1Gearbox Fault Diagnosis using Adaptive Zero Phase Time-varying Filter Based on Multi-scale Chirplet Sparse Signal Decomposition显示文摘When used for separating multi-component non-stationary signals, the adaptive time-varying filter(ATF) based on multi-scale chirplet sparse signal decomposition(MCSSD) generates phase shift and signal distortion. To overcome this drawback, the zero phase filter is introduced to the mentioned filter, and a fault diagnosis method for speed-changing gearbox is proposed. Firstly, the gear meshing frequency of each gearbox is estimated by chirplet path pursuit. Then, according to the estimated gear meshing frequencies, an adaptive zero phase time-varying filter(AZPTF) is designed to filter the original signal. Finally, the basis for fault diagnosis is acquired by the envelope order analysis to the filtered signal. The signal consisting of two time-varying amplitude modulation and frequency modulation(AM-FM) signals is respectively analyzed by ATF and AZPTF based on MCSSD. The simulation results show the variances between the original signals and the filtered signals yielded by AZPTF based on MCSSD are 13.67 and 41.14, which are far less than variances (323.45 and 482.86) between the original signals and the filtered signals obtained by ATF based on MCSSD. The experiment results on the vibration signals of gearboxes indicate that the vibration signals of the two speed-changing gearboxes installed on one foundation bed can be separated by AZPTF effectively. Based on the demodulation information of the vibration signal of each gearbox, the fault diagnosis can be implemented. 2Application of next-generation sequencing technology to precision medicine in cancer: joint consensus of the Tumor Biomarker Committee of the Chinese Society of Clinical Oncology显示文摘Next-generation sequencing(NGS) technology is capable of sequencing millions or billions of DNA molecules simultaneously.Therefore, it represents a promising tool for the analysis of molecular targets for the initial diagnosis of disease, monitoring of disease progression, and identifying the mechanism of drug resistance. On behalf of the Tumor Biomarker Committee of the Chinese Society of Clinical Oncology(CSCO) and the China Actionable Genome Consortium(CAGC), the present expert group hereby proposes advisory guidelines on clinical applications of NGS technology for the analysis of cancer driver genes for precision cancer therapy. This group comprises an assembly of laboratory cancer geneticists, clinical oncologists, bioinformaticians,pathologists, and other professionals. After multiple rounds of discussions and revisions, the expert group has reached a preliminary consensus on the need of NGS in clinical diagnosis, its regulation, and compliance standards in clinical sample collection. 4A Multicenter Study of Viral Aetiology of Community-Acquired Pneumonia in Hospitalized Children in Chinese Mainland显示文摘Community-acquired pneumonia(CAP) is one of the leading causes of morbidity and mortality in children worldwide.In this study,we aimed to describe the aetiology of viral infection of pediatric CAP in Chinese mainland.During November2014 to June 2016,the prospective study was conducted in 13 hospitals.The hospitalized children under 18 years old who met the criteria for CAP were enrolled.The throat swabs or nasopharyngeal aspirates(NPAs) were collected which were then screened 18 respiratory viruses using multiplex PCR assay.Viral pathogens were present in 56.6%(1539/2721) of the enrolled cases,with the detection rate of single virus in 39.8% of the cases and multiple viruses in 16.8% of the cases.The most frequently detected virus was respiratory syncytial virus(RSV)(15.2%,414/2721).The highest detection rate of virus was in <6-month-age group(70.7%,292/413).RSV,human metapneumovirus(HMPV),human parainfluenza viruses(HPIVs) and influenza B virus(Flu B) showed the similar prevalence patterns both in north and south China,but HPIVs,Flu A,human bocavirus(HBoV),human adenovirus(HAdV) and human coronaviruses(HCoVs) showed the distinct circulating patterns in north and south China.Human enterovirus/human rhinovirus(HEV/HRV)(27.6%,27/98),HBoV(18.4%,18/98),RSV(16.3%,16/98) and HMPV(14.3%,14/98) were the most commonly detected viruses in severe pneumonia cases with single virus infection.In conclusion,viral pathogens are frequently detected in pediatric CAP cases and may therefore play a vital role in the aetiology of CAP.RSV was the most important virus in hospitalized children with CAP in Chinese mainland.Yun Zhu Baoping Xu Changchong Li Zhimin Chen Ling Cao Zhou Fu Yunxiao Shang Aihuan Chen Li Deng Yixiao Bao Yun Sun Limin Ning Shuilian Yu Fang Gu Chunyan Liu Ju Yin Adong Shen Zhengde Xie Kunling Shen 2021Virologica Sinica2021,36,6:7
5Evapotranspiration of a Populus euphratica Oliv. forest and its controlling factors in the lower Heihe River Basin,Northwest China显示文摘Evapotranspiration(ET) within an ecosystem is crucial for the water-limited environment that currently lacks adequate quantification in the arid region of Northwest China, mainly covered by phreatophytes, such as the Populus euphratica Oliv. tree and the Tamarix ramosissima Ledeb. shrub species. Accordingly, ET was measured for an entire year using eddy covariance(EC) in P. euphratica stands in the lower Heihe River Basin, Northwest China. During the growing season,the total ET was 850 mm, with a mean of 4.0 mm/d, which is obviously more than that observed at tree-level and standlevel scales, which was likely due to the different level of soil evaporation induced by irrigation via water conveyance.Factors associated with ET fall into either environmental or plant eco-physiological categories. Environmental factors account for at least 79% variation of ET, and the linear relationship between ET and the groundwater table(GWT) revealed the potential water use of P. euphratica forests under the non-water stress condition with the GWT less than 3 m deep.Plant eco-physiological parameters, specifically the leaf area index(LAI), have direct impact on the seasonal pattern of ET, which provides a valuable reference to the wide-area estimates of ET for riparian forests by using LAI. 6Mycorrhizal symbiosis modulates the rhizosphere microbiota to promote rhizobia–legume symbiosis显示文摘Plants establish symbioses with mutualistic fungi,such as arbuscular mycorrhizal(AM)fungi,and bacteria,such as rhizobia,to exchange key nutrients and thrive.Plants and symbionts have coevolved and represent vital components of terrestrial ecosystems.Plants employ an ancestral AM signaling pathway to establish intracellular symbioses,including the legume–rhizobia symbiosis,in their roots.Nevertheless,the relationship between the AM and rhizobial symbioses in native soil is poorly understood.Here,we examined how these distinct symbioses affect root-associated bacterial communities in Medicago truncatula by performing quantitative microbiota profiling(QMP)of 16S rRNA genes.We found that M.truncatula mutants that cannot establish AM or rhizobia symbiosis have an altered microbial load(quantitative abundance)in the rhizosphere and roots,and in particular that AM symbiosis is required to assemble a normal quantitative root-associated microbiota in native soil.Moreover,quantitative microbial co-abundance network analyses revealed that AM symbiosis affects Rhizobiales hubs among plant microbiota and benefits the plant holobiont.Through QMP of rhizobial rpoB and AM fungal SSU rRNA genes,we revealed a new layer of interaction whereby AM symbiosis promotes rhizobia accumulation in the rhizosphere of M.truncatula.We further showed that AM symbiosis-conditioned microbial communities within the M.truncatula rhizosphere could promote nodulation in different legume plants in native soil.Given that the AM and rhizobial symbioses are critical for crop growth,our findings might inform strategies to improve agricultural management.Moreover,our work sheds light on the co-evolution of these intracellular symbioses during plant adaptation to native soil conditions.Xiaolin Wang Huan Feng Yayu Wang Mingxing Wang Xingguang Xie Huizhong Chang Like Wang Jicheng Qu Kai Sun Wei He Chunyan Wang Chuanchao Dai Zhaohui Chu Changfu Tian Nan Yu Xuebin Zhang Huan Liu Ertao Wang 2021Molecular Plant2021,14,3:6

8Effects of the Fhb1 gene on Fusarium head blight resistance and agronomic traits of winter wheat显示文摘The gene Fhb1 has been used in many countries to improve wheat Fusarium head blight(FHB) resistance. To make better use of this gene in the Yellow-Huai River Valleys Winter Wheat Zone(YHWZ), the most important wheat-producing region of China, it is desirable to elucidate its effects on FHB resistance and agronomic traits in different genetic backgrounds. Based on a diagnostic marker for Fhb1, six BC2 populations were developed by crossing dwarf-male-sterile(DMS)-Zhoumai 16 to three Fhb1 donors(Ningmai 9, Ningmai 13, and Jianyang 84) and backcrossing to Zhoumai 16 and Zhoumai16’s derivative cultivars(Lunxuan 136 and Lunxuan 13) using marker-assisted backcross breeding. The progenies were assessed for FHB resistance and major agronomic traits.The Fhb1 alleles were identified using the gene-specific molecular marker. The plants with the Fhb1-resistant genotype(Fhb1-R) in these populations showed significantly fewer infected spikelets than those with the Fhb1-susceptible genotype(Fhb1-S). When Lunxuan 136 was used as the recurrent parent, Fhb1-R plants showed significantly fewer infected spikelets per spike than Fhb1-R plants produced using Lunxuan 13 as the recurrent parent, indicating that the genetic backgrounds of Fhb1 influence the expression of FHB resistance. Fhb1-R plants from the DMS-Zhoumai 16/Ningmai 9//Zhoumai 16/3/Lunxuan 136 population showed the highest FHB resistance among the six populations and a significantly higher level of FHB resistance than the moderately susceptible control Huaimai 20. No significant phenotypic differences between Fhb1-R and Fhb1-S plants were observed for the eight agronomic traits investigated. 9ULK1 and JNK are involved in mitophagy incurred by LRRK2 G2019S expression显示文摘Mutations in LR RK2(Leucine rich repeat kinase 2)are a major cause of Parkinson’s disease(PD).We and others reported recently that expression of the pathogenic gain-of-function mutant form of LRRK2,LRRK2 G2019S,induc-es mitochondrial fi ssion in neurons through DLP1.Here we provide evidence that expression of LRRK2 G2019S stimulates mitochondria loss or mitophagy.We have char-acterized several LRRK2 interacting proteins and found that LRRK2 interacts with ULK1 which plays an essential role in autophagy.Knockdown of either ULK1 or DLP1 expression with shRNAs suppresses LRRK2 G2019S expression-induced mitochondrial clearance,suggesting that LRRK2 G2019S expression induces mitochondrial fi ssion through DLP1 followed by mitophagy via an ULK1 dependent pathway.In addition to ULK1,we found that LRRK2 interacts with the endogenous MKK4/7,JIP3 and coordinates with them in the activation of JNK signaling.Interestingly,LRRK2 G2019S-induced loss of mitochon-dria can also be suppressed by 3 different JNK inhibitors,implying the involvement of the JNK pathway in the path-ogenic mechanism of mutated LRRK2.Thus our fi ndings may provide an insight into the complicated pathogenesis of PD as well as some clues to the development of novel therapeutic strategies.Yuangang Zhu Chunyan Wang Mei Yu Jie Cui Liang Liu Zhiheng Xu 2013Protein & Cell2013,4,9:6

11Effect of acupuncture at Fengchi(GB 20)on the activity of myosin light chain kinase in the middle meningeal artery of migraine modeled rats显示文摘OBJECTIVE:To study the effect of acupuncture at Fengchi(GB 20) on the activation of myosin light chain kinase(MLCK) in the middle meningeal artery of migraine modeled rats.METHODS:Forty-four clean grade healthy female Sprague-Dawley(SD) rats were randomly divided into four groups:the control group,blank control group,Fengchi(GB 20) acupuncture group,and Fengchi(GB 20) prevention group.Neurogenic inflammation of these rats was induced by electrical stimulation.The γ-^(32)P infiltration method was then used to detect MLCK activation in the middle meningeal artery,and immunocytochemistry was applied to detect the structural protein expression of MLCK.RESULTS:The migraine model was successfully established in the rats.Compared with the control group,MLCK activation was significantly decreased in the blank control group(P < 0.01).CONCLUSION:The activation of MLCK in the middle meningeal artery was increased by acupuncture at Fengchi(GB 20),indicating its effectiveness in preventing and curing on acute migraine attacks.Zhou Peijuan Wang Aicheng Li Bai Liu Chunyan Wang Yu 2015Journal of Traditional Chinese Medicine2015,35,3:5

13Validation of the Nanjing Criteria for Diagnosing Pyrrolizidine Alkaloids-induced Hepatic Sinusoidal Obstruction Syndrome显示文摘Background and Aims:Hepatic sinusoidal obstruction syndrome(HSOS)is caused by toxic injury to sinusoidal endothelial cells in the liver.The intake of pyrrolizidine alkaloids(PAs)in some Chinese herbal remedies/plants remains the major etiology for HSOS in China.Recently,new diagnostic criteria for PA-induced HSOS(i.e.PA-HSOS)have been developed;however,the efficacy has not been clinically validated.This study aimed to assess the performance of the Nanjing criteria for PA-HSOS.Methods:Data obtained from consecutive patients in multiple hospitals,which included 86 PA-HSOS patients and 327 patients with other liver diseases,were retrospectively analyzed.Then,the diagnostic performance of the Nanjing criteria and simplified Nanjing criteria were evaluated and validated.The study is registered in www.chictr.org.cn(ID:ChiCTR1900020784).Results:The Nanjing criteria have a sensitivity and specificity of 95.35%and 100%,respectively,while the simplified Nanjing criteria have a sensitivity and specificity of 96.51%and 96.33%,respectively,for the diagnosis of PA-HSOS.Notably,a proportion of patients with Budd-Chiari syndrome(11/49)was misdiagnosed as PA-HSOS on the basis of the simplified Nanjing criteria,and this was mainly due to the overlapping features in the enhanced computed tomography/magnetic resonance imaging examinations.Furthermore,most of these patients(10/11)had occlusion or thrombosis of the hepatic vein,and communicating vessels in the liver were found in 8/11 patients,which were absent in PA-HSOS patients.Conclusions:The Nanjing criteria and simplified Nanjing criteria exhibit excellent performance in diagnosing PA-HSOS.Thus,both could be valuable diagnostic tools in clinical practice.Wei Zhang Lu Liu Ming Zhang Feng Zhang Chunyan Peng Bin Zhang Jun Chen Lin Li Jian He Jiangqiang Xiao Yanhong Feng Xunjiang Wang Aizhen Xiong Li Yang Xiaoping Zou Yuecheng Yu Yuzheng Zhuge 2021Journal of Clinical and Translational Hepatology2021,9,3:4
14Study on the stability of non-cohesive river bank显示文摘Frequent bank collapse in nature highlights the need to study the mechanism of bank stability. This paper presents a theoretical analysis and a flume experimental study on the interaction of hydrodynamic conditions and non-cohesive banks of meandering and straight rivers. No bank collapse occurs if the bank angle is smaller than a critical value. The critical angle is a function of a dimensionless parameter K UD, which is directly proportional to the square of flow velocity near river bed and inversely proportional to the median diameter of bank material. Furthermore, the critical angle reduces with flow velocity and is higher in meandering rivers than in straight rivers. Formulas for estimating the threshold of bank stability are obtained by curve fitting method with experimental data. 15Nighttime transpiration of Populus euphratica during different phenophases显示文摘Evidence exists of nighttime transpiration and its potential impact on plant/water relations for species in a diversity of ecosystems. However, relevant data related to typical desert riparian forest species remains limited Accordingly, we measured sap flow velocity of Populus euphratica using the heat ratio method between 2012 and2014. Nocturnal stem sap flow was separated into nighttime and stem refilling using the ‘‘forecasted refilling''method. Nighttime transpiration was observed for each phenophase. The highest value was during the full foliation period but lowest during leaf expansion and defoliation periods. The contribution of nighttime transpiration to daytime transpiration was an average of 15% but this was comparatively higher during the defoliation period. Relationships between nighttime transpiration, vapor pressure deficits, and air temperatures were more closely associated than with wind speed in all phenophases. Moreover, we found that nighttime transpiration linearly correlated to vapour pressure deficit during the first and the full foliation periods, but nighttime transpiration showed exponential correlations to air temperatures during the same phenophases. Additionally, environmental drivers of transpiration were significantly different between nighttime and daytime(P \ 0.05). Driving forces behind nighttime transpiration were characterized by many factors, and integrated impacts between these multiple environmental factors were complex. 16Chronic intermittent hypoxia promotes the development of experimental non-alcoholic steatohepatitis by modulating Treg/Th17 differentiation显示文摘The present study aims to characterize the effect of chronic intermittent hypoxia and HIF1α on the non-alcoholic steatohepatitis (NASH)process in mice,and to explore the role of the Treg/Th17 balance in the formation of NASH inflammation and fibrosis.To achieve this purpose,simple steatosis was induced in mice by high-fat diet administration.Subsequently,chronic intermittent hypoxia was simulated by intraperitoneally injecting sodium nitrite.The changes of inflammation, fibrosis,and Treg/Th17 balance in the liver were.quantified under chronic intermittent hypoxia condition and after tail vein injection of HIF1α-siRNA.In addition,T cells were cultured in vitro, and HIF1α expression was either blocked or overexpressed under chronic intermittent hypoxia or normal conditions.Then,the changes of Treg/Th17 balance,inflammatory factors,and cell pathways were measured in each group.Our results demonstrated that chronic intermittent hypoxia accelerates the NASH process,while tail vein injection of HIF1α-siRNA improves liver histology and function.Chronic intermittent hypoxia alters the ratio of Th17 and Treg cells through HIF1α and roTOR signaling,and increases the expressions of NF-κB,IL-6,and IL-17,but decreases IL-10 expression.Inhibition of the mTOR-HIF1α-TLR4-IL-6 pathway increases the ratio of Treg/Th17. 17Serum uric acid and risk of incident diabetes in middle-aged and elderly Chinese adults: prospective cohort study显示文摘The association between serum uric acid and the risk of incident diabetes in Chinese adults remains unknown.This study aimed to investigate this association in a community-dwelling population aged≥40 years in Shanghai,China.Oral glucose tole3rance test was conducted during baseline and follow-up visits.Relative risk regression was utilized to examine the associations between baseline gender-specific serum uric acid levels and incident diabetes risk.A total of 613(10.3%)incident diabetes cases were identified during the follow-up visit after 4.5 years.Fasting plasma glucose,postload glucose,and glycated hemoglobin A1c during the follow-up visit progressively increased across the sex-specific quartiles of serum uric acid(all Ps<0.05).The incidence rate of diabetes increased across the quartiles of serum uric acid(7.43%,8.77%,11.47%,and 13.43%).Multivariate adjusted regression analysis revealed that individuals in the highest quartile had 1.36-fold increased risk of diabetes compared with those in the lowest quartile of serum uric acid(odds ratio(95%confidence interval)=1.36(1.06−1.73)).Stratified analysis indicated that the association was only observed in women.Accordingly,serum uric acid was associated with the increased risk of incident diabetes among middle-aged and elderly Chinese women.Di Cheng Chunyan Hu Rui Du Hongyan Qi Lin Lin Xueyan Wu Lina Ma Kui Peng Mian Li Min Xu Yu Xu Yufang Bi Weiqing Wang Yuhong Chen Jieli Lu 2020Frontiers of Medicine2020,14,6:3
18Comparative Cyto-molecular Analysis of Repetitive DNA Provides Insights into the Differential Genome Structure and Evolution of Five Cucumis Species显示文摘The genus Cucumis, includes the cucumber(2n = 14), melon(2n = 24), and other wild species, which is a good model for studying genome organization and evolution due to their variation in genome size and basic chromosome number. In this study, five Cucumis species with different geographical origins and basic chromosome numbers(i.e., C. sativus, C. hystrix, C. melo, C. anguria, and C. metuliferus) were used to identify and characterize the repetitive DNA in detail using a phylogenetic method. Comparative cyto-molecular genetic analysis of repetitive DNA was carried out using a graph-based clustering method, construction of Neighbor-Joining tree and fluorescence in situ hybridization(FISH).The results revealed that the five Cucumis species had differences in the repeat content of their genome, as well as in the composition of repetitive DNA and their genomic proportions. Three species from subgenus Melo showed a decreased tendency in both repeat types and genomic proportions, while two species from subgenus Cucumis also showed a decreased tendency in repeat types, but an increased tendency in genomic proportions. Phylogenic analysis of Cucumis Ty1/Copia, Ty3/Gypsy, and 45 S rDNA ITS regions revealed that C. sativus, C. hystrix, and C. melo were closely related species, which C. sativus and C. hystrix were closer, while C. anguria and C. metuliferus were closer to each other and further from the other three species. Differential accumulation and elimination of different repeat types divergently shaped the genomic architecture of these five Cucumis species, contributing to the genome’s evolution and diversification.
