization of an E3-ubiquitin ligase,HIGH EXPRESSION OF OSMOTICALLY RESPONSIVE GENES1(HOS1),which destabilizes CO protein in the morning period.Taken together,these results provide a mechanistic understanding of Nup function in the transition from vegetative to reproductive growth,suggesting that deposition of HOS1 at NPCs by Nup160 is essential for preventing precocious flowering in response to photoperiod in Arabidopsis.Chunying Li Lu Liu Zhi Wei Norman Teo Lisha Shen Hao Yu 2020Plant Communications2020,1,2:0
14LIMK2 is required for membrane cytoskeleton reorganization of contracting airway smooth muscle显示文摘Airway smooth muscle(ASM)has developed a mechanical adaption mechanism by which it transduces force and responds to environmental forces,which is essential for periodic breathing.Cytoskeletal reorganization has been implicated in this process,but the regulatory mechanism remains to be determined.We here observe that ASM abundantly expresses cytoskeleton regulators Limk1 and Limk2,and their expression levels are further upregulated in chronic obstructive pulmonary disease(COPD)animals.By establishing mouse lines with deletions of Limk1 or Limk2,we analyse the length-sensitive contraction,F/Gactin dynamics,and F-actin pool of mutant ASM cells.As LIMK1 phosphorylation does not respond to the contractile stimulation,LIMK1-deficient ASM develops normal maximal force,while LIMK2 or LIMK1/LIMK2 deficient ASMs show approximately 30%inhibition.LIMK2 deletion causes a significant decrease in cofilin phosphorylation along with a reduced F/G-actin ratio.As LIMK2 functions independently of cross-bridge movement,this observation indicates that LIMK2 is necessary for F-actin dynamics and hence force transduction.Moreover,LIMK2-deficient ASMs display abolishes stretching-induced suppression of 5-hydroxytryptamine(5-HT)but not acetylcholine-evoks force,which is due to the differential contraction mechanisms adopted by the agonists.We propose that LIMK2-mediated cofilin phosphorylation is required for membrane cytoskeleton reorganization that is necessary for ASM mechanical adaption including the 5-HT-evoked length-sensitive effect.Yeqiong Li Yuwei Zhou Pei Wang Tao Tao Lisha Wei Ye Wang Wei Wang Yanyan Zheng Zhihui Jiang Tiantian Qiu Wei Zhao Jie Sun Xin Chen Xue-Na Zhang Min-Sheng Zhu 2021Journal of Genetics and Genomics2021,48,6:0

16A nomogram-based immune-serum scoring system predicts overall survival in patients with lung adenocarcinoma显示文摘Objective:The immunoscore,which is used to quantify immune infiltrates,has greater relative prognostic value than tumor,node,and metastasis(TNM)stage and might serve as a new system for classification of colorectal cancer.However,a comparable immunoscore for predicting lung adenocarcinoma(LUAD)prognosis is currently lacking.Methods:We analyzed the expression of 18 immune features by immunohistochemistry in 171 specimens.The relationship of immune marker expression and clinicopathologic factors to the overall survival(OS)was analyzed with the Kaplan-Meier method.A nomogram was developed by using the optimal features selected by least absolute shrinkage and selection operator(LASSO)regression in the training cohort(n=111)and evaluated in the validation cohort(n=60).Results:The indicators integrated in the nomogram were TNM stage,neuron-specific enolase,carcino-embryonic antigen,CD8 center of tumor(CT),CD8 invasive margin(IM),Fox P3 CT,and CD45 ROCT.The calibration curve showed prominent agreement between the observed 2-and 5-year OS and that predicted by the nomogram.To simplify the nomogram,we developed a new immune-serum scoring system(I-SSS)based on the points awarded for each factor in the nomogram.Our I-SSS was able to stratify same-stage patients into different risk subgroups.The combination of I-SSS and TNM stage had better prognostic value than the TNM stage alone.Conclusions:Our new I-SSS can accurately and individually predict LUAD prognosis and may be used to supplement prognostication based on the TNM stage.Qiujuan Huang Tongyuan Qu Lisha Qi Changxu Liu Yuhong Guo Qianru Guo Guangning Li Yalei Wang Wenshuai Zhang Wei Zhao Danyang Ren Leina Sun Shengguang Wang Bin Meng Baocun Sun Bin Zhang Wenjuan Ma Wenfeng Cao 2021Cancer Biology & Medicine2021,18,2:0
17WiFi6 Dynamic Channel Optimization Method for Fault Tolerance in Power Communication Network显示文摘As the scale of power networks has expanded,the demand for multi-service transmission has gradually increased.The emergence of WiFi6 has improved the transmission efficiency and resource utilization of wireless networks.However,it still cannot cope with situations such as wireless access point(AP)failure.To solve this problem,this paper combines orthogonal fre-quency division multiple access(OFDMA)technology and dynamic channel optimization technology to design a fault-tolerant WiFi6 dynamic resource optimization method for achieving high quality wireless services in a wirelessly covered network even when an AP fails.First,under the premise of AP layout with strong coverage over the whole area,a faulty AP determination method based on beacon frames(BF)is designed.Then,the maximum signal-to-interference ratio(SINR)is used as the principle to select AP reconnection for the affected users.Finally,this paper designs a dynamic access selection model(DASM)for service frames of power Internet of Things(IoTs)and a schedul-ing access optimization model(SAO-MF)based on multi-frame transmission,which enables access optimization for differentiated services.For the above mechanisms,a heuristic resource allocation algorithm is proposed in SAO-MF.Simulation results show that the method can reduce the delay by 15%and improve the throughput by 55%,ensuring high-quality communication in power wireless networks.Hong Zhu Lisha Gao Lei Wei Guangchang Yang Sujie Shao 2023Computers, Materials & Continua2023,,6:0
