| 1 | A complete sequence and comparative analysis of a SARS-associated virus(Isolate BJ01)显示文摘The genome sequence of the Severe Acute Respiratory Syndrome (SARS)-associated virus provides essential information for the identification of pathogen(s), exploration of etiology and evolution, interpretation of transmission and pathogenesis, development of diagnostics, prevention by future vaccination, and treatment by developing new drugs. We report the complete genome sequence and comparative analysis of an isolate (BJ01) of the coronavirus that has been recognized as a pathogen for SARS. The genome is 29725 nt in size and has 11 ORFs (Open Reading Frames). It is composed of a stable region encoding an RNA-dependent RNA polymerase (composed of 2 ORFs) and a variable region representing 4 CDSs (coding sequences) for viral structural genes (the S, E, M, N proteins) and 5 PUPs (putative uncharacterized proteins). Its gene order is identical to that of other known coronaviruses. The sequence alignment with all known RNA viruses places this virus as a member in the family of Coronaviridae. Thirty putative substitutions have been identified by comparative analysis of the 5 SARS- associated virus genome sequences in GenBank. Fifteen of them lead to possible amino acid changes (non-synonymous mutations) in the proteins. Three amino acid changes, with predicted alteration of physical and chemical features, have been detected in the S protein that is postulated to beinvolved in the immunoreactions between the virus and its host. Two amino acid changes have been detected in the Mprotein, which could be related to viral envelope formation. Phylogenetic analysis suggests the possibility of non-human origin of the SARS-associated viruses but provides noevidence that they are man-made. | 3 | Records of the East Asian winter monsoon from the mud area on the inner shelf of the East China Sea since the mid-Holocene显示文摘AMS14C dating and analysis of grain size,major elements and clay minerals were applied to Core MZ01 from the mud area on the inner shelf of the East China Sea.Based on the environmentally sensitive grain size,clay mineral and major element assemblages,the history of the East Asia winter monsoon since the mid-Holocene could be reconstructed.These three proxies,mean grain size(>9.71μm),chemical index of alteration(CIA)and ratio of smectite to kaolinite in particular,show similar fluctuation patterns. | 5 | Biological soil crust distribution in Artemisia ordosica communities along a grazing pressure gradient in Mu Us Sandy Land, Northern China显示文摘This study investigated the distribution pattern of biological soil crust (BSC) in Artemisia ordosica communities in Mu Us Sandy Land. Three experimental sites were selected according to grazing pressure gradient. In each experimental site, the total vegetation cover, A. ordosica cover, BSC cover, litter-fall cover, BSC degree of fragmentation, BSC thickness and soil properties were investigated in both fixed and semi-fixed sand dunes and simultaneously analyzed in the laboratory. The results showed that at the same grazing pressure, BSC cover and composition were significantly affected by the fixation degree of sand dunes. In addition, BSC cover in the fixed sand dunes was 83.74% on average, whereas it is proportionally dominated by 28% mosses, 21% lichens, and 51% algae. Meanwhile, BSC cover in the semi-fixed sand dunes was 23.54% on average, which is proportionally dominated by 6.3% mosses, 2.5% lichens, and 91.2% algae. Fine sand, organic matter, and total nitrogen (N) contents in the fixed sand dunes were all significantly higher than those in the semi-fixed sand dunes. Litter-fall cover decreased along the grazing pressure gradient, whereas BSC fragmentation degree increased. Fine sand content decreased along with the increase of grazing pressure, whereas medium sand content increased in both fixed and semi-fixed dunes. The organic matter and total N contents in the no grazing site were significantly higher than those in light and normal grazing sites. However, there were no significant differences between the light and normal grazing sites. In addition, there were also no significant differences in BSC thickness between the light and normal grazing sites in the fixed sand dunes. However, a significant decrease was observed in both BSC cover and thickness in the normal grazing site. | 8 | Clinical characterization and diagnosis of cystic fibrosis throughexome sequencing in Chinese infants with Bartter-syndrome-like hypokalemia alkalosis显示文摘Cystic fibrosis (CF) is a fatal autosomal-recessive disease caused by mutations in the CFtransmembrane conductance regulator (CFTR) gene. CF is characterized by recurrent pulmonary infectionwith obstructive pulmonary disease. CF is common in the Caucasian population but is rare in the Chinesepopulation. The symptoms of early-stage CF are often untypical and may sometimes manifest as Bartter syndrome(BS)-like hypokalemic alkalosis. Therefore, the ability of doctors to differentiate CF from BS-like hypokalemicalkalosis in Chinese infants is a great challenge in the timely and accurate diagnosis of CF. In China, sporadic CFhas not been diagnosed in children younger than three years of age to date. Three infants, who were initiallyadmitted to our hospital over the period of June 2013 to September 2014 with BS-like hypokalemic alkalosis, werediagnosed with CF through exome sequencing and sweat chloride measurement. The compound heterozygousmutations of the CFTR gene were detected in two infants, and a homozygous missense mutation was found in oneinfant. Among the six identified mutations, two are novel point mutations (c.1526G 〉 C and c.3062C 〉 T) that arepossibly pathogenic. The three infants are the youngest Chinese patients to have been diagnosed with sporadic CFat a very early stage. Follow-up examination showed that all of the cases remained symptom-free after earlyintervention, indicating the potential benefit of very early diagnosis and timely intervention in children with CF.Our results demonstrate the necessity of distinguishing CF from BS in Chinese infants with hypokalemic alkalosisand the significant diagnostic value of powerful exome sequencing for rare genetic diseases. | 11 | Recent progress in Chinese polar upper-atmospheric physics research: review of research advances supported by the Chinese Arctic and Antarctic expeditions显示文摘It has been more than 30 years since the first Chinese Antarctic Expedition took place. Polar upper atmospheric observations started at this time. First began at Great Wall Station and then at Zhongshan Station in Antarctica, and later in the Arctic at Yellow River Station, Kjell Henriksen Observatory on Svalbard, and at the China-Iceland Joint Aurora Observatory in Iceland. In this paper, we reviewed the advances in polar upper atmosphere physics(UAP) based on the Chinese national Arctic and Antarctic research over the last five years. These included newly deployed observatories and research instruments in the Arctic and Antarctic; and new research findings, from ground-based observations, about polar ionosphere dynamics, aurora and particle precipitation, polar plasma convection, geomagnetic pulsations and space plasma waves, space weather in the polar regions, simulations of the polar ionosphere-magnetosphere. | 15 | Characteristics of Surface Solar Radiation under Different Air Pollution Conditions over Nanjing, China: Observation and Simulation显示文摘Surface solar radiation (SSR) can affect climate, the hydrological cycle, plant photosynthesis, and solar power. The values of solar radiation at the surface reflect the influence of human activity on radiative climate and environmental effects, so it is a key parameter in the evaluation of climate change and air pollution due to anthropogenic disturbances. This study presents the characteristics of the SSR variation in Nanjing, China, from March 2016 to June 2017, using a combined set of pyranometer and pyrheliometer observations. The SSR seasonal variation and statistical properties are investigated and characterized under different air pollution levels and visibilities. We discuss seasonal variations in visibility, air quality index (AQI), particulate matter (PM10 and PM2.5), and their correlations with SSR. The scattering of solar radiation by particulate matter varies significantly with particle size. Compared with the particulate matter with aerodynamic diameter between 2.5 μm and 10 μm (PM2.5-10), we found that the PM2.5 dominates the variation of scattered radiation due to the differences of single-scattering albedo and phase function. Because of the correlation between PM2.5 and SSR, it is an effective and direct method to estimate PM2.5 by the value of SSR, or vice versa to obtain the SSR by the value of PM2.5. Under clear-sky conditions (clearness index ≥0.5), the visibility is negatively correlated with the diffuse fraction, AQI, PM10, and PM2.5, and their correlation coefficients are ?0.50,?0.60,?0.76, and ?0.92, respectively. | 17 | The osmolyte-producing endophyte Streptomyces albidoflavus OsiLf-2 induces drought and salt tolerance in rice via a multi-level mechanism显示文摘Drought and salinity are major environmental stresses that impair crop growth and productivity worldwide. Improving drought and salt tolerance of crops with microbial mutualists is an effective and environmentally sound strategy to meet the demands of the ever-growing world population. In the present study, we found that the Streptomyces albidoflavus Osi Lf-2, a moderately salt-tolerant endophytic actinomycete, produced abundant osmolytes, including proline, polysaccharides, and ectoine. Inoculation with Osi Lf-2 increased the osmotic-adjustment ability of the rice host by increasing the proline content(by250.3% and 49.4%) and soluble sugar(by 20.9% and 49.4%) in rice under drought and salt conditions, relative to the uninoculated control. Osi Lf-2 increased stress responses in the rice host at the physiological and biochemical levels(photosynthesis efficiency, osmolytes and antioxidant content), and the gene level(osmolytes synthesis, stress-responsive and ion-transport related genes), raising rice yields under both greenhouse and saline–alkaline soil conditions. The nonlinear optimization problem is decomposed to two linear least squares problems, and solved in an alternating way. Then, a new algorithm for numerical derivative calculation using discrete Fourier transform is proposed to attenuate the effect of noises in the process of parameter estimation. Based on the estimated dynamic load characteristics, the application on voltage stability is analyzed.
