| 3 | The level of bile salt-stimulated lipase in the milk of Chinese women and its association with maternal BMI显示文摘This study aimed to investigate the bile salt-stimulated lipase(BSSL) concentration in the milk of Chinese women and its correlation with maternal body mass index(BMI), gestational diabetes mellitus(GDM) and gestational hypertensive disorder(GHD). The BSSL levels in the milk samples were measured by enzyme-linked immunosorbent assay(ELISA). BSSL level in colostrum milk of mothers with full-term infants was positively correlated with pregnancy week and negatively correlated with maternal pre-pregnancy BMI and BMI late in pregnancy. Moreover, the BSSL concentration in mature milk was positively correlated with BMI gain during pregnancy. The BSSL concentration in colostrum milk was lower in GDM mothers than in normal mothers. The BSSL helps infants digest fat in early life and its level was associated with lactation. | 7 | Numerical study of the flowing sequence of a pouring liquid显示文摘Liquid flows in a particular sequence when it is poured out from an open-top receptacle. Since the sequence is hard to catch by experiment, a numerical simulation was performed in this pouring process with the moving particle semi-implicit (MPS) method. A modified solid-liquid boundary condition was verified and employed with a new definition of static liquid layers. The whole system was discretized by a set of particles and the liquid particles were marked and tracked in the pouring process. The flowing sequence of the liquid can be calculated by restoring the liquid particles back to their initial positions before it is poured. The mass transfer property is found to depend on the position of the rotation axis and the rotation speed, as well as the viscosity of the liquid. The mechanism of the flowing sequence results from a temporal vortex and its motion during the process. The character vortex is generated by the rotation of the container. | 9 | Targeting iron metabolism using gallium nanoparticles to suppress ferroptosis and effectively mitigate acute kidney injury显示文摘Ferroptosis plays a critical pathophysiological role in several types of acute kidney injury(AKI).The development of nanomaterials targeting iron metabolism and ferroptosis is a promising approach for AKI treatment.Herein,we synthesized gallic acid-gallium polyvinyl pyrrolidone nanoparticles(GGP NPs)as a potential iron-scavenging agent because of their nearly ionic radius and chemical similarity with iron.The results indicated that GGP NPs accumulated in tubular epithelial cells and showed good biocompatibility.GGP NPs significantly inhibited cisplatin(CP)-induced ferroptosis in HK-2 cells by reducing the accumulation of intracellular free iron and mitochondrial dysfunction,and suppressing the perturbations of ferroptosis processes,including lipid peroxidation,nicotinamide adenine dinucleotide phosphate(NADPH)and glutathione(GSH)levels,glutathione peroxidase 4(GPX4)activity,and ferritinophagy.An in vivo study demonstrated that treatment with GGP NPs significantly ameliorated the renal tubular injury and mitochondrial damage induced by CP treatment or ischemia-reperfusion injury.Our study suggests that GGP NPs may be an effective and promising candidate for AKI treatment and enable potential clinical translation. | Xishao Xie Yunjing Zhang Xinwan Su Junni Wang Xi Yao Dou Lv Qin Zhou Jianhua Mao Jianghua Chen Fei Han Yangyang Li Weiqiang Lin | 2022 | Nano Research2022,15,7: | 0 |
| 10 | Accurate quantification of 3'-terminal 2'-O-methylated small RNAs by utilizing oxidative deep sequencing and stem-loop RT-qPCR显示文摘The continuing discoveries of novel classes of RNA modifications in various organisms have raised the need for improving sensitive,convenient,and reliable methods for quantifying RNA modifications.In particular,a subset of small RNAs,including microRNAs(miRNAs)and Piwi-interacting RNAs(piRNAs),are modified at their 3'-terminal nucleotides via 2'-0-methylation.However,quantifying the levels of these small RNAs is difficult because 2'-0-methylation at the RNA 3'-terminus inhibits the activity of polyadenylate polymerase and T4 RNA ligase.These two enzymes are indispensable for RNA labeling or ligation in conventional miRNA quantification assays.In this study,we profiled 3'-terminal 2'-0-methyl plant miRNAs in the livers of rice-fed mice by oxidative deep sequencing and detected increasing amounts of plant miRNAs with prolonged oxidation treatment.We further compared the efficiency of stem-loop and poly(A)-tailed RT-qPCR in quantifying plant miRNAs in animal tissues and identified stem-loop RT-qPCR as the only suitable approach.Likewise,stem-loop RT-qPCR was superior to poly(A)-tailed RT-qPCR in quantifying 3'-terminal 2'-0-methyl piRNAs in human seminal plasma.In summary,this study established a standard procedure for quantifying the levels of 3'-terminal 2'-0-methyl miRNAs in plants and piRNAs.Accurate measurement of the 3'-terminal 2'-0-methylation of small RNAs has profound implications for understanding their pathophysiologic roles in biological systems. | Yan Kong Huanhuan Hu Yangyang Shan Zhen Zhou Ke Zen Yulu Sun Rong Yang Zheng Fu Xi Chen | 2022 | Frontiers of Medicine2022,16,2: | 0 |
