1Biosynthesis of Triacylglycerol Molecules with a Tailored PUFA Profile in Industrial Microalgae显示文摘The composition of polyun saturated fatty acids (PUFAs) in triacylglycerols (TAGs) is key to health ben efits and for oil applications, yet the underlying genetic mechanism remains poorly understood. In this study, by in si Iico, ex vivo, and in vivo profili ng of type-2 diacylglycerol acyltransferases (DGAT2s) in Nannochloropsis oceanica we revealed two novel PUFA-preferring enzymes that discriminate individual PUFA species in TAG assembly, with NoDGAT2J for linoleic acid (LA) and NoDGAT2K for eicosapentaenoic acid (EPA). The LA and EPA composition of TAG molecules is mediated in vivo via the functional partitioning between NoDGAT2J and 2K, both of which are localized in the chloroplast envelope. By modulating transcript abundance of the DGAT2s, an N. oceanica strain bank was created, where proportions of LA and EPA in TAG vary by 18.7-fold (between 0.21% and 3.92% dry weight) and 34.7-fold (between 0.09% and 3.12% dry weight), respective!y. 3Adenomyosis-derived extracellular vesicles endow endometrial epithelial cells with an invasive phenotype through epithelial-mesenchymal transition显示文摘Extracellular vesicles from highly metastatic tumor cells have been shown to mediate epithelial-mesenchymal transition(EMT)-related events in recipient cells.In endometrial epithelial cells,EMT processes are known to be involved in the development of adenomyosis.We aimed to investigate whether adenomyosis-derived extracellular vesicles(AMEVs)are able to induce an EMT process in endometrial epithelial cells.In this study,AMEVs were isolated from patients with adenomyosis and characterized by transmission electron microscopy,Western blot,and nanoparticle tracking.Primary endometrial epithelial cells(EECs)were derived from normal endometrium tissues from patients with leiomyoma and co-cultured with AMEVs in vitro.AMEV uptake was examined by fluorescence confocal microscopy.The invasion of EECs was confirmed by Transwell assay.Immunohistochemistry,Western blot,and qRT-PCR were performed on EECs to illustrate the expression levels of cytokeratin 19,E-cadherin,vimentin,and zinc finger E-boxbinding homeobox 1(ZEB1).The results indicated that the cellular fluorescence intensity gradually increased after 48 h of co-culture,but decreased after 72 h.After co-culturing with AMEVs for 72 h,EECs expressed significantly lower levels of cytokeratin 19 and E-cadherin,and significantly higher levels of vimentin and ZEB1.Together these results demonstrated that AMEVs induce an EMT process and enhance the invasion of EECs.These changes may contribute to the pathogenesis and progression of adenomyosis.Dayong Chen Hai Qiao Yiting Wang Ling Zhou Na Yin Liaoqiong Fang Zhibiao Wang 2020Genes & Diseases2020,7,4:8
4Salmonella flagella confer anti-tumor immunological effect via activating Flagellin/TLR5 signalling within tumor microenvironment显示文摘Salmonella:mediated cancer therapy has achieved remarkable anti-tumor effects in experimental animal models,but the detailed mechanism remains unsolved.In this report,the active involvement of the host immune response in this process was confirmed by comparing the tumor-suppressive effects of Salmonella in immunocompetent and immunodeficient mice bearing melanoma allografts.Since flagella are key inducers of the host immune response during bacterial infection,flagella were genetically disrupted to analyse their involvement in Salmonella-mediated cancer therapy.The results showed that flagellum-deficient strains failed to induce significant anti-tumor effects,even when more bacteria were administered to offset the difference in invasion efficiency.Flagella mainly activate immune cells via Flagellin/Toll-like receptor 5(TLR5)signalling pathway.Indeed,we showed that exogenous activation of TLR5 signalling by recombinant Flagellin and exogenous expression of TLR5 both enhanced the therapeutic efficacy of flagellum-deficient Salmonella against melanoma.Our study highlighted the therapeutic value of the interaction between Salmonella and the host immune response through Flagellin/TLR5 signalling pathway during Salmonella-mediated cancer therapy,thereby suggesting the potential application of TLR5 agonists in the cancer immune therapy.Jianxiang Chen Yiting Qiao Guo Chen Cunjie Chang Heng Dong Bo Tang Xiawei Cheng Xiufeng Liu Zichun Hua 2021Acta Pharmaceutica Sinica B2021,11,10:6
5Gap-surface plasmon metasurfaces for linear-polarization conversion, focusing,and beam splitting显示文摘Gap-surface plasmon(GSP)metasurfaces have attracted progressively increasing attention due to their planar configurations,ease of fabrication,and unprecedented capabilities in manipulating the reflected fields that enable integrating diverse bulk-optic-based optical components into a single ultrathin flat element.In this work,we design and experimentally demonstrate multifunctional metalenses that perform simultaneous linear-polarization conversion,focusing,and beam splitting,thereby reproducing the combined functionalities of conventional halfwave plates,parabolic reflectors,and beam splitters.The fabricated single-focal metalens incorporates properly configured distinct half-wave-plate-like GSP meta-atoms and exhibits good performance under linearly polarized incidence in terms of orthogonal linear-polarization conversion(>75%)and focusing(overall efficiency>22%)in the wavelength spectrum ranging from 800 to 950 nm.To further extend the combined functionalities,we demonstrate a dual-focal metalens that splits and focuses a linearly polarized incident beam into two focal spots while maintaining the capability of orthogonal linear-polarization conversion.Furthermore,the power distribution between two split beams can readily be controlled by judiciously positioning the incident beam.The demonstrated multifunctional GSP-based metalenses mimic the combined functionalities of a sequence of discrete bulk optical components,thereby eliminating the need for their mutual alignment and opening new perspectives in the development of ultracompact and integrated photonic devices.FEI DING YITING CHEN SERGEY IBOZHEVOLNYI 2020Photonics Research2020,8,5:5
6A waveguide metasurface based quasi-far-field transverse-electric superlens显示文摘The imaging capability of conventional lenses is mainly limited by the diffraction of light,and the so-called superlens has been developed allowing the recovery of evanescent waves in the focal plane.However,the remarkable focusing behavi-or of the superlens is greatly confined in the near-field regime due to the exponential decay of evanescent waves.To tackle this issue,we design a waveguide metasurface-based superlens with an extraordinary quasi-far-field focusing capability beyond the diffraction limit in the present work.Specifically,we analyze the underlying physical mechanism and provide experimental verification of the proposed superlens.The metasurface superlens is formed by an array of gradient nanoslits perforated in a gold slab,and supports transverse-electric(TE)waveguide modes under linearly polar-ized illumination along the long axis of the slits.Numerical results illustrate that exciting such TE waveguide modes can modulate not only optical phase but also evanescent waves.Consequently,some high-spatial-frequency waves can con-tribute to the focusing of the superlens,leading to the quasi-far-field super-resolution focusing of light.Under 405 nm illu-mination and oil immersion,the fabricated superlens shows a focus spot of 98 nm(i.e.λ/4.13)at a focal distance of 1.49μm(i.e.3.68λ)using an oil immersion objective,breaking the diffraction limit ofλ/2.38 in the quasi-far field regime.The developed metasurface optical superlens with such extraordinary capabilities promises exciting avenues to nanolitho-graphy and ultra-small optoelectronic devices.Yechuan Zhu Xiaolin Chen Weizheng Yuan Zhiqin Chu Kwok-yin Wong Dangyuan Lei Yiting Yu 2021Opto-Electronic Advances2021,4,10:3
7Distinguishing Between Natural and GAN-Generated Face Images by Combining Global and Local Features显示文摘With the development of face image synthesis and generation technology based on generative adversarial networks(GANs), it has become a research hotspot to determine whether a given face image is natural or generated. However, the generalization capability of the existing algorithms is still to be improved. Therefore,this paper proposes a general algorithm. To do so, firstly,the learning on important local areas, containing many face key-points, is strengthened by combining the global and local features. Secondly, metric learning based on the Arc Face loss is applied to extract common and discriminative features. Finally, the extracted features are fed into the classification module to detect GAN-generated faces.The experiments are conducted on two publicly available natural datasets(Celeb A and FFHQ) and seven GANgenerated datasets. Experimental results demonstrate that the proposed algorithm achieves a better generalization performance with an average detection accuracy over 0.99 than the state-of-the-art algorithms. 9Gas Occurrence and Accumulation Characteristics of Cambrian–Ordovician Shales in the Tarim Basin, Northwest China显示文摘The Tarim Basin is located in northwestern China and is the biggest basin in China with huge oil and gas resources. Especially the Lower to Middle Cambrian and Middle to Upper Ordovician possess the major marine source rocks in the Tarim Basin and have large shale gas resource potential. The Cambrian–Ordovician shales were mainly deposited in basin–slope facies with thicknesses between 30–180 m. For shales buried shallower than 4500 m, there is high organic matter abundance with TOC(total organic carbon) mainly between 1.0% and 6.0%, favorable organic matter of Type I and Type II, and high thermal maturity with RoE as 1.3%–2.75%. The mineral composition of these Cambrian–Ordovician shale samples is mainly quartz and carbonate minerals while the clay minerals content is mostly lower than 30%, because these samples include siliceous and calcareous shale and marlstone. The Cambrian and Ordovician shales are compacted with mean porosity of 4% and 3%, permeability of 0.0003×10^(-3)–0.09×10^(-3) μm^2 and 0.0002×10^(-3)–0.11×10^(-3) μm^2, and density of 2.30 g/m^3 and 2.55 g/m^3, respectively. The pores in the shale samples show good connectivity and are mainly mesopore in size. Different genetic types of pores can be observed such as intercrystal, intergranular, dissolved, organic matter and shrinkage joint. The reservoir bed properties are controlled by mineral composition and diagenesis. The maximum adsorption amount to methane of these shales is 1.15–7.36 cm^3/g, with main affecting factors being organic matter abundance, porosity and thermal maturity. The accumulation characteristics of natural gas within these shales are jointly controlled by sedimentation, diagenesis, hydrocarbon generation conditions, reservoir bed properties and the occurrence process of natural gas. The natural gas underwent short-distance migration and accumulation, in-place accumulation in the early stage, and adjustment and modification in the later stage. 10Structural Characteristics and their Significance for Hydrocarbon Accumulation in the Northern Slope of the Central Sichuan Paleo-uplift显示文摘Based on 2D and 3D seismic data,the latest drilling data and field outcrop data of the northern slope of the Central Sichuan paleo-uplift,the structural analysis method is used to analyze unconformity development characteristics and fault characteristics during the key structural transformation period,discussing the influence of the structural characteristics on the hydrocarbon accumulation of deep carbonate rocks.The results show that:(1)The two key unconformities of the Tongwan and Caledonian periods were primarily developed in deep carbonate rocks.Firstly,Tongwan’s unconformities are characterized by regional disconformities between the second and third members of the Dengying Formation,the top formation of the Sinian and the lower Cambrian,strips of which zigzag through the north and south sides of the study area.Secondly,the Caledonian unconformity is characterized by a regional unconformable contact between the lower Permian and the ower Paleozoic strata.From NE to SW,the age of the strata,which were subject to erosion,changes from new to old,the denudation distribution showing as a nose-shaped structure which inclines towards the ENE.(2)Boundary fault and transtensional strike-slip faults developed in the Sinian to Paleozoic strata.In profile,there are three types of structural styles:steep and erect,flower structures,’Y’and reversed’Y’type faults.In plane view,the Sinian developed extensional boundary faults extending in an almost NS direction,strike-slip faults developing and extending linearly in approximately EW,WNW and NE strikes in the Cambrian,with characteristically more in the south and less in the north.(3)The faults in the northern slope show obvious zonal deformations in transverse view as well as significant stages and stratified activity in a longitudinal direction.Among them,the activity of faults in the Sinian was the strongest,followed by the activity in the Cambrian period,the activity intensity of faults in the Permian period being the weakest.This fault activity can be divided into four periods:Sinian,Cambrian-Permian,the early Indosinian period and the late Indosinian-Himalayan period,the transtensional strikeslip faults being the products of oblique extensions of pre-existing weak zones in the Xingkai and Emei taphrogenesis,with a particular inheritance in the main faults.(4)Combined with hydrocarbon accumulation factors,it is considered that the epigenetic karstification of the Tongwan and Caledonian unconformities in the northern slope controlled the formation and distribution of carbonate karst reservoirs over a large area,also acting as a good pathway for oil and gas migration.The extensional faults developed at the margin of the NS trending rift,controlling the sag-platform sedimentary pattern in the Dengying Formation of the Sinian.Strike-slip faults in NE,WNW and ENE directions may control the microgeomorphological pattern inside the platform and intensify the differential distribution of grain beach facies.The multi-stage hereditary activity of strike-slip faults not only improved the porosity and permeability of the reservoirs,but also acted as the main channel of oil and gas migration,providing favorable conditions for the development of the current multi-layer gasbearing scenario in the northern slope of the Central Sichuan Basin.TIAN Xingwang LIU Guangdi LUO Bing YANG Yu WEN Long ZHANG Benjian CHEN Xiao SU Guiping SONG Zezhang PENG Hanlin ZHOU Gang YAN Wei YANG Dailin WANG Yunlong SUN Yiting DOU Shuang 2022Acta Geologica Sinica(English Edition)2022,96,4:2

15Zinc-substituted hemoglobin with specific drug binding sites and fatty acid resistance ability for enhanced photodynamic therapy显示文摘Precisely designed protein-based nanodrugs, as a kind of colloidal drug system, have attracted significant attention in tumor therapy because of their refined drug loading ratio, controlled delivery efficacy and natural biocompatibility. However, most drugs are conjugated to the protein carriers randomly without specific binding sites. Moreover, such sites could easily be replaced by lipophilic molecules in the physiological environment and result in low delivery efficiency. With strong and specific binding locations especially comparatively narrow spatial binding sites and nonflexible structure, hemin (FePPIX)-free hemoglobin or apohemoglobin (apoHb), as a natural metalloporphyrin protein carrier, represents great potential in bioapplication. Therefore, we herein introduce a folate acid (FA) modified, zinc-substituted hemoglobin (ZnPHb-FA) as a naturally occurring protein matrix-based photosensitizer for cancer photodynamic therapy (PDT). Noncovalent inserted ZnPPIX molecules in apoHb possess an extremely stable property and significant recovered photoproperties with superior biocompatibility and phototoxicity, both in vitro and in vivo. This stability was verified by molecular docking analysis and calculation of binding constant, representing a total of five drug binding sites of apoHb for ZnPPIX molecules, four of which are energetically favorable (△G value of -11.9 kcal/mol), and one which is energetically acceptable (△G value of -9 kcal/mol). Folate acid modification has been shown to efficiently enhance the internalization and retention time of ZnPHb nanodrug. ZnPHb-FA is also an efficient depressor of hemin oxygenase-1 (HO-1), which could, in turn, lower the antioxidant ability of cancer cells by decreasing the production of biiirublin. 17Efficacy of gefitinib as a first-line single agent treatment in patients with advanced non-small cell lung cancer显示文摘Objective: To assess the efficacy and toxicity of gefitinib as a single agent treatment in Chinese patients with advanced non-small cell lung cancer (NSCLC). Methods: Forty-five patients with advanced NSCLC were treated with gefitinib at 250 mg daily until the disease progressed or the patient could not tolerate the toxicity. Results: None of the patients achieved a complete response (CR), while 15 patients achieved a partial remission (PR) and 17 experienced a stable disease (SD). Thirteen patients continued to have a progressive disease (PD). The response rate and the disease control rate were 33.3% and 71.1%, respectively. The symptom remission rate was 72.5%, and the median remission time was 8 days. The median survival time was 15.3 months. The median progression-free survival time was 6.0 months. The most common toxicities included rash (53.3%) and diarrhea (33.3%). Dehydration and pruritus of the skin developed in 26.7% and 22.2% of the patients, respectively. Hepatic toxicity occurred in 6.7% of patients and oral ulceration occurred in 4.4% of patients.
