1Low-Temperature Selective Catalytic Reduction of NO with NH_3 over Fe–Ce–O_x Catalysts显示文摘In this study,we used a simple impregnation method to prepare Fe-Ce-O_x catalysts and tested them regarding their low-temperature(200-300 ℃) selective catalytic reduction(SCR) of NO using NH_3.We investigated the effects of Fe/Ce molar ratio,the gas hourly space velocity(GHSV),the stability and SO_2/H_2O resistance of the catalysts.The results showed that the FeCe(1:6)O_x(Ce/Fe molar ratio is 1:6) catalyst,which has some ordered parallel channels,exhibited good SCR performance.The FeCe(1:6)O_x catalyst had the highest NO conversion with an activity of 94-99%at temperatures between 200 and300 ℃ at a space velocity of 28,800 h^(-1).The NO conversion for the FeCe(1:6)O_x catalyst also reached 80-98%between 200 and 300 ℃ at a space velocity of204,000h^(-1).In addition,the FeCe(1:6)O_x catalyst demonstrated good stability in a 10-h SCR reaction at200-300 ℃.Even in the presence of SO_2 and H_2O,the FeCe(1:6)O_x catalyst exhibited good SCR performance.Yan Sun Ying Guo Wei Su Yajuan Wei 2017Transactions of Tianjin University2017,23,1:5
2miR-7/TGF-β2 axis sustains acidic tumor microenvironment-induced lung cancer metastasis显示文摘Acidosis,regardless of hypoxia involvement,is recognized as a chronic and harsh tumor microenvironment(TME)that educates malignant cells to thrive and metastasize.Although overwhelming evidence supports an acidic environment as a driver or ubiquitous hallmark of cancer progression,the unrevealed core mechanisms underlying the direct effect of acidification on tumorigenesis have hindered the discovery of novel therapeutic targets and clinical therapy.Here,chemical-induced and transgenic mouse models for colon,liver and lung cancer were established,respectively.miR-7 and TGF-β2 expressions were examined in clinical tissues(n=184).RNA-seq,miRNA-seq,proteomics,biosynthesis analyses and functional studies were performed to validate the mechanisms involved in the acidic TME-induced lung cancer metastasis.Our data show that lung cancer is sensitive to the increased acidification of TME,and acidic TME-induced lung cancer metastasis via inhibition of miR-7-5 p.TGF-β2 is a direct target of miR-7-5 p.The reduced expression of miR-7-5 p subsequently increases the expression of TGF-β2 which enhances the metastatic potential of the lung cancer.Indeed,overexpression of miR-7-5 p reduces the acidic p H-enhanced lung cancer metastasis.Furthermore,the human lung tumor samples also show a reduced miR-7-5 p expression but an elevated level of activated TGF-β2;the expressions of both miR-7-5 p and TGF-β2 are correlated with patients’survival.We are the first to identify the role of the miR-7/TGF-β2 axis in acidic p H-enhanced lung cancer metastasis.Our study not only delineates how acidification directly affects tumorigenesis,but also suggests miR-7 is a novel reliable biomarker for acidic TME and a novel therapeutic target for non-small cell lung cancer(NSCLC)treatment.Our study opens an avenue to explore the p H-sensitive subcellular components as novel therapeutic targets for cancer treatment.Tao Su Suchao Huang Yanmin Zhang Yajuan Guo Shuwei Zhang Jiaji Guan Mingjing Meng Linxin Liu Caiyan Wang Dihua Yu Hiu-Yee Kwan Zhiying Huang Qiuju Huang Elaine Lai-Han Leung Ming Hu Ying Wang Zhongqiu Liu Linlin Lu 2022Acta Pharmaceutica Sinica B2022,12,2:2
3Insulin sensitivity,βcell function,and adverse pregnancy outcomes in women with gestational diabetes显示文摘Background: The potential impact of β cell function and insulin sensitivity on adverse pregnancy outcomes in women with gestational diabetes mellitus (GDM) remains uncertain. We aimed to investigate the association between β cell dysfunction, insulin resistance, and the composite adverse pregnancy outcomes.Methods: This observational study included 482 women diagnosed with GDM during pregnancy. Quantitative metrics on β cell function and insulin sensitivity during pregnancy were calculated using traditional equations. The association of β cell dysfunction and insulin resistance with the risk of the composite adverse pregnancy outcomes was investigated using multivariable-adjusted logistic regression models.Results: Multivariable-adjusted odds ratios (ORs) of adverse pregnancy outcomes across quartiles of homeostatic model assessment for insulin resistance (HOMA-IR) were 1.00, 0.95, 1.34, and 2.25, respectively (P for trend = 0.011). When HOMA-IR was considered as a continuous variable, the multivariable-adjusted OR of adverse pregnancy outcomes was 1.34 (95% confidence interval 1.16-1.56) for each 1-unit increase in HOMA-IR. Multivariable-adjusted ORs of adverse pregnancy outcomes across quartiles of homeostatic model assessment for β cell function (HOMA-β) were 1.00, 0.51, 0.60, and 0.53, respectively (P for trend = 0.068). When HOMA-β was considered as a continuous variable, the multivariable-adjusted OR of adverse pregnancy outcomes was 0.57 (95% CI 0.24-0.90) for each 1-unit increase in HOMA-β. However, other quantitative metrics were not associated with the composite adverse pregnancy outcomes.Conclusions: We demonstrated a significant association of β cell function and insulin sensitivity with the risk of adverse pregnancy outcomes. 6Unique and complementary suppression of cGAS-STING and RNA sensing- triggered innate immune responses by SARS-CoV-2 proteins显示文摘The emergence of SARS-CoV-2 has resulted in the COVID-19 pandemic,leading to millions of infections and hundreds of thousands of human deaths.The efficient replication and population spread of SARS-CoV-2 indicates an effective evasion of human innate immune responses,although the viral proteins responsible for this immune evasion are not clear.In this study,we identified SARS-CoV-2 structural proteins,accessory proteins,and the main viral protease as potent inhibitors of host innate immune responses of distinct pathways.In particular,the main viral protease was a potent inhibitor of both the RLR and cGAS-STING pathways.Viral accessory protein 0RF3a had the unique ability to inhibit STING,but not the RLR response.On the other hand,structural protein N was a unique RLR inhibitor.0RF3a bound STING in a unique fashion and blocked the nuclear accumulation of p65 to inhibit nuclear factor-KB signaling.3CL of SARS-CoV-2 inhibited K63-ubiquitin modification of STING to disrupt the assembly of the STING functional complex and downstream signaling.Diverse vertebrate STINGs,including those from humans,mice,and chickens,could be inhibited by 0RF3a and 3CL of SARS-CoV-2.The existence of more effective innate immune suppressors in pathogenic coronaviruses may allow them to replicate more efficiently in vivo.Since evasion of host innate immune responses is essential for the survival of all viruses,our study provides insights into the design of therapeutic agents against SARS-CoV-2.Yajuan Rui Jiaming Su Si Shen Ying Hu Dingbo Huang Wenwen Zheng Meng Lou Yifei Shi Meng Wang Shiqi Chen Na Zhao Qi Dong Yong Cai Rongzhen Xu Shu Zheng Xiao-Fang Yu 2021Signal Transduction and Targeted Therapy2021,6,4:1

14TRAF3 activates STING-mediated suppression of EV-A71 and target of viral evasion显示文摘Innate immunity represents one of the main host responses to viral infection.1,2,3 STING(Stimulator of interferon genes),a crucial immune adapter functioning in host cells,mediates cGAS(Cyclic GMP-AMP Synthase)sensing of exogenous and endogenous DNA fragments and generates innate immune responses.4 Whether STING activation was involved in infection and replication of enterovirus remains largely unknown.In the present study,we discovered that human enterovirus A71(EV-A71)infection triggered STING activation in a cGAS dependent manner.EV-A71 infection caused mitochondrial damage and the discharge of mitochondrial DNA into the cytosol of infected cells.However,during EV-A71 infection,cGAS-STING activation was attenuated.EV-A71 ^(pro)teins were screened and the viral ^(pro)tease 2A^(pro) had the greatest capacity to inhibit cGAS-STING activation.We identified TRAF3 as an important factor during STING activation and as a target of 2A^(pro).Supplement of TRAF3 rescued cGAS-STING activation suppression by 2A^(pro).TRAF3 supported STING activation mediated TBK1 phosphorylation.Moreover,we found that 2A^(pro) ^(pro)tease activity was essential for inhibiting STING activation.Furthermore,EV-D68 and CV-A16 infection also triggered STING activation.The viral ^(pro)tease 2A^(pro) from EV-D68 and CV-A16 also had the ability to inhibit STING activation.As STING activation prior to EV-A71 infection generated cellular resistance to EV-A71 replication,blocking EV-A71-mediated STING suppression represents a new anti-viral target.Wenwen Zheng Zhenbang Zhou Yajuan Rui Runxin Ye Fengyan Xia Fei Guo Xiaoman Liu Jiaming Su Meng Lou Xiao-Fang Yu 2023Signal Transduction and Targeted Therapy2023,8,3:0

16Which metal represents the greatest risk to freshwater ecosystem in bohai region of china?显示文摘Metals discharged from industrial effluents,agricultural wastewater,and sewage runoff by rapid urbanization are of concern as contaminants of freshwater ecosystem because of their persistence and high toxicity to aquatic organisms.This study attempted to identify which metal posed the greatest risk to freshwater ecosystem in the Bohai Region,China.The metals arsenic(As),cadmium(Cd),chromium(Cr),copper(Cu),zinc(Zn),lead(Pb),nickel(Ni),mercury(Hg),iron(Fe),and manganese(Mn)were compared against norfloxacin and gamma-hexachlorocyclohexane(lindane).By comparing the median reported environmental and ecotoxicity concentrations,it showed that Cu,Fe,Zn,Mn,and Cr were the top five metals of concern.Of these,Cu was deemed to represent the highest risk and Hg the lowest risk.The risks for all metals were higher than those for norfloxacin and lindane.Almost all the metals except Hg had water concentrations that exceeded levels where ecotoxicity effects had been recorded in the literature.A comparison with the measurements across the UK rivers suggested that all metals examined had water concentrations about 5-to 10-fold higher than the UK median values except for Cu,Fe,Cd,and Pb.The Fuyang River,a tributary of the Haihe River Basin,seemed to be the location with the highest metal concentrations.However,comparing the post-2010 period to 2000-2009,concentrations of all the metals had fallen except for Fe and Mn,so risks have decreased over the last 7 yr with the greatest improvements for Cd and Pb.While metals still pose high risks to freshwater ecosystem in this region,there is encouragement that some control measures are taken into effect.Chao Su Yonglong Lu Andrew C.Johnson Yajuan Shi Meng Zhang Yueqing Zhang Monika D.Juergens Xiaowei Jin 2017Ecosystem Health and Sustainability2017,3,2:0
