| 11 | The effect of hematoma puncture drainage before decompressive craniectomy on the prognosis of hypertensive intracerebral hemorrhage with cerebral hernia at a high altitude显示文摘Purpose:Rapid decompressive craniectomy(DC)was the most effective method for the treatment of hypertensive intracerebral hemorrhage(HICH)with cerebral hernia,but the mortality and disability rate is still high.We suspected that hematoma puncture drainage(PD)+DC may improve the therapeutic effect and thus compared the combined surgery with DC alone.Methods:From December 2013 to July 2019,patients with HICH from Linzhi,Tibet and Honghe,Yunnan Province were retrospectively analyzed.The selection criteria were as follows:(1)altitude≥1500 m;(2)HICH patients with cerebral hernia;(3)Glascow coma scale score of 4-8 and time from onset to admission≤3 h;(4)good liver and kidney function;and(5)complete case data.The included patients were divided into DC group and PD+DC group.The patients were followed up for 6 months.The outcome was assessed by Glasgow outcome scale(GOS)score,Kaplan-Meier survival curve and correlation between time from admission to operation and prognosis.A good outcome was defined as independent(GOS score,4-5)and poor outcome defined as dependent(GOS score,3-1).All data analyses were performed using SPSS 19,and comparison between two groups was conducted using separatet-tests or Chi-square tests.Results:A total of 65 patients was included.The age ranged 34-90 years(mean,63.00±14.04 years).Among them,31 patients had the operation of PD+DC,whereas 34 patients underwent DC.The two groups had no significant difference in the basic characteristics.After 6 months of follow-up,in the PD+DC group there were 8 death,4 vegetative state,4 severe disability(GOS score 1-3,poor outcome 51.6%);8 moderate disability,and 7 good recovery(GOS score 4-5,good outcome 48.4%);while in the DC group the result was 15 death,6 vegetative state,5 severe disability(poor outcome 76.5%),4 moderate disability and 4 good recovery(good outcome 23.5%).The GOS score and good outcome were significantly less in DC group than in PD+DC group(Z=-1.993,p=0.046;χ2=4.38,p=0.043).However,there was no significant difference regarding the survival curve between PD+DC group and DC group.The correlation between the time from admission to operation and GOS at 6 months(r=-0.41,R2=0.002,p=0.829)was not significant in the PD+DC group,but significant in the DC group(r=-0.357,R2=0.128,p=0.038).Conclusion:PD+DC treatment can improve the good outcomes better than DC treatment for HICH with cerebral hernia at a high altitude. | Lin-Jie Wei Chi Lin Xing-Sen Xue Guo-Dong Dun Jian-Bo Zhang Yan-Xiang Tong Jia-Xiong Wang Shi-Ji Yang Ling Wang Zhi Chen Hua Feng Gang Zhu | 2021 | Chinese Journal of Traumatology2021,24,6: | 11 |

| 17 | The effect of a secretion-enhanced heavy chain on improving intein-based dual-vector co-delivery of a full-length factor Ⅷ gene显示文摘Treatment of hemophilia A by gene therapy is adversely affected by inefficient FVIII secretion and the large FVIII gene,which is difficult to package in the promising adeno-associated virus (AAV) vectors.Inhibited secretion of FVIII is caused mainly by inefficient secretion of its heavy chain.Previously,we have employed a protein splicing-based dual-vector to co-transfer a B-domain-deleted FVIII (BDD-FVIII) gene,suggesting that the light chain,covalently ligated to a co-expressed heavy chain can improve the secretion of spliced BDD-FVIII.However,its level of secretion was affected by inefficient secretion the heavy chain.Here,we studied the effect of a mutant heavy chain with L303E/F309S substitutions,which enhance FVIII secretion on the heavy chain itself and spliced FVIII when using a protein splicing-based split-delivery of a full-length FVIII gene.Eukaryotic vectors expressing Ssp DnaB intein-fused mutant heavy and light chains were transiently co-transfected into cultured COS-7 cells.A spliced FVIII protein was seen in co-transfected cells by Western blot analysis.The heavy chain was secreted by cells transfected with the mutant heavy chain gene alone at (39±11) ng/mL and this secretion increased to (123±13) ng/mL when cells were co-transfected with the light chain gene,which was greater than the secretion of wild-type heavy chain.The amount of spliced FVIII in the culture supernatant of co-transfected cells was (86±14) ng/mL,with an activity of (0.61±0.08) IU/mL,which was greater than that of wild-type FVIII co-transfected cells.Spliced FVIII and bioactivity were also detected in the combined culture supernatant of cells individually transfected with mutant heavy and light chain gene at a higher level than that of combined wild-type heavy and light chain transfections.This suggested that the heavy chain with improved secretion markedly increased the efficacy of protein splicing-based split delivery of the full-length FVIII gene using a dual-vector.These results encourage the transfer of this technology to an animal model using a dual-AAV vector. | ZHU FuXiang YANG ShuDe LIU ZeLong MIAO Jing QU HuiGe CHI XiaoYan | 2011 | Chinese Science Bulletin2011,56,2: | 8 |

Their rare earth partitionings are strongly enriched in light rare earths, where CeO2 is50% and the light rare earth partitioning is totally over 95%. The Mianning rare earth ore as well as the Weishan rare earthis a kind of rare earth ore only having bastnaesite. Their rare earth partitionings are also strongly enriched in light rare earths,in which CeO2 is 47% and the light rare earth partitioning is totally over 94%. For the weathering crust type rare earth ore,there are the Longnan rare earth ore, the Xunwu rare earth ore, and the middle yttrium and rich europium ore. In the Long-nan rare earth ore, which is strongly enriched in heavy rare earths, Y2O3 is 64.83%, and the heavy and light rare earthpartitionings are 89.40% and 10.53%, respectively. In the Xunwu rare earth ore, which is strongly enriched in light rareearths, CeO2 is 47.16%, and the light rare earth partitioning is totally 93.25%. Y and Eu are enriched in the middle yttriumand rich europium ore.
