| 4 | Disruption of the white matter structural network and its correlation with baseline progression rate in patients with sporadic amyotrophic lateral sclerosis显示文摘Objective:There is increasing evidence that amyotrophic lateral sclerosis(ALS)is a progressive neurodegenerative disease impacting large-scale brain networks.However,it is still unclear which structural networks are associated with the disease and whether the network connectomics are associated with disease progression.This study was aimed to characterize the network abnormalities in ALS and to identify the network-based biomarkers that predict the ALS baseline progression rate.Methods:Magnetic resonance imaging was performed on 73 patients with sporadic ALS and 100 healthy participants to acquire difusion-weighted magnetic resonance images and construct white matter(WM)networks using tractography methods.The global and regional network properties were compared between ALS and healthy subjects.The single-subject WM network matrices of patients were used to predict the ALS baseline progression rate using machine learning algorithms.Results:Compared with the healthy participants,the patients with ALS showed signifcantly decreased clustering coefcient C_(p)(P=0.0034,t=2.98),normalized clustering coefcientγ(P=0.039,t=2.08),and small‐worldnessσ(P=0.038,t=2.10)at the global network level.The patients also showed decreased regional centralities in motor and non-motor systems including the frontal,temporal and subcortical regions.Using the single-subject structural connection matrix,our classifcation model could distinguish patients with fast versus slow progression rate with an average accuracy of 85%.Conclusion:Disruption of the WM structural networks in ALS is indicated by weaker small-worldness and disturbances in regions outside of the motor systems,extending the classical pathophysiological understanding of ALS as a motor disorder.The individual WM structural network matrices of ALS patients are potential neuroimaging biomarkers for the baseline disease progression in clinical practice. | Wenbin Li Qianqian Wei Yanbing Hou Du Lei Yuan Ai Kun Qin Jing Yang Graham J.Kemp Huifang Shang Qiyong Gong | 2021 | Translational Neurodegeneration2021,10,3: | 2 |
| 5 | Analgesic effect of intrathecal bumetanide is accompanied by changes in spinal sodium-potassium-chloride co-transporter 1 and potassium-chloride co-transporter 2 expression in a rat model of incisional pain显示文摘Accumulating evidence has demonstrated that the sodium-potassium-chloride co-transporter 1 and potassium-chloride co-transporter 2 have a role in the modulation of pain transmission at the spinal level through chloride regulation in the pain pathway and by effecting neuronal excitability and pain sensitization.The present study aimed to investigate the analgesic effect of the specific sodium-potassium-chloride co-transporter 1 inhibitor bumetanide,and the change in spinal sodium-potassium-chloride co-transporter 1 and potassium-chloride co-transporter 2 expression in a rat model of incisional pain.Results showed that intrathecal bumetanide could decrease cumulative pain scores,and could increase thermal and mechanical pain thresholds in a rat model of incisional pain.Sodium-potassium-chloride co-transporter 1 expression increased in neurons from dorsal root ganglion and the deep laminae of the ipsilateral dorsal horn following incision.By contrast,potassium-chloride co-transporter 2 expression decreased in neurons of the deep laminae from the ipsilateral dorsal horn.These findings suggest that spinal sodium-potassium-chloride co-transporter 1 expression was up-regulated and spinal potassium-chloride co-transporter 2 expression was down-regulated following incision.Intrathecal bumetanide has analgesic effects on incisional pain through inhibition of sodium-potassium-chloride co-transporter 1. | Yanbing He Shiyuan Xu Junjie Huang Qingjuan Gong | 2014 | Neural Regeneration Research2014,9,10: | 2 |

| 7 | Development of a double antibodies sandwich ELISA for the detection of avian leukosis virus subgroup J based on monoclonal antibodies against gp85显示文摘Avian leukosis virus(ALV)belongs to the genus Alpharetrovirus of the family Retroviridae and can cause various types of neoplastic diseases,even mortality in infected chickens[1].ALV is categorized into seven subgroups,including A,B,C,D,E,J,and K,based on the pathogenicity and antibody neutralization characteristics[2].The subgroup E is endogenous and transmitted by Mendelian inheritance.The subgroups A,B,C,D,J,and K are exogenous and oncogenic,transmitted horizontally and vertically and caused different pathological lesions[3].Among them,subgroups A and B were most common in commercial poultry and subgroup J is the major cause for myeloid leukosis and nephromas in broilers.In 1980,ALV subgroup J(ALV-J)was first isolated and identified from infected broilers in Britain.It caused significant economic losses in the poultry industry because of increased myelocytomas and other tumors[4].In China,ALV-J was first detected in broilers in 1999 and,subsequently,spread to other chicken types.Compared to other ALV subgroups,ALV-J has been a serious issue in China poultry industry since 2007.Currently,there is no effective vaccine and drugs against ALV-J infection;thus,early detection and elimination of ALV-positive breeder chickens are key measures to block vertical and horizontal ALV-J transmission.Given the importance of ALV to the poultry industry,many ALV detection methods,including virus isolation,immunofluorescence assays,neutralization tests,reverse transcription polymerase chain reaction(PCR)or real-time fluorescent quantitative PCR-based molecular diagnosis ways,and enzyme-linked immunosorbent assays(ELISA)have been developed.Among them,ELISA is widely used for its efficiency,convenience,and sensitivity.Yun et al.[5]developed an antigen-capture ELISA for the detection of ALV based on highly conserved antigen p27.Yu et al.[6]exploited a colloidal gold test strip specific for the detection of ALV p27 antigen.These antigen capture-based serological detection methods share obviously simple,accurate,and rapid advantages in comparison with conventional time-consuming methods of indirect immunofluorescence assay or neutralization tests.However,it is difficult to identify different subgroups of ALV using p27-coated antibodies. | Lili Cao Panpan Zhao He Ding Nan Wang Xue Zhang Shuxian Yuan Hang Dong Yanbing Guo Xinhua Yao Qinlei Yu Hongze Shao Pengtao Gong | 2021 | Acta Biochimica et Biophysica Sinica2021,53,10: | 1 |
| 8 | Fracture characteristics of iron ore under uncoupled blast loading显示文摘A blasting experiment was conducted on iron ore samples by considering multiple coupling charge coefficients.The resulting internal fracture and damage characteristics were quantitatively analyzed via computerized tomography(CT),scanning,and three-dimensional(3D)model reconstruction.The results show that the iron ore primarily displayed radial and circumferential cracks along the blast hole under an explosive load.When the decoupling coefficient was small,the crack surface was dominated by transgranular fractures in the form of intracrystalline fractures.As the uncoupling coefficient increased,the crack surface exhibited transgranular and intergranular coupled fracture modes.Using fractal theory to analyze crack distribution characteristics,as the decoupled coefficient increased,the body fractal dimension tended to decrease,and the degree of damage gradually decreased.The degree of damage reached a turning point when the decoupling charge coefficient was approximately 1.33.A numerical simulation suggested that the explosion energy transmitted to the iron ore and the effective stress decrease sharply when the decoupling coefficient exceeds 1.33.In some optimal uncoupling coefficient range,excessive fragmentation of the ore body is prevented,thereby allowing full use of the explosive energy. | Jinjing Zuo Renshu Yang Min Gong Xinmin Ma Yanbing Wang | 2022 | International Journal of Mining Science and Technology2022,32,4: | 1 |

| 11 | From tuberculosis bedside to bench:UBE2B splicing as a potential biomarker and its regulatory mechanism显示文摘Alternative splicing(AS)is an important approach for pathogens and hosts to remodel transcriptome.However,tuberculosis(TB)-related AS has not been sufficiently explored.Here we presented the first landscape of TB-related AS by long-read sequencing,and screened four AS events(S100A8-intron1-retention intron,RPS20-exon1-alternaitve promoter,KIF13B-exon4-skipping exon(SE)and UBE2B-exon7-SE)as potential biomarkers in an in-house cohort-1.The validations in an in-house cohort-2(2274 samples)and public datasets(1557 samples)indicated that the latter three AS events are potential promising biomarkers for TB diagnosis,but not for TB progression and prognosis.The excellent performance of classifiers further underscored the diagnostic value of these three biomarkers.Subgroup analyses indicated that UBE2B-exon7-SE splicing was not affected by confounding factors and thus had relatively stable performance.The splicing of UBE2B-exon7-SE can be changed by heat-killed mycobacterium tuberculosis through inhibiting SRSF1 expression.After heat-killed mycobacterium tuberculosis stimulation,231 ubiquitination proteins in macrophages were differentially expressed,and most of them are apoptosis-related proteins.Taken together,we depicted a global TB-associated splicing profile,developed TB-related AS biomarkers,demonstrated an optimal application scope of target biomarkers and preliminarily elucidated mycobacterium tuberculosis-host interaction from the perspective of splicing,offering a novel insight into the pathophysiology of TB. | Mengyuan Lyu Jian Zhou Yanbing Zhou Weelic Chong Wei Xu Hongli Lai Lu Niu Yang Hai Xiaojun Yao Sheng Gong Qinglan Wang Yi Chen Yili Wang Liyu Chen Zengwanggema Jiongjiong Zeng Chengdi Wang Binwu Ying | 2023 | Signal Transduction and Targeted Therapy2023,8,3: | 0 |
| 12 | In vitro leishmanicidal activity of antimicrobial peptide KDEL against Leishmania tarentolae显示文摘Leishmaniasis,caused by the intracellular protozoan parasite Leishmania,remains an important neglected tropical infectious disease.Infection may be lethal if untreated.Currently,the available drugs for the disease are limited by high toxicity and drug resista nee.There is an urgent n eed to develop novel anti-leishmanial strategies.Antimicrobial peptides(AMPs)have been described as the first-line immune defense against pathogenic microbes and are being developed as emerging anti-parasitic therapies.In the present study,we showed the anti-leishmanial activity of the synthetic 4-amino acid peptide lysine,aspartic acid,glutamic acid,and leucine(KDEL),the endoplas?mic reticulum retention sequence,against Leishmania tarentolae promastigote and amastigote.Different concentrations of KDEL peptides were incubated with promastigotes,MTT viability assay,and promastigote assay were carried out.Macrophages infected with GFP-transfected L.tarentolae promastigotes were incubated with KDEL peptides,and the anti-amastigote activity of the KDEL peptides was measured by fluoresce nee microscopy.The damage of L.tarentolae was observed by light microscopy and electron microscopy.The cell apoptosis was analyzed using the Annexin V-FITC/PI apoptosis detection kit and mitochondrial membrane potential assay kit and by flow cytometry.Results showed that L.tarentolae was susceptible to KDEL peptides in a dosedepende nt manner,and KDEL peptides disrupted the surface membrane integrity and caused cell apoptosis.In our study,we found for the first time an AMP KDEL from Pseudomonas aeruginosa and proved its significant therapeutic potential as a novel anti-leishmanial drug. | Lili Cao Weina Jiang Songgao Cao Panpan Zhao Juan Liu Hang Dong Yanbing Guo Quan Liu Pengtao Gong | 2019 | Acta Biochimica et Biophysica Sinica2019,51,12: | 0 |
