Erianin,a novel dibenzyl compound in Dendrobium extract, inhibits lung cancer cell growth and migration via calcium/calmodulin-dependent ferroptosis显示文摘Ferroptosis,a novel form of programmed cell death,is characterized by iron-dependent lipid peroxidation and has been shown to be involved in multiple diseases, including cancer.Stimulating ferroptosis in cancer cells may be a potential strategy for cancer therapy.Therefore,ferroptosis-inducing drugs are attracting more attention for cancer treatment. Here,we showed that erianin,a natural product isolated from Dendrobium chrysotoxum Lindl, exerted its anticancer activity by inducing cell death and inhibiting cell migration in lung cancer cells.Subsequently,we demonstrated for the first time that erianin induced ferroptotic cell death in lung cancer cells, which was accompanied by ROS accumulation, lipid peroxidation, and GSH depletion. The ferroptosis inhibitors Fer-1 and Lip-1 but not Z-VAD-FMK,CQ,or necrostatin-1 rescued erianin-induced cell death, indicating that ferroptosis contributed to erianin-induced cell death.Furthermore,we demonstrated that Ca^(2+)/CaM signaling was a critical mediator of erianin-induced ferroptosis and that blockade of this signaling significantly rescued cell death induced by erianin treatment by suppressing ferroptosis.Taken together, our data suggest that the natural product erianin exerts its anticancer effects by inducing Ca^(2+)/CaMdependent ferroptosis and inhibiting cell migration, and erianin will hopefully serve as a prospective compound for lung cancer treatment. | Peng Chen Qibiao Wu Jiao Feng Lili Yan Yitian Sun Shuiping Liu Yu Xiang Mingming Zhang Ting Pan Xiaying Chen Ting Duan Lijuan Zhai Bingtao Zhai Wengang Wang Ruonan Zhang Bi Chen Xuemeng Han Yicong Li Liuxi Chen Ying Liu Xingxing Huang Ting Jin Wenzheng Zhang Hong Luo Xiaohui Chen Yongqiang Li Qiujie Li Guohua Li Qin Zhang Lvjia Zhuo Zuyi Yang Huifen Tang Tian Xie Xiaoping Ouyang Xinbing Sui | 2020 | Signal Transduction and Targeted Therapy2020,5,1: | 42
Atypical clinical and pathological findings in a patient with isolated cortical vein thrombosis显示文摘Isolated cortical vein thrombosis often produces a focal lesion. Because of the rapid development of collateral circulation, increased intracranial pressure has never been reported in a patient with isolated cortical vein thrombosis. The diagnosis of isolated cortical vein thrombosis is based mainly on MRI, catheter digital subtraction angiography, and histological findings, but may be challenging. We report a patient who presented with intermittent seizures and left-sided limb weakness. Her symptoms gradually progressed, and she eventually developed signs of increased intracranial pressure. Imaging studies showed a space-occupying lesion in the right frontal lobe of the brain. As we could not diagnose isolated cortical vein thrombosis based on the preoperative findings, surgical excision of the lesion was performed under general anesthesia. Histological examination showed destruction of the brain parenchyma with infiltration of macrophages, proliferation of reactive astrocytes and small vessels, and foci of hemorrhage. Further examination found that a number of small vessels in both the subarachnoid space and brain parenchyma were filled with thrombus, some of which was organized. Elastic fiber staining showed that the obstructed vessels were veins. We diagnosed isolated cortical vein thrombosis with atypical clinical features. | Yan Ding Vance Fredrickson Yicong Lin Yueshan Piao Xiangbo Wang Dehong Lu Cunjiang Li | 2012 | Neural Regeneration Research2012,7,31: | 3
Medroxyprogestogen enhances apoptosis of SKOV-3 cells via inhibition of the PI3K/Akt signaling pathway显示文摘We sought to assess the effect of progestin on the apoptosis of epithelial ovarian cancer cell line SKOV-3 and via regulation of phosphorylation signaling in.Epithelial ovarian cancer cell line SKOV-3 was treated with medroxyprogestogen, phosphatidylinositol 3-kinase inhibitor LY294002 and vehicle control.Akt,phospho-Akt,Bcl-2 and phospho-Bad proteins were examined by immunoblotting assays.Medroxyprogestogen-induced apoptosis was assessed by MTT assays and Annexin V apoptosis assay. We found no significant difference in Akt and Bad expression in both the medroxyprogestogen groups and the control group. The levels of phospho-Akt,Bcl-2 and phospho-Bad were decreased in all the medroxyprogestogen groups and significantly decreased in the high dose mitogen-activated protein (MAP) group (10 μmol/L).Viability of SKOV-3 was reduced and apparent apoptosis of SKOV-3 cells was observed with increased doses of MAP.The findings suggest that medroxyprogestogen can induce SKOV-3 cell apoptosis by inhibiting Akt phosphorylation. | Yan Li Yi Jiang Yicong Wan Lin Zhang Weiwei Tang Jingjing Ma Shan Wu Wenjun Cheng | 2013 | The Journal of Biomedical Research2013,27,1: | 2
Valence electron structure analysis of refining mechanism of Sc and Ti additions on aluminum显示文摘The mechanism of the difference of refining effect between Sc and Ti adding to aluminum can not be explained substantially with traditional theory. Valence electron structures of Al-Ti and Al-Sc alloys have been studied by using the empirical electron theory of solids and molecules (EET). The covalent bond electron numbers and interfacial electron density differences are calculated. The conclusion is that, in the two alloys, different covalent bond electron numbers of nucleation particles, and different electron densities on the interface between the second phase particles and the matrix, fundamentally lead to the difference of refining effect between Sc and Ti adding to aluminum. | LI PieJie YE YiCong HE LiangJu | 2009 | Chinese Science Bulletin2009,54,5: | 2
Characterization of a Novel Weak Allele of RGA1/D1 and Its Potential Application in Rice Breeding显示文摘Semi-dwarfing improves the lodging resistance and yield of rice, and the vast majority of modern rice varieties harbor the sd1 allele to decrease plant height, resulting in reduced genetic diversity and negative agronomic traits. Thus, exploring alternative sources of dwarfism is imperative for rice breeding. Here,we identified a novel RGA1 allele,d1-w,from a local indica variety Xiaolixiang(XLX)using a map-based cloning approach.Compared with other rice varieties,RGA1 in XLX contained a unique single nucleotide polymorphism that resulted in an additional transcript and reduced functional RGA1 transcript level. The RGA1 from Nipponbare was introduced into XLX to estimate the value of d1-w in rice breeding.Compared with transgenic XLX plants(XLX^(D1)),XLX exhibited reduced plant height, increased stem strength, lower reactive oxygen species accumulation, delayed senescence, stronger photosynthesis, higher grain yield and quality(including external, milling and nutritional qualities),and enhanced resistance to drought and Rhizoctonia solani. Therefore,we proposed that the d1-w allele has potential as an excellent dwarfism resource for rice breeding. | LIU Yantong LI Ting JIANG Zhishu ZENG Chuihai HE Rong QIU Jiao LIN Xiaoli PENG Limei SONG Yongping ZHOU Dahu CAI Yicong ZHU Changlan FU Junru HE Haohua XU Jie | 2022 | Rice science2022,29,6: | 2

Herein, the first asymmetric synthesis of TBAJ-876 has been realized using synergistic Li/Li catalysis with excellent yield of 95% and 88 : 12 er (99.6 : 0.4 er, 10 : 1 dr after simple recrystallization). Furthermore, DFT calculations and 7Li-NMR analysis illustrated the mechanism of the synergistic reaction: a chiral Li-complex activates the nucleophile to control the stereoselectivity, while the other achiral Li-complex activates the electrophile to catalyze the carbonyl addition reaction. | 10 | Asymmetric synthesis of bedaquiline based on bimetallic activation and non-covalent interaction promotion strategies显示文摘Bedaquiline(BDQ),approved by Food and Drug Administration(FDA)in 2012 as the first anti-tuberculosis-specific drug in the last 40 years,is viewed as one of the world’s most promising treatments for tuberculosis(TB).Due to the stereoselective construction of the Csp^(3)–Csp^(3)bond with vicinal stereocenters of BDQ and its analogues being an unsolved challenge, there have not been any reports concerning its asymmetric synthesis for the current industrial production process until now.Herein,we have successfully developed a cooperative bimetallic system for the asymmetric synthesis of BDQ under the guidance of density functional theory(DFT)computations.Based on the optimized conditions,BDQ could be synthesized with excellent enantioselectivity(>99%ee)and diastereoselectivity(16:1 dr).A 5-g scale reaction was also conducted with comparably excellent results, showing its potential for industrial application. | Feng Gao Jing Li Tanveer Ahmad Yicong Luo Zhenfeng Zhang Qianjia Yuan Xiaohong Huo Tao Song Wanbin Zhang | 2022 | Science China Chemistry2022,65,10: | 1 |
Steady-migration retention characteristics of peptides under gradient elution: application towards a dynamic separation method for minor-adjustments of the retention of peptides in RPLC显示文摘Minor-adjustment of the retention of peptides, induced by varying the mobile phase flow-rate(MPF-R), is a new dynamic separation method for simultaneously and rapidly identifying and improving the selectivity of hidden and overlapping peptide peaks. It can also-stabilize the reverse elution order of some pair-peaks under gradient elution in reverse phase liquid chromatography. The retention characteristics of peptides under gradient elution in RPLC was firstly found to be dominated by two variables of the steady region(SR) and migration region(MR). The changes in peptide retention induced by varying the MPF-R can be attributed to changes in the rate of bond breaking of multiple molecular interactions of peptides from the SR and of the mass transfer of peptides from the stationary phase to the mobile phase in the MR. The two dynamic variables were also found to independently depend on the type of peptide. Desirable results were obtained using six standard oligopeptides and a real sample of trypsin-digested lysozyme. It is expected that the quality control of peptide drugs, high dispersion of peptide peaks in peptide mapping and 'bottom-up MS'in proteomics will be improved by this method, even enabling peptide purification on a preparative scale in industry. | Min Li Yongjun Lu Yicong Yang Jianjun Li Lili Wang Wei Tuo Xiaohui Ning Xin-Du Geng | 2017 | Science China Chemistry2017,60,6: | 0
Comparison of enhancement techniques based on neural networks for attenuated voice signal captured by flexible vibration sensors on throats显示文摘Wearable flexible sensors attached on the neck have been developed to measure the vibration of vocal cords during speech. However, highfrequency attenuation caused by the frequency response of the flexible sensors and absorption of high-frequency sound by the skin are obstacles to the practical application of these sensors in speech capture based on bone conduction. In this paper, speech enhancement techniques for enhancing the intelligibility of sensor signals are developed and compared.Four kinds of speech enhancement algorithms based on a fully connected neural network(FCNN),a long short-term memory(LSTM),a bidirectional long short-term memory(BLSTM),and a convolutional-recurrent neural network(CRNN)are adopted to enhance the sensor signals, and their performance after deployment on four kinds of edge and cloud platforms is also investigated.Experimental results show that the BLSTM performs best in improving speech quality, but is poorest with regard to hardware deployment. It improves short-time objective intelligibility(STOI)by 0.18 to nearly 0.80,which corresponds to a good intelligibility level, but it introduces latency as well as being a large model. The CRNN,which improves STOI to about 0.75,ranks second among the four neural networks. It is also the only model that is able to achieves real-time processing with all four hardware platforms, demonstrating its great potential for deployment on mobile platforms. To the best of our knowledge, this is one of the first trials to systematically and specifically develop processing techniques for bone-conduction speed signals captured by flexible sensors. The results demonstrate the possibility of realizing a wearable lightweight speech collection system based on flexible vibration sensors and real-time speech enhancement to compensate for high-frequency attenuation. | Shenghan Gao Changyan Zheng Yicong Zhao Ziyue Wu Jiao Li Xian Huang | 2022 | Nanotechnology and Precision Engineering2022,5,1: | 0
Adhesive chitosan-based hydrogel assisted with photothermal antibacterial property to prompt mice infected skin wound healing显示文摘Bacterial infection of wounds is an escalating medical problem, issuing threats to both global public health and personal health.Photothermal antibacterial technology as a novel sterilization strategy has outstanding sterilization efficiency, high safety and low risk of emergence of drug-resistant bacteria.By combining inherent antibacterial activity and light-assisted antibacterial treatment, developing novel multifunctional dressings with synergistic high-efficiency antibacterial effects and also promoting wound healing possesses attractive advantages in the field of treating bacterial wound infections in clinical care.Herein,a multifunctional hydrogel formed by in situ photo-cross linking was designed and prepared by first grafting methacrylic anhydride as a photosensitizer onto chitosan, and then introducing oxidatively synthesized polydopamine(PDA).The physicochemical characterizations of the synthesized hydrogels demonstrated their tunability certainly associated with PDA concentration, including pore size, water swelling, rheological properties and in vitro degradability. In addition, the composite hydrogels exhibited good adhesion,anti-oxidation and photothermal properties due to the existence of PDA.Within 10min upon exposure to 808 nm near-infrared(NIR)light irradiation, this hydrogel system displayed outstanding antibacterial activity against Staphylococcus aureus with almost 100%killing efficiency,of which rapid efficient sterilization plays a significant role in wound healing. Moreover, the hydrogel is capable of cytocompatibility and has low toxicity to murine fibroblasts(L929 and NIH/3T3).In the full-thickness wound defect infection model in mice, the wound closure ratio, inflammatory response, fibroblasts, neovascularization and epithelialization were measured.Animal experiments also reveal that the hydrogel assisted with NIR laser irradiation can inhibit effectively infection at an early stage and accelerate the wound healing process. In summary, this novel multifunctional injectable hydrogel exhibits excellent swelling capacity,bio-adhesion,antioxidant property, photothermal activity, efficient antibacterial property and facilitates skin healing, which has great promising application as a medical dressing biomaterial in infected wound carefields. | Shuang Cheng Meng Pan Danrong Hu Ruxia Han Lang Li Zhongwu Bei Yicong Li Ao Sun Zhiyong Qian | 2023 | Chinese Chemical Letters2023,34,12: | 0

Natural variation of an autophagy-family gene among rice subspecies affects grain size and weight显示文摘Elucidating the genetic basis of natural variation in grain size and weight among rice varieties can help breeders develop high-yielding varieties. We identified a novel gene,GW3a(Grain Weight 3a)(LOC_Os03g27350),that affects rice grain size and weight.gw3a mutants showed higher total starch content and dry matter accumulation than the wild type(WT),Nipponbare, suggesting that GW3a negatively regulates grain size and weight. Moreover, our study found that GW3a interacted with OsATG8 by cleaving it,suggesting that GW3a may be involved in the assembly of autophagosomes and starch degradation in plants. The haplotype analysis of GW3a showed functional differences between indica and japonica rice.Taken together,we conclude that GW3a is expressed in the autophagosome pathway regulating starch metabolism in rice, affecting yield-related traits, such as grain size, grain weight and thousand grain weight(TGW).Our findings also shed new light on autophagy-mediated yield trait regulation, proposing a possible strategy for the genetic improvement of high-yield germplasm in rice. | Shiying Huang Peng Wang Caijing Li Qingwen Zhou Tao Huang Yicong Cai Qin Cheng Hui Wang Qi Zhong Zhihao Chen Liping Chen Haohua He Jianmin Bian | 2024 | The Crop Journal2024,12,1: | 0
AMMONIA DISPERSION FROM MULTI-FLOOR VERSUS STANDARD SINGLE-FLOOR PIG PRODUCTION FACILITIES BASED ON COMPUTATIONAL FLUID DYNAMICS SIMULATIONS显示文摘Multi-floor buildings for raising pigs have recently attracted widespread attention as an emerging form of intensive livestock production especially in eastern China, due to the fact that they can feed a much larger number of animals per unit area of land and thus alleviate the shortage of land available for standard single-floor pig production facilities. However, this more intensive kind of pig building will pose new challenges to the local environment in terms of pollutant dispersion. To compare the dispersion air pollutants(ammonia as a representative)emitted from multi-versus single-floor pig buildings, ammonia dispersion distance and concentration gradients were investigated through three-dimensional simulations based on computational fluid dynamics. The validation of an isolated cubic model was made to ensure the simulation method was effective. The effects of wind direction, wind speed and emission source concentration at 1.5 m(approximate human inhalation height)during summer were investigated. The results showed that the ammonia dispersion distance of the multi-floor pig building was far greater than that of the single-floor building on a plane of Z=1.5 m.When the wind direction was 67.5°,the wind speed was 2 m·s^(−1) and the emission source concentration was 20 ppmv, the dispersion distance of the multi-floor pig building could reach 1380 m.Meanwhile, the ammonia could accumulate in the yard to 7.68 ppmv. Therefore, future site selection, wind speed and source concentration need to be given serious consideration.Based on the simulation used in this study with source concentration is 20 ppmv, the multi-floor pig buildings should be located 1.4 km away from residential areas to avoid affecting residents. The results of this study should guidance for any future development of multi-floor pig buildings. | Yicong XIN Li RONG Gunther SCHAUBERGER Dejia LIU Xiusong LI Zhihua YANG Songming ZHU Dezhao LIU | 2023 | Frontiers of Agricultural Science and Engineering2023,10,3: | 0

How light affect the magnetotactic behavior and reproduction of ellipsoidal multicellular magnetoglobules?显示文摘Magnetotactic bacteria(MTB)synthesize intracellular magnetic organelles, magnetosomes, which consist of magnetic crystals that are enveloped in a membrane.Magnetosomes are organized into a chain(s)and confer on cells a magnetic dipolar moment. This magnetic property allows MTB cells to align and swim along geomagnetic field lines,a movement referred to as magnetotaxis. Some MTB species change their swim direction in response to illumination by UV,violet and blue light. Here we analyzed the polarity of morphology, magnetism, and motion in Mediterranean multicellular magnetotactic prokaryotes, also called, magnetoglobules or MMP.The magnetoglobules were assembled from 60-80 cells into an asymmetric ellipsoidal morphology with a relative narrow and large end. They swam dominantly northward, parallel to the direction of the magnetic field, with the narrow-end as the leading side.In response to a reversal in the direction of the magnetic field, they aligned quickly along the magnetic field lines and kept swimming northward.Interestingly, under constant illumination,385-nm UV light, magnetoglobules changed their swimming direction southward anti-parallel to the direction of the magnetic field, with the large-end as the leading side. The change from a northward to southward direction occurred along with an increase of swimming speed.A minimum of 35-mW/cm^(2) irradiance of UV light was sufficient to trigger the swimming re-orientation.UV radiation also triggered the unidirectional division of magnetoglobules.Together these results revealed a coordination of the polarity of magnetoglobule morphology, magnetic moment, and swimming orientation,in response to magnetic and optical stimuli. The UV triggered the reversal of magnetotaxis and magnetoglobule division indicating the ecological significance of light for multicellular magnetotactic prokaryotes. | Xinxin QIAN Yicong ZHAO Claire-Lise SANTINI Hongmiao PAN Tian XIAO Haitao CHEN Tao SONG Jinhua LI Francois ALBERTO Sophie BRUSTLEIN Long-Fei WU | 2021 | Journal of Oceanology and Limnology2021,39,6: | 0
Trogocytosis of CAR molecule regulates CAR-T cell dysfunction and tumor antigen escape显示文摘Chimeric antigen receptor(CAR)T-cell therapy has demonstrated clinical response in treating both hematologic malignancies and solid tumors.Although instances of rapid tumor remissions have been observed in animal models and clinical trials, tumor relapses occur with multiple therapeutic resistance mechanisms.Furthermore, while the mechanisms underlying the long-term therapeutic resistance are well-known,short-term adaptation remains less understood. However, more views shed light on short-term adaptation and hold that it provides an opportunity window for long-term resistance. In this study,we explore a previously unreported mechanism in which tumor cells employ trogocytosis to acquire CAR molecules from CAR-T cells,a reversal of previously documented processes. This mechanism results in the depletion of CAR molecules and subsequent CAR-T cell dysfunction, also leading to short-term antigen loss and antigen masking.Such type of intercellular communication is independent of CAR downstream signaling,CAR-T cell condition, target antigen, and tumor cell type. However,it is mainly dependent on antigen density and CAR sensitivity, and is associated with tumor cell cholesterol metabolism.Partial mitigation of this trogocytosis-induced CAR molecule transfer can be achieved by adaptively administering CAR-T cells with antigen density-individualized CAR sensitivities.Together, our study reveals a dynamic process of CAR molecule transfer and refining the framework of clinical CAR-T therapy for solid tumors. | You Zhai Yicong Du Guanzhang Li Mingchen Yu Huimin Hu Changqing Pan Di Wang Zhongfang Shi Xu Yan Xuesong Li Tao Jiang Wei Zhang | 2024 | Signal Transduction and Targeted Therapy2024,9,1: | 0
