| 1 | Identification and Characterization of microRNA during Bemisia tabaci Infestations in Solanum lycopersicum and Solanum habrochaites显示文摘MicroRNAs(miRNAs) are a class of small non-coding RNAs that can regulate target gene expression during many plant growth and development processes. In recent years, several studies identified the miRNAs involved in fruit development, leaf development, and abiotic responses in tomato. However, little is known about the miRNAs that respond to insect attack. Here, miRNAs were identified by high-throughput sequencing at different stages after infections by the whitefly Bemisia tabaci in Solanum lycopersicum and Solanum habrochaites, which are susceptible and resistant to whitefly, respectively. A total of 44 known miRNA families were identified, and 33 were shared between the two species. Among these, 13 miRNA families were newly reported in tomato. After strict filtering, some novel miRNAs were also discovered. The global expression patterns of the miRNAs exhibited were different between the two species, reflecting their different responses and resistance levels to whitefly attack. Some of the predicted target genes of differentially expressed miRNAs may be involved in responding to, and defending against, diseases and insects. | 4 | Age-related Resistance and the Defense Signaling Pathway of Ph-3 Gene Against Phytophthora infestans in Tomatoes显示文摘Resistance(R) genes against plant pathogens often have age-related resistance(ARR) effects. However, the mechanism involved in this phenomenon remains unknown. In this paper, Solanum lycopersicum ‘CLN2037B' and S. pimpinellifolium ‘L3708' harboring the Ph-3 gene, as well as S. habrochaites ‘LA2099', ‘LA1777' and ‘LA1033' harboring quantitative trait loci(QTLs), were tested to investigate age-related resistance against late blight(LB; caused by Phytophthora infestans) in the three-leaf stage of the plants. The results demonstrated that the QTL-related LB resistance showed the same age-related resistance as the Ph-3-mediated resistance at the six- and nine-leaf stages compared with the three-leaf stage. This indicated that there is a common defense mechanism in tomatoes against P. infestans via ARR. In addition, we combined ethylene(ET), salicylic acid(SA) and jasmonic acid(JA) mutants with virus-induced gene silencing(VIGS) to study the Ph-3-dependent resistance signaling pathway. | 5 | OBV(obscure vein),a C_(2)H_(2)zinc finger transcription factor,positively regulates chloroplast development and bundle sheath extension formation in tomato(Solanum lycopersicum)leaf veins显示文摘Leaf veins play an important role in plant growth and development,and the bundle sheath(BS)is believed to greatly improve the photosynthetic efficiency of C_(4) plants.The OBV mutation in tomato(Solanum lycopersicum)results in dark veins and has been used widely in processing tomato varieties.However,physiological performance has difficulty explaining fitness in production.In this study,we confirmed that this mutation was caused by both the increased chlorophyll content and the absence of bundle sheath extension(BSE)in the veins.Using genome-wide association analysis and map-based cloning,we revealed that OBV encoded a C_(2)H_(2) L domain class transcription factor.It was localized in the nucleus and presented cell type-specific gene expression in the leaf veins.Furthermore,we verified the gene function by generating CRISPR/Cas9 knockout and overexpression mutants of the tomato gene.RNA sequencing analysis revealed that OBV was involved in regulating chloroplast development and photosynthesis,which greatly supported the change in chlorophyll content by mutation.Taken together,these findings demonstrated that OBV affected the growth and development of tomato by regulating chloroplast development in leaf veins.This study also provides a solid foundation to further decipher the mechanism of BSEs and to understand the evolution of photosynthesis in land plants. | Jinghua Lu Chunyang Pan Xin Li Zejun Huang Jinshuai Shu Xiaoxuan Wang Xiaoxiao Lu Feng Pan Junling Hu Hui Zhang Wenyue Su Min Zhang Yongchen Du Lei Liu Yanmei Guo Junming Li | 2021 | Horticulture Research2021,8,1: | 1 |
| 6 | Bioresponsive cisplatin crosslinked albumin hydrogel served for efficient cancer combination therapy显示文摘Combination therapy is one of the potential strategies for tackling complicated tumor treatments like drug resistance.In this work,we have generated a therapeutic cisplatin-crosslinked albumin hydrogel(BC-Gel)that allows the local release of L-Buthionine-sulfoximine(BSO),cisplatin,and glucose oxidase(GOx)with distinct release kinetics.The BC-Gel with favorable biostimuli degradability and injectability could release therapeutic agents in a programmed manner within the tumor microenvironment(TME).The preferentially released BSO significantly suppressed the glutathione(GSH)-related cisplatin resistance and sensitized the tumor cells to cisplatin by inhibiting theγ-glutamylcysteine synthetase.Meanwhile,cisplatin achieved a sequential release and long-term treatment following the bioresponsive gel degradation under the combined action of chloride ions(Cl−)and proteinase in the body.In addition,the overproduced H_(2)O_(2)of GOx-catalyzed glucose oxidation accelerated the depletion of existed GSH within cells and further weakened the cisplatin resistance,achieving enhanced tumor treatment together with a strong cell-killing effect.The above sequential drug release strategy based on the dual GSH depletion effect breaks the balance of the GSH-mediated redox TME and enhances the sensitivity of A549 cells to cisplatin forcefully,and provides a promising way for temporal control of drug release as well as efficient cancer combination therapy. | An Yan Zherui Zhang Jiamei Gu Xiaoran Ding Yongchen Chen Jingjing Du Shu Wei Hongcheng Sun Jiayun Xu Shuangjiang Yu Junqiu Liu | 2023 | Nano Research2023,16,2: | 1 |

An introgression line population of Solanum pennellii LA0716 in a fresh market tomato inbred line 1052 was developed by our group. This population was composed of 214 lines. In the present study, fi e quantitative trait loci(QTLs)for fruit weight, two QTLs for soluble solids content(SSC), three QTLs for plant height, and one QTL for leaf size were identifie using this introgression line population. Among these, fw3 a and fw4 a for fruit weight, ssc7 a for SSC, h4t2 a, h4t3 a, and h4t7 a for plant height, and lz12 a for leaf size were determined to be novel loci.
