1Quantitative analysis of vascular endothelial growth factor, microvascular density and their clinicopathologic features in human hepatocellular carcinoma显示文摘BACKGROUND: Angiogenesis is known to be essential to the survival, growth, invasion, and metastasis of tumor cells. Vascular endothelial growth factor (VEGF) are an important angiogenic factor regulating tumor angiogenesis, but its significance and tumor pathologic features are un- clear in hepatocellular carcinoma (HCC). In the present study, we analyzed expression of tissue VEGF, alteration of microvascular density (MVD) in microvessel angiogenesis, development and metastasis of HCC, and level of serum VEGF in differential diagnosis of benign and malignant liv- er diseases. METHODS: Tumor specimens were prospectively collected from HCC patients undergoing resection. Total RNAs were extracted and the expression levels were detected from different parts of HCC tissues. The cellular distributions of VEGF and MVD of liver tumors and their paracancerous and distal cancerous tissues were investigated by streptavi- din peroxidase (S-P) immunohistochemistry, respectively. The VEGF levels of circulating blood and hepatoma tissues were measured by enzyme-linked immunosorbent assay. RESULTS: The incidence of VEGF expression was 63.9% in HCCs (23/36 cases), 78.3% in non-encapsulated HCCs (18/23), and 90.9% in HCCs with extrahepatic metastasis (10/11), respectively. The VEGF expression was tightly correlated with MVD (P <0.01). The MVD in HCC with metastasis, low differentiation or non-encapsulation was significantly higher than that in HCC with intact capsule, high differentiation, or no metastasis. No significant diffe- rence was found between VEGF, MVD, tumor size, and hepatitis virus infection. The level of total RNA in HCC tis- sues was significantly lower but the VEGF level significantly higher than those in paracancerous or distal cancerous ones (P<0.01). The abnormal expression levels of VEGF in sera of HCC patients were directly correlated with the me- tastasis and recurrence of tumors. 2Ulinastatin for acute lung injury and acute respiratory distress syndrome: A systematic review and meta-analysis显示文摘AIM: To investigate the efficacy and safety of ulinastatin for patients with acute lung injury(ALI) and those with acute respiratory distress syndrome(ARDS).METHODS: A systematic review of randomized controlled trials(RCTs) of ulinastatin for ALI/ARDS was conducted. Oxygenation index, mortality rate [intensive care unit(ICU) mortality rate, 28-d mortality rate] and length of ICU stay were compared between ulinastatin group and conventional therapy group. Meta-analysis was performed by using Rev Man 5.1.RESULTS: Twenty-nine RCTs with 1726 participants were totally included, the basic conditions of which were similar. No studies discussed adverse effect. Oxygenation index was reported in twenty-six studies(1552 patients). Ulinastatin had a significant effect in improving oxygenation [standard mean difference(SMD) = 1.85, 95%CI: 1.42-2.29, P < 0.00001, I2 = 92%]. ICUmortality and 28-d mortality were respectively reported in eighteen studies(987 patients) and three studies(196 patients). We found that ulinastatin significantly decreased the ICU mortality [I2 = 0%, RR = 0.48, 95%CI: 0.38-0.59, number needed to treat(NNT) = 5.06, P < 0.00001], while the 28-d mortality was not significantly affected(I2 = 0%, RR = 0.78, 95%CI: 0.51-1.19, NNT = 12.66, P = 0.24). The length of ICU stay(six studies, 364 patients) in the ulinastatin group was significantly lower than that in the control group(SMD =-0.97, 95%CI:-1.20--0.75, P < 0.00001, I2 = 86%). 3Mapping wetland changes in China between 1978 and 2008显示文摘Four wetland maps for all China have been produced,based on Landsat and CBERS-02B remote sensing data between 1978 and 2008 (1978,1990,2000 and 2008).These maps were mainly developed by manual interpretation and validated by substantial field investigation in 2009.Based on these maps,we analyzed the 2008 wetland distribution in China and discussed wetland changes and their drivers over the past 30 years.(i) There were about 324097 km 2 of wetlands in 2008,for which inland marshes or swamps were the most common wetland type (35%),with lakes (26%) second.Most of the wetlands were in Heilongjiang,Inner Mongolia,Qinghai and Tibet,occupying about 55% of the national wetland area.(ii) From 1978 to 2008,China's wetland area continually and significantly decreased,by about 33% based on changes in the wetland map.This was in sharp contrast to the increase in artificial wetlands,which increased by about 122%.Inland marshes accounted for the main loss of total wetlands from 1978 to 2000.From 2000 through 2008,riverine and lacustrine wetlands constituted the main wetland loss.Fortunately however,the rate of wetland loss decreased from 5523 to 831 km 2 /a.(iii) The change ratio of lost natural wetlands (including inland and coastal wetlands) to non-wetlands has decreased slightly over the past 30 years.From 1978 to 1990,nearly all natural wetlands (98%) lost were transformed into non-wetlands.However,the ratio declined to 86% from 1990 to 2000,and to 77% from 2000 to 2008.(iv) All Chinese provinces were divided into three groups according to patterns of wetland changes,which could relate to the driving forces of such changes.Tibet was completely different from other provinces,as it was one representative example in which there was a net wetland increase,because of global warming and decreased human activity since 1990.Increased economic development caused considerable wetland loss in most eastern provinces,and artificial wetlands increased.NIU ZhenGuo ZHANG HaiYing WANG XianWei YAO WenBo ZHOU DeMin ZHAO KuiYi ZHAO Hui LI NaNa HUANG HuaBing LI CongCong YANG Jun LIU CaiXia LIU Shuang WANG Lin LI Zhan YANG ZhenZhong QIAO Fei ZHENG YaoMin CHEN YanLei SHENG YongWei GAO XiaoHong ZHU WeiHong WANG WenQing WANG Hong WENG YongLing ZHUANG DaFang LIU JiYuan LUO ZhiCai CHENG Xiao GUO ZiQi GONG Peng 2012Chinese Science Bulletin2012,57,22:51
4Expression and alteration of insulin-like growth factor II-messenger RNA in hepatoma tissues and peripheral blood of patients with hepatocellular carcinoma显示文摘AIM: To investigate the clinical values of serum free insulin-like growth factor Ⅱ (IGF-Ⅱ) levels and IGF-Ⅱ mRNA in hepatocellular carcinoma (HCC) tissues and peripheral blood for diagnosis of HCC and monitoring of extrahepatic metastasis.METHODS: Total RNAs were extracted from HCC tissues or peripheral blood mononuclear cells from patients with HCC, liver diseases devoid of cancer, non-hepatic tumors,and healthy controls, respectively. IGF-Ⅱ cDNAs were synthesized through random primers and reversetranscriptase, amplified by polymerase chain reaction (PCR), and confirmed by DNA sequencing analysis. Serum free IGF-Ⅱ levels in patients with different liver diseases were analyzed by an enzyme-linked immunosorbent assay.RESULTS: The amplified fragments of IGF-Ⅱ mRNA by RT-PCR were identical to originally designed ones with a size of 170 bp and confirmed by sequencing analysis.The dilution experiments revealed that the lowest sensitivity of our system was 2 ng/L of total RNA. The positive frequencies of IGF-Ⅱ mRNA were 100% in HCC tissues,53.3% in para-cancerous tissues, and 0% in non-cancerous tissues, respectively. The serum free IGF-Ⅱ levels were significantly higher in HCC than those in chronic hepatitis or liver cirrhosis. The positive frequency of circulating IGF-Ⅱ mRNA was 34.2% in HCC, no amplified fragment was found in other liver diseases, extrahepatic tumors,and normal controls, respectively. The circulating IGF-Ⅱ mRNA correlated with the stage of HCC, and its positive rate was 100% in HCC with extrahepatic metastasis and 35.5% in HCC with AFP-negative. 5前列腺癌在东亚——十年来的发展现状显示文摘近年来,前列腺癌(PCa)已越发成为东亚地区的健康隐患。目前我们对于PCa的认识大部分来自基于西方人群的研究,而东、西方人群中的差异仍然存在。本文讨论并对比了十年来东亚与欧美国家PCa在流行病学、发病因素、疾病特征和诊疗预后等方面的状况。更多的亚洲证据表明,遗传和环境因素在PCa的发生发展中有重要的作用。生活方式、饮食西方化,医疗水平的提高与发病率的提升有一定的联系。诊疗方面的相关指南主要基于西方研究。十年来PCa的预后有了很大的改善,但不同地区疾病的诊断准确率,治疗有效率和不良反应发生率仍存在一定差异。为了能制定更加适合亚洲人的治疗策略,需要谨慎对待西方人群的相关研究结果。综上,本文展现十年来PCa的发展现状,为改善东亚地区前列腺癌的预防及疾病控制提供证据。Yao Zhu 2015Asian Journal of Andrology2015,17,1:43

7Multispherical interactions and their effects on the Tibetan Plateau's earth system: a review of the recent researches显示文摘The Tibetan Plateau(TP) is a regional Earth system showing very strong interactions among its lithosphere, hydrosphere, cryosphere, biosphere, atmosphere, and anthrosphere. hese interactions manifest TP's impact on surrounding regions and relect TP's response to the global change. Quantifying the multispherical interactions is critically important to understand the TP environment. Our recent years researches including the ongoing program entitled ‘Tibetan Multi-Spheres Interactions and heir Resource-Environment Signiicance(TIMI)', the completed program entitled. ‘Paleo-Altitudes of Tibetan Plateau and Environment(PATE)', as well as the other relating projects have focused on multidisciplinary research approaches and emphasized on three major pathways: Eurasia-Indian plates collision on deep-Earth dynamics, uplit impact on Earth's mantle–crust dynamics, and contemporary interface on land surface and atmospheric dynamics. Our researches have taken in situ measurement as priority and developed several platforms of data acquisition and analysis, including the platforms of water-phase transformations, and ecosystem observations. Our ield investigations have been conducted to obtain data about stratum, paleontology, paleoenvironment, genetic diferentiation of animals and plants. We have developed conceptual and mathematical models for crust uplit formation, paleoclimate, glacial melt,water–air interface lux, vegetation climate, and soil erosion. We have also assessed the anthropogenic impacts on environment. 10Experimental Study of the Pressure Fluctuations in a Pump Turbine at Large Partial Flow Conditions显示文摘Frequent shifts of output and operating mode require a pump turbine with excellent stability. Current researches show that large partial flow conditions in pump mode experience positive-slope phenomena with a large head drop. The pressure fluctuation at the positive slope is crucial to the pump turbine unit safety. The operating instabilities at large partial flow conditions for a pump turbine are analyzed. The hydraulic performance of a model pump turbine is tested with the pressure fluctuations measured at unstable operating points near a positive slope in the performance curve. The hydraulic performance tests show that there are two separated positive-slope regions for the pump turbine, with the flow discharge for the first positive slope from 0.85 to 0.91 times that at the maximum efficiency point. The amplitudes of the pressure fluctuations at these unstable large partial flow conditions near the first positive slope are much larger than those at stable operating condtions. A dominant frequency is measured at 0.2 times the impeller rotational frequency in the flow passage near the impeller exit, which is believed to be induced by the rotating stall in the flow passage of the wicket gates. The test results also show hysteresis with pressure fluctuations when the pump turbine is operated near the first positive slope. The hysteresis creates different pressure fluctuations for those operation points even though their flow rates and heads are similar respectively. 11Lakes' state and abundance across the Tibetan Plateau显示文摘Understanding the changes in number and areal extent of lakes,as well as their abundance and size distribution is important for assessments of regional and global water resources,biogeochemical cycles,and changes in climate.In this study,changes in lake area greater than 1 km2are mapped using Landsat datasets,spanning the 1970s,1990,2000,and 2010.In addition,high-resolution images(GeoCover Landsat mosaic 2000,with a pixel size of 14.25 m)are used for the first time to map lakes as small as 0.001 km2across the entire Tibetan Plateau(TP).Results show that the numbers and areal extent of individual lakes[1 km2in size show a slight decrease between the 1970s and 1990,followed by a clear increase from 1990 to 2010.Ninety-nine new lakes are identified between the 1970s and 2010,71 of which are found between 1990 and 2010.This indicates the accelerated glacier melt and/or increased difference of precipitation minus evaporation since the 1990s.More than 80%of the lakes show an increase in their area between the 1970s and2010.The lake census,using 2000 imagery,shows that there are 32,843 lakes with a total area of 43,151.08±411.49 km2,which makes up 1.4%of the total area of the TP.Around 96%of all lakes are small,with an area\1 km2,while the 1,204 large lakes([1 km2)account for96%of the total lake area.The TP is subdivided into 12greater drainage basins,and of these the inner TP dominates in terms of the number of lakes(55.03%),the total area of lakes(66%),and lake density(0.026/km2compared to the mean,0.011/km2).A plot of lake abundance against size shows that the size distribution of lakes departs from a typical power-law distribution,but displays such a distribution at the mean elevation(4,715 m),with an r2value of 0.97 and a slope of-0.66.The slopes of the abundance-size equations from each of the 12 greater basins,and from all basins together,are larger than-1,supporting the inference that larger lakes,rather than the small lakes,contribute more to the total lake surface area across the TP.The lake inventory provided in this study,along with the assessment of lake size distribution,have important implications for estimates of water balance,for water resource management,and for lake area estimations in the TP.Guoqing Zhang Tong Yao Hongjie Xie Kexiang Zhang Fujing Zhu 2014Chinese Science Bulletin2014,59,24:33
12Effect of cholecystokinin octapeptide on tumor necrosis factor α transcription and nuclear factor-κB activity induced by lipopolysaccharide in rat pulmonary interstitial macrophages显示文摘AIM: To elucidate the anti-inflammatory mechanism ofan intestinal neuropeptide, sulfated cholecystokininoctapeptide (sCCK-8), the effects of sCCK-8 onlipopolysaccharide (LPS)-induced tumor necrosis factorpulmonary interstitial macrophages (PIMs) werestudied.METHODS: PIMs from rat were stimulated with LPS(:1mg @ L-1) in the presence or absence of sCCK-8 (10-8-:10-6mol@ L-1) or/and CCK receptor antagonistproglumide (2 mg @ L-1). The expression of TNF-α mRNAwas assayed by reverse transcription polymerase chainreaction (RT-PCR) at 3h of the stimulation, and nuclearelectrophoretic mobility shift assay (EMSA) at 1 h ofat 30 min of the stimulation was detected by Westernblot.RESULTS: sCCK-8, at concentrations from 10-8 mol @ L-1to 10-6 mol @ L-1 obviously inhibited LPS-induced TNF-αdependent manner, P<0.05, P<0.01. Stimulation PIMsP<0.01, which was elevated by sCCK-8, P<0.05. 14Clinical Research on Alzheimer's Disease: Progress and Perspectives显示文摘Alzheimer's disease(AD), the most common type of dementia, is becoming a major challenge for global health and social care. However, the current understanding of AD pathogenesis is limited, and no early diagnosis and disease-modifying therapy are currently available. 15Engineering a New Chloroplastic Photorespiratory Bypass to In crease Photosynthetic Efficiency and Productivity in Rice显示文摘Over the past few years, three photorespiratory bypasses have been introduced into plants, two of which led to observable in creases in photos yn thesis and biomass yield. However, most of the experiments were carried out using Arabidopsis under controlled environmental conditions, and the increases were only observed under low-light and short-day conditions. In this study, we designed a new photorespiratory bypass (called GOC bypass), characterized by no reducing equivalents being produced during a complete oxidation of glycolate into CO2 catalyzed by three rice-self-originating enzymes, i.e., glycolate oxidase, oxalate oxidase, and catalase. We successfully established this bypass in rice chloroplasts using a multi-gene assembly and transformation system. Transgenic rice plants carrying GOC bypass (GOC plants) showed significant increases in photosynthesis efficiency, biomass yield, and nitrogen content, as well as several other CO2-enriched phe no types under both greenhouse and field conditions .Grain yield of GOC plants varied depending on seeding season and was increased significantly in the spring. We further demonstrated that GOC plants had significant advantages under high-light conditions and that the improvements in GOC plants resulted primarily from a photosynthetic CO2-concentrating effect rather than from improved energy balance. 16Accuracy Analysis of Stewart Platform Based on Interval Analysis Method显示文摘A Stewart platform is introduced in the 500 m aperture spherical radio telescope(FAST) as an accuracy adjustable mechanism for feed receivers.Accuracy analysis is the basis of accuracy design.However,a rapid and effective accuracy analysis method for parallel manipulator is still needed.In order to enhance solution efficiency,an interval analysis method(IA method) is introduced to solve the terminal error bound of the Stewart platform with detailed solution path.Taking a terminal pose of the Stewart platform in FAST as an example,the terminal error is solved by the Monte Carlo method(MC method) by 4 980 s,the stochastic mathematical method(SM method) by 0.078 s,and the IA method by 2.203 s.Compared with MC method,the terminal error by SM method leads a 20% underestimate while the IA method can envelop the real error bound of the Stewart platform.This indicates that the IA method outperforms the other two methods by providing quick calculations and enveloping the real error bound of the Stewart platform.According to the given structural error of the dimension parameters of the Stewart platform,the IA method gives a maximum position error of 19.91 mm and maximum orientation error of 0.534°,which suggests that the IA method can be used for accuracy design of the Stewart platform in FAST.The IA method presented is a rapid and effective accuracy analysis method for Stewart platform.YAO Rui ZHU Wenbai HUANG Peng 2013Chinese Journal of Mechanical Engineering2013,26,1:27
17Puerarin protects brain tissue against cerebral ischemia/reperfusion injury by inhibiting the inflammatory response显示文摘Puerarin, a traditional Chinese medicine, exerts a powerful neuroprotective effect in cerebral ischemia/reperfusion injury, but its mechanism is unknown. Here, we established rat models of middle cerebral artery ischemia/reperfusion injury using the suture method. Puerarin(100 mg/kg) was administered intraperitoneally 30 minutes before middle cerebral artery occlusion and 8 hours after reperfusion. Twenty-four hours after reperfusion, we found that puerarin significantly improved neurological deficit, reduced infarct size and brain water content, and notably diminished the expression of Toll-like receptor-4, myeloid differentiation factor 88, nuclear factor kappa B and tumor necrosis factor-α in the ischemic region. 18Estimating Leaf Chlorophyll Content Using Red Edge Parameters显示文摘Hyperspectral remote sensing makes it possible to non-destructively monitor leaf chlorophyll content(LCC).This study characterized the geometric patterns of the first derivative reflectance spectra in the red edge region of rapeseed(Brassica napus L.) and wheat(Triticum aestivum L.) crops.The ratio of the red edge area less than 718 nm to the entire red edge area was negatively correlated with LCC.This finding allowed the construction of a new red edge parameter,defined as red edge symmetry(RES).Compared to the commonly used red edge parameters(red edge position,red edge amplitude,and red edge area),RES was a better predictor of LCC.Furthermore,RES was easily calculated using the reflectance of red edge boundary wavebands at 675 and 755 nm(R675 and R755) and reflectance of red edge center wavelength at 718 nm(R718),with the equation RES =(R718-R675)/(R755-R675).In addition,RES was simulated effectively with wide wavebands from the airborne hyperspectral sensor AVIRIS and satellite hyperspectral sensor Hyperion.The close relationships between the simulated RES and LCC indicated a high feasibility of estimating LCC with simulated RES from AVIRIS and Hyperion data.This made RES readily applicable to common airborne and satellite hyperspectral data derived from AVIRIS and Hyperion sources,as well as ground-based spectral reflectance data.JU Chang-Hua TIAN Yong-Chao YAO Xia CAO Wei-Xing ZHU Yan D. 19Chinese Trauma Surgeon Association for management guidelines of vacuum sealing drainage application in abdominal surgeries-Update and systematic review显示文摘Vacuum sealing drainage (VSD) is frequently used in abdominal surgeries. However, relevant guidelines are rare. Chin ese Trauma Surge on Associati on orga nized a committee composed of 28 experts across China in July 2017, aiming to provide an evidence-based recommendation for the application of VSD in abdominal surgeries.Eleven questions regarding the use of VSD in abdominal surgeries were addressed:(1) which type of materials should be respectively chosen for the intraperitoneal cavity, retroperitoneal cavity and superficial incisions?(2) Can VSD be preventively used for a high-risk abdominal incision w让h primary suture?(3) Can VSD be used in severely contaminated/infected abdominal surgical sites?(4) Can VSD be used for temporary abdominal cavity closure under some special conditions such as severe abdominal trauma, infection, liver transplantation and intra-abdominal volume increment in abdominal compartment syndrome?(5) Can VSD be used in abdominal organ inflammation, injury, or postoperative drainage?(6) Can VSD be used in the treatment of intestinal fistula and pancreatic fistula?(7) Can VSD be used in the treatment of intra-abdominal and extra-peritoneal abscess?(8) Can VSD be used in the treatment of abdominal wall wounds, wound cavity, and defects?(9) Does VSD in crease the risk of bleeding?(10) Does VSD increase the risk of intestinal wail injury?(11) Does VSD increase the risk of peritoneal adhesion? Focusing on these questions, evidence-based recommendations were given accordingly. VSD was strongly recommended regarding the questions 2-4. Weak recommendations were made regarding questions 1 and 5-11. 20LOW DOSE PIRFENIDONE SUPPRESSES TRANSFORMING GROWTH FACTOR BETA-1 AND TISSUE INHIBITOR OF METALLOPROTEINASE-1, AND PROTECTS RATS FROM LUNG FIBROSIS INDUCED BY BLEOMYCIN显示文摘Objective To investigate the optimal dosage of pirfenidone for the treatment of pulmonary fibrosis induced by bleomycin in Wistar rats, and the alteration of expressions of transforming growth factor beta-1 (TGF-β_ 1), tissue inhibitor of metalloproteinase-1 (TIMP-1), and matrix metalloproteinase-13 (MMP-13) in lung tissue. Methods Male Wistar rats were endotracheally instilled with bleomycin or normal saline. Pirfenidone (25-[KG*8]800 mg·kg -1·d -1), dexamethasone (3 mg/kg), or 1% carboxymethylcellulose sodium were given daily by feed 2 days before instillation of bleomycin. Groups T7 and T14 were fed pirfenidone 50 mg·kg -1·d -1 at 7 days or 14 days after bleomycin instillation. Lungs were harvested at 28 days after bleomycin instillation. Patholological changes in lung tissues were evaluated with HE staining. Lung collagen was stained by sirius red and measured by content of hydroxyproline. Expression of proteins of TGF-β_ 1, TIMP-1, and MMP-13 were detected by Western blotting. Results At doses of 25, 50, and 100 mg·kg -1·d -1, pirfenidone had significant anti-fibrotic effects for bleomycin-induced rat pulmonary fibrosis, and these effects were most significantly attenuated at the dosage of 50 mg·kg -1·d -1 (HE: P<0.01, P<0.01, and P=0.064; sirius red: P<0.05, P<0.01, and P<0.05; hydroxyproline: P=0.595, P<0.01, and P=0.976). Pirfenidone at a dosage of[KG*3]50 mg·kg -1·d -1 inhibited protein expression of TGF-β_ 1 and TIMP-1 in lung tissue in the early phase (0.79 and 0.75 times of control group), but had no effect on expression of MMP-13. Conclusion Low dose pirfenidone, especially at dosage of 50 mg·kg -1·d -1, has significant anti-fibrotic effects on bleomycin-induced rat pulmonary fibrosis.
