A Novel Coronavirus Genome Identified in a Cluster of Pneumonia Cases--Wuhan,China 2019−2020显示文摘Emerging and re-emerging pathogens are great challenges to the public health(1).A cluster of pneumonia cases with an unknown cause occurred in Wuhan starting on December 21,2019.As of January 20,2020,a total of 201 cases of pneumonia in China have been confirmed.A team of professionals from the National Health Commission and China CDC conducted epidemiological and etiological investigations. On January 3,2020,the first complete genome of the novelβgenus coronaviruses(2019-nCoVs)was identified in samples of bronchoalveolar lavage fluid(BALF)from a patient from Wuhan by scientists of the National Institute of Viral Disease Control and Prevention(IVDC)through a combination of Sanger sequencing,Illumina sequencing, and nanopore sequencing.Three distinct strains have been identified, the virus has been designated as 2019-nCoV,and the disease has been subsequently named novel coronavirus-infected pneumonia(NCIP). | Wenjie Tan Xiang Zhao Xuejun Ma Wenling Wang Peihua Niu Wenbo Xu George F.Gao Guizhen Wu | 2020 | China CDC weekly2020,2,4: | 95
Cold-chain transportation in the frozen food industry may have caused a recurrence of COVID-19 cases in destination:Successful isolation of SARS-CoV-2 virus from the imported frozen cod package surface显示文摘Coronavirus disease 2019(COVID-19)pandemic has spread in 220 countries/regions towreak havoc to human beings around theworld. At present, the second wave of COVID-19 has begun in many European countries. The complete control of COVID-19 is very urgent.Although China quickly brought the virus under control, there have been eight sporadic outbreaks in China since then. Both in Xinfadi of Beijing and Dalian outbreak of COVID-19,environmental swab samples related to imported cold chain food were tested nucleic acid positive for SARS-CoV-2.In this outbreak in Qingdao,we directly isolated SARS-CoV-2 from the cod outer package's surface swab samples. This is the first time worldwide,SARS-CoV-2 were isolated from the imported frozen cod outer package's surface, which showed that imported frozen food industry could import SARS-CoV-2 virus. | Peipei Liu Mengjie Yang Xiang Zhao Yuanyuan Guo Liang Wang Jing Zhang Wenwen Lei Weifang Han Fachun Jiang William J.Liu George F.Gao Guizhen Wu | 2020 | Biosafety and Health2020,2,4: | 41
Age-related rhesus macaque models of COVID-19显示文摘Background:Since December 2019,an outbreak of the Corona Virus disease 2019(COVID-19)caused by severe acute respiratory syndrome coronavirus(SARS-CoV-2)in Wuhan,China, has become a public health emergency of international concern. The high fatality of aged cases caused by SARS-CoV-2 was a need to explore the possible age-related phenomena with non-human primate models. Methods:Three 3-5 years old and two 15 years old rhesus macaques were intratracheally infected with SARS-CoV-2,and then analyzed by clinical signs, viral replication, chest X-ray,histopathological changes and immune response. Results:Viral replication of nasopharyngeal swabs, anal swabs and lung in old monkeys was more active than that in young monkeys for 14 days after SARS-CoV-2 challenge.Monkeys developed typical interstitial pneumonia characterized by thickened alveolar septum accompanied with inflammation and edema, notably, old monkeys exhibited diffuse severe interstitial pneumonia.Viral antigens were detected mainly in alveolar epithelial cells and macrophages.Conclusion:SARS-CoV-2 caused more severe interstitial pneumonia in old monkeys than that in young monkeys.Rhesus macaque models infected with SARS-CoV-2 provided insight into the pathogenic mechanism and facilitated the development of vaccines and therapeutics against SARS-CoV-2 infection. | Pin Yu Feifei Qi Yanfeng Xu Fengdi Li Peipei Liu Jiayi Liu Linlin Bao Wei Deng Hong Gao Zhiguang Xiang Chong Xiao Qi Lv Shuran Gong Jiangning Liu Zhiqi Song Yajin Qu Jing Xue Qiang Wei Mingya Liu Guanpeng Wang Shunyi Wang Haisheng Yu Xing Liu Baoying Huang Wenling Wang Li Zhao Huijuan Wang Fei Ye Weimin Zhou Wei Zhen Jun Han Guizhen Wu Qi Jin Jianwei Wang Wenjie Tan Chuan Qin | 2020 | Animal Models and Experimental Medicine2020,3,1: | 22
Closed loop control design for the sense mode of micromachined vibratory gyroscopes显示文摘This paper presents a novel design method of force rebalance control for the sense mode of micromachined vibratory gyroscopes.Specific theoretical deductions are performed to identify a precise linear model of the open loop system of the sense mode, which is crucial for the PI controller design. The frequency responses obtained by experimental tests agree well with those calculated with the theoretical model, indicating the accuracy of the theoretical analyses.Experimental results demonstrate that the bandwidth of the closed loop is extended to 94.8 Hz from 2.3 Hz in the open loop and the quadrature signal is suppressed by about 64 dBV in the closed loop system. The overshoot and stable time in the step response of the closed loop system are measured to be about 15% and 35 ms,respectively. The mode-splitting gyroscope with the closed loop controlled sense mode achieves a scale factor of 41.0 mV/deg/s with nonlinearity of 0.09% and asymmetry of 1%,and a bias instability of 4.0 °/h with angle random walk of 0.171 deg/h1/2. | HE ChunHua ZHAO QianCheng LIU YuXian YANG ZhenChuan YAN GuiZhen | 2013 | Science China(Technological Sciences)2013,56,5: | 10

Genomic characterization of SARS-CoV-2 identified in a reemerging COVID-19 outbreak in Beijing's Xinfadi market in 2020显示文摘After 56 days without coronavirus disease 2019(COVID-19)cases,reemergent cases were reported in Beijing,China on June 11,2020.Here,we report the genetic characteristics of severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)sequenced from the clinical specimens of 4 human cases and 2 environmental samples. The nucleotide similarity among six SARS-CoV-2 genomes ranged from 99.98%to 99.99%.Compared with the reference strain of SARS-CoV-2(GenBank No.NC_045512),all six genome sequences shared the same substitutions at nt241(C→T),nt3037(C→T),nt14408(C→T),nt23403(A→G),nt28881(G→A),nt28882(G→A),and nt28883(G→C),which are the characteristic nucleotide substitutions of L-lineage European branch I.This was also proved by themaximum likelihood phylogenetic tree based on the full-length genome of SARS-CoV-2.They also have a unique shared nucleotide substitution,nt6026(C→T),which is the characteristic nucleotide substitution of SARS-CoV-2 in Beijing's Xinfadi outbreak. It is noteworthy that there is an amino acid D614Gmutation caused by nt23403 substitution in all six genomes, which may enhance the virus's infectivity in humans and help it become the leading strain of the virus to spread around the world today. It is necessary to continuously monitor the genetic variation of SARS-CoV-2,focusing on the influence of key mutation sites of SARS-CoV-2 on viral transmission, clinical manifestations, severity, and course of disease. | Yong Zhang Yang Pan Xiang Zhao Weifeng Shi Zhixiao Chen Sheng Zhang Peipei Liu Jinbo Xiao Wenjie Tan Dayan Wang William J.Liu Wenbo Xu Quanyi Wang Guizhen Wu | 2020 | Biosafety and Health2020,2,4: | 3
Identification of Histoplasma causing an unexplained disease cluster in Matthews Ridge, Guyana显示文摘Here,we report the identification of Histoplasma causing an unexplained disease cluster in Matthews Ridge,Guyana. In March 2019,14 employees of Chongqing Bosai Mining Company,China, working in a manganese mining of Guyana, had unexplained fever, and two of them died.We obtained lung and brain tissues as well as the blood samples from the two deceased cases(patient No.1 and 2),and bronchoscopy lavages and cerebrospinal fluid samples from one severe case(patient No.3),respectively.All samples were tested by pathological examination,high-throughput sequencing, and real-time PCR.Pathological detection showed the presence of spore-like structures in the lung tissue of patient No.1,indicating a fungal infection in this patient.Nanopore sequencing identified the existing of H.capsulatum in the lung tissue sample within 13 h.Next-generation sequencing identified specific fragments of H.capsulatum in all of the samples tested(lung, brain and blood serum from the deceased cases, and plasma from the severe case).Real-time PCR assays did not reveal any viral infection related to transmission from bat feces. We conclude that H.capsulatum was the causative pathogen of this disease cluster based on epidemiologic, clinical, pathological and nucleic acid evidence. | Ji Wang Weimin Zhou Hua Ling Xiaoping Dong Yi Zhang Jiandong Li Yong Zhang Jingdong Song William J.Liu Yang Li Ruiqing Zhang Wei Zhen Kun Cai Shuangli Zhu Dongyan Wang Jinbo Xiao Yigang Tong Wenli Liu Lihua Song Wei Wu Yang Liu Xiang Zhao Ruihuan Wang Sheng Ye Jing Wang Roujian Lu Baoying Huang Fei Ye Wenwen Lei Rongbao Gao Qi Shi Cao Chen Jun Han Wenbo Xu George F.Gao Xuejun Ma Guizhen Wu | 2019 | Biosafety and Health2019,1,3: | 3
How Much Do We Know about Atopic Asthma: Where Are We Now?显示文摘Asthma is a common disease in the worldwide and it affects over 3.5 million adults and children in the UK. Asthma is a chronic disease characterized by airway hyperresponsiveness, airway inflammation, airway remodelling and reversible airway obstruction. Inflammatory cells, cytokines, chemokines, adhesion molecules, and mediators are involved in pathogenesis of asthma. Chronic airway inflammation and remodelling are the major characters in asthma, which result in decreased pulmonary function. The precise processes are far understood at moment. Although corticosteroid therapy plus other exiting drugs (bronchodilators and oral leukotriene receptor antagonists) influences many different inflammatory and structural cell types and continues to be as the 'gold standard' of therapy in asthma, many thousands have chronic, severe diseases and suffer daily symptoms which make their lives a misery. There remains a clear need for novel approaches to therapy, which will be informed by a clearer understanding of disease pathogenesis, particularly in the target organ where airway inflammation and remodelling, the hallmarks of asthma occur. | Sun Ying Guizhen Zhang Shuyan Gu Jisheng Zhao | 2006 | Cellular & Molecular Immunology2006,3,5: | 3
Isolation and Characterization of Monkeypox Virus from the First Case of Monkeypox—Chongqing Municipality,China,2022显示文摘Introduction:The first imported case of monkeypox(MPX)from the mainland of China was reported in September 2022.Herein,the study reports the isolation and characterization of MPX virus(MPXV)in this case. Methods:Clinical specimens including skin blister fluid, oropharyngeal and nasopharyngeal swabs, and blood were collected and inoculated onto Vero cells. The isolated virus was identified as MPXV using quantitative polymerase chain reaction(qPCR),cytopathic effects(CPEs),immunofluorescence assay(IFA)and transmission electron microscopy(TEM).Plaque assays were employed to quantify infectious plaque-forming units(PFUs).The plaque reduction neutralization test(PRNT)was developed to determine the neutralizing antibody(nAb)against MPXV.Results:MPXV replication was confirmed with qPCR.Typical CPEs were observed 48 h postincubation. The isolated virus was named MPXV-B.1-China-C-Tan-CQ01.IFA showed that MPXV reacted with serum of MPX case.Orthopoxvirus morphology was observed using TEM.The virus titer increased to>106 PFU/mL after three passages. The serum PRNT 50%neutralization titer(NT50)was 35 for the MPX patient 6 days after symptom onset.Discussion:The study successfully isolated the first MPXV strain in the mainland of China,MPXV-B.1-China-C-Tan-CQ01.Infectious titration and PRNT methods have been developed. The study provides key resources and technical platforms for further research as well as anti-viral drug and vaccine development against MPX. | Baoying Huang Hua Zhao Jingdong Song Li Zhao Yao Deng Wen Wang Roujian Lu Wenling Wang Jiao Ren Fei Ye Houwen Tian Guizhen Wu Hua Ling Wenjie Tan | 2022 | China CDC weekly2022,4,46: | 3

Cartilage oligomeric matrix protein is an endogenous β-arrestin-2-selective allosteric modulator of AT1 receptor counteracting vascular injury显示文摘Compelling evidence has revealed that biased activation of G protein-coupled receptor(GPCR)signaling, including angiotensin II(AngII)receptor type 1(AT1)signaling,plays pivotal roles in vascular homeostasis and injury, but whether a clinically relevant endogenous biased antagonism of AT1 signaling exists under physiological and pathophysiological conditions has not been clearly elucidated. Here,we show that an extracellular matrix protein, cartilage oligomeric matrix protein(COMP),acts as an endogenous allosteric biased modulator of the AT1 receptor and its deficiency is clinically associated with abdominal aortic aneurysm(AAA)development.COMP directly interacts with the extracellular N-terminus of the AT1 via its EGF domain and inhibits AT1-β-arrestin-2 signaling, but not Gq or Gi signaling,in a selective manner through allosteric regulation of AT1 intracellular conformational states.COMP deficiency results in activation of AT1a-β-arrestin-2 signaling and subsequent exclusive AAA formation in response to AngII infusion.AAAs in COMP–/–or ApoE–/–mice are rescued by AT1a orβ-arrestin-2 deficiency,or the application of a peptidomimetic mimicking the AT1-binding motif of COMP.Explorations of the endogenous biased antagonism of AT1 receptor or other GPCRs may reveal novel therapeutic strategies for cardiovascular diseases. | Yi Fu Yaqian Huang Zhao Yang Yufei Chen Jingang Zheng Chenfeng Mao Zhiqing Li Zhixin Liu Bing Yu Tuoyi Li Meili Wang Chanjuan Xu Yiwei Zhou Guizhen Zhao Yiting Jia Wei Guo Xin Jia Tao Zhang Li Li Ziyi Liu Shengchao Guo Mingliang Ma Heng Zhang Bo Liu Junbao Du Wengong Wang Chaoshu Tang Pei Gao Qingbo Xu Xian Wang Jianfeng Liu Jinpeng Sun Wei Kong | 2021 | Cell Research2021,31,7: | 2
Identification of a potential tumor suppressor gene, UBL3, in non-small cell lung cancer显示文摘Objective: Oncogenes have been shown to be drivers of non-small cell lung cancer(NSCLC), yet the tumor suppressing genes involved in lung carcinogenesis remain to be systematically investigated. This study aimed to identify tumor suppressing ubiquitin pathway genes(UPGs) that were critical to lung tumorigenesis. Methods: The 696 UPGs were silenced by an siRNA screening in NSCLC cells;the potential tumor suppressing UPGs were analyzed, and their clinical significance was investigated. Results: We reported that silencing of 11 UPGs resulted in enhanced proliferation of NSCLC cells, and four UPGs(UBL3, TRIM22, UBE2 G2, and MARCH1) were significantly downregulated in tumor samples compared to that in normal lung tissues and their expression levels were positively associated with overall survival(OS) of NSCLC patients. Among these genes, UBL3 was the most significant one. UBL3 expression was decreased in tumor samples compared to that in paired normal lung tissues in 59/86(68.6%) NSCLCs, was correlated with TNM stage and sex of NSCLC patients, and was significantly higher in non-smoking patients than in smoking patients. Silencing UBL3 accelerated cell proliferation and ectopic expression of UBL3 suppressed NSCLC in vitro and in vivo. Conclusions: These results showed that UBL3 represented a tumor suppressor in NSCLC and may have potential for use in therapeutics and for the prediction of clinical outcome of patients. | Xinchun Zhao Yongchun Zhou Qian Hu Sanhui Gao Jie Liu Hong Yu Yanfei Zhang Guizhen Wang Yunchao Huang Guangbiao Zhou | 2020 | Cancer Biology & Medicine2020,17,1: | 2
Genetic tracing of HCoV-19 for the re-emerging outbreak of COVID-19 in Beijing, China显示文摘The ongoing pandemic of coronavirus disease 2019(COVID-19)caused by a novel severe acute respiratory syndrome coronavirus 2(SARS-CoV-2,also named as 2019-nCoV or HCoV-19)poses an unprecedented threat to public health(Zhu et al.,2020;Wang et al.,2020;Jiang et al.,2020).The novel HCoV-19 virus has rapidly spread into multiple countries across the world since it was first reported in December 2019.The World Health Organization(WHO)declared COVID-19 as a pandemic on 11th March 2020.As of 4th July, over 10 million confirmed COVID-19 cases have been reported in over 200 countries/regions with more than 0.5 million deaths, including 85,287 documented cases and 4,648 deaths in China(WHO,2020a). | Jing Yang Peihua Niu Lijuan Chen Liang Wang Li Zhao Baoying Huang Juncai Ma Songnian Hu Linhuan Wu Guizhen Wu Chun Huang Yuhai Bi Wenjie Tan | 2021 | Protein & Cell2021,12,1: | 2
Immunogenicity and safety of NVSI-06-07 as a heterologous booster after priming with BBIBP-CorV:a phase 2 trial显示文摘The increased coronavirus disease 2019(COVID-19)breakthrough cases pose the need of booster vaccination. We conducted a randomised,double-blinded,controlled,phase 2 trial to assess the immunogenicity and safety of the heterologous prime-boost vaccination with an inactivated COVID-19 vaccine(BBIBP-CorV)followed by a recombinant protein-based vaccine(NVSI-06-07),using homologous boost with BBIBP-CorV as control.Three groups of healthy adults(600 individuals per group)who had completed two-dose BBIBP-CorV vaccinations 1–3 months,4–6 months and≥6 months earlier, respectively, were randomly assigned in a 1:1 ratio to receive either NVSI-06-07 or BBIBP-CorV boost.Immunogenicity assays showed that in NVSI-06-07 groups, neutralizing antibody geometric mean titers(GMTs)against the prototype SARS-CoV-2 increased by 21.01–63.85 folds on day 28 after vaccination, whereas only 4.20–16.78 folds of increases were observed in control groups. For Omicron variant, the neutralizing antibody GMT elicited by homologous boost was 37.91 on day 14,however,a significantly higher neutralizing GMT of 292.53 was induced by heterologous booster.Similar results were obtained for other SARS-CoV-2 variants of concerns(VOCs),including Alpha,Beta and Delta. Both heterologous and homologous boosters have a good safety profile.Local and systemic adverse reactions were absent, mild or moderate in most participants, and the overall safety was quite similar between two booster schemes. Our findings indicated that NVSI-06-07 is safe and immunogenic as a heterologous booster in BBIBP-CorV recipients and was immunogenically superior to the homologous booster against not only SARS-CoV-2 prototype strain but also VOCs,including Omicron. | Nawal Al Kaabi Yun Kai Yang Jing Zhang Ke Xu Yu Liang Yun Kang Ji Guo Su Tian Yang Salah Hussein Mohamed Saif ElDein Shuai Shao Sen Sen Yang Wenwen Lei Xue Jun Gao Zhiwei Jiang Hui Wang Meng Li Hanadi Mekki Mekki Walid Zaher Sally Mahmoud Xue Zhang Chang Qu Dan Ying Liu Jing Zhang Mengjie Yang Islam Eltantawy Peng Xiao Zhao Nian Wang Jin Liang Yin Xiao Yan Mao Jin Zhang Ning Liu Fu Jie Shen Liang Qu Yun Tao Zhang Xiao Ming Yang Guizhen Wu Qi Ming Li | 2022 | Signal Transduction and Targeted Therapy2022,7,7: | 2
